The stability of monascin yellow pigment isolated from Monascus purpureus was determined over a period of storage for the wide range of pH, various metal ions and antioxidants. The absorption maximum of monascin pigment was 385 nm. Monascin pigment was more stable in acid solutions than in alkaline (pH 9 and pH 11) during storage period. It was also observed the reduction of absorption was occur after 3 days storage. The stability of monascin pigment was not changed by adding the various metal ions of the concentration of 10-4 M, however, it was unstable by adding the Zn2+, Al3+ and Fe3+ of 103- M concentration. The antioxidants. BHA, BHT, cysteine and L-ascorbic acid, have no effects on the stability of monascin yellow pigment. Thus, it may be concluded that the monascin pigment is stable and useful food additives as the natural colorant except for the alkaline food and food containing the Zn2+, Al3+ and Fe3+.
The growth characteristics and production of color pigments by Monascus strains were investigated during liquid culture, and production of monacolin K in red mold rice was carried out by solid state fermentation. Four different Monascus ruber strains were cultured in potato dextrose yeast extract broth (PDYB) media at $25^{\circ}C$ for 15 days. The high producing strain for red pigment was not corresponded to the strain for yellow pigment. Production of red pigment was high in the strain causing the fast pH change in culture broth. Production of monacolin K in red mold rice by solid state fermentation was influenced by a combination of wet cell weight and spore density in inoculum by liquid culture. Most strains showed the high production of monacolin K in red mold rice, when submerged fermentation was carried out for 5 days as inoculum for solid state fermentation. These results suggest that submerged fermentation period of inoculum have an effect on the production of monacolin K in red mold rice by solid state fermentation, and monacolin K in red mold rice could be increased by controlling the condition of submerged fermentation for inoculum.
The optimum cultural conditions for production of red pigment from Monascus rubber KCTC 6122 is liquid culture were studied. Monascus robber KCTC 6122 was shown to give the maximum production of red pigment in the medium containing 4% rice powder, 0.2% NaNO$_3$, 0.3% Na$_2$HP0$_4$ and 0.15% MgSO$_4$. The optimum culture conditions, temperature, initial pH and shaking speed were 30$^{\circ}C$, 6.5 and 150 rpm, respectively. The red pigment production reached a maximum level at 8days of cultivation.

We had screened the Monascus strain capable of producing monacolin K dominantly among 29 Monascus strains. Red yeast rice was prepared by culturing each Monascus sp. with 200 g of steamed rice (12$0^{\circ}C$, 20 min) at 3$0^{\circ}C$ for 10 days and drying at 8$0^{\circ}C$ for 20 min. As a result, red yeast rice cultured by M. purpureus ATCC16457, M. purpureu IFO 32316, M. purpureus IFO 32228, M. kaoliang ATCC 46595 and M. kaoliang ATCC 46596 produced lots of red pigment and monacolin K. An unidentified Monascus sp. showed the highest productivityof red pigment and monacolin K among 29 Monascus strains. Its production of red pigment and monacolin K was 1.3∼39 times and 2.4∼8 times higher than other strains, respectively. Although the morphological characteristics of unidentified Monascus strain were a little different from the typical M. purpureus, it was identified as M. purpureus CBS 281.34 from the result of sequencing of ITS (Internal transcribed spacer) and 28S ribosomal RNA (partial).
Monascus is a source of food colorant with high productivity of the pigment azaphilone. Monascus azaphilone (MAz) is biosynthesized through a single non-reducing polyketide pathway, the major components of which are ankaflavin (1), monascin (2), rubropunctatin (3) and monascorubrin (4); valuable biological activities have been reported for these compounds. Thus, various culture conditions were explored to reduce the cost of culture ingredients, enhance productivity and modulate compound composition. In the present study, we examined an extractive fermentation (EF) method with Diaion HP-20 resin (HP20) in direct comparison to a previously explored method involving Triton X-100 (TX100) to explore the modulated production of the major MAzs. We employed wild-type Monascus purpureus as well as two derivative recombinant strains (${\Delta}mppG$ and ${\Delta}mppE$) that are known to have differential MAz profiles as that of the wild-type strain. The HP20 resin was capable of modulating the MAz profile in favor of orange MAzs 3 and 4, oxidized congeners in this class, as was TX100-a phenomenon not previously observed for TX100 EF with Monascus anka. These finding substantiate that HP20 can be employed for the selective production of oxidized MAz and for diversifying the culture conditions used for Az production.
Growth and pigment formation in Genus Monascus(M. pilosus and M purpureus) related 15 kinds of culture media(Lin medium, SP medium, YM medium, YE medium, GMIN medium, SMO medium, MY medium, GY medium, Nishikawa medium, sucrose medium, stock culture, Mizutani medium, modified Lin medium, Toya medium and rice medium) were investigated. Mizutani medium and Lin medium among 15 kinds of the culture media showed good growth for M. pilosus, M purpureus, fresh mycelium weight cultivated for 10 days at 30$^{\circ}C$ was each 24.5∼26.9 or 15.9∼17.2 g/100 mL. The culture media which showed higher content of pigment production in two fungi were Lin medium(OD: 1.2 ∼ 1.6 and Mizutani medium(OD: 0.8 ∼ 1.0) that showed higher in M. pilosus.

In general, pigment production can be influenced by the medium composition, pH and physical factors such as aeration, agitation, and visible light. The influence of gaseous environments on the pigment production by Monascus purpureus ATCC 16365 was investigated by controlling the DO (dissolved oxygen) concentration through aeration and agitation. When the DO concentration was controlled below 20%, the production of red pigment significantly increased whereas the biomass production decreased. Therefore, the dissolved oxygen concentration could significantly affect the biosynthesis of red pigment as a secondary metabolite by a wild-type filamentous fungus under the anaerobic condition. The results indicate a high potential of enhancing the productivity of the red pigment as a secondary metabolite through controlling the DO concentration.
Optimization of culture conditions for pigment production of Monascus purpureus P-57 mutant was investigated in liquid culture. The optimum composition of medium for the pigment production was $4\%$ rice power, $0.1\%$ beef extract, $0.03\%$ glutamic acid, $0.1\%\;MgSO_4{\cdot}7H_2O,\;0.25\%\;KH_2PO_4$, the optimum initial pH was 5.0. And the optimum culture conditions was at $30^{\circ}C$ for 8 days under 150 rpm with shaking. M. purpureus P-57 mutant produced the highest pigment as 356.04 units at red pigment and as 268.20 units at yellow pigment, and produced high cell mass as 15.00 g/L in liquid culture under the optimum conditions.

The hyphal growth, production of color pigments and monacolin K by Monascus strains were investigated in liquid medium. Thirty five different strains were collected and cultured in potato dextrose yeast extract broth (PDYB), potato dextrose broth (PDB) and malt extract broth (MEB) medea at $25^{\circ}C$ for 7 days. The growth rates of most of strains were highest in PDYB medium. Growth rate as well as pigment production were influenced by suspension conditions of mycelia during liquid cultivation. Most of strains producing monacolin K corresponded to strains producing red pigment highly and showing more pH changes of liquid media. Monacolin K produced from strains was detected in culture broth as well as mycelia. Any citrinin was not detected in monacolin K producing strains. These results imply that the selection of the strains producing red pigment highly and showing more pH changes in liquid cultivation could be applied for primary screening of Monascus strains for preparation of red mold rice.
To investigate the mechanism for the main chain-elongation process and the possibility of putative precursors as stater units in the biosynthesis of the Monascus pigments, feeding experiments with possible $poly-{\beta}-ketide$ intermediates were carried out. Both crotonic acid and sorbic acid, especially in low concentrations, enhanced the pigment production while not increasing the dried mycelium weight appreciably. Also, it was observed that the feeding of sorbic acid and its ethyl ester was about two folds efficient in the pigment production than the feeding of crotonic acid and its ethyl ester. In addition to these acids, cinnamic acid and vinylacrylic acid were examined for their possibility as starter units. It was observed that the color of the culture fed with cinnamic acid was dominantly dark-red, but with overall decrease in the pigment production. Whey its ethyl ester was administered to the culture, however, the pigment production increased significantly. Also noted in 2D TLC study of the pigments was the increased production of red pigment and the formation of new red pigments.
