Lack of Mitochondrial DNA Sequence Divergence between Two Subspecies of the Siberian Weasel from Korea: Mustela sibirica coreanus from the Korean Peninsula and M. s. quelpartis from Jeju Island

  • The objective of this study was to determine the degree of mitochondrial DNA (mtDNA) divergence between two subspecies of $Mustela$ $sibirica$ from Korea ($M.$ $s.$ $coreanus$ on the Korean Peninsula and ($M.$ $s.$ $quelpartis$ on Jeju Island) and to examine the taxonomic status of ($M.$ $s.$ $quelpartis$. Thus, we obtained complete sequences of mtDNA cytochrome $b$ gene (1,140 bp) from the two subspecies, and these sequences were compared to a corresponding haplotype of ($M.$ $s.$ $coreanus$, downloaded from GenBank. From this analysis, it was observed that the sequences from monogenic ($M.$ $s.$ $quelpartis$ on Jeju Island were identical to the sequences of four ($M.$ $s.$ $coreanus$from four locations across the Korean Peninsula, and that the two subspecies formed a single clade; the average nucleotide distance between the two subspecies was 0.26% (range, 0.00 to 0.53%). We found that the subspecies $quelpartis$ is not genetically distinct from the subspecies $coreanus$, and that this cytochrome $b$ sequencing result does not support the current classification, distinguishing these two subspecies by pelage color. Further systematic analyses using morphometric characters and other DNA markers are necessary to confirm the taxonomic status of ($M.$ $s.$ $quelpartis$.

Maturation and Spawning of the Korean Anchovy Coilia nasus on the West Coast of Korea (한국 서해산 웅어, Coilia nasus 암컷의 성숙과 산란)

  • The gonadosomatic index (GSI), fatness, ovarian development, first sexual maturity, and fecundity of the Korean anchovy Coilia nasus were investigated by histological observations and morphometric analysis from January to December, 2007. The GSI and fatness began to increase in February, and reached the maximum in June when the ovary was getting mature and spawning occurred. Thereafter these parametes rapidly decreased in July when spawning occurred. Therefore, monthly changes in the GSI and fatness were closely related to ovarian maturation and spawning. The duration of ovarian development in females can be classified into five successive stages: early growing stage (February to March), late growing stage (March to April), mature stage (May to June), ripe and spent stage (June to July), and recovery and resting stage (December to January). Maturation and spawning of this species occurred between June and July during the period of high seawater temperature-long day length. Percentages of first sexual maturity in female individuals were over 50% for fish ranging 24.1 to 27.0 cm in total length, and 100% for fish over 30.1 cm in total length. The number of total eggs and mature eggs in the absolute fecundity were increased with the increase of total length and body weight, respectively. The number of total eggs and mature eggs in relative fecundity were also proportional to total length, but rather these numbers decreased in the maximum body weight (126.0${\sim}$150.0 g).

Desalinated underground seawater of Jeju Island (Korea) improves lipid metabolism in mice fed diets containing high fat and increases antioxidant potential in t-BHP treated HepG2 cells

  • This study was performed to investigate the effect of desalinated underground seawater (named as 'magma seawater', MSW) of Jeju Island in Korea on lipid metabolism and antioxidant activity. MSW was collected from underground of Han-Dong in Jeju Island, and freely given to high fat diet (HFD)-fed C57BL/6 mice for 10 weeks. Although there were no significant differences in the body weight changes and plasma lipid levels, hepatic triglyceride levels were significantly lower in the MSW group than in the normal tap water (TW)-drunken control group. Furthermore, the activity of fatty acid synthase (FAS) was significantly decreased and carnitine palmitoyltransferase (CPT) activity was increased in MSW group compared to TW group. Similarly, real-time PCR analysis revealed that mRNA expressions of lipogenic genes were lowered in MSW groups compared to the control group. In a morphometric observation on the liver tissue, accumulation of fats was remarkably reduced in MSW group. Meanwhile, in vitro assay, tree radical scavenging activity measured by using diphenylpicrylhydrazyl (DPPH) was increased in MSW group. The 2'-7'-dichlorofluorescein diacetate (DCF-DA) staining followed with fluorescent microscopy showed a low intensity of fluorescence in MSW-treated HepG2 cells, compared to TW-treated HepG2 cells, which indicated that the production of reactive oxygen species by tert-butyl hydroperoxide (t-BHP) in HepG2 cells was decreased by MSW treatment. The antioxidant effect of MSW on t-BHP-induced oxidative stress in HepG2 cells was supported by the increased activities of intracellular antioxidant enzymes such as catalase and glutathione reductase. From these results, we speculate that MSW has an inhibitory effect on lipogenesis in liver and might play a protective role against cell damage by t-BHP-induced oxidative stress.

Variation Pattern of mtDNA among Six Subspecies of Apodemus agrarius(Mammalia, Rodentia) in Korea, China, and Russia (한국, 중국, 러시아에 서식하는 등줄쥐, Apodemus agrarius(포유강, 설치목), 6아종의 미토콘도리아 DNA 절단단편의 변이)

  • One hundred and eleven samples of six subspecies of striped field mouse, Apodemus agrarius Pallas from Korea, China and Russia, were used for the analysis of mitochondrial DNA(mtDNA) fragment patterns resulted from the digestion with eight restriction enzymes by blot hybridization technique. All 32 fragments, nine mtDNA haplotypes, and four major subgroups with the mean divergence value of 0.896 to 1.150% were revealed. In summary, three forms are recognized: [I, subspecies chejuensis (Chejudo island, Korea)], [II, subspecies pallescens (southwestern Korea), coreae (central Korea), and septentrionalis (Russia)], and [III, subspecies manchuricus (northeastern China) and pallidior (northern China)], although some samples of subspecies coreae are somewhat different from almost all samples of six subspecies, and some samples of subspecies pallidior are similar with all samples of subspecies septentrionalis to form same haplotype. It is confirmed that A. agrarius chejuensis is a distinct subspecies, that subspecies coreae (including pallescens) is also a distinct subspecies, that subspecies manchuricus and pallidior are synonyms of subspecies ningpoensis, and that subspecies septentrionalis is a synonym of subspecies ningpoensis, and that subspecies septentrionalis is a synonym of subspecies agrarius. Moreover, it seems that A. agrarius shows constant karyotype, minimal variation in mtDNA genotype, and considerable divergence in morphometric characters, although further analyses with additional samples of A. agrarius in Eurasia will be necessary to determine the degree of variation of these taxonomic characters and to clarify subspecies classification as well.

Reproductive Cycle of Sulf clam, Coecella chinensis (Mesodesmatidae: Bivalvia) (퇴조개, Coecella chinensis (Mesodesmatidae: Bivalvia)의 생식주기)

  • The reproductive cycle, gonad index, condition index, and the sex ratio in female and male Coecella chinensis, which were collected from the coastal waters of Namhae, the South Coast of Korea, were investigated by histological analysis and morphometric data. Monthly variations of the gonad index and condition index of this species showed similar patterns to that of the reproductive cycle. The reproductive cycle in female and male C. chinensis can be classified into six successive stages: early active stage (April), late active stage (May), ripe stage (June), partially spawned/spent stage (June to August), degeneration stage (August) and inactive stage (September to March). According to monthly changes in relative frequency distributions of ovarian egg diameters of this species, in particular, a number of ripe eggs ranging about 70 ${\mu}m$ appear in June, however, in July and August, although the mode of egg diameters ranging about 50 ${\mu}m$ appear about 80%, these ripe eggs ranging from 60 ${\mu}m$ to 70 ${\mu}m$ are gradually decreased during the period from June to August. Therefore, the spawning period of this species is continued from June to August with a peak between July and August when the seawater temperature exceeds about $24.5^{\circ}C$. The sex ratio of female and male individuals was not significantly different from 1:1 (${\chi}^2$=1.20, p>0.05). No hermaphrodites were found.

A Geomorphological Classification System to Chatacterize Ecological Processes over the Landscape (생태환경 특성 파악을 위한 지형분류기법의 개발)

  • The shape of land surface work as a cradle for various environmental processes and human activities. As spatially distributed process modelings become increasing important in current research communities, a classification system that delineates land surface into characteristic geomorphological units is a pre-requisite for sustainable land use planning and management. Existing classification systems are either morphometric or generic, which have limitations to characterize continuous ecological processes over the landscape. A new classification system was developed to delineate the land surface into different geomorphological units from Digital Elevation Models(DEMs). This model assumes that there are pedo-geomorphological units in which distinct sets of hydrological, pedological, and consequent ecological processes occur. The classification system first divides the whole landsurface into eight soil-landscape units. Possible energy and material nows over the land surface were interpreted using a continuity equation of mass flow along the hillslope, and subsequently implemented in terrain analysis procedures. The developed models were tested at a 12$\textrm{km}^2$ area in Yangpyeong-gun, Kyeongi-do, Korea. The method proposed effectively delineates land surface into distinct pedo-geomorphological units, which identify the geomorphological characteristics over a large area at a low cost. The delineated landscape units mal provide a basic information for natural resource survey and environmental modeling practices.

Assessment of buccal bone thickness of aesthetic maxillary region: a cone-beam computed tomography study

  • Purpose: The aim of this study was to analyze the anatomical dimensions of the buccal bone walls of the aesthetic maxillary region for immediate implant placement, based upon cone-beam computed tomography (CBCT) scans in a sample of adult patients. Methods: Two calibrated examiners analyzed a sample of 50 CBCT scans, performing morphometric analyses of both incisors and canines on the left and right sides. Subsequently, in the sagittal view, a line was traced through the major axis of the selected tooth. Then, a second line (E) was traced from the buccal to the palatal wall at the level of the observed bone ridges. The heights of the buccal and palatal bone ridges were determined at the major axis of the tooth. The buccal bone thickness was measured across five lines. The first was at the level of line E. The second was at the most apical point of the tooth, and the other three lines were equidistant between the apical and the cervical lines, and parallel to them. Statistical analysis was performed with a significance level of $P{\leq}0.05$ for the bone thickness means and standard deviations per tooth and patient for the five lines at varying depths. Results: The means of the buccal wall thicknesses in the central incisors, lateral incisors and canines were $1.14{\pm}0.65mm$, $0.95{\pm}0.67mm$ and $1.15{\pm}0.68mm$, respectively. Additionally, only on the left side were significant differences in some measurements of buccal bone thickness observed according to age and gender. However, age and gender did not show significant differences in heights between the palatal and buccal plates. In a few cases, the buccal wall had a greater height than the palatal wall. Conclusions: Less than 10% of sites showed more than a 2-mm thickness of the buccal bone wall, with the exception of the central incisor region, wherein 14.4% of cases were ${\geq}2mm$.

The First Record on the Rhynchocypris kumgangensis from Taehwa River, Korea (태화강에서 처음 채집된 금강모치(Rhynchocypris kumgangensis))

  • Morphological characteristics and population characteristics of the Rhynchocypris kumgangensis were investigated at Taehwa River from April 2014 to April 2015. This is the first record on R. kumgangensis not only from Guksu stream (the upstream region of Taehwa River, Ulsan-si Ulchu-gun Beomseo-eup Kuksu-ri) but also from the stream flowing into the Gangneungnamdae stream, in the southern part of East Sea, Korea. The morphometric characteristics of R. kumgangensis in the Taehwa River and the Han River are very similar except that the population at Taehwa River showed shorter caudal peduncle length than caudal peduncle depth when compared with the population at Han River. In terms of meristic character the population at Taehwa River showed fewer numbers of scales at the lateral line, above the lateral line and below the lateral line. Other than that, the two populations showed the same meristic characters. The fish cohabiting with R. kumgangensis mainly composed of Zacco platypus (25.2%), Rhynchocypris oxycephalus (24.1%) and Zacco koreanus (21.8%). Length frequency distribution analysis indicated that the length of the group ranged between 45~65 mm and the average length was 54.3 mm. Calculation of the length-weight relation of R. kumgangensis showed the value of constant a as 0.000002 and b as 3.34, and the Condition factor (K) was 1.10 an on average.

Improved Early Survival in Backcrosses of Male Mud Loach (Misgurnus mizolepis)$\times$Cyprinid Loach (M. anguillicaudatus) Hybrids to Femal Cyprinid Loach (미꾸리, Misgurnus anguillicaudatus의 생존율 향상을 위한 역교배체 생산)

  • Backcross hybridization between female cyprinid loach(Misgurnus anguillicaudatus) any male mud loach (M. mizolepis)$\times$cyjprinid loach hybrid were made, and its effect on early survival, cytogenetic traits, and gonad development were examined. Mean fertilization rate and hatching success of backcross hybrids were similar to those found in the maternal species, the cyprinid loach. However, the backcross hybrids revealed significantly improved early survival up to yolk sac absorption (86.0%) compared to cyprinid loach (0%) under low culture temperature ($15^{\circ}C$). the erythrocytic size and DNA content of backcross hybrids were nearly intermediate to those of the parents. Karyological analysis of backcross hybrids displayed two kinds of modal chromosome number of 2n=48 or 2n=49. Growth of backcross hybrids over 6 months after hatching was proven to be intermediate between their parental species. Although backcross hybrids had intermediate values between those of thier parents in most morphometric traits, overall external morphology of backcross hybrids was morer simlar to cyprinid loach than mud loach$\times$cyprinid loach hybrid. Histological examination of gonads at 2 and 4 months of age revealed that no female was obseved in backcross hybrid groups, suggesting the possibility of production of monosex male population.

Morphological and Genetic Stock Identification of Todarodes pacificus in Korean Waters (한국 주변해역에 서식하는 살오징어(Todarodes pacificus)의 형태 및 유전학적 계군분석)

  • Stock identification of Todarodes pacificus collected in the East Sea, Yellow Sea and East China Sea during the period from September to December in 2011 was analyzed by morphometric characters and mitochondrial DNA (mtDNA) cytochrome oxidase subunit I (COI) gene nucleotide variations. Frequency distributions of mantle length was analyzed by morphological method with measuring size of T. pacificus. Then each stock was estimated to confirm their maturation for mean mantle length comparing with mean mature mantle length 20-22 cm. According to morphologic stock identification, it is estimated that the northern part of East Sea is categorized as summer stock and the rest parts, including mid /southern part of the East Sea, northern part of the East China Sea and northern part of the West Sea were autumn stock. For genetic analysis, a total 49 haplotypes were defined by 33 variable nucleotide sites. From the extensive haplotype diversity, limited nucleotide diversity and star-like shape of haplotype network, T. pacificus appears to have undergone rapid population expansion from an ancestral population with a small effective population size. Although pair-wise Fst estimates which represent genetic difference among groups were low, there are relatively remarkable difference of Fst between middle and southern part of the East Sea. Although middle part of the East Sea and southern part of the East Sea were situated at the East Sea, genetically separated groups were appeared.
