상엽(桑葉)추출물의 항산화효과에 대한 연구 (Studies on the Antioxidant Effects of Mori Folium Extract)

  • Objective : The objective of this study was to investigate the antioxidant effects of Mori Folium extract. Methods Total antioxidant status was examined by total antioxidant capacity(TAC) and total antioxidant response(TAR) against potent free radical reactions. The effect of Mori Folium extract was examined by measuring total phenolic content, concentration at which 1,1-dipheny1-2-picrylhydrazyl(DPPH) radical scavenging activity was inhibited, inhibitory effect on lipid peroxidation, and the effect on reactive oxygen species(ROS) generation. Results : 1. TAC and TAR of Mori Folium extract at the concentration of 5 mg/ml were 1.61 and 1.24 mM Trolox equivalents, respectively. 2. Total phenolic content of Mori Folium extract at the concentration of 5 mg/Ml was 1.70 mM gallic acid equivalent. 3. Concentration of Mori Folium extract at which DPPH radical scavenging activity was inhibited by 50% was 2.29 m9/m4 as compared to 100% by Pyrogallol solution as a reference. 4. The inhibitory effect of the extract on lipid peroxidation was examined using rat liver mitochondria induced by FeSO$_4$/ascorbic acid. Mori Folium extract at the concentration of 10 mg/ml significantly decreased thiobarbituric acid reactive substances(TBARS) concentration. The extract prevented lipid peroxidation in a dose-dependent 5. The effect of Mori Folium extract on reactive oxygen species(ROS) generation was examined using a celt-free system induced by hydrogen peroxide FeSO$_4$. Addition of 1 mg/ml of Mori Folium extract significantly reduced dichlorofluorescein(DCf) fluorescence. The extract caused concentration-dependent attenuation of the increase in DCF fluorescence, indicating that the extract significantly prevented ROS generation in vitro. Conclusion ; The antioxidant effects of Mori Folium extract seem to be due, at least in part, to the prevention offree radical-induced oxidation, fllowed by inhibition of lipid peroxidation.

Bioconversion enhances anti-oxidant and anti-inflammation activities of different parts of the Mulberry Tree (Morus alba L.), especially the leaf (Mori Folium)

  • The mulberry tree (Morus alba L.) has been traditionally used in Chinese medicine to treat inflammatory diseases. We investigated the effects of bioconversion on different components of the mulberry tree, and determined changes in the physiological activities. Ethyl acetate-soluble fractions of five different segments (fruit, Mori Fructus; leaf, Mori Folium; twig, Mori Ramulus; root, Mori Cortex; and mistletoe, Loranthi Ramulus) of the mulberry tree show enhanced anti-oxidant effects in the 2,2-diphenyl-1-picrylhydrazyl, and 2,2'-azinobis-(3-ethylvenzothiazoline-6-sulfonic acid) assays, and enhanced anti-inflammatory effects of lipopolysaccharide (LPS)-stimulated nitric oxide (NO) production in RAW 264.7 macrophages, after being treated with a crude enzyme extract from Aspergillus kawachii, in the following order of activity: Mori Folium>Mori Cortex>Mori Ramulus>Mori Fructus>Loranthi Ramulus. Ethyl acetate- soluble fraction of mulberry leaves (Mori Folium) that underwent bioconversion was most effective, and was devoid of any cytotoxicity. The fraction was also effective against mRNA expression of LPS-induced pro-inflammatory cytokines, such as inducible nitric oxide synthase, cyclooxygenase-2, tumor necrosis $factor-{\alpha}$, $interleukin-1{\beta}$, and interleukin-6. In addition, the fraction was effective in LPS-induced phosphorylation of mitogen-activated protein kinases and IKK, and $I{\kappa}B$ degradation, followed by translocation of the nuclear $factor-{\kappa}B$ from the cytoplasm to the nucleus. Thus, bioconversion increased the anti-oxidative and anti-inflammatory activities of the mulberry leaf.

  • In order to investigate the effect of Mori Folium on immunologic control function, the various immune responses were studied. The results were obtained as follows : 1. The transformation of lymphocyte was increased significantly in all concentration of Mori Folium treated group as compared with negative and positive control. 2. In FACS analysis, the population of helper T cell and T cell were increased significantly in Mori Folium treated group as compared with control. 3. IL-12(p35) and INF-${\gamma}$ were upregulated in Mori Folium treated group 4. NO and carbon clearance were significantly increased in Mori Folium treated group as compared with control. 5. The hemagglutinin titer and hemolysin titer were increased but insignificantly in Mori Folium treated group as compared with control.

탈모증에 대한 상백피 복합물의 모발성장 효과 (Hair growth effects of Mori Cortex Radicis Mixture on the hair of rat)

  • This study was carried out to investigate the hair growth effect of Mori cortex radicis mixture on the hair of rat. Cytarabine(50mg/kg) was injected to eight-day-old rats for 7 days. When the mixture of Mori cortex radicis was administered to the rat by route of skin, it promoted the growth of hair. These data suggest that Mori cortex radicis mixture have an effect on the hair growth in cytarabine-induced alopecia rat.

Effect of Mori Cortex in the Cardiac Injury Induced by Skin Burn

  • This study was conducted to investigate an effect of Mori Cortex in the cardiac injury following dermal scald burn in rats. Sprague-Dawley rats were induced scald bum (15% of total body surface area). Heart was removed at 5 h postburn and examined with biochemical assay, ultrastructural observations and stereological analysis. The activity of serum aspartate aminotransferase and creatinine was increased at 5 h postburn compared with them of control. Administration of heat extracts of Mori Cortex after scald burn inhibited the production of KC (neutrophil chemoattractant cytokine) and increased the activity of protein kinase C (PKC) in heart tissue. The activity of myeloperoxidase (MPO) in heart tissue was decreased both at 5 h postburn and in case of Mori Cortex administration after scald burn. Ultrastructurally, many contraction bands and separation of intercalated disk induced by scald burn were decreased by administration of heat extracts of Mori Cortex. In stereological analysis, administration of Mori Cortex after scald burn resulted the volume densities of myofibril and mitochondria were increased compared with them of burn control. These data suggest that Mori Cortex may be a useful stuff to the range of available treatments for cardiac injury induced by skin burn.

Molecular Cloning of a Profilin cDNA from Bombyx mori

  • The actin-binding protein profilin cDNA was firstly isolated from the lepidopteran insect, silkworm Bombyx mori. The B. mori profilin cDNA contains an open reading frame of 378 bp encoding 126 amino acid residues and possesses three cysteine residues. The deduced amino acid sequence of the B. mori profilin cDNA showed 80% identity to Apis mellifera profilin and 72% to Drosophila melanogaster profilin. Northern blot analysis showed that B. mori profilin is highly expressed in epidermis and less strongly in silk gland. In addition, Northern blot analysis revealed the presence of B. mori profilin transcripts in all tissues examined, suggesting that B. mori profilin gene is expressed in most, if not all, body tissues.

Cortex Mori Inhibits the CGG-specific IgE-Dependent Histamine Release

  • Cortex Mori, the root bark of mulbery tree has been used as an antiphlogistic, diuretic, and expectorant in herbal medicine. The purpose of this study is to evaluate chicken gamma globulin (CGG)-specific IgE-induced morphologic and functional changes in rat peritoneal mast cells (RPMC), and to determine whether Cortex Mori could inhibit the CGG-specific IgE-depeildent mast cell degranulation and histamine release from RPMC. Results are 1) the degranuration and histamine release from RPMC were not induced within 1 hour after addition of Cortex Mori alone, 2) the CGG and CGG-specific IgE-Induced degranulation from RPMC was observed within 10 minutes, 3) the histamine release from RPMC sensitised with CGG-specific IgE was induced by tile addition of CGG, 4) CGG-specific IgE-dependent degranulation rate in RPMC pretreated with Cortex Mori was significantly Inhibited, compared to that of control group without Cortex Mori pretreatment, and 5) the CGG-specific IgE-dependent histamine release from RPMC was significantly inhibited by pretreatment with Cortex Mori. These data suggest that Cortex Mori contains some substances with capabilities to inhibit CGG-specific IgE-dependent degranulation and histamine release from RPMC.

Comparative Analysis of $\alpha$-glucosidase Activity in Bombyx mori and Antheraea yamamai

  • [ $\alpha$ ]Glucosidase (EC 3.2.1.20) is a glycosidase that hydrolyzes disaccharides, oligosaccharides, and polysaccharides resulting in the release of α-D-glucose. In this study, $\alpha$-glucosidase activity in the hemolymph and midgut of the mulberry silkworm Bombyx mori and Japanese oak silkmoth Antheraea yamamai was measured using maltose, sucrose, trehalose, and p-nitrophenyl $\alpha$-D-glucopyranoside as substrates. In general, hemolymph $\alpha$-glucosidase activity was higher in B. mori than in A. yamamai. In contrast, midgut $\alpha$-glucosidase activity was higher in A. yamamai than in B. mori for all of the substrates tested. $\alpha$-Glucosidase activity in the midgut of both B. mori and A. yamamai showed similar responses to changes in pH and temperature for all of the substrates tested. Native (7.5%) PAGE of hemolymph and midgut proteins from B. mori and A. yamamai followed by staining with 4-methylumbelliferyl $\alpha$-D-glucoside (MUG) indicated that the $\alpha$-glucosidases of these related lepidopterans are functionally similar but structurally different. In comparison to $\alpha$-glucosidase activity from A. yamamai, $\alpha$-glucosidase activity from B. mori was generally less sensitive to the $\alpha$-glucosidase inhibitors, 1-deoxynojirimycin (DNJ), acarbose, and voglibose when the activity was determined using maltose, sucrose, and trehalose.

Inhibitory Effect of Cortex Mori on Ovalbumin-induced Late Asthmatic Reaction in Guinea pigs.

  • Cortex mori (Morus alba L.), the root bark of mulberry tree, has been used as an antiphlogistic, diuretic, and expectorant in herbal medicine. The purpose of this study was to determine whether Cortex mori could inhibit the ovalbumin (OA) -induced late asthmatic reaction in guinea pigs. Guinea pigs were sensitized by two exposures to an aerosol of OA(1.0%) and then challenged with aerosolized antigen(2.0%), The animals were pretreated by three inhalations of the aerosoled Cortex mori either before antigen sensitization or cahllenge. Bronchoalveolar lavage fluid(BALF) and peripheral blood were collected at 17 hours after OA challenge. The cell populations in BALF and peripheral blood were examined to determine the changes of the relative proportions of eosinophils,neutrophils and mononuclear cells etc. Beta-glucuronidase activity in BALF was measured to evaluate the alveolar macrophage activation. OA-induced histamine release from guinea pig peritoneal fluid cells was measured by radioisotope enzymatic asssay. Results were as follows. The number of eosinophils, neutriphils and lymphocytes recovered in BALF were significantly increased in the 17h following aerosol challenge with OA. Among them, eosinophil and neutriphils were decreased remarkably in group that had been preinhalated with Cortex mori. The number of lymphocytes in BALF were not decreased in group pretreated with CM before sensitization but decreased in Group pretreated with CM before challenge. After OA challenge, the number of eosinophils in peripheral blood were markedly increased, but Cortex mori inhibited significantly the OA-induced eosinophilia. Beta-glucuronidase activity in the supernatants of BALF were significantly increased in the 17h following aerosol challenge with OA, however, pretreatment of Cortex mori had no influence on Beta-glucuronidase activity, suggesting that Cortex mori had no inhibitory effect on OA-induced alveolar macrophage activation. Cortex mori inhibited the OA-induced histamine release from guinea pig peritoneal fluid cells. From the above results, it is suggested that Cortex mori contains some substances with an activity to inhibit the the OA-induced late phase reaction of the bronchial asthma in guinea pigs.

상엽(桑葉)의 고지방식이 유도 비만 쥐에 대한 조직학적 및 혈청학적 항비만 효과 연구 (Histological and Serological anti-Obesity Effect of Mori Folium in Obesity Induced SD Rat by High Fat Diet)

  • Objectives : Mori Folium is used for hyperlipidemia or diabetes. The study was designed to test the anti-obesity effect of Mori Folium on body weight, hepatic fat accumulation and serum lipid level. Methods : The extract from Mori Folium was made by the pharmacy department of Kyung-hee oriental medical hospital. Normal diet, high-fat diet, 30% reduced diet and Mori Folium groups were set. The normal group was administered normal rat food, but the other three groups were administered high fat food. We measured body weight once a week. After 3 weeks experiment, hepatic lipid accumulations were measured. Also we compared serum lipid levels among the 4 groups after 3 weeks. Results : Mori Folium had no effect on body weight, but the 30% reduced diet had an inhibitory effect on body weight gain. Mori Folium had more inhibitory effect on hepatic fat accumulation than a 30% reduced diet. Mori Folium also has more inhibitory effect on serum triglyceride than 30% reduced diet. Conclusions : Mori Folium has anti-obesity effect on hepatic lipid accumulaion and serum triglyceride level in high fat diet induced rats.
