Expression patterns of PRDM10 during mouse embryonic development

  • It is well known that PR/SET family members participate in transcriptional regulation via chromatin remodeling. PRDM10 might play an essential role in gene expression, but no such evidence has been observed so far. To assess PRDM10 expression at various stages of mouse development, we performed immunohistochemistry using available PRDM10 antibody. Embryos were obtained from three distinct developmental stages. At E8.5, PRDM10 expression was concentrated in the mesodermal and neural crest populations. As embryogenesis proceeded further to E13.5, PRMD10 expression was mainly in mesoderm-derived tissues such as somites and neural crest-derived populations such as the facial skeleton. This expression pattern was consistently maintained to the fetal growth period E16.5 and adult mouse, suggesting that PRDM10 may function in tissue differentiation. Our study revealed that PRDM10 might be a transcriptional regulator for normal tissue differentiation during mouse embryonic development.

Chromosomal band pattern of black-striped field mouse (Apodemus agrarius) (한국 야생 등줄쥐의 고해상도 염색체분염상)

  • We investigated the cytogenetic characteristics of male black-striped field mouse (Apodemus agrarium) in Korea. Chromosome slides were obtained from blood cell cultures which were synchronized with thymidine blocking or not. In the chromosome slide which synchronization with thymidine blocking was employed on, the GTG(G bands by trypsin using Giemsa)-bands of high resolution were observed. The male black-striped field mouse has 48 chromosomes composed 46 autosomes and XY sex chromosomes. The centromeric regions of autosomes were positive to GTG-banding. According to this investigation, thymidine blocking in cell culture process was useful to get lengthened chromosomes. It may be necessary to employ RBG-banding technique to investigate complementary band patterns between R- and G-banding in black-striped field mouse.

Effects of Various Kinds and Concentrations of Cryoprotectants on Viabilities of Ultrarapidly Frozen 4-cell Mouse Embryos and Morulae (생쥐 4세포기 및 상실배기 수정란의 초급속동결에 있어서 동결보호제의 종류와 농도가 생존성에 미치는 영향)

  • This study was carried out to select the best cryoprotectant and to establish optimal concentration of the cryoprotectant in ultrarapid freezing of mouse 4-cell embryos and morulae, respectively. We investigated survival of ultrarapid frozen embryos according to various cryoprotectants such as glycerol, ethylene glycol, propylene glycol and dimethyl sulfoxide (DMSO). Suvival of the embryos frozen at different concentrations (3.0, 4.0 and 5.0 M) of indivisual cryoprotectant was also tested. Preimplantation developmental rate (96.3%, 83/86) of 4-cell mouse embryos treated with 4.0 M ethylene glycol after ultrarapid freezing and thawing was higher than those of other cryoprotectants (glycerol, propylene glycol and DMSO). In the ultrarapid freezing of mouse morulae, the highest developmental rate (98.8%, 89 /90) of the embryos to blastocysts was shown in the group of 5.0 M glycerol. Thus, these results demonstrate that 4.0 M ethylene glycol and 5.0 M glycerol are optimal for ultrarapid freezing of 4-cell mouse embryos and morulae, respectively.

  • The purpose of this research was to investigate effect of water extract of DangKwi-Eum-Ja ka Sumsoo(DESE) on the cytotoxicity of human epidemloid cell, A431 cells. The effects of DESE on the proliferation of A431 cells, Balb/c 3T3 cells, mouse thymocytes and splenocnes were estimated by MTT colorimetric assay, and nitric oxide production from mouse peritoneal macrophage was estimated by Griess method. DESE inhibited the proliferation of A431 cells at $10{\mu}g/ml$, and did not affect the proliferation of Balb/c 3T3 cells. DESE decreased the cytotoxicity of mitomycin C or cisplatin on A431 cells, increased the cytotoxicity of mitomycin C or cisplatin on Balb/c 3T3 cells. DESE inhibited the proliferation of mouse thymocytes and splenocytes at $100{\mu}g/ml$. DESE did not affect the nitric oxide production from mouse peritoneal macrophage in vitro, but decreased the nitric oxide production from DESE-treated mouse peritoneal macrophage.

Isolation and Characterization of a CDNA Encoding a Protein Homologous to the Mouse 70 kDa Heat Shock Protein (생쥐 섬 유아세포에서 70 kDa 고온충격 단백질의 CDNA 클로닝과 염기서열 분석)

  • Hsp70, a 70 kDa protein, is the maior protein expressed when cells are heat-shocked. A cDNA library from mouse ID13 cells was screened with the human hsp70 gene as a probe, and a positive clone was obtained. The positive clone was subcloned into puc19 and the precise restriction was obtained. The CDNA was sequenced by the Sanger's dideoxv termination method. Single open reading frame that codes for a protein of 70 kDa was found. The DNA sequence of the cloned mouse DNA shows great homology (66-90%) with other mouse hsp70 genes and somewhat less homology (50",) with E. coli hsp70 gene (dnak). With the exception of one amino acid, the protein sequence deduced from the CDNA is identical to the mouse that shock cognate protein 70 (hsc70) that is constitutivelv expressed at normal temperature. The result suggests that the cloned CDNA encodes a hsc70 family rather than a heatinducible family.mily.

Developmental potential of aggregated mouse embryos and production of chimeras after freezing (동결보존한 마우스 집합배의 생존성과 chimera의 생산에 관한 연구)

  • The present study was carried out to investigate the viability of frozen-thawed aggregated mouse embryos and to produce the chimeras. Different phenotypic embryos were obtained by mating ICR female mice with ICR or CBA male mice. The aggregated morulae were produced following aggregation of embryos at 4-, 8- and 12- to 16-cell stage. The desirable stage for the aggregation of the mouse embryos was 8- to 16-cell stage. The post-thawed in vitro survival rates of aggregated embryos in glycerol, DMSO and ethylene glycerol were 51.5, 78.6 and 69.4%, respectively. Although the higher survival was obtained in DMSO, there were no significant differences in the survival rates among the three cryoprotectants. A total of 155 frozen-thawed aggregated embryos were transferred to 11 recipient mice, 3 out of 7 offsprings were born to overt chimera. These experiments have proven that mouse chimeras can be obtained from frozen-thawed aggregated embryos.

Quantifying Inundation Analysis in Misari motorboat racing stadium using MOUSE (MOUSE를 활용한 미사리 조정경기장의 정량적 침수해석)

  • Recently, heavy rainfalls due to the climate change in Korea have caused inundation problems in urban sewer networks. In july 2006, a flooding accident at Misari motorboat racing stadium near the Han river occurred due to the effect of record-breaking outflow discharge from Paldang-dam. The purpose of this study was to simulate and analyze the flooding accident at Misari stadium by MOUSE model. The results of simulation analysis indicated that the total flood volume was $1,313,450m^3$. The effect of back water was 85.9% of the total volume which was caused by the manhole accident, and the effect of accumulated runoff was 14.1% of total volume which was caused by non-return valve shutdown. The simulation results of this MOUSE modeling that was linked to the boundary condition of the dynamic flows in the river by DWOPER model showed the potential of successful inundation analysis for sewer networks.

Design and Implementation of a Multimodal Input Device Using a Web Camera

  • We propose a novel input pointing device called the multimodal mouse (MM) which uses two modalities: face recognition and speech recognition. From an analysis of Microsoft Office workloads, we find that 80% of Microsoft Office Specialist test tasks are compound tasks using both the keyboard and the mouse together. When we use the optical mouse (OM), operation is quick, but it requires a hand exchange delay between the keyboard and the mouse. This takes up a significant amount of the total execution time. The MM operates more slowly than the OM, but it does not consume any hand exchange time. As a result, the MM shows better performance than the OM in many cases.

  • In this paper, we design and implement a mouse movement measurement system using an infrared sensor called MAMS (Mouse Activity Measurement System). Unlike existing systems, MAMS can measure movements between IREDs. MAMS is removable allowing convenient portability and is also low cost. MAMS automatically measures mouse activity during a pre-set time interval. Since the measured data can be easily stored in a computer system, it is much simpler and more efficient than the Ugo 47420 model. Moreover, MAMS can be used in medical and veterinary field to eliminate manual observations.

  • Objective: To verify the effect of Matrigel, a ECM complex from Engelbreth-Holm-Swarm (EHS) mouse sarcoma on the preimplantation development and apoptosis of mouse fertilized eggs. Method: Late pronucleus stage eggs were cultured through the blastocyst stage in the presence of Matrigel (0.5%, v/v). Characteristics of apoptosis and cell number assesed by Hoecst staining and TUNEL labeling at the blastocyst stage, respectively. Results: Morphological development, number of cells per embryo was significantly increased but rate and number of TUNEL positive nuclei of the embryo were decreased in the presence of Matrigel. Conclusion: This result suggested that at low concentration of Matrigel improves both viability and morphological development in the preimplantation mouse embryos.
