Investigation on Antibacterial and Antioxidant Activities, Phenolic and Flavonoid Contents of Some Thai Edible Plants as an Alternative for Antibiotics

  • This study was aimed to examine the antibacterial and antioxidative properties of seven edible plants from Thailand to develop alternative antibiotics as feed additives. The plants include Citrus aurantifolia Swingle (Lime) fruits and its leaves, Sesbania grandiflora L. (Agati sesbania) leaves, Piper sarmentosum Roxb (Wild betal) leaves, Curcuma domestica Valeton (Turmeric) roots, Morinda citrifolia L. (Beach mulberry) leaves, Cassia siamea britt (Siamea cassia) leaves, and Cocos nucifera L. (Coconut) peels. The plants were extracted by methanol, n-hexane, chloroform, ethyl acetate, butanol and water. Antibacterial activities with minimum inhibitory concentration (MIC) were determined by agar diffusion assay against Escherichia coli, Burkholderia sp., Haemopilus somnus, Haemopilus parasuis, and Clostridium perfringens that were considered pathogenic strains in livestock infection. Methanol extracts of C. aurantifolia Swingle fruits and leaves showed the broadest spectrum of antibacterial activities except for C. perfringens. Butanol extract of S. grandiflora L. leaves showed the strongest activity against Burkholderia sp. with MIC, $135{\mu}g/mL$. P. sarmentosum Roxb leaves showed antibacterial activities against E. coli, Burkholderia sp. and H. parasuis. Ethyl acetate and water extracts from C. domesitca Valeton roots showed MIC of $306{\mu}g/mL$ and $183{\mu}g/mL$, respectively against only C. perfringens. Antioxidative activity was determined by 2-diphenyl-2-picryl hydrazyl photometric assay. The methanol extracts of C. aurantifolia Swingle fruits and P. sarmentosum Roxb leaves showed the highest antioxidant activity among all the extracts with 3.46 mg/mL and 2.70 mg/mL effective concentration 50% ($EC_{50}$) values, respectively. Total contents of phenolics and flavonoids were measured from the plant extracts. Methanol extracts of S. grandiflora L. and chloroform extracts of C. domestica Valeton were found to have the highest amount of total phenolics, 41.7 and $47.8{\mu}g/mL$, respectively. Flavonoid content of methanol extracts in S. grandiflora L. T was $22.5{\mu}g/mL$ and the highest among plant extracts tested. These results indicated that C. aurantifolia Swingle, S. grandiflora L., P. sarmentosum Roxb, and C. domestica Valeton have antibacterial and antioxidant activities and can be used as alternative antibiotics or potential feed additives for the control of animal pathogenic bacteria.

Effects of Water Extracts from Mulberry Leaves on Hepatic HMG-CoA Reductase and Acyl-CoA-Cholesterol Acyl Transferase Activity in Rats Fed High Cholesterol Diets

  • This study investigated the effects of mulberry leaf extract on lipid metabolism in rats fed a high cholesterol diet. Sprague-Dawley male rats weighing $100{\pm}10g$ were randomly assigned either to one of two normal diet groups, with (NE group) or without (N group) mulberry extract, or one of four high cholesterol groups containing 1% cholesterol and various levels of dietary mulberry leaf extract. The rats fed high cholesterol diets were subdivided into 4 groups according to level of mulberry extract; Mulberry extract free group (HC group), 0.8% mulberry leaf extract group (HCL group), 1.6% mulberry leaf extract (HCM group) and 3.2% mulberry leaf extract (HCH group). The rats were fed their respective diets ad libitum for 4 weeks. The levels of serum triglyceride, total cholesterol and LDL-cholesterol of the HC group were higher than mulberry leaf extract supplemented groups. In contrast, the levels of serum HDL-cholesterol in groups supplemented with mulberry leaf extract were significantly lower than that of HC group. Hepatic total lipids, triglycerides, and cholesterol were significantly higher in the high cholesterol groups compared to those of the normal group, but were lower in the HCL, HCM and HCH groups than in the HC group. HMG-CoA reductase activity was significantly decreased in the HC and HCL groups compared to the normal and NE groups. However, the activities in the HCM and HCH group were similar to that of the normal group. The activity of acyl-CoA-cholesterol acyl transferase (ACAT) was increased in high cholesterol groups compared to the normal group. However, the activity was lower for all of the high cholesterol groups fed mulberry leaf extracts, and was lowest for the highest supplemented group (HCH), with no significantly difference from the normal group. In conclusion, the reduction in serum and hepatic lipid composition by mulberry leaf extract may be due to its modulation of HMG-CoA reductase and ACAT activities.

상심자추출복합물의 퇴행성관절염 개선 효능에 대한 동물실험 및 임상연구 (Animal and Clinical Study of the Efficacy of Mulberry Extract Complex on Degenerative Arthritis)

  • Objectives : This study was performed to investigate the efficacy and safety of the Mulberry Extract Complex in a placebo-controlled randomized clinical trial and animal study on degenerative arthritis. Methods : Animal study: Mulberry Extract Complex is composed of extracts of mulberry (Morus alba L.) fruit, mulberry leaves and black beans (Glycine max (L.) Merr.). To evaluate the serum level of interleukin-2, interferon-$\gamma$, and prostaglandin E2, an animal model of degenerative arthritis induced by monosodium iodoacetate was employed. Clinical study: The efficacy index (VAS, K-WOMAC) was compared among patients with symptoms of degenerative arthritis before and after Mulberry Extract Complex ingestion as well as the one in groups. Evaluations of the improvement by the subjects and by doctor assessment were also performed. Results : Animal study: Mulberry Extract Complex reduced the serum level of interferon-$\gamma$ and prostaglandin E2 in an animal model with degenerative arthritis. Clinical study: The VAS change showed statistical significance in the experimental groups after 4 weeks (PP set) and 8 weeks (ITT set) of ingestion. When the K-WOMAC was analyzed using a modified ITT set to determine the effectiveness, statistically significant results were obtained in the fields of pain & symptom within the Mulberry Extract Complex group as well as between the Mulberry Extract Complex and placebo groups after 8 weeks of ingestion. Results from the improvement evaluation by subjects and the assessment of improvement by doctors showed statistical significance in the experimental groups (PP set) after 8 weeks. Conclusions : Mulberry Extract Complex could be useful for the improvement of various symptoms of degenerative arthritis based on its anti-inflammatory activity and its reduction of VAS and K-WOMAC pain scores.

청목노상 뽕잎으로부터 Helicobacter pylori 억제물질의 정제 및 동정 (Purification and Identification of Inhibitory Compounds from Cheongmoknosang Mulberry Leaves (Morus alba. L.) on Helicobacter pylori)

  • 본 연구에서는 청목노상 추출물로부터 Helicobacter pylori를 억제하는 물질을 정제 및 동정을 실시하였다. 80% 에탄올 추출물의 페놀성 물질의 함량은 16.1 mg/g으로 측정되었으며, H. pylori 저해활성은 clear zone이 15 mm로 나타났다. 저해물질의 분리는 $C_{18}$ column을 이용하여 분리한 결과 9개의 fraction으로 분리되었으며, H. pylori 저해활성을 나타낸 fraction 7과 8을 MCI-gel CHP-20 column을 이용하여 normal phase type인 ethanol 100%에서 0%로 농도를 감소시키며 10 mL/min의 유속으로 용출한 결과 6개의 단일 물질 분획을 얻을 수 있었다. 그 중 세 가지의 물질이 H. pylori저해활성을 보여 FAB-Mass, $^1H$$^{13}C$ NMR과 IR spectrum을 사용하여 구조 동정한 결과 chlorogenic acid, caffeic acid 및 rosmarinic acid인 것으로 확인되었다.

상엽(桑葉)(Morus alba Linne)의 혈압강하작용(血壓降下作用) (Depressor Activity of Water Extracts of the Mulberry(Morus alba Linne))

  • The present study was undertaken to investigate effect of water extracts of the mulberry leaves (MWE) on arterial blood pressure and respiration in cats. And also studied were depressor responses to intravenously administered MWE in the animals pretreated separately with atropine(2.5 mg/kg), propranolol(2 mg/kg), dibenamine (15 mg/kg), and hexamethonium (1.5 mg/kg) in order to find out the mechanism of depressor activity of MWE. The results obtained were as follows; 1) Following intravenous administration of 0.25 ml/kg and 0.5 ml/kg of MWE into the cat the maximum depressor responses observed were $60.2{\pm}2.3\;mmHg$ and $72.3{\pm}1.7\;mmHg$ respectively. 2) Since depressor responses to intravenously administered MWE were partially inhibited by hexamethonium and markedly by atropine, it is strongly suggested that depressor activity of MWE mainly results from its vagal effects. 3) After administration of MWE respiratory rate was invariably increased following a short period of apnea.

뽕잎의 당침 및 유산발효에 의한 추출물의 기능성 품질 특성 (Functional quality characteristics of extracts by sugar-leaching and lactic acid fermentation of mulberry leaves (Morus alba L.))

  • 이 연구는 뽕잎을 전통방식의 당침 추출액과 Lactobacillus plantarum TO- 2100 유산균을 이용한 유산발효 추출액의 기능성 품질 특성을 조사하였다. 당침 추출액은 12주 동안 추출하였으며, 유산발효 추출액은 8주간 추출하였다. 당침 추출액 및 유산발효 추출액의 수율은 각각 27% 와 166% 였으며, 당 함량은 각각 43 $^{\circ}Brix$와 33 $^{\circ}Brix$였다. pH와 총 산도는 4.6, 0.45% 와 3.6, 1.17%였다. 유산발효 추출액의 유산균 생균수는 $1.2{\times}10^{10}$ $CFU/m{\ell}$으로 당침 추출액의 $2.8{\times}10^2$ $CFU/m{\ell}$ 보다 월등히 많았다. 유산발효 추출액이 amylase, cellulase, pectinase, protease 등 분해효소의 활성을 보인 반면, 당침 추출액은 분해효소의 활성을 나타내지 않았다. 유기산의 경우 malic acid, acetic acid, citric acid의 함량은 당침 추출액이 높은 반면, lactic acid 의 함량은 유산발효 추출액이 2.6배 높았다. 당침 추출액의 유리당은 glucose (200.93mg/g), fructose (236.32mg/g), sucrose (18.41mg/g) 등 GI값이 높은 단당으로 구성된 반면, 유산발효 추출액에서는 유리당이 검출되지 않았다. 기능성 성분인 polyphenol, anthocyanin 및 당질대사 저해물질인 piperidine alkaloid의 함량 또한 유산발효 추출액이 당침 추출액보다 높았다. 당질대사의 주 효소인 ${\alpha}$-glycosidase의 활성을 당침 추출액이 3.4% 저해한 반면 유산발효 추출액은 16.2% 저해 활성을 보였다. 이 결과로부터 전통방식의 당침 추출법에 비해 유산발효 추출법이 뽕잎의 수율 향상 및 기능성 성분의 함량 증진에 더 효과적이며, 이로 인한 대사기능장애의 예방 및 기능증진에 더 유효할 것으로 사료된다.

청목노상 뽕잎으로부터 분리한 Helicobacter pylori 저해물질에 대한 In Vivo 안전성 평가 (Evaluation of In Vivo Safety of Inhibitory Compounds from Cheongmoknosang Mulberry Leaves against Helicobacter pylori)

  • 뽕잎에서 분리한 유효성분의 혼합물에 대한 안전성을 확인하기 위하여 경구 투여하여 시험한 결과, 투여량 4000 mg/kg에서 암수 mouse 모두가 생존하여 급성독성은 없을 것으로 판단하였으며, 13주 반복 독성 시험에서도 투여량 2000 mg/kg에서 암수 mouse 모두가 생존하여 약물의 독성에 의한 문제는 없을 것으로 판단되었다. 육안으로는 대조군과 비교 시 이상이 확인되지 않았으며 부검 후 H. pylori 의 감염에 영향을 받을 수 있는 위의 무게 역시 대조군과의 차이를 확인할 수 없었다. 면역세포에 대한 영향은 약물 투여 시 매체대조군에 비해 대식세포의 독성 물질인 NO의 생성이 감소하는 것을 확인하였다. 또한 암컷군에서 48시간에서만이 LPS와 Con A에 의해 비장세포가 약간 증식되었을 뿐 다른 실험군에서는 대조구와 비슷한 수치를 나타내었으며, 자연살해세포의 세포독성은 감소되었다. 뽕잎에서 분리한 유용 성분인 caffeic acid, rosemarinic acid와 chlorogenic acid가 H. pylori의 감염환자의 치료용 또는 예방의 목적으로 임상적으로 사용할 경우 아무런 독성이 없을 것으로 판단되어 뽕잎을 건강기능성 식품으로 개발하는데 있어서 안전성 문제는 없는 것으로 판단되었다.

강원도산 상엽과 누에의 혈당강하 활성물질의 특성

  • It was proved that both ethanol extracts from Mori Folium from Kangwon do and silk worm had higher inhibition acitivity on $\alpha$-glucohydrolase than the water extracts. In adding above 8.5 (mg/L) of silkworm extracts, the inhibition rate on $\alpha$-1,4 glucosidase was saturated while the inhibition rate was continuously increased in adding the extracts from Moli Folium. It was also found that the diethyl ether fraction showed much better inhibition activity than water fraction from ethanol extracts, yielding ca. 85% of inhibition rate for the extract of Moli Folium, compared to 91% for a commercially available hypoglycemic drug, Chloropropamide. In separating the diethyl ether fractions by Consecutive Sephadex gel filtration and Thin layer chromatography, three and four active spots were obtained from Moli Folium and silkworm, respectively. It is interesting that the similar Rf spots from both species among several spots in TLC have the highest inhibition acitivity on a target enzyme, which can imply that the active substances from both species are same or similar molecular weight and structure. Glucose-lowering activities of both speciese were also examined in vivo, showing that the fraction from Moli Folium had better activity than that from silkworm, and its activity was similar to that of a commercial drug.

당뇨 쥐에서 상엽추출물을 첨가한 보리면의 혈당조절효과 (Effects of Barley Noodles Contained Mulberry Leave Extracts on Blood Glucose Regulation in Diabetic Mice)

  • 본 연구는 상엽추출물이 포함된 보리면의 혈당조절능력을 입증하기 위하여 4주간 고지방/고탄수화물 식이 후 streptozotocin과 nicotinamide로 당뇨를 유발하고 음성대조군, 양성대조군, 보리면대조군, 상엽대조군, 보리면+상엽추출물 2.5%군, 보리면+상엽추출물 5%군의 6군으로 나누었다. 그 후 2주간 식이를 진행하며 체중, 혈당, 간 기능 및 혈액 내 지질의 변화를 측정하였다. 그 결과 보리면과 상엽 단독 섭취군보다 혼합섭취군에서 체중 감소가 적었고 혈당조절효과도 뛰어났으며 이는 경구 당 부하검사에서도 동일한 결과를 보였다. 또한 간기능, 혈중 지질 개선 효과도 혼합 섭취군에서 강하게 나타났다. 따라서, 보리면과 상엽추출물을 함유한 식이는 복합식이에서 단독식이보다 강한 혈당조절효과와 간 기능 개선, 지질 강하효과도 나타났다. 결과적으로 상엽을 포함하는 보리면은 혈당을 조절하고, 간 기능, 지질 강하 기능을 보여 만성질환 관리에도 도움이 될 것으로 사료된다.

Protective Effect of Mulberry Leaf and Yacon Extract Induced Hyperlipidemia in Obese Rats

  • There has been a lot of interest in medicinal effects for hyperlipidemia from the natural product since the therapy of atherosclerosis has emerged as social concern. Rats were acclimated to the feeding environment for one week and induced to obesity with high fat diet during 4 weeks until their body weight were more than 30% of normal range. To evaluate the effect of hyperlipidemia of mulberry leaf of Morus alba and yacon tuber of Smallanthus sonchifolia extracts (MLYT), 1%, 3%, 5% of MLYT were treated to Sprague-Dawley rats. Aspartate transaminase (AST) was significantly increased 26.2% in high fat diet group (G2) compared to normal diet group (G1) (P<0.05). But AST were significantly decreased in high fat diet with 0.5% hydroxycitric acid diet group (G3) and all the MLYT treated groups compared to G2. Total cholesterol, low-density lipoprotein (LDL), triglycerides and phospholipids were observed significant improvements in the MLYT-treated groups (P<0.05). These improvements in liver and feces were further supported by the lipid content. MLYT remarkably suppressed the level of lipid peroxidation caused by high-fat diet in rats. The level of 3-hydroxy-3-methyl-glutaryl-coenzyme A (HMG-CoA) reductase activity. As a result, the proper combination of mulberry leaves and yacon tuber extract will have a synergistic effect on hyperlipidemia. These results indicate that MLYT could be a candidate for the improvement of hyperlipidemia.
