Determination of Total Aflatoxins in Foods by Parallelism of ELISA and LC/MS/MS (ELISA-LC/MS/MS 병행에 의한 식품 중 aflatoxins 분석)

  • High performance liquid chromatography (HPLC) and liquid chromatography mass spectrometry (LC/MS) have been widely used to quantify aflatoxins in food, but these methods are expensive, time-consuming, unsuitable for analysis of the routine screening of large sample numbers and require derivatization and high level techniques to perform. The objective of this study is to detect aflatoxins in a large number of foods by a high efficient analytical system of combined enzyme linked immunosorbent assay (ELISA) for screening and LC/MS/MS for confirmation. The samples spiked individually with aflatoxin $B_1$ (0.5 and 1.0 ng/g) and total aflatoxins (10 ng/g) were analyzed by ELISA and LC/MS/ MS, and the recoveries for ELISA and LC/MS/MS were 71.8~119.2% and 70.8~135.3%, respectively. A total of 378 samples (grains, nuts, soybean and fermented soybean foods, pepper and fermented pepper foods) were purchased from the six major cities in Korea and analyzed by ELISA-LC/MS/MS system. Twenty two (5.8%; peanut: 11, pistachio: 2, walnut: 6, almond: 1, pepper powder: 1, pepper paste: 1) out of 378 samples were screened as aflatoxin B1 positive by ELISA, but, 4 (1.1%; peanut: 2, pistachio:1, pepper powder: 1) out of the 22 samples screened were confirmed as aflatoxins positive at levels of 1.02~52.79 ng/g by LC/MS/MS. ELISA-LC/MS/MS system provides a more rapid, accurate and cost-effective method for the detection of aflatoxins in large number of samples.

A Remote User Authentication Scheme Preserving Anonymity and Traceability with Non-Tamper Resistant Smart Cards (정보추출 가능한 스마트카드 환경에서 익명성과 추적성을 제공하는 원격 사용자 인증 기법)

  • Recently, because the interest and needs in privacy protection are growing, smartcard-based remote user authentication schemes have been actively studied to provide the user anonymity. In 2008, Kim et al. first proposed an authentication scheme in order to ensure the user anonymity against both external attackers and the remote server and track malicious users with the help of a trusted trace sever. However, in 2010, Lee et al. showed that Kim et al.'s scheme cannot provide the user anonymity against remote server, which is because the server can trace users without any help of the trace server, and then proposed a improved scheme. On the other hand, in 2010, Horng et al. proposed an authentication scheme with non-tamper resistant smart cards, in which the non-tamper resistant smart card means that an attacker may find out secret information stored in the smart card through special data analysis techniques such as monitoring power consumption, to be secure against a variety of attacks and to provide the user anonymity against external attackers. In this paper, we will propose a remote user authentication scheme with non-tamper resistant smart cards not only to ensure the user anonymity against both external attackers and the remote server but also to track malicious users with only the help of a trusted trace sever.

A Toolbox Approach for the Environmental Site Assessment of a Chemical Plant in a Coastal Area (연안지역 화학공장부지의 부지환경평가를 위한 복합조사기법의 적응)

  • Recently, the branch-out of foreign companies into domestic markets through M&A and the opened followed by the Free Trade Agreement(FTA) with America have made the environmental site assessments of specific site more necessary. In this study, through case study of conducting actual environmental site assessment by use of a toolbox approach at a large scale of chemical plant with various contaminants located in a coastal area, the problems of guideline of domestic environmental assessment of soil were complemented. And an efficient and economical assessment was achieved. All six steps such as basic investigation, environmental site history survey, sampling and analysis, installation of monitoring wells and hydrogeological survey, and data interpretation were conducted in this study. All results of document survey, geological lineament analysis, field geology survey of surrounding area, geophysical prospecting of the site, hydraulic conductivity, measurement of groundwater flow rate and direction, sampling and analysis at each step were associated and estimated as an integrated tool box approach. As a consequence of this study, toolbox approaches were very useful techniques for contamination level and site characterization of subsurface media. The given conditions to conduct a basic survey for domestic soil environment assessment of site by use of existing documents, as well as interviews with the owner/manager/user of all adjacent properties and thorough review of all practically reviewable records pertaining to the property and surrounding properties within "Guideline for Soil Environment Assessment" radii are very poor. As a result, the application of toolbox approach in the environment site assessment of site is not only more efficient and economical, but also could be very useful assessment to integrate the soil and groundwater contamination.

  • Numerous scientists who are engaged in compute-intensive researches require more computing facilities than before, while the computing resource and techniques are increasingly becoming more advanced. For this reason, many works for e-Science environment have been actively invested and established around the world, but still the scientists look for an intuitive experimental environment, which is guaranteed the improved environmental facilities without additional configurations or installations. In this paper, we present an integrated scientific workflow execution environment for Scientific applications supporting workflow design with high performance computing infrastructure and accessibility for web browser. This portal supports automated consecutive execution of computation jobs in order of the form defined by workflow design tool and execution service concerning characteristics of each job to batch over distributed grid resources. Workflow editor of the portal presents a high-level frontend and easy-to-use interface with monitoring service, which shows the status of workflow execution in real time so that user can check the intermediate data during experiments. Therefore, the scientists can take advantages of the environment to improve the productivity of study based on HTC.

Molecular Cloning and Expression Analysis of Red-spotted Grouper, Epinephelus akaara Hsp70 (수온변화에 따른 붉바리(Epinephelus akaara)의 heat shock protein (Hsp) 70 mRNA 발현)

  • A new heat shock protein 70 was identified in red-spotted grouper (Epinephelus akaara) based on an expression analysis. The cDNA of red-spotted grouper Hsp70 (designated RgHsp70) was cloned by the rapid amplification of cDNA ends (RACE) techniques. The full-length of RgHsp70 cDNA was 2,152 bp, consisting of a 5'-terminal untranslated region (UTR) of 105 bp, a 3'-terminal UTR of 274 bp, and an open reading frame (ORF) of 1,773 bp that encode a polypeptide of 590 amino acids with a theoretical molecular weight of 64.9 kDa and an estimated isoelectric point of 5.2. Multiple alignment and phylogenetic analyses revealed that the RgHsp70 gene shares a high similarity with other Hsp70 fish genes. RgHsp70 contained all three classical Hsp70 family signatures. The results indicated the RgHsp70 is a member of the heat shock protein 70 family. RgHsp70 mRNA was predominately expressed in the liver, with reduced expression noted in the head-kidney tissues. The expression analysis of different water temperatures (21, 18, 15 and $12^{\circ}C$) for sampled livers revealed that expression gradually increased at $12^{\circ}C$ compared to $21^{\circ}C$. In this study, the effects of water temperature lowering on the physiological conditions were investigated, and the results revealed that novel RgHsp70 may be an important molecule involved in stress responses.

Real-time monitoring for blending uniformity of trimebutine CR tablets using near-infrared and Raman spectroscopy (근적외분광분석법과 라만분광분석법을 이용한 트리메부틴말레인산 서방정의 혼합 과정 모니터링)

  • Chemometrics using near-infrared (NIR) and Raman spectroscopy have found significant uses in a variety quantitative and qualitative analyses of pharmaceutical products in complex matrixes. Most of the pharmaceutical can be measured directly with little or no sample preparation using these spectroscopic methods. During pharmaceutical manufacturing process, analytical techniques with no or less sample preparation are very critical to confirm the quality. This study showed NIR and Raman spectroscopy with principal component analysis (PCA) was very effective for the blending processing control. It is of utmost importance to evaluate critical parameters related to quality of products during pharmaceutical processing. The blending is confirmed by off-line determination of active pharmaceutical ingredient (API) by a conventional method such as high performance liquid chromatography (HPLC) and UV spectroscopy. These analytical methods are time-consuming and ineffective for real time control. This study showed the possibility for the determination of blend uniformity end-point of CR tablets with the use of both NIR and Raman spectroscopy. The samples were acquired from six positions during blending processing with U-type blender from 0 to 30 min. Using both collected NIR and Raman spectral data, principal component analysis (PCA) was used to follow the uniformity of blending and finally determine the end-point. The variation of homogeneity of six samples during blending was clearly found and blend uniformity end-point was successfully confirmed in the domains of principal component (PC) scores.

A Study on the Improvement of Image-Based Water Level Detection Algorithm Using the Region growing (Region growing 기법을 적용한 영상기반 수위감지 알고리즘 개선에 대한 연구)

  • In this study, the limitations of the existing water level detection algorithm using CCTV images were recognized and the water level detection algorithm was improved by applying the Region growing technique. It applied three techniques (Horizontal projection profile, Texture analysis, and Optical flow) to estimate the water area, and the results were analyzed in a comprehensive analysis to select the initial water area. The water level was then continuously detected by the Region growing technique, referring to the initial water area. As a result, it was possible to confirm that the exact level of water was detected without being affected by environmental factors compared to the existing level detection algorithm, which had frequent mis-detection phenomena depending on the surrounding environmental factors. In addition, the water level was detected in the video showing flooded roads in urban areas, not in the video of the river. These results are believed to be able to supplement the difficulty of monitoring at all times with limited manpower by automatically detecting the level of water through numerous CCTV footage installed throughout the country, and to contribute to laying the foundation for preventing disasters caused by torrential rains and typhoons in advance.

Surgical Complications of Cerebral Arterivenous Malformation and Their Management (뇌동정맥기형의 외과적 수술합병증과 그 처치)

  • Objectives : The goal of surgical management of cerebral arteriovenous malformation(AVM) is elimination of the lesion without development of new neurological deficits. To improve the management results of cerebral AVMs in the future, this article discusses about surgical complications of the AVM and their management. Material and Methods : During the past 18 years, 116 patients with cerebral AVMs were managed by surgery. Among these cases, 7 cases died, 7 cases developed new neurological deficits, 11 cases residual AVM and 5 cases intracerebral hematoma(ICH) after surgery. The author analyzes the causes of those complications and investigates the methods to minimized those complications based on the review of the literatures. Results : One stage removal of AVM and ICH in the poor neurological state were performed in 5 of 7 death cases. Subtotal removal of ICH followed by delayed AVM surgery after recovery is regard as one method to improve the outcome of patient with large ICH. Postoperative new neurological deficits developed owing to normal perfusion pressure breakthrough(NPPB) in 3, judgement error in 2, preoperative embolization in 1 and cortical injury in 1 case(s). Proper management of NPPB, accurate anatomical knowledge and physiological monitoring during operation, and well trained skill for embolization are regard as methods to minimize those complications. Residual AVMs after surgery were noticed in 11 cases, in which unintended 6 cases due to inaccurate dissection of peripheral margin of AVM, and intended 3 cases due to massive brain swelling during operation, 1 cases due to diffuse type and 1 case due to multiple type of AVM. Accurate dissection of peripheral margin of AVM and mild hypotension during operation may help to avoid this complication. Postoperative hemorrhage occurred in 3 cases due to rupture of the residual AVM and in 2 cases due to oozing from the AVM bed. Complete resection of AVM, complete control of bleeding points at AVM bed and mild hypotension during early postoperative period are the methods to avoid this complication. Conclusion : A precise but flexible therapeutic strategy and refined skill for endovascular, radiosurgical and microsurgical techniques are required to successful treatment of cerebral AVM. Adequate timing of AVM resection, accurate anatomical knowledge, proper management of NPPB and accurate dissection of peripheral margin of AVM are the key points for avoiding complications of the AVM surgery.

Analysis of the Forest Road Cut-slope Erosion Control and Rehabilitation Techniques using Gabion Systems with Vegetation Base Materials (임도비탈면에 시공한 식생기반재돌망태의 침식방지 및 녹화효과 분석)

  • Analysis of new approaches to achieve naturally good ecological potential of forest road cut-slope by making the best use of advantages of gabion systems with vegetation base materials to prevent slope failure and erosion, in the area with highly erodible soil. Existing gabion systems can be divided into monolithic and modular system and can be divided into ten subtypes according to the purpose of establishment and combination of other measures. As a result on the monitoring of erosion amount from forest road cut-slopes in the test applications, the order of erosion amount from largest to smallest is as follows: the curved road cut-slope site where normal gabion system was established 5,840 $cm^3$; the control site 5,833 $cm^3$; the straight road cut-slope site where normal gabion system was established 5,621 $cm^3$; the curved road cut-slope site where the new gabion system was established 4,298 $cm^3$; and the straight road cut-slope site where the new gabion system 4,117 $cm^3$. Therefore, the result shows that the new gabion system is more effective than the normal gabion system to reduce erosion amount from forest road cut-slopes. During the study period, vegetation coverages of the straight and curved road cut-slope site where the new gabion system was established were about 56(30~85)% and about 45(28~65)%, so average vegetation coverage of the sites where the new gabion systems was established was higher than the sites where the normal gabion systems was established. Therefore, it was concluded that the new gabion system can be more effective for cut-slope revegetation.

Intertidal DEM Generation Using Satellite Radar Interferometry (인공위성 레이더 간섭기술을 이용한 조간대 지형도 작성에 관한 연구)

  • High resolution intertidal DEM is a basic material for science research like sedimentation/erosion by ocean current, and is invaluable in a monitoring of environmental changes and practical management of coastal wetland. Since the intertidal zone changes rapidly by the inflow of fluvial debris and tide condition, remote sensing is an effective tool for observing large areas in short time. Although radar interferometry is one of the well-known techniques for generating high resolution DEM, conventional repeat-pass interferometry has difficulty on acquiring enough coherence over tidal flat due to the limited exposure time and the rapid changes in surface condition. In order to overcome these constraints, we tested the feasibility of radar interferometry using Cosmo-SkyMed tandem-like one-day data and ERS-ENVISAT cross tandem data with very short revisit period compared to the conventional repeat pass data. Small temporal baseline combined with long perpendicular baseline allowed high coherence over most of the exposed tidal flat surface in both observations. However the interferometric phases acquired from Cosmo-SkyMed data suffer from atmospheric delay and changes in soil moisture contents. The ERS-ENVISAT pair, on the other hand, provides nice phase which agree well with the real topography, because the atmospheric effect in 30-minute gap is almost same to both images so that they are cancelled out in the interferometric process. Thus, the cross interferometry with very small temporal baseline and large perpendicular baseline is one of the most reliable solutions for the intertidal DEM construction which requires very accurate mapping of the elevation.
