Background & Objective : Naeso-san(NSS) has been used for the treatment of functional dyspepsia, regarded as a gastric dysmotility disease. A main cause of gastric dysmotility is antral dilatation or antroduodenal uncoordination. Therefore, we investigated the effect of NSS on gastric motility and its mechanism of action, as well as the morphologic changes in antral dilatated rats. Methods : Antral dilatated rats were induced by wrapping a nonabsorbable rubber ring(D:6mm, W:4mm, T:1mm) around the 1st portion of the duodenum for 8 weeks. Then morphologic changes were investigated and compared with normal intact rats before and after 8 weeks. Gastric emptying was measured by administration of normal saline(NS) or NSS in normal intact and antral dilatated rats. In another series of experiments to evaluate the mechanism of NSS under delayed conditions, normal intact rats were treated with atropine sulfate(1mg/kg, s.c.), quinpirole HCl(0.3mg/kg, i.p.), $NAME(N^{G}-nitro-L-arginine$ methyl ester, 75mg/kg, s.c.) and cisplatin(10mg/kg, i.p.), respectively. The myoelectrical activity of the gastric smooth muscle was recorded in normal intact and antral dilatated rats. The contractile waves were measured for 30 minutes before and after administration of each solution(NS, NSS). Results : Body weight gain of antral dilatated rats was significantly lower than that of the controls. Futhermore, we found the thickness of the mucosal and muscular layers and surface area of the stomach increased significantly compared with controls. NSS 278㎎/㎏ improved gastric emptying more than normal saline or NSS 93mg/kg in normal intact(p=0.026) and antral dilatated rats(p=0.03). NSS enhanced gastric emptying significantly in the NAME treated group(p=0.002). NSS 278mg/kg increased the significant postprandial dominant power than that of NS in normal intact rats, whereas there was no statistical significance in antral dilatated rats. Conclusions : NSS stimulates gastric motility through the cholinergic pathway. We expect that pathologic model with antral dilatation can be used as an exprimental tool which is similar to dyspepsia and NSS would be effective especially in dysmotility-like functional dyspepsia with antral dilatation or impaired reservoir functions such as gastric adaptive relaxation.
본 연구는 수컷 생식기관내 에스트로겐 작용을 구명하기 위한 과정의 일환으로 항에스트로겐 물질인 ICI 182,780을 투여하고, 이로 인한 수출소관의 형태적 변화를 관찰하였다. 대조군과 처리군 간에 수출소관의 관강 직경은 처리후 8일째에 팽창하기 시작하여 59일째까지 지속되었고, 처리군 마우스의 수출소관내의 상피세포 높이는 대조군과 비교해 볼 때, 처리 후 경과기간 내내 감소되었다. 사이세포 높이가 최대로 감소된 곳은 처리된 마우스 수출소관의 근위부로 38% 감소하였으며, 수출소관 상피세포내의 핵상 세포질의 높이는 처리군 마우스의 근위부, 원추부, 원위부에서 각각 46%, 39%, 33% 감소된 것으로 나타났다. 항에스트로겐, ICI 182,780을 수컷 마우스에 투여한 본 실험에서, ${\alpha}ERKO$ 유전자 변형 동물에서 관찰되었던 것과 유사한 생식기관의 형태적 변화가 나타났다. 즉 항에스트로겐이 에스트로겐 수용체와 결합하여 에스트로겐의 기능을 차단하고 이로 인해 수출관의 관강 분비물 재흡수가 억제되어 분비물이 축적됨으로써 다양한 형태적 변환가 초래된 것으로 사료된다. 따라서 수컷 생식기관내의 중요한 에스트로겐 기능중의 하나는 수출소관 부위의 분비물 재흡수 작용이라고 할 수 있다.
The author analyzed the morphologic changes of bone structures from 1256 radiographs of 314 patients with temporomandi.bular joint arthrosis, which were obtained by the oblique-lateral projection and orthopantomography. The interrelations of the bone changes and clinical symptoms were examined. Also, the positional relationships of condylar head, articular fossa and articular eminence in the mouth open and closed state were observed in the patients with bone changes. The results were as follows; 1. The most frequent bone change in the TMJ arthorsis was eburnation of cortical bone (35. 64%) of total cases). Then came bone surface erosion and localized radiolucency (26.18%), marginal proliferation (9.7%) and flattening of articular surface (9.58%) in that order. 2. The most frequent site of bone change was articular eminence (41.70%). The came condylar head (21.09%) and articular fossa (20.73%) in that order. 3. In the patients with bone changes, their clinical symptoms were pain (51.55%), clicking sound during mandibular movement (37.71%) and limited mandibular movement (10.73%). In the patients complaining pain, their radiographs showed eburnation of cortical bone (30.68%), bone surface erosion and localized radiolucency (27.45%) and flattening in the (30.68%), bone surface erosion and localized radiolucency (27.45%) and flattening of articular surface (10.68%). 4. The condylar positional changes in the TMJ arthrosis patients with bone changes were as follows: in the mouth closed !tate, there were the widening of joint space in 624 cases (50.00%), the narrowing of joint space in 543 cases (43.47%) and bone on bone relatioships in 82 cases (6.57%). In the mouth open state, there were forward positioning of the condyle in 332 cases (28.55%), limitation of movenent in 332 cases (28.55%), bone on bone relation- ships in 248 cases (21.31%) and downward positioning of condyle in 217 cases (18.66%). bone on bone relationships in 243 cases (21.32%) and downward positioning of condyle in 217 cases (18.66%). 5. In the TMJ arthrosis patients with bone changes, 1249 cases of abnormal condylar position in the mouth closed state and 1163 cases of abnormal condylar position in the mouth open state could be interpreted. so, for the radiographic interpretation of TMJ arthrosis, the reading of condylar positional changes as well as that of bond changes should be performed and their interrelations should be profoundly considered.
Objective : Gypsum fibrosum has been traditionally used in treatment of febrile diseases and recently been shown to have anti-inflammatory effect. Chronic renal failure has a serious clinical symptoms including proteinuria, azotemia, anemia, and hyperlipidemia and has characteristic histopathological changes, glomerular hypertrophy, infiltration of inflammatory cells, and crescentic sclerosis, We investigated the effects of gypsum fibrosum on renal functional and histopathological disorder in chronic renal failure rat model induced 5/6 nephrectomy. Methods : Using Sprague-Dawley rats, CRF was induced by 5/6 nephrectomy. The rats were divided into 3 groups, normal, conrol, and gypsum administered orally with gypsum fibrosum 500mg/kg/day. Body weight, 24 hr proteinuria, hematologic analysis, and histological morphologic changes were followed up after 8 weeks. The glomerular macrophage/monocyte infiltration, $TGF-{\beta}_1$, type IV collagen, and angiotensin II type1 receptor($AT_1$) were evaluated by immunohistochemistry. Resuls : In the CRF control group, functional parameters and histopathologic changes clearly indicated the development of CRF. 24 hr proteinuria significantly increased in the CRF control group over the normal group, and serum creatinine level was lower in the gypsum group than in the control group, LDL-cholesterol was significantly lower in the gypsum group than in the control group. Morphological investigations showed a variety of characteristic features of CRF, glomerular hypertrophy, increasing cellular density of glomerulus, deposition of extra-cellular matrix, fibrotic change, and glomerular sclerosis in the control group, but in the gypsum group, these features diminished significantly. In observation of renal type IV collagen and $AT_1$ expression, positive area significantly increased in the control group over the normal group, and it significantly decreased in the gypsum group compared to the control group. Conclusions : Our findings suggest that gypsum fibrosum inhibits $AT_1$ and type IV collagen expression in renal tissues and attenuates progression of glomerulosclerosis and interstitial fibrosis in chronic renal failure rats, which lead to amelioration of renal function. From these results, we suggest that gypsum fibrosum may have renoprotective effects and could be a useful remedy agent for treating chronic renal failure.
The purpose of this in vitro study was to evaluate the efficiency of Er:YAG laser on calculus removal and the morphologic changes and hardness of the irradiated surface at different power settings. This experiment used human teeth which were extracted due to periodontal disease and had a band of calculus. Forty root slabs ($5{\times}5mm$) were made and divided into control group and irradiated groups. Experimental groups were as follows; Control group (root planing), Group1 (irradiated with laser at 30mJ), Group2 (irradiated with laser at 60mJ), Group3 (irradiated with laser at 100mJ). Twelve root slab embedded in resin block were used in each group. Er:YAG laser was applied under water irrigation with the tip held perpendicular to the root surface in contact mode. The treatment time was measured until the calculus was removed completely under naked eyes. The efficiency of calculus removal was evaluated by the time for removal. Morphological changes of laser irradiated site were observed under SEM and the surface hardness was measured using a VH tester. The results were as follows; 1. The efficiency of laser scaling was increased with increasing the energy level of irradiation(p<0.05). 2. The morphological changes such as carbonization, crater and scale-like defects in the irradiated root surface were frequently observed with increasing the energy level. 3. The surface hardness tended to increase at 60mJ and 100mJ irradiated groups than that of control group. From the results evaluting on the efficiency, morphological change and surface hardness, lower energy level was suggested for the clinical application of the Er:YAG laser in scaling.
Anatomic and physiological changes of the larynx with advancing age result in morphologic changes of the vocal fold and reduced control of the phonatory mechanism in elderly individuals and are reflected in increased unstability of fundamental frequency (Fo). The purpose of this study is to increase current understanding of acoustic and stroboscopic characteristics of normal elderly persons voices. First, phonated /a/ vowel productions by 40 normal adults (20 to 40 years, 20 men and 20 women) and 40 normal elderly persons (60 to 80 years,20 men and 20 women) were analyzed, using CSL (model 4300B) acoustic analysis software, to obtain acoustic measures related to fundamental frequency stability nd vocal resonance characteristics. Second, stroboscopic images of the vocal fold behavior in all subjects were analyzed by experienced specialists. In the men, fundamental frequency variation (vFe) (p<0.01), jitter. (p<0.05), and shimmer (p<0.05) for the older group were significantly higher than the value for the adult group. In the stroboscopic findings, edema of vocal fold had a significant finding in aged men (15%). In the women, vFo (p<0.05), jitter (p<0.05), and noise to harmonic ratio (NHR) (p<0.05) for the older group were significantly higher than the value for e adult group and first formant frequency (F1) (p<0.01) and second formant frequency (F2) (p<0.01) for. the older group were significantly lower than the value for the adult group. In the stroboscopic findings, vocal fold atrophy had a significant finding in aged women (25%). Frequency stability, as reflected by vFo, jitter, shimmer, and NHR, decreases with advancing age in men and women and spectral analysis of phonated /a/ vowel productions reveals the lowering of the frequency of F1 and second F2 with advancing age, especially in aged women. Change in the mass of vocal folds, due to atrophy or edema, is considered to be the greatest factor in these acoustic changes.
수제군 주변의 유동장 및 지형학적 변화에 대한 수리학적 효과를 분석하기 위하여, 본 연구는 영천강 만곡부에 설치된 저수 수제군 주변의 하도지형 특성의 변화에 대하여 모니터링과 수치모의를 수행하였다. 연구 대상지는 연장 190m로서 2008년 3월에 저수 수제군이 설치되었다. 현장 모니터링을 2008년 5월부터 2014년 4월까지 수행하였다. 설계된 수제 높이 보다 홍수시 수위가 더 높아져도 수제는 하안으로부터 흐름을 하도의 중심으로 밀어낸다. 따라서, 만곡부의 하도 안정을 위하여 설치된 수제군은 어느 정도 효과적으로 수리적 기능을 수행하고 있다. 시간의 경과에 따라 주수로가 하도의 중심으로 이동하면서 안정되었으며, 국소세굴에 의하여 수제 선단부 주변에 소가 형성되었다. 또한, 수제 사이에는 유사가 퇴적되었고, 하도에는 여울-소가 형성됨으로써 부수적으로 수제가 하도의 지형적 측면에서 다양한 물리적 특성을 창출함을 확인하였다.
The purpose of this study is to observe the morphological changes of prepared dentin surfaces by 5 dentin conditioning agents. Freshly extracted 48 healthy human molars were used in this study. The teeth were stored at $4^{\circ}C$ physiologic saline solution befor experiment. The teeth were cross-sectioned to expose dentin below 3.0mm at the cusp tip and above 2.0mm at the cemento-enamel junction with Crystal Cutter (MC411 D, Maruto Co., Japan). The specimens were then divided into 12 groups. The sectioned dentin surfaces in group 1, 3, 5, 7, 9, and 11 were prepared with No. 301 diamond point under air-water spray and those in group 2, 4, 6, 8, 10, and 12 were prepared with No. 700 carbide bur. The prepared dentin surfaces were conditioned with Nitric acid, Citric acid, Poly acrylic acid, EDTA, and Phosphoric acid. All the specimens were gold-coated with Eiko ion coater (Eiko-engineering Co.) and observed in Hitachi S-2300 Scanning electron microscope at 20 KV. The following results from this study were obtained; 1. The dentinal smear layers prepared with diamond point were compacted than those prepared with the carbide bur. 2. The dentinal smear layers prepared with diamond point or carbide bur were thick but after treatment of dentin conditioning agents smear layers were removed almost. 3. Irrespective of the uses of the diamond point or the carbide bur the morphological changes of dentin surfaces treated with the same conditioning agents were similar. 4. Treatment of nitric acid and EDTA was a little effect in removing dentinal smear layer. 5. Treatment of citric acid and phosphoric acid removed the smear layer very effectively and showed dissolution of peritubular dentin and opening of dentinal tubules.

Ginseng has an anti-cancer effect in several cancer models. This study was to characterize active constituents of ginseng and their effects on proliferation of prostate cancer cell lines, LNCaP and PC3. Cell proliferation was measured by $[^3H]$thymidine incorporation, the intracellular calcium concentration by a dual-wavelength spectrophotometer system, effects on mite-gen-activated protein (MAP) kinases by Western blotting, and cell attachment and morphologic changes were observed under a microscope. Among 11 ginsenosides tested, ginsenosides $Rg_3\;and\;Rh_2$ inhibited the proliferation of prostate cancer cells. $EC_{50}s\;of\;Rg_3\;and\;Rh_2$ on PC3 cells were $8.4{\mu}M\;and\;5.5{\mu}M$, respectively, and $14.1{\mu}M\;and\;4.4{\mu}M$ on LNCaP cells, respectively. Both ginsenosides induced cell detachment and modulated three modules of MAP kinases activities differently in LNCaP and PC3 cells. These results suggest that ginsenosides $Rg_3\;and\;Rh_2$-induced cell detachment and inhibition of the proliferation of prostate cancer cells may be associated with modulation of three modules of MAP kinases.
Objective: The prevalence of metabolic syndrome and type 2 diabetes is increasing worldwide. Mitochondrial dysfunction is known to be involved in insulin resistance and obesity, researches have been increasing highly. Astragali Radix extract (ARE) or its main components have been shown to perform comparably to insulin by significantly reducing blood glucose levels in animal models however, the influence on mitochondrial dysfunction are not well understood. Methods: ARE (0.2, 0.5 and 1.0 mg/ml) or metformin (2.5 mM) were treated in C2C12 after 6 day-differentiation. The expressions of adenosine monophosphate (AMP)-activated protein kinase (AMPK) and phosphorylation AMPK, peroxisome proliferators-activated receptror ${\gamma}$ coactivator $1{\alpha}$ ($PGC1{\alpha}$), nuclear respiratory factors 1 (NRF1), mitochondrial transcription factor (Tfam) and myosin heavy chain were detected with western blotting or polymerase chain reaction analysis. The morphological changes were also investigated. Results: ARE dose dependently increased phosphorylation of AMPK and respectively activated mRNA expressions of $PGC1{\alpha}$, NRF1 and Tfam which are mitochondrial biogenesis regulators. Furthermore, there were some morphologic differences of differentiated cells between ARE treatment and control. Conclusions: This study suggests that ARE has the potential to increase muscle mitochondrial function by activating AMPK and $PGC1{\alpha}$.
