Background: Lung cancer and chronic obstructive lung disease often coexist in the same person who are elderly and cigarette smoking. There are several reports that the presence of chronic obstructive pulmonary disease constitutes an independent risk factor for the development of lung cancer. Moreover, the association between mucus hypersecrtion and lung cacer has been reported. Method: In 72 cases with primary lung cancer which were confirmed histopathologically at Chonbuk University Hospital from August 1986 to July 1991, We evaluated the relationship between spirometry and lung cancer characteristics. Results: Six cases(8.3%) showed normal lung function, 16(22.2%) cases showed pure restrictive lung disease, 46(63.9%) cases showed moderated obstructive lung disease and 4(5.6%) cases showed severe obstructive lung disease. $FEV_1$(%) was lower in central type than in peripheral type, lower in advanced non-small cell cancer and lower in subjects with phlegm. $FEV_1$/FVC(%) was higher in small cell cancer than in squamous cell cancer and higher in patients without previous pulmonary disease than with previous pulmonary disease. But there was no statistically significant difference in lung function according to histologic types and smoking history. Lung cancers with $FEV_1$/FVC less than 75% consisted of 35 cases of squamous cell cancer, 7 of small cell cancer(14%), 5 of adenocarcinoma(10%), 2 of large-cell carcinoma and 1 of unclassified carcinoma. Squamous cell carcinoma occured more in patients with $FEV_1$/FVC<75% than with $FEV_1$/FVC$\geq$75%(p<0.05). Conclusion: It was suggest that low $FEV_1$/FVC, as reflection of obstructive lung disease, may be at greater risk for squamous cell carcinoma in cigarette smoker.

In the present study, the author intended to investigate whether two oriental medical prescriptions named socheongryong-tang(SCRT) and Kamichihyo-san(KCHS) significantly affect mucin release from cultured hamster tracheal surface epithelial(HTSE) cells. Confluent HTSE cells were metabolically radiolabeled with ³H-glucosamine for 24 hrs and chased for 30 min in the presence of SCRT or KCHS to assess the effect of each agent on ³H-mucin release. Possible cytotoxicities of each agent were assessed by measuring lactate dehydrogenase(LDH) release. Also, the effects of SCRT and KCHS on contractility of isolated tracheal smooth muscle were investigated. The results were as follows: (1) SCRT significantly inhibited mucin release from cultured HTSE cells, without cytotoxicity; (2) KCHS significantly increased mucin release without cytotoxicity; (3) SCRT and KCHS did not affect contractility of isolated tracheal smooth muscle. We suggest that the effects of SCRT and its components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have the possible inhibitory effects on mucin release from the viewpoint of management of hypersecretion of airway mucus.
Roots of Astragalus membranaceus (A. membranaceus) (Astragali Radix, AR) has been used as a herbal medicine for gastrointestinal disorders in China, Korea, Japan, and other Asia countries. In this study we investigated the effects of the AR 70% ethanol extract and compounds isolated from AR on gastritis in rats and growth of human gastric cancer cells. AR 70% ethanol extract showed the potent acid-neutralizing capacities and partly cytotoxicity of Helicobacter pylori and human AGS gastric cancers cell. Astragaloside I and daucosterol, which were isolated from AR, significantly inhibited HCl/ethanol-induced gastric lesions. In addition, daucosterol increased the mucus content to almost the same as the positive control. The results of this study suggest that astragaloside I and daucosterol may be good candidates for the development of new drugs or neutraceuticals which can be used for the treatment or prevention of gastritis.
Using the electron microscopical method, this study was attempted to investigate the ultrastructural characteristics of glomeruli in FGS/Kist mice showing high proteinuria. The mucus-like substances were observed in glomerular capillary lumen which were usually narrowed by the widening of mesangial area filled with electron-dense deposits. Some portions of basal lamina in capillary wall were projecting to urinary space. Moreover, many vacuoles were observed in the cytoplasm of visceral epithelial cells, and an irregular-shaped fusion or effacement of foot processes were often shown in areas adjacent to basal lamina of glomeruli.
Objective : Allergic asthma is a chronic airway disease that affects millions of people in the developed world. The disease is characterized by concurring airway inflammation, Th2 cytokine production, increased mucus secretion, airway hyperresponsiveness (AHR) to inhaled antigen, and pulmonary fibrosis. To investigate the therapeutic and anti-asthmatic effects of Drynariae Rhizoma (DR), we examined the influence of DR on the development of pulmonary eosinophilic inflammation and airway hyperresponsiveness in a mouse model of allergic asthma. Methods : In this study, BALB/c mice were systemically sensitized to ovalbumin (OVA) followed intratracheally, intraperitoneally, and by aerosol allergen challenges. We investigated the effect of DR on airway hyperresponsiveness, pulmonary eosinophilic infiltration, various immune cell phenotypes, Th2 cytokine production and OVA specific IgE production in a mouse model of asthma. Results : In asthmatic mice, we found that DR.treated groups had suppressed eosinophil infiltration, allergic airway inflammation and AHR by suppressing the production of IL-5, IL-13 and OVA specific IgE. Conclusions : Our data suggest that the therapeutic mechanism by which DR effectively treats asthma is based on reductions of Th2 cytokines (IL-5), eotaxin, OVA-specific IgE production and eosinophil infiltration.

Objective : For various reasons, Obesity & Rhinitis are constantly rising. So Interest of treatment has been expanding. We want to verify The Chungyeol (fire extinguishing) Lisup (Draining) effect of Bangpungtongsungsan on obese model of allergy rhinitis.Material and Methods : BALB/c mouse were divided four groups: control(CON), allergic induction(ARE), Bangpungtongsungsan extract administration(BTT), Bangpungtongsungsan double concentration extract administration (BT2T). Every group except control group were caused allergic rhinitis by Ovalbumin. BTT & BT2T were orally administered the Bangpungtongsungsan for 21days. Since then we observed the liver tissue cell and the nasal mucous membrane. Results : In comparison with ARE, experimental groups show relief of the nasal mucous membrane damage(secretion of mucus decrease, Itching decrease), Th2 eruption control(IL-4 decline), effect of anti-inflamatory(reducing TNF-α creation, decreasing of iNOS through NF-κB activation-inhibition). In addition, experimental groups show a loss in weight, diminished accumulation of fat. (decreasing within liver tissue, reducing TNF-α creation) BT2T is more effective to BTT.Conclusion : Bangpungtongsungsan treat obese model on allergy rhinitis thereby control fat augmentation, relieving inflammation.

In the present study, the author intended to investigate whether two oriental medical prescriptions named GSGT and GCPT significantly affect mucin release from cultured hamster tracheal surface epithelial (HTSE) cells. Confluent HTSE cells were metabolically radiolabeled with 3H-glucosamine for 24 hrs and chased for 30 min in the presen+ce of GSGT or GCPT to assess the effect of each agent on 3H-mucin release. Possible cytotoxicities of each agent were assessed dy measuring lactate dehydrogenase(LDH) release. Also, the effects of GSGT and GCPT on contractility of isolated tracheal smooth muscle were investigated. (1) GSGT did not affect mucin release without cytotoxicity ; (2) GCPT significantly stimulated mucin release from cultured HTSE cells, with significant cytotoxicity ; (3) GSGT and GCPT did not affect contractility of isolated tracheal smooth muscle. We suggest that the effects of GCPT and its components should be further investigated and it is of great value to find, from oriental medical prescriptions, novel agents which have potent expectorant effects on mucin secretion from airway goblet cells.
Microbial dysbiosis in the gut is associated with human diseases, and variations in mucus alter gut microbiota. Therefore, we explored the effects of mucin on the gut microbiota using a community of 19 synthetic gut microbial species. Cultivation of these species in modified Gifu anaerobic medium (GAM) supplemented with mucin before synthetic community assembly facilitated substantial growth of the Bacteroides, Akkermansia, and Clostridium genera. The results of 16S rRNA microbial relative abundance profiling revealed more of the beneficial microbes Collinsella, Bifidobacterium, Ruminococcus, and Lactobacillus. This increased acetate levels in the community cultivated with, rather than without (control), mucin. We identified differences in predicted cell function and metabolism between microbes cultivated in GAM with and without mucin. Mucin not only changed the composition of the gut microbial community, but also modulated metabolic functions, indicating that it could help to modulate microbial changes associated with human diseases.

Gene expression in five tissues of the abalone (Haliotis discus hannai) was investigated using an expressed sequence tag (EST) analysis. Randomly selected clones were obtained from cDNA libraries constructed with gill (GI), digestive diverticula(DD), hepatopancreas (HP), foot/mucus (FM) and rectangular muscle (RM). Of 1,235 clonesanalyzed (288 clones for GI, DD, HP each,166 for FM, and 205 for RM), 741 (60.0%) clones in total turned out to share significant similarity with the sequences from NCBI GenBank (less than 10/sup -3/ of e-values), 423 sequences showed poor similarity (> 10/sup -3/), and 71 sequences didn't match with any sequences in GenBank. The percent unique sequence (singleton) was ranged from 56.1% (RM) to 74.7% (FM) among libraries. On the other hand, overall percent singleton was 55.3% when all the ESTs from five libraries were assembled into contigs. Analysis of the organisms represented by the best hit for each EST (e-values < 10/sup -3/) showed that 23.8% matched with mammalian entries, 24.0% with mollusks, 14.4% with insects, 11.6% with fish and 26.2% with others. The expressed patterns differed among the tissues when judged by the categorization of the sequences from each library into 10 broad functional classes. In all the libraries, the class I (no hit o. poor similarity) was the largest category with an average of 40.1%. This largest class was followed by class V (general metabolisms) in DD (21.9%), GI (14.6%) and HP (16.7%), while the 'cell structure and motility'(class VI) was the second largest class in remaining two libraries (31.2% for RM and 9.6% for FM). The class IX (cell division and proliferation) was the smallest class in all the libraries (less than 3%). This report provides the first tissue-specific lists of expressed abalone genes, which could be a fundamental basis for genomics program of abalone species.
Background and Objectives: Sinusitis, referring to an inflammatory disease that occurs on the mucus membrane inside the sinus, is one of the most common diseases in the otorhinolaryngology area. In oriental clinic, Sunbanghwalmyungeum (SHE) has been used as a primary prescription to treat inflammatory diseases and intumescence and to treat sinusitis. The aim of this study was to investigate the anti-inflammatory and anti-allergic effects of SHE on acute sinusitis induced mice. Materials and Methods: BALB/c mice were divided into three groups: the normal group, the group inoculated with S. pneumoniae which caused them allergic rhinitis (control group), and the group treated with the SHE extract after it was treated the same as the control group (sample group). We investigated the inhibition of Th 2 cell differentiation by SHE and the suppression of NF-${\kappa}B$ activation. Results: NF-${\kappa}B$ activation was suppressed, and iNOS & COX-2 production were inhibited by SHE in acute sinusitis. IL-4 and STAT 6 also appeared to be suppressed. The number of eosinophils in the sample group noticeably decreased when compared to the control group. In the general morphologic change, the increase of damaged respiratory ciliated epithelium & eosinophil's infiltration were decreased in the sample group. Goblet cells were maintained in the sample group. MIP-2 and HSP-70 decreased in the sample group. Apocrine secretion decreased in the sample group. Conclusion: The results suggest that SHE is significantly effective in the treatment of inflammation caused by acute sinusitis through the suppression of NF-${\kappa}B$ activation and the inhibition of Th 2 cell differentiation.
