SH21B is a natural composition composed of seven herbs: Scutellaria baicalensis Georgi, Prunus armeniaca Maxim, Ephedra sinica Stapf, Acorus gramineus Soland, Typha orientalis Presl, Polygala tenuifolia Willd and Nelumbo nucifera Gaertner (Ratio 3:3:3:3:3:2:2). In our previous study, we reported that SH21B inhibited adipogenesis and fat accumulation in 3T3-L1 cells through modulation of various regulators in the adipogenesis pathway. The aim of this study was to analyze the transcriptome profiles for the anti-adipogenic effects of SH21B in 3T3-L1 cells. Total RNAs from SH21B-treated 3T3-L1 cells were reverse-transcribed into cDNAs and hybridized to Affymetrix Mouse Gene 1.0 ST array. From microarray analyses, we identified 2,568 genes of which expressions were changed more than two-fold by SH21B, and the clustering analyses of these genes resulted in 9 clusters. Three clusters among the 9 showed down-regulation by SH21B (cluster 4, cluster 6 and cluster 9), and two clusters showed up-regulation by SH21B (cluster 7 and cluster 8) during the adipogenesis of 3T3-L1 cells. It was found that many genes related to cell proliferation and adipogenesis were included in these clusters. Clusters 4, 6 and 9 included genes which were related with adipogenesis induction and cell cycle arrest. Clusters 7 and 8 included genes related to cell proliferation as well as adipogenesis inhibition. These results suggest that the mechanisms of the anti-adipogenic effects of SH21B may be the modulation of genes involved in cell proliferation and adipogenesis.

${\beta}$-(1,3)-D-Glucans have been known to exhibit antitumor and antimicrobial activities. The presence of dectin-1,${\alpha}$, ${\beta}$-glucan receptor of dendritic cell, on macrophage has been controvertial. RT-PCR analysis led to the detection of dectin-1${\alpha}$ and ${\beta}$ in murine macrophage Raw264.7 cell line. Among the various organs of mouse, dectin-1${\alpha}$ and ${\beta}$ were detected in the thymus, lung, spleen, stomach and intestine. To analyze gene expression modulated by ${\beta}$-glucan treated murine Raw264.7 macrophage, total mRNA was applied to cDNA microarray to interrogate the expression of 7,000 known genes. cDNA chip analysis showed that ${\beta}$-glucan of P. osteatus increased gene expressions of immunomodulating genes, membrane antigenic proteins, chemokine ligands, complements, cytokines, various kinases, lectin associated genes and oncogenes in Raw 264.7 cell line. When treated with ${\beta}$-glucan of P. osteatus and LPS, induction of gene expression of TNF-${\alpha}$ and IFN-R1 was confirmed by RT-PCR analysis. Induction of TNF-R type II expression was confirmed by FACS analysis. IL-6 expression was abolished by EDTA in ${\beta}$-glucan and LPS treated Raw264.7 cell line, indicating that ${\beta}$-glucan binds to dectin-l in a Ca$\^$++/ -dependent manner. To increase antitumor efficacy of ${\beta}$-glucan, ginsenoside Rh2 (GRh2) was co-treated with ${\beta}$-glucan in vivo and in vitro tests. IC$\sub$50/ values of GRh2 were 20 and 25 $\mu\textrm{g}$/$m\ell$ in SNU-1 and B16 melanoma F10 cell line, respectively. Co-treatment with ${\beta}$-glucan and GRh2 showed synergistic antitumor activity with cisplatin and mitomycin C both in vitro and in vivo. Single or co-treatment with ${\beta}$-glucan and GRh2 increased tumor bearing mouse life span. Co-treatment with ${\beta}$-glucan and GRh2 showed more increased life span with mitomycin C than that with cisplatin. Antitumor activities were 67% and 72 % by co-injection with ${\beta}$-glucan and GRh2 in the absence or presence of mitomycin C, respectively.
This study was carried out to investigate the anticancer effect of extracts from sulfur fed duck carcass. Growth inhibition of cancer cell lines was measured by MTT assay. Eleven cancer cell lines, such as Calu-3(human lung carcinoma), SK-MES-1(human lung carcinoma), HL6O(human leukemia), KB(human epidermoid of mouth carcinoma), Farrow(human melanoma), HEP-2(human larynx carcinoma), SNU-1(human stomach carcinoma), K-562 (human leukemia), WiDr(human colon carcinoma), P388(mouse leukemia) and 3LL(mouse lung carcinoma) showed the growth inhibition higher than 50%, but those, such as SF-188(human brain carcinoma), A-549(human lung carcinoma) and HEC-lB(human uterus carcinoma) showed the growth inhibition lower than 50% in the extract of sulfur fed duck carcass at the concentration of 10 mg/㎖. The sulfur fed duck carcass extract had better growth inhibition than the normal counterpart against various cancer cell lines at the concentration of 10 mg/㎖. When the effect of growth inhibition of an effluent by different concentrations of methyl alcohol(25, 50, 75 and 100%) tested on Diaion HP-20 column chromatography, an effluent by concentration of 100% methyl alcohol showed the most strong effect of growth inhibition against HEP-2(human larynx carcinoma).

We present here the effect of extracellular matrix (ECM) on the proliferation and physiology of HL-1 cardiac cells. HL-1 cell is from AT-1 mouse atrial cardiomyocyte tumor lineage. HL-1 cell can be serially passaged, yet they maintain the ability to contract which is a promising character of HL-1 cell for the cell based biosensors. HL-1 cells grow up on the ECM which can affect on the attachment and growth of HL-1. In this paper, we discuss HL-1 cell-ECM interactions with three different ECMs and non-treated surface. HL-1 cells are grown for 4 days after seeding then observed their attachment. Also they were immunostained by hoechst and EthD-1 for proliferation, phalloidin for Factin, and DAPI for nuclei. Fibronectin was revealed as the proper ECM material for HL-1 cell culture. This study can provide basic information for understanding the cell-ECM interactions and growth of HL-1 cells.
Osteoblast(OBL) cells were isolated from ICR Swiss neonatal mouse calvarial tissues and cultured in a CO2 incubator with minimum essential medium (MEM) containing 0.25g BSA. The cells were cultured for 7 days and were treated with bovine parathyroid hormone (bPTH, 1-34) and calcitonin(CT). Enzyme activities related to mineral metabolism and other biochemical actions within the bone cells including protein phosphorylation were investigated. In other experiments using cultured calvarial bone tissues, hormones were treated for 24, 48, 72 or 96 hours. The activities of $\beta$-glucuronidase enzymes involved in bone collagen synthesis and mineral deposits were increased by 8% with bPTH and were inhibited with CT treatment, while those were 67% increase treated with bPTH and CT together. On the other hand, alkaline phophatase(AP) activities were inhibited by PTH hormone at all the time courses observed. Protein phosphorylation reaction in OBL was mediated by bPTH, cAMP and ionized Ca. Phosphorylation was observed in different cell fractions including homogenate, membrane and cytosol. The number of proteins phosphorylated by PTH, cAMP, and Ca were 10, 5, and 9, respectively. Most of the protein kinases(PKs) were existed in cytosolic compartment. In membrane fractions, two bPTH-dependent-PKs (70K, 50K Da) were observed of which 70K Da protein was also Ca-dependent. Most of the cAMP-dependent PKs were regulated via bPTH. 70K, 50K, 5K, 19K, 16K, 10.5K phosphoproteins regulated by Ca share the same pathways as those by bPTH-dependent proteins. Ca seems to regulate PK activities differently from cAMP.
Mizoribine (MZR) has been shown to possess immunosuppressive activity that selectively inhibits the proliferation of lymphocytes by interfering with inosine monophosphate dehydrogenase. The efficacy of MZR is not only in patients who have had renal transplantation, but also in patients with rheumatoid arthritis (RA), lupus nephritis, and primary nephritic syndrome. Because the exact mechanism of its immunosuppressive action is not clear, the object of this study was to examine the ability of MZR to regulate the antigen presenting cells (APCs), dendritic cells (DCs). In this work, we tested whether MZR ($1{\sim}10\;{\mu}g/mL$) could inhibit the cross-presentation of DCs. DC2.4 cells ($H-2K^{b}$) or bone marrow-derived DCs (BM-DCs) generated from BM cells of C57BL/6 mouse ($H-2K^{b}$) were cultured in the presence of MZR with OVA-microspheres, and the amount of OVA peptide-class I MHC complexes was measured by a T cell hybridoma, B3Z, that recognizes OVA (257-264 : SIINFEKL)-$H-2K^{b}$ complex and expresses-galactosidase. MZR profoundly inhibited the expression of SIINFEKL-$H-2K^{b}$ complexes. This inhibitory activity of MZR appeared to affect the phagocytic activity of DCs. MZR also decreased IL-2 production when we examined the effects of MZR on $CD4^{+}$ T cells. These results provide an understanding of the mechanism of immunosuppressive activity of MZR on the inhibition of MHC-restricted antigen presentation and phagocytic activity in relation to their actions on APCs.

NF-${\kappa}B$ is a transcriptional factor which is involved in many biological processes including immunity, inflammation, and cell survival. Many investigators studied on the mechanism involved in activation of NF-${\kappa}B$ signalling pathway via ubiquitination and degradation of $I{\kappa}B$ regarding skin disease. Some specific molecules including Akt, MEK, p38 MAP Kinase, Stat3, et al. represent convergence points and key regulatory proteins in signaling pathways controlling cellular events such as growth and differentiation, energy homeostasis, and the response to stress and inflammation. Ultraviolet (UV) irradiation has many adverse effects on skin, including inflammation, alteration in the extracellular matrix, cellular senescence, apoptosis and skin cancer. Prunus mume, a naturally derived plant extract, has beneficial biological activities as blood fluidity improvement, anti-fatigue action, antioxidative and free radical scavenging activities, inhibiting the motility of Helicobacter pyolri. Previous reports on various beneficial function prompted us to investigate UVB-induced or other immunostimulated biological marker regarding P. mume extract. P. mume extract suppresses UVB-induced cyclooxygenase-2 (COX-2) expression in mouse skin epidermal JB6 P+ cells. The activation of activator protein-1 and nuclear factor-${\kappa}B$ induced by UVB was dose-dependently inhibited by P. mume extract treatment. This results suggest that P. mume extracts might be used as a potential agents for protection of inflammation or UVB induced skin damage.

The use of pluripotent stem cells has tremendous advantages for various purposes but these cell lines with proven germ-line transmission have been completely established only in the mouse. Embryonic germ (EG) cell lines are also pluripotent and undifferentiated stem cells established from primordial germ cells (PGCs). This study was conducted to establish and characterize the chicken EG cells derived from gonadal primordial germ cells. We isolated gonadal PGCs from 5.5-day-old (stage 28) White leghorn (WL) embryos and established chicken EG cells lines with EG culture medium supplemented with human stem cell factor (hSCF), murine leukemia inhibitory factor (mLIF), bovine basic fibroblast growth factor (bFGF), human interleukin-11 (hIL-11), and human insulin-like growth factor-I (hIGF-I). These cells grew continuously for 4 months (10 passages) on a feeder layer of mitotically active chicken embryonic fibroblasts. These cells were characterized by screening with the Periodic acid-Shiff's reaction, anti-SSEA-1 antibody, and a proliferation assay after several passages. As the results, the chicken EG cells maintained characteristics of undifferentiated stem cells as well as that of gonadal PGCs. When cultured in suspension, the chicken EG cells successfully formed an embryoid body and differentiated into a variety of cell types when re-seeded onto culture dish. The chicken EG cells were injected into blastodermal layer at stage X and dorsal aorta of recipient embryo at stage 14 (incubation of 53hrs) and produced chimeric chickens with various differentiated tissues derived from the EG cells. The germline chimeras were also successfully induced by using EG cells. Thus, Chicken EG cells will be useful for the production of transgenic chickena and for studies of germ cell differentiation and genomic imprinting.
Activating and inhibitory cell surface receptors play important roles in regulation of immune responses. Recent progress has demonstrated that many inhibitory receptors pair with activating, as well as inhibitory, isoforms, both of whose genes are located in small clusters on a chromosome. We and others identified paired activating and inhibitory immunoglobulin-like receptors, designated myeloid-associated immunoglobulin-like receptors (MAIR) (CD300). MAIR is a multigene family consisting of nine genes on a small segment of mouse chromosome 11. MAIR family receptors are preferentially expressed on myeloid cells, including macrophages, dendritic cells, granulocytes, and bone-marrow-derived cultured mast cells, and a subset of B cells and regulate activation of these cells. Thus, MAIR plays an important role in innate immunity mediated by myeloid cells.

Refreshable Braille displays have already been developed in the past, but they remain quite bulky and expensive. Small displays contain only a single or a few Braille cells, with pins pushing into the fingertips. However, they are not as successful as larger displays because the skin is more sensitive to lateral movement than to orthogonal pressure. This, paper presents the design of linear electromagnetic actuators, which allows a considerable reduction in size and cost for such refreshable Braille displays. Different actuator concepts are compared. A new actuator is proposed and optimised by means of finite element simulations, which are verified with measurements on a prototype.
