The Effect of HCl-AlCl3(Catalyst) Concentrations on Acid Hydrolysis of Ricestraw Cellulose (염산(鹽酸)과 촉매 AlCl3의 농도가 볏짚 Cellulose의 산가수분해(酸加水分解)에 미치는 영향)

  • 81.9% of the cellulose delignified by acetosolv process was hydrolyzed in HCl-$AlCl_3$ hydrolysis system when $AlCl_3$ was used as catalyst in breaking down of glycosidic bond of cellulose. It was well compared that the HCl hydrolysis system without $AlCl_3$ as catalyst showed only 60~61% of the hydrolyzed yield. Also monosaccharide yield including glucose clearly increased when $AlCl_3$ was use. When concentration of HCl and $AlCl_3$ was increased, the hydrolyzed monosaccharide was increased within certain range. The monosaccharid yield out of the hydrolyzed reached 55.4% at optimum conditions which were identified as 20% of Hel solution, 0.03 Mol of $AlCl_3$, $120^{\circ}C$ of reaction temperature and 7 hours of reaction time employed in this study.

Composition of Crude Polysaccharides in Some Phezllinus Species (수 종 상황버섯의 조다당 분획물 조성)

  • Various Phellinus species (Hymenochaetaceae) including P. linteus, P. baumii, P. pini, and P. igniarius of domestic and foreign products, were examined by gas chromatography to validate their composition of crude polysaccharides. The crude polysaccharide fraction, which is known to be one of the physiologically active fractions, were prepared by a cold ethanol precipitation method and the yield of the crude polysaccharide fractions of P. linteus, P. baumii, P. pini, and P. igniarius were 15.0%, 5.1%, 6.3% and 5.7%, respectively. The total monosaccharide amount of the crude polysaccharide fraction of P. linteus was 148.713 mg/g (dry weight of mushroom) while those of P. baumii, P. pini and P. igniarius were 29.022 mg/g, 29.222 mg/g and 1.546 mg/g, respectively. Although the major monosaccharide of all Phellinus mushrooms was glucose, there were remarkable differences in the absolute quantity of each monosaccharide. P. linteus included plenty of monosaccharides except mannose and ribose in comparison with P. baumii and P. pini. The absolute quantities of monosaccharides of P. baumii and P. pini were nearly similar except arabinose. The total monosaccharides of P. igniarius were minimal, but the portions of galactose and mannose in P. igniarius were greater than in the other species.

Neutral and Amino Sugars Composition of a Lectin from Maackia fauriei (Maackia fauriei 유래 렉틴의 중성당 및 아미노당 조성)

  • The glycosylation of therapeutic glycoproteins can affect their efficacy, stability, solubility, and half-life. Analyzing the composition of monosaccharides, such as that of neutral and amino sugars, is the first step for elucidating the structure of glycan attached to glycoproteins. In the present study, neutral and amino sugars of lectin obtained from Maackia fauriei were analyzed using an enzyme-linked lectinsorbent assay (ELLA) and high-performance anion exchange chromatography with pulsed amperometric detection (HPAEC-PAD). Peroxidase-labeled lectins such as concanavalin A, Ricinus communis agglutinin, and soybean agglutinin were used for ELLA, since they specifically bind to the monosaccharide residue most frequently encountered in a glycan. The hydrosylate of lectin was prepared by treatment with trifluoroacetic acid, which resulted in the lectin mainly possessing the N-glycan consisting of 98.1 pmol Fuc, 342.1 pmol GlcN, 51.9 pmol Gal, 678.9 pmol Man, and 330.7 pmol Xyl. The present results demonstrate that ELLA and HPAEC-PAD are very effective methods for rapidly estimating the types and relative amounts of monosaccharides in intact glycoproteins.

  • The oligosaccharide metabolism in soymilk was investigated by mixed culture with Lactobacillus acidophilus and Saccharomyces uvarum. When Saccharomyces uvarum was cultured in soywhey, change of oligosaccharide could be shown apparently. However, Lactobacillus acidophilus could not utilize oligosaccharide in soywhey for growth and lactic acid production. During the fermentation of mixed culture with Lactobacillus acidophilus and Saccharomyces uvarum, Saccaharomyces uvarum was supposed to convert oligosaccharide to monosaccharide first and then Lactobacillus acidophilus to convert these produced monosaccharide to lactic acid.

Factors Affecting the Survival Rates of Frozen-Thawed Spermatozoa in Jindo Dog by Monosaccharides and Freezing Rate (진도개 정액 동결 시 단당류 및 동결 속도가 생존율에 미치는 영향)

  • This studies were conducted to investigate the survival rate of frozen-thawed spermatozoa of Jindo Dog by monosaccharide and freezing rates. Experimental animals were prepared 12 males within 1~8 year's old and collected once in a couple of weeks by digital manuplation methods. Collected semen was diluted 1:1 with Tris-egg yolk extender and added 4, 6 or 8% of glycerol and none, 4 mM glucose or 4 mM fructose as cryoprotectant and was equilibrated for 2 hrs in $4^{\circ}C$. In monosaccharide groups, the freezing rate was 5 cm-5 min. above $LN_2$. The survival rates without monosaccharide were $50.7{\pm}19.0%$, $58.6{\pm}18.0%$, $40.0{\pm}10.0%$ in 4, 6 or 8% glycerol, respectively. In addition of glucose, the survival rates were $43.1{\pm}14.7%$, $38.1{\pm}16.5%$, $33.3{\pm}4.0%$ in 4, 6 or 8% glycerol, respectively and in fructose, were $47.9{\pm}21.1%$, $61.3{\pm}6.2%$, $34.3{\pm}12.6%$ in 4, 6 or 8% glycerol, respectively. There showed significantly different between glycerol groups and monosaccharides groups (p<0.05). The survival rates of freezing rate in 5 cm-5 min. group was $64.5{\pm}15.8%$, $51.9{\pm}27.6%$, $29.7{\pm}24.8%$ and in 10 cm-10 min. group was $62.5{\pm}20.3%$, $64.9{\pm}23.6%$, $34.5{\pm}27.4%$ in 4, 6 or 8% glycerol, respectively. There were significantly different between freezing rates (p<0.05). These results suggest that the addition of fructose with 6%-glycerol and slow freezing improve the survival of frozen-thawed sperm in Jindo Dog.

Monosaccharide as a Central Scaffold Toward the Construction of Salicylate-Based Bidentate PTP1B Inhibitors via Click Chemistry

  • The discovery of carbohydrate-based bioactive compounds has recently received considerable interest in the drug development. This paper stresses on the application of 1-methoxy-O-glucoside as the central scaffold, whereas salicylic pharmacophores were introduced with diverse spatial orientations probing into the structural preference of an enzymatic target, i.e. protein tyrosine phosphatase 1B (PTP1B). By employing regioselective protection and deprotection strategy, 2,6-, 3,4-, 4,6- and 2,3-di-O-propynyl 1-methoxy-O-glucosides were previously synthesized and then coupled with azido salicylate via click chemistry in forming the desired bidentate salicylic glucosides with high yields. The inhibitory assay of the obtained triazolyl derivatives leads to the identification of the 2,3-disubstituted salicylic 1-methoxy-O-glucoside as the structurally privileged PTP1B inhibitor among this bidentate compound series with micromole-ranged $IC_{50}$ value and reasonable selectivity over other homologous PTPs tested. In addition, docking simulation was conducted to propose a plausible binding mode of this authorized inhibitor with PTP1B. This research might furnish new insight toward the construction of structurally different bioactive compounds based on the monosaccharide scaffold.

Effect of Cultural Conditions on Polysaccharide Production and its Monosaccharide Composition in Phellinus linteus L13202 (배양 조건에 따른 상황 버섯의 다당류 생산 및 단당류 구성 변화)

  • The effect of cultural conditions on mycelial growth, polysaccharide production in Phellinus linteus and its monosaccharide composition was studied. P. linteus showed the highest growth (0.9 g/100 ml) on glucose but the polysaccharide production was the highest (13.7%) on mannose. The fungus grew very well at neutral pH (0.9 g/100 ml) but the growth was reduced to 0.47 g per 100 ml at alkaline pH. For the different pH, the yield of polysaccharide was in the range of $5{\sim}8%$. The highest yield of 7.94% was obtained at pH 5. Also a variation in monosaccharide composition was observed for different carbon sources and pH. The composition ranges of glucose, mannose, and galactose of polysaccharide were $80{\sim}95%,\;3{\sim}12%,\;and\;2{\sim}10%$ depending on carbon sources, respectively. In contrast, the variation of composition range of three monosaccharides was narrower for different pH than that for carbon sources. These results suggested the possibility of the improvement of production and the physiological modification of the polysaccharide.

  • Complex viscosities of dense alumina slurries over 45% volume density measured with the oscillating method were correlated well with Casson model. Among several monosaccharides and disaccharides studied here, fructose and sucrose showed good rheological properties in making dense alumina slurry plastic compared to other monosaccharides and disaccharides like glucose, galactose, arabinose, xylose and maltose. Sucrose content or additional water content in dense alumina slurry with sucrose contributed to the plasticity of the slurries.

Effects of Carbohydrates on Change in Blood Glucose Levels (당류유형이 혈당변화에 미치는 효과)

  • Purpose: This study was to investigate the effects of Carbohydrates on blood glucose levels in healthy adults after taking the monosaccharide glucose and disaccharide candies. Methods: This study was experimental research using a randomized controlled trial. Participants were college students who could agree the purpose of the study and participated voluntarily and met the selection criteria. Considering the dropout rate, 25 subjects in each group were included. The assignments of the experimental group and the control group were randomly assigned, and this study used the allocation concealment. Glucose tablets of 15g in the experimental group, and 15g sugar of candies in the control group were orally ingested. Blood glucose was measured before ingestion, 10 minutes, 15 minutes, and 30 minutes after ingestion. Results: There were no statistically significant differences in blood glucose of 10 minutes (U=406.00, p=.069), 15 minutes (U=370.00, p=.264), and 30 minutes after ingestion (U=337.00, p=.634) between experimental (glucose tablet oral ingestion) and control groups (mint candy oral ingestion). Conclusion: There was no difference in the blood glucose level up to 30 minutes after ingestion of monosaccharide glucose and disaccharide candy. Through this study, the decision to use either candy or glucose tablets in the event of hypoglycemia can be chosen according to the patient's preference.
