Toxic effect of Hexavalent chromium $(CrO_3)$ on various cells and organs has been well recognized. However, the mechanism and degree of cytotoxicity of $CrO_3$ remain unclear. This study was performed to examine the cytotoxicity of $CrO_3$ on $C_6$ glioma cells by measuring cell viability. The XTT assay, one of the sensitive methods to determine the cell viability, was taken to examine the viability of glioma cells treated with $CrO_3$. In this study, not only decreased the number of glioma cells but morphologic changes of them were noted and cell viability decreased in a time and dose-dependent manner after treated with various concentrations of $CrO_3$ for 48hours. $IC_{90}\;and\;IC_{50}$ values in XTT assay were determined at $25{\mu}M\;and\;55{\mu}M$$CrO_3$, respectively. These results suggest that Hexavalent chromium has a highly cytotoxic effect and has a time and dose-dependent cytotoxicity on $C_6$ glioma cells.
The Korean Chinese Chives(Allium tuberosum Rottler) are a perennial herb and have been used from ancient as food and medical purpose in Korea. They are known to posses diuresis, sthemia activities and used for the treatment or anemia, haematemesis, and eczema. Recently the Korean Chinese Chives have been studied on the morphologic property, the classification of origin and chemical constituent. However there has not been studied on kg) (or 6 days and observed the hematological effects in dogs. The results are as follows, 1. The red blood cell count was significantly (P < 0.05) decreased on day 2. 2. The reduced glutathion(GSH) concentration was significantly (P< 0.05) increased on day 4 and Met-hemoglobin concentration was significantly (P< 0.05) increased on 3 hrs after administration. 3. Packed cell volume(PCV) and hemoglobin concentration were not showed significant difference during experimental periods. 4. The mean corpuscular volume(MCV) was significantly(P < 0.05) increased on 3 hours after administration.
The Schizonepeta tenuifolia spikes (STS) have been used as a traditional folk medicine for antiinflammatory, analgesic, anti-pyretic and anti-spasmodic purpose in Korea. Phytosterols (mixture of campesterol 3.68%, stigmasterol 2.30% and ${\beta}$-sitosterol 94.02%) and hesperidin were isolated by chromatography from ether and n-BuOH fractions of STS respectively. These compounds significantly reduced the blood glucose level and lessened the loss of body weight and water consumption dose-dependently when administered at a i.p. doses of 10 and 20 mg/kg for 4 days after the i.v. injection of streptozotocin (I 80 mg/kg). In the morphologic study, these compounds showed protective activity on the pancreatic islets, especially .betha.-cells, from the degenerative changes by streptozotocin.
Waterline extraction is the one of widely used methods for studying changes in tidal flat environment and coastlines using multi-temporal optical images such as Landsat TM and Landsat ETM+. High dynamics of tidal currents and land reclamation which accelerate sedimentation and/or erosion cause waterline change in tidal flats. The amount of sediment deposited or eroded can be evaluated by precisely estimating waterline changes in tidal flats. The objective of this study is to detect the change of waterlines during 17 years and analyze the trends of erosion and sedimentation in the study areas. The Ganghwado tidal flat on the west coast of the Korean Peninsula was selected. The study area is famous for high dynamics of tidal currents and vast tidal flats. Land reclamation which has been carried out on a large scale is also considered as one of elements that have accelerated the environmental changes in this tidal flat. In this study, we acquired 26 waterlines from Landsat TM and Landsat ETM+ images. We extracted the waterline from each satellite image to generate a digital elevation map (DEM) which was used for reference and to compare with the other waterline which was extracted from DEM having a same tide. The result of comparison well depicted the areas of dominant sedimentation and erosion, and general trends of sedimentation and erosion according to sub-regions are also revealed during the investigation time. Results showed that erosion during a decade was dominant at the west of the Southern Ganghwado tidal flat, while sedimentation was dominant at the wide channel between the Southern Ganghwado tidal flat and the Yeongjongdo tidal flat. This area has been commonly affected by high currents and sedimentation energy. Although we were not able to verify the accuracy of the waterline changes, this result clearly showed the waterline change and therefore, the waterline extraction method used in this study has proven as an effective tool for long term tidal change estimation.

The author analysed the interrelation between the morphologic changes of bone Structures and the position of condylar head from the routine radiographs of 134 cases of the temporomandibular joint arthrosis. The frequencies of coincidence between the site of bone defrmity and condylar head positional change were examined. Also, the positional changes of condylar head and the direction of condylar movement in relation to the kind of bone deformities were observed. The results obtained were as follows; 1. In 52.65 per cent of total cases, the site, of positional change of condylar head was coincided with the site of bone deformans. The frequencies of the coincidence between these in the five items among seven items examined were above 53 per cent. From the results, it seems that the positional changes of condylar head were related with the morphological change of bone structure. 2. Eburnation and erosion. revealed frequently positional changes in the opening and closing position of the mouth, although in the early stages of the TMJ arthrosis. 3. In the bone deformans, during opene position of the mough 44.81 per cent of total cases revealed backword movement and 37.74 per cent showed forward movement. In closed position of the mouth, downward movement was revealed in 35.23 per cent of total cases and upward movement 28.41 per ,cent of total cases. 4. In the cases showing eburnation, the frequencies of coincidence between the site of positional change and bone deformans were 58.57 per cent of the total cases. that means it was high in the early stages of the TMJ arthrosis.
Neuronal apoptotic events, which result in cell death, are occurred in hypoxic/ischemic conditions. Estradiol is a female sex hormone with steroid structure known to provide neuroprotection through multiple mechanisms in the central nervous system. This study was aimed to investigate the signal transduction pathway of $CoCl_2$-induced neuronal cell death and the inhibitory effects of estradiol. Administration of $CoCl_2$ decreased cell viability in both a dose- and time-dependent manner in PC12 cells. $CoCl_2$-induced cell death produced genomic DNA fragmentation and morphologic changes such as cell shrinkage and condensed nuclei. It was found that $CoCl_2$-treated cells increased the reactive oxygen species (ROS) as well as caspase-8, -9 and -3 activities. However, pretreatment with estradiol before exposure to $CoCl_2$ prevented the reduction in cell viability reduction and attenuated DNA fragmentation and morphologic changes caused by $CoCl_2$. Furthermore, the $CoCl_2$-induced increases of ROS levels and caspases activities were attenuated by estradiol. Gene expression analysis revealed that estradiol blocked the underexpression of the Bcl-2 and ameliorated the increase in the release of cytochrome c from mitochondria into cytoplasm and Fas-ligand (Fas-L) upregulated by $CoCl_2$. These results suggest that $CoCl_2$ induce apoptosis in PC12 cells through both mitochondria- and death receptor-mediated cell death pathway. Estradiol was found to have a neuroprotective effect against $CoCl_2$-induced apoptosis through the inhibition of ROS production and by modulating apoptotic effectors associated with the mitochondria- and death-dependent pathway in PC12 cells.
In order to study the morphologic changes of the unfixed odontoblasts suspended in phenol solution of several different concentrations, the author carried out the extraction of lower incisor of S-D strain rats to collect the odontoblasts, and the cells obtained were suspended immediately in saline solution. After observing the odontoblasts in fresh state, the saline solution was substituted with 0.125%, 0.25% 0.5%, 1% and 2% diluted phenol solutions. The morphologic changes were examined with phase contrast microscope at intervals of 10, 30, and 60 minutes. The results were as follows: 1. In saline solution the odontoblast showed cytoplasmic swelling, slender cytoplasmic process, thick rim nuclear membrane with increased dark contrast, and prominent nucleoli and chromatin granules with lapse of time intervals. In accordance with time intervals, blisters appeared in the supranuclear zone and increased its size and moved outward of the cytoplasmic membrane resulting detachment from the cell membrane. The phase dark cytoplasmic granules were increased in its dark contrast and in its size. 2. In 0.125% and 0.25% phenol solution, the odontoblasts and its nucleus shrunk immeidately and its contrast of cellular components was increased. With the lapse of time, the phase-dark granules in cytoplasm were aggregated, and several blisters were formed in and out of the cells. The outline of cytoplasmic membrane was also obscured. 3. In 0.5% phenol solution, the necleus shrunk at once, but soon after it revealed karyolysis accompanying dark contrast of neclear components such as nuclear membrane, nucleoli, and chromatin granules. On the contrary, the cytoplasmic granules showed aggregation and increased dark contrast, small and large blisters were formed in and out of the odontblasts and the outline of cytoplasmic membrane became obscured. 4. In 1% phenol solution, it showed shrinkage of odontblasts and its nuclei with thick rim nuclear membrane, aggregation of chromatin granules and occasional karyorrhexis. The dark contrast of cytoplasmic granules was increased and aggregated each other. But the blister formation could not be found. 5. In 2% phenol solution, it showed the shrinkage of odontoblasts and pyknotic nuclei with increased dark contrast of nucleoli and chromatin granules. The number of cytoplasmic granules was decreased by aggregation. But the blister formation could not be found as in 1% phenol solution.
The purpose of this study was to analyze the change of morphologic pelvis according to age and gender. Pelvis AP(anterior-posterior) projection were taken from 382 individuals (men: 194, women: 188) at the age of twenty and older. We measured four values of pelvis using CR (computed radiological system) including maximum pelvic breadth (MPB), sacral breadth (SB), transverse diameter of pelvic inlet (TD), and distance between femoral heads (DFH). Average (t-test, ANOVA) and Pearson correlation were calculated for comparison. While there was no significant difference of gender in case of MPB, significant differences were observed in SB (men 97.76 mm, women 102.00 mm), TD (men 138.0 mm, women 149.79 mm), and DFH (men 141.94 mm, women 150.39 mm). The DFH values were negatively correlated with the age (men r=-0.280, women r=-0.207). The morphological changes of pelvis were measured high in the men of 50's and in the women of 60's and 70's. The SB, TD and DFH were greater in the women than in the man. As the ages get higher, the deviation of pelvis were gradually increased, which indicates that the morphological changes of pelvis with respect to the age reflects an aging process.

The corneal epithelium is constantly being shed. The mechanism of corneal desquamation is not fully understood. Apoptosis, programmed cell death, may play a role. Apoptosis can be induced by a number of factors and different mechanisms. The study was performed to examine the apoptotic index induced in human corneal epithelial cells maintained in tissue culture by various apoptotic inducers. Various inducers, recombinant human cytokines($INF{\gamma}$, $TNF{\alpha}$, FASAb), actinomycin D. camptothecin, cycloheximide, dexamethasone and etoposide, were purchased from commercial suppliers. Inducers at manufacturer-recommended concentration were added to the corneal epithelial cells for 48 hours. Cell viability was measured using MTT assay. The cells were then assessed for the level of apoptosis. Morphologic changes and quantification of apoptotic cells were determined and counted under fluorescence microscope after inducers-treated human corneal epithelial (HCE) cells for 48 hours with Hoechst 33342 staining. Annexin V-FITC/PI staining and DePsipher assay. The expression of Fas protein was studied by immunocytochemistry. All inducers induced apoptosis in HCE cells in a dose dependent manner. Actinomycin D. camptothecin and etoposide induced apoptosis at lower than manufacturer-recommended concentration, while cytokines, cycloheximide and dexamethasone induced apoptosis at higher concentrations at the end of 48 hours. All inducers elicited typical apoptotic morphologic changes (chromatin condensation, nucleus fragmentations non-orange-red colored mitochondria) and expresses Fas protein highly. Apoptotic index of HCE cells by these inducers was different from the other cell lines. RNA synthesis inhibitor and topoisomerase inhibitors induced apoptosis at lower concentration than manufacturer-recommended concentration. Cytokines, cycloheximide and dexamethasone were able to produce apoptosis at 10 times higher concentrations. RNA synthesis inhibitor and topoisomerase inhibitors are more sensitive than intracellular receptor-activators in apoptotic induction of HCE cells.
Background & Objective : Naeso-san(NSS) has been used for the treatment of functional dyspepsia, regarded as a gastric dysmotility disease. A main cause of gastric dysmotility is antral dilatation or antroduodenal uncoordination. Therefore, we investigated the effect of NSS on gastric motility and its mechanism of action, as well as the morphologic changes in antral dilatated rats. Methods : Antral dilatated rats were induced by wrapping a nonabsorbable rubber ring(D:6mm, W:4mm, T:1mm) around the 1st portion of the duodenum for 8 weeks. Then morphologic changes were investigated and compared with normal intact rats before and after 8 weeks. Gastric emptying was measured by administration of normal saline(NS) or NSS in normal intact and antral dilatated rats. In another series of experiments to evaluate the mechanism of NSS under delayed conditions, normal intact rats were treated with atropine sulfate(1mg/kg, s.c.), quinpirole HCl(0.3mg/kg, i.p.), $NAME(N^{G}-nitro-L-arginine$ methyl ester, 75mg/kg, s.c.) and cisplatin(10mg/kg, i.p.), respectively. The myoelectrical activity of the gastric smooth muscle was recorded in normal intact and antral dilatated rats. The contractile waves were measured for 30 minutes before and after administration of each solution(NS, NSS). Results : Body weight gain of antral dilatated rats was significantly lower than that of the controls. Futhermore, we found the thickness of the mucosal and muscular layers and surface area of the stomach increased significantly compared with controls. NSS 278㎎/㎏ improved gastric emptying more than normal saline or NSS 93mg/kg in normal intact(p=0.026) and antral dilatated rats(p=0.03). NSS enhanced gastric emptying significantly in the NAME treated group(p=0.002). NSS 278mg/kg increased the significant postprandial dominant power than that of NS in normal intact rats, whereas there was no statistical significance in antral dilatated rats. Conclusions : NSS stimulates gastric motility through the cholinergic pathway. We expect that pathologic model with antral dilatation can be used as an exprimental tool which is similar to dyspepsia and NSS would be effective especially in dysmotility-like functional dyspepsia with antral dilatation or impaired reservoir functions such as gastric adaptive relaxation.
