Benzo(a)pyrene에 의해 유도된 간기능 장해에 미치는 쑥의 효과 (The Effect of Mugwort Extracts on the Benzo(a)pyrene-induced Hepatotoxicity in Rats)

  • Mugwort has been used as a Korean folk medicine in treating liver diseases acting as an analgesics, sedative, diuresis, choleretics. This study was perfomed to evaluate the effect of mugwort extracts on the changes of enzyme activities, lipid accumulation of the serum and liver, when hepatotoxicity was induced by benzo(a)pyrene. The results are as follows: 1. Mugwort water extract administration prevented the increase of serum and liver AST, ALT, LDH, ${\gamma}$-GTP, liver ALP activities and bilirubin content caused by B(a)P injection. 2. The increase of serum and liver ALT, LDH, ${\gamma}$-GTP, serum AST activities and liver bilirubin contents in B(a)P treated group were decreased by mugwort methanol extract treatment. 3. Serum and liver total cholesterol, phospholipid, triglyceride level and serum HDL-cholesterol level were increased by B(a)P treatment. After combined treatment of mugwort water and methanul extracts, these lipid content were significantly decreased. 4. The hepatotropic effect of mugwort water extract and after-treatment against B(a)P induced hepatotoxicity was superior to that of methanol extract and pretreatment.

Preparation of Mulberry Leaf Extract by Adding Mugwort and Pine Needle and Effects on Lipid Composition in Rats Fed High Cholesterol Diets

  • This study investigated the effects of feeding mulberry leaf extracts on lipid composition in rats fed high cholesterol diets. An initial 30-person sensory evaluation of preparations containing various concentrations of mulberry leaf extract showed that a preparation containing 9% mulberry leaf extracts was the most highly preferred. In addition, subsidiary materials of pine needle extracts and mugwort extracts were added to weaken the unpleasant smell of mulberry leaf extract A preparation containing 9% mulberry leaf extract with 3% mugwort extract and 7% pine needle extract was given highest preference scores by the 30-person panel. When comparing the functional ingredients contents of the various preparations of mulberry leaf extracts, such as GABA, DNJ and flavonoids, no significant differences were found as a result of adding subsidiary materials (pine needle and mugwort extracts). Sprague-Dawley male rats weighing l00$\pm$10g were randomly assigned to one normal diet group, and to four high cholesterol diet groups containing 1% cholesterol, to elucidate the functionality of the mulberry leaf extract The four high cholesterol diet groups were classified into: a mulberry leaf extract diet group free of subsidiary materials (EB group); a mulberry extract diet group with pine needle extracts (EP group); a mulberry leaf extract diet group with mugwort extracts (EM group); and a control group (HC group). The mulberry leaf extracts were provided as drinking water; the diet and water were fed ad libitum. Hepatic cholesterol and triglyceride levels were higher, by 279% to 475%, in the high cholesterol groups compared to the normal diet groups, but were significantly lower in the three groups supplied with mulberry leaf extracts, compared with the high cholesterol control. There were no changes in functionality of the mulberry leaf extract preparations due to the addition of subsidiary materials. In conclusion, preparations of mulberry leaf extracts were shown to improve lipid metabolism in rats fed a high cholesterol diet, by reducing hepatic and plasma triglyceride and cholesterol levels. Also human palatability of the mulberry leaf preparation was improved by adding subsidiary materials such as pine needle and mugwort extracts.

들깨잎과 쑥 추출물의 구강병 원인균에 대한 항균 및 KB 세포 증식 억제효과 (Anti-microbial Activities Against Oral Microbes and Growth-Inhibitory Effect on Oral Tumor Cell of Extracts of Perilla and Mugwort)

  • Methanol extracts of Perilla and Mugwort were stepwise extracted with hexane, chloroform, ethyl acetate, butanol and water. Anti-microbial activities and inhibitory effect on growth of oral tumor cell of each extract were investigated. Each extracts of Perilla and Mugwort were investigated to anti-microbial effects on oral microbes by means of agar diffusion method and MIC. These results suggest that the hexane extracts of Perilla and Mugwort have antimicrobial activities against S. mutans and potent inhibitory Effect to KB cell growth.

쑥 추출물의 사람 Low Density Lipoprotein에 대한 항산화능 (Antioxidative Activity of Mugwort extracts(Artemisia Princeps Var. Orientalis) on Human Low Density Lipoprotein)

  • The antioxidative effect of mugwort extracts was measured by DPPH and LDL with four different solutions (70% acetone, ethanol, hot water, cold water). Mugwort extracts contained 3.2% of polyphenol, 380 RE/l00 g of vitamin A, 16.2 mg/100 g of vitamin C, and 5.1 ${\alpha}$-TE/100 g of vitamin E. DPPH revealed the effect in the order of 70% acetone, hot water, ethanol, and cold water. In particular, 70% acetone showed outstandingly stronger activity than the control group. Also, when 10 ${\mu}\ell$ was added, the effect was well noticed. But the antioxidative activity was hardly seen at 15 ${\mu}\ell$. LDL exhibited the same order of strength in proportion to mugwort's concentration. Against the control group, the activity of 70% acetone was 7 times, hot water and ethanol 6 times, and cold water 2 times. This result is attributable to the antioxidative increase of polyphenol and antioxidative vitamins.

쑥과 솔잎의 항산화작용 및 아질산염 소거작용 (Antioxidative and Nitrite Scavenging Activities of Mugwort and Pine Needle Extracts)

  • 쑥과 솔잎을 각각 물과 70% 에탄올로서 추출하여 전자공여능과 아질산염 소거능을 조사하였다. 쑥 추출물의 전자공여능은 300-1,000ppm 농도에서 물추출물은 50-57%였으며, 에탄올추출물은 51-64%로서 에탄을 추출물이 우수하였다. 솔잎 추출물의 전자공여능은 물추출물은 100-500ppm에서 52-60%, 1,000ppm에서는 70%의 전자공여능을 나타내었으며 에탄을 추출물은 100-500ppm에서 68-71%, 1,000ppm에서는 약 77%로서 물추출물보다 높은 활성을 나타내었다. 아질산염 소거능은 pH 1.2, 3.0, 4.2, 6.0에서 측정한 결과, pH 1.2에서 가장 높았으며 pH 증가에 따라 감소하였다. 쑥추출물 1,000ppm의 아질산염 소거능은 pH 1.2에서 물추출물은 37%, 에탄올추출물은 27%였다. 솔잎 추출물 1,000ppm의 경우에는 pH 1.2에서 물추출물은 65%, 에탄올추출물은53%의 아질산염 소거능을 나타내었다. 추출물의 전자공여능과 아질산염소거능은 솔잎추출물이 쑥추출물에 비하여 우수하였다. 쑥과 솔잎 추출물에서 전자공여능은 에탄을 추출물이 물추출물보다 우수하였으나 아질산염소거능은 물추출물이 에탄을 추출물보다 우수하였다.

Evaluation of extraction methods for essential oils in mugwort (Artemisia montana) using gas chromatography-mass spectrometry

  • Mugwort (Artemisia montana), which is a perennial plant mainly distributed throughout Northeast Asian regions, has been used as a preferred source of various foods and traditional medicines in Korea. In particular, as essential oils extracted from mugwort were reported to be biologically active, its steam distillate has been widely used to treat various conditions, such as itching, hemorrhoids, and gynecological inflammation. Therefore, efforts have been devoted to develop effective methods for the collection of bioactive essential oils from mugwort. In this study, five mugwort extracts were obtained using different extraction conditions, namely, 6 % ethanol at room temperature and at 80 ℃, pure ethanol, n-hexane, and an adsorbent resin. To evaluate the five extracts of mugwort, area-under-the-curve values (AUCs), chemical profiles, and major bioactive essential oil contents were investigated using gas chromatography-mass spectrometry (GC-MS). An overall assessment of the volatile components, including essential oils, in the five extracts was conducted using AUCs, and the individual essential oil in each extract was identified. Furthermore, the four major essential oils (1,8-cineole, camphor, borneol, and α-terpineol), which are known to possess anti-microbial and anti-inflammatory activities, were quantified using authentic chemical standards. Based on the evaluation results, pure ethanol was the best extractant out of the five used in this study. This study provides evaluation results for the five different mugwort extracts and would be helpful for developing extraction methods to efficiently collect the bioactive oil components for medical purposes using chemical profiles of the extracts.

쑥의 수용성추출성분(水溶性抽出成分)이 백서영양(白鼠營養)에 미치는 영향(影響) (A Study on the Nutritional Effects of Boiling Water Extracts of Mugwort Powder in Rats)

  • This study was intended to observe some nutritional effects of the boiling water extracts of mugwort powder in albino rats. Forty young male albino rats of Sprague-Dawley strain, body weight of $77{\pm}5g$ were employed for the experiment. They were divided into 5 dietary group, 0 (control), 1, 2, 4, and 8g% mugwort powder extract supplemented into the control diet, and fed each corresponding diet to the rats for 4 weeks. They were observed on intake of food and protein, efficiencies of food and protein, amount of energy intake, growing rate, and levels of hematocrit and hemoglobin in the blood. The amount of intakes of food, protein and energy was the highest in the group fed on 4g% mugwort extracts supplemented diet than any other groups studied. The efficiencies of food and protein in the groups fed on mugwort extracts supplemented diets were all lower than that of the control group. The growing rate of the group fed on 1g% mugwort extracts supplemented diet was the highest, however, there was no significant difference among them. The levels of hemoglobin of the groups fed 4g% or 8g% mugwort extracts supplemented diet was higher than that of the control, but not significant.

쑥추출물이 혈중 에탄올 농도와 간기능에 미치는 효과 (The Effects of Extracts from Mugwort on the Blood Ethanol Concentration and Liver Function)

  • This study was conducted to investigate the effects of mugwort extracts on the blood ethanol concentration and liver function in rats. Sprague-Dawley rats were used, the rats administered with 25% ethanol (5g/kg$.$B.W.) were devided into three groups (CON-E ; 25% ethanol administered to the CON-E) according to the administered ethanol concentration and the levels of administered mugwonts. Mugwont roots extracts were administered via the caudal vein. Ethanol concentration was measured at the time of 0, 1, 2 and 3hr by gas chromatography. GOT(Glutamic Oxaloacetic Transaminase) and GPT(Glutamic Pyruvic Transaminase) were measured at the time of 0 and 5hr. Components of each extracts were analyzed by using high performance liquid chromatography. Cell number, GOT and GPT were investigated by using rat hepatocyte culture. Megwort extracts were added at the levels of 1% or 2%. Hepatocyte culture were into five groups according to the addition levels. The results were summarized as follows ; 1. Catechin contents of 8∼10mg/100g and the contents of (-)-epigallocatechin was high in the water extracts. 2. Ethanol degradation efficiency declines in the following order : MDW-E>MOH-E>CON-E. 3. The numbers of rat hepatocytes declines in the following order : 2% MDW-L>1%MDW-L>1%MOH-L>CON-L>2%MOH-L. These results suggest that crude catechin of mugwort extracts may play important roles to degrade ethanol and recover liver function in rats.

쑥 추출물이 첨가된 Set-Type Yoghurt의 발효 특성 (Fermentation Characteristics of Set-Type Yoghurt from Milk Added with Mugwort Extract)

  • This study was carried out to investigate the fermentation characteristics and storage of set-type yoghurt added mugwort extracts(AME) such as pH, growth of lactic acid bacteria, number of viable cells, viscosity, and sensory characteristics during 24 hours fermentation and 15 days storage. Addition of mugwort extracts was grown rapidly of lactic acid bacteria rather than that of control and also 4 or 8% AME groups were grown similar to control. The drop of AME pH of broth was less compared with control during incubation of lactic acid bacteria. The growth of lactic acid bacteria during incubation of AME yoghurt was not different of viable cell count between AME group and control in beginning time, but the viable cell count of AME groups were increased depended opon addition quantity of AME in ending time. Addition of mugwort extracts was not affect on pH change during yoghurt fermentation and increased a lactic acid bacteria number as well as no effect of yoghurt fermentation in ending time. The viscosity of yoghurt was almost not changed 3 hours after yoghurt mix and increased rapidly 6 hours after yoghurt mix. Although control and 0.5% AME group showed maximum viscosity at 18 hours of fermentation, 1 and 2% AME group showed linear increase until 24 hours of fermentation. Mugwort did not affect pH and viable cel number of lactic acid bacteria during 15 days storage 24 hours after fermentation. Sensory evaluation of the AME yoghurt showed that flavour, texture and acid taste were not affected by addition of mugwort. However, the appearance and taste were dropped by addition of mugwort.

축산식품에 이용하기 위한 쑥 성분중의 생리활성에 관한 연구 (The Studies on the Physiological Active Substances of Mugwort Components for the Utilization to the Foods of Animal Husbandry)

  • 건국대학교본 연구는 쑥의 추출물을 이용해 생체내에서 에탄올의 분해효능, 간기능 및 Cd 독성 저하작용에 미치는 효과를 알아보기 위하여 실시하였다. 본 실험에서 에탄올 분해효능은 Rat 1kg 당 25% 에탄을 5g을 경구투여한 후 쑥추출물의 카테킨 농도를 측정하여 이를 기준으로 일일 사료섭취량의 2% 원료수준으로 각각 미정맥주사를 하였다. 에탄올만 경구투여한 대조군과 에탄올을 경구투여한 후 쑥추출물을 미정맥주사한 실험군은 시간별(0, 1, 2, 3시간)로 미정맥에서 혈액을 채취해 3000rpm에서 10분간 원심분리하여 혈장을 분리시킨 후 GC(Gas Chromatography)로 혈중 에탄올 농도를 측정하였다. 또한 같은 방법으로 시간만 달리해 (0시간, 5시간) 혈장을 취해 GOT, GPT의 활성치를 측정하여 간손상정도를 측정하였다. 간기능에 미치는 효과는 간실질세포를 기본배지에 배양한 대조군과 쑥추출물을 배지의 1% 및 2% 원료수준으로 기본배지에 첨가하여 간실질세포를 배양한 실험군으로 나누어 36시간 배양한 후 현미경으로 형태를 관찰하고 생존 세포수와 GOT, GPT의 활성치를 함께 측정하였다. Cd 독성 저하작용은 4주령의 Sprague-Dawley 수컷 흰쥐를 구입하여 대조군(CON,0.85% saline 10ml/kg B.W.)과 Cd단독 투여군(CD, 0.4mg $CdCl_2/kg$ B.W.), Cd과 쑥추출물을 복강내 투여한 군(IP, 쑥 추출물은 일일사료섭취량의 1% 수준) 그리고 쑥추출물을 구강내 투여한 군(PO, 쑥 추출물은 일일사료섭취량의 1%수준)으로 분류하여 4주간 사육하였다. 사육 후 각종 장기 및 혈액을 채취하여 장기에서의 GSH 농도, GSH-Px 및 GR의 활성과 혈장 GOT, GPT 활성을 측정하였고 더불어 각 조직에서 Cd축적량을 조사하였다. 실험결과를 요약하면 다음과 같다. (1) 쑥추출물의 카테킨의 함량을 Iwasa와 Torri의 방법을 변형하여 측정한 결과 네 가지의 추출물중에서 카테킨의 함량은 쑥의 열수추출물이 $8{\sim}10mg/100g$으로 가장 많았으며 쑥의 에탄올 추출물은 $3{\sim}4mg/100g$인 것으로 나타났다. (2) 쑥 추출물을 HPLC를 이용해 주요 카테킨의 성분을 비교한 결과 쑥의 열수추출물 및 에탄올추출물은 (-)-Epigallocatechin (EGC)의 함량이 가장 높았다. (3) 시간에 따른 혈중 에탄올 농도를 GC로 측정한 결과 각각 에탄올을 투여한지 1시간 후의 에탄올 농도에서는 CON-E의 경우가 가장 높은 것으로 나타났고 MDW-E, MOH-E군은 CON-E에 비해 유의하게(p<0.05) 감소하는 것을 알 수 있었다. (4) 간실질세포 배양 36시간 후 배지를 걷어내고 밑면에 모인 세포를 모아 현미경으로 수를 측정하여 평균을 낸 결과는 2% MDW-L가 가장 많았으며 2% MOH-L을 제외하고는 CON-L의 세포수가 가장 적었다. 또한 배양 후 취한 세포내의 GOT, GPT의 활성치는 실험군간의 유의한 차이는 없었다. (6) 간 조직중의 GSH 함량수준은 쑥추출물 투여군들(IP, PO)이 CD군에 비해 유의적으로 증가(p<0.05)하여 대조군과 같은 정상수준으로 되었다. (7) 간 신장에서의 GSH-Px 활성은 저농도의 Cd에 의해서는 영향을 받지 않은 것으로 나타났다. (8) 혈장 GOT활성은 IP군이 대조군에 비해 증가하였으나 CD군에 비해 유의적으로 감소(p<0.05)하였다. 이상의 결과에서 쑥추출물이 체내에서 에탄올 분해를 촉진시키고 간기능을 개선시킴은 물론 GSH농도의 정상회복과 GR의 증가 및 혈장에서의 GOT활성의 감소 등으로 보아 Cd의 간독성도 감소시키는 작용이 있는 것으로 사료되었다.
