Syntheses and Evaluations of Antitumor and Antiangiogenic Phthalate Polymers Containing 5-Fluorouracil and Carboxylates

  • New antitumor active polymers, poly(methacryloyl-2-oxy-1,2,3-propanetricarboxylic acid-co-exo-3,6-epoxy-l,2,3,6-tetrahydrophthalic acid) [poly(MTCA-co-ETAc)], poly(methacryloyl-2-oxy-l,2,3-propanetricarboxylic acid-co-hydrogen ethyl-exo-3,6-epoxy-l,2,3,6-tetrahydrophthalate) [poly(MTCA-co-HEET)], and poly(methacryloyl-2-oxy-l,2,3-propanetricarboxylic acid-co-a-ethoxy-exo-3,6-epoxy-1,2,3,6-tetrahydrophthaloyl-5-fluorouracil) [poly(MTCA-co-EETFU)] were synthesized and characterized. Their antitumor activity, inhibition of DNA replication and antiangiogenesis were examined. The structures of the polymers were identified by FT-IR, $^1H$ and $^{13}C$-NMR spectroscopy. The number average molecular weights of the fractionated polymers determined by GPC ranged from 9,400 to 14,900, and polydispersity indices were less than 1.7. The in vitro cytotoxicity of these polymers was determined and their antitumor activity was evaluated. The $IC_{50}$ values (the drug concentration at inhibition of 50% tumor growth) indicated that the synthesized polymers were much better inhibitors of cancer cells and showed lower cytotoxicity than the free 5-FU. The in vivo antitumor activity of the conjugates was examined using mice bearing the sarcoma 180 tumor cell line. The life spans (TIC) of the mice treated with the conjugates were higher than those treated with the free 5-FU. In addition, the synthesized conjugates showed excellent antiangiogenic activity based on an embryo chorioallantoic membrane assay.

Identification of Allergens of Dermatophagoides farinae on Canine Atopic Dermatitis in Korea (국내 개에서 발생한 아토피성 피부염에서 Dermatophagoides farinae의 항원 확인)

  • Dermatophagoides farinae plays important role in the pathogenesis of canine atopic dermatitis as environmental allergens. Also, many studies revealed that D. farinae was the main causative allergen for Korean dogs with atopic dermatitis. To identify major allergens of D. farinae in Korean atopic dogs allergic to D. farinae by immunoblot using commercial allergenic extracts, 26 dogs from two groups were enrolled in the study. Control group consists of 10 dogs with no clinical signs of disease and atopic group consists of 16 dogs diagnosed as atopic dermatitis. Sera from Korean dogs with atopic dermatitis showed six allergens of D. farinae extract by procedure of immunoblot. The molecular weights of identifying protein bands were 177, 109, 75, 44, 27, 15 kDa. The major allergens showing reactivity with greater than 50% of atopic dogs were detected at approximately 44, 109 and 177 kDa. Subsequent investigations will be carried out to verify the identity of the allergens detected in this study.

Localization and isozyme patterns of phosphatase in Fibricola seoulensis (Fibricola seoulensis에서 phosphatase의 분포와 동위효소유형)

  • The present study was carried out to investigate the localization and isozyme patterns of acid phosphatase and alkaline phosphatase in metacercariae and in adults of F. seoulensis by enzyme-histochemistry method and electrophoresis. Acidphosphatase showed a strong activity at pH 5 in the intestinal caecum of adults, but showed no reactions in the nonsubstrate control and in the inhibitor-treated control. Alkaline phosphatase showed a strong activity at pH 8 in the intestinal caecum and the tribocytic organ of adults, and in the intestinal caecum and in the genital anlagen of metacercariae. In non-denature PAGE, ten bands of protein fraction from the extracts of metacercariae and twenty-two bands from adults were detected. In denature PAGE, two protein bands having molecular weights of 192 kDa and 123 kDa were detected in the metacercariae, but absent from adult stage. In adults, protein fractions of 27.5 kDa, 24.5 kDa, 21.4 kDa, 18 kDa, 16 kDa and 15 kDa were detected. In non-denature PAGE, isozymes of acid phosphatase showed the most strong activity at pH 5, whereas no activity was shown at pH 2 and pH 7. One isozyme 85 kDa, 73 kDa and 62 kDa) in adults.

Anticariogenic Effects and Inhibition of Glucosyltransferase Activity of Chrysanthemun indicum L. Extracts (감국(Chrysanthemun indicum L.) 추출물의 항충치효과와 Glucosyltransferase 저해활성 탐색)

  • The anticariogenic effects and inhibition of glucosyltransferase activity of Chrysanthemun indicum L. extracts were investigated against 5 strains of microorganisms. The ethanol extracts of Chrysanthemun indicum L. showed the growth inhibition effects on the Streptococcus mutans and oral bacteria. The inhibition rate of glucosyltransferase activity of Chrysanthemun indicum L. extracts showed 78.4 to 92.3% range. The extracts were analysed by using solvent fractionation, silica gel adsorption chromatography, Sephadex LH-20 column chromatography, TLC, HPLC techniques. Three components whose molecular weights ranged from 200 to 400 were confirmed to have the anticariogenic activity.

  • Chitin was isolated from red snow crab (Chinonecetes japonicus) and deacetylated by boiling alkaline solution to produce chitosan. The physical properties of chitosan solution and its film properties was examined, As the molecular weights of chitosan was increased from 297Kpa to 319Kpa, the tensile strength, degree of elongation and water permeability of chitosan film were increased. As the degree of deacetylation of chitosan was increased, the tensile strength, the degree of elongation and water permeability of chitosan film were increased. As the concentration of chitosan solution was increased, the degree of elongation and water permeability of film were increased, whereas the tensile strength of film was decreased, As the pH of chitosan solution was increased, the tensile strength and water permeability of film were decreased, whereas the degree of elongation of film was increased.

Protein Profiles in Response to Salt Stress in Seedling of Salt Tolerant Rice Mutants

  • Proteomic analysis was performed in order to identify proteomic changes by salt stress between the Japonica cv. Donganbyeo (WT) and two salt-tolerant (ST) mutant lines by using the SDS-PAGE and 2-DE. Two salt tolerant rice mutant lines, ST-87 and ST-301, were selected by in vitro mutagenesis with gamma-ray. Three-week-old seedlings were treated with 171 mM NaCl for 7 days. In the SDS-PAGE, three proteins with molecular weights of 27, 46 and 58 kDa were highly increased under salt treatment. Total proteins from shoots of both WT and ST-lines were separated by two-dimensional gel electrophoresis. In 2-DE, 201, 226, 217 and 213 protein spots were detected in the untreated-or treated-WT and untreated- or treated-ST-87, respectively. Of theses, 17 and 10 protein spots were up- and down-regulated under salt stress in the WT, respectively. While, 16 and 8 protein spots were up- and down-regulated under salt stress in the ST-87, respectively, compared with the untreated plants. High intensity or de novo synthesized proteins were analyzed by MALDI-TOF/MS analysis.

  • Poly(ethylene glycol) (PEG), isophoron diisocyanate (IPDI), and 1,1,1-tris(hydroxymethyl)propane (THMP) were used to synthesize polyurethane microgel. The formulation to form a microgel was determined, and the shape and the properties of the microgel were characterized with SEM and particle size analyzer. The microgel was only formed when PEG was used more than THMP exceedingly. Therefore, it is believed that PEG plays an important role in the synthesis of the microgel. During the formation of the microgel, the viscosity of the microgel solution was decreased. Molecular weights of the PEGs which were used in this research were 2,000, 6,000, and 10,000. The microgel synthesized with PEG 6000 showed the best property in comparison with others. The size of the microgel measured by particle size analyzer were about 130~230 nm.

  • This review shows the design and the development of new $CO_2$-soluble hydrocarbon copolymers which can be used as effective stabilizers for successful dispersion polymerizations of bio-compatible materials in supercritical carbon dioxide ($scCO_2$). The basic concepts of supercritical fluid including its solvent properties and applications in polymer synthesis are described. We report the facile synthesis of highly soluble hydrocarbon based copolymers, prepared with good control via controlled free radical polymerization from readily accessible and commercially available monomers. The phase behaviour of these materials was monitored in pure $CO_2$ to investigate how the molecular weights and the composition of the copolymers affect their solubility in $CO_2$. Their activity as a stabilizer was then tested in dispersion polymerization of N-vinyl pyrrolidone in $CO_2$ at various reaction conditions to identify the key parameters required for a successful dispersion stabilization of growing PVP particles. Some prospective potentials of this research which can be applied in developing new polymer materials in an environmentally-friendly fashion for use in cosmetics are also discussed.

  • UV-screening agents have some adverse effects that raise consumers' concern. The organic agents often cause irritation and may penetrate into human body while the inorganic agents raise aesthetic issues because they often turn opaque. Organic agents with high molecular weights and nano-sized inorganic agents have been developed respectively to minimize transdermal intrusion into human body and suppress turning opaque. Recently, we reported preparation of powdery UV-screening agents made of polysilsesquioxane, an organic-inorganic hybrid material. Powders would not penetrate into epidermis and organic-inorganic hybrid nature would suppress the opaqueness problem. In this study, we continued our research on this powdery polysilsesquioxane UV-screening agent, SESQUV, regarding its chemical composition, its synergic UV-screening effects when mixed with other organic agents, and its applicability in practical formulation. Results showed SESQUV was promising UV-screening agents useful in sunscreen formulation.

Quality Characteristics of Gamma-Irradiated Chickens (감마선 조사 계육의 품질특성)

  • This research was conducted to investigate changes in quality characteristics of gamma-irradiated chickens during storage at different temperature and periods. In heme pigment(myoglobin) content, metmyoglobin content of chicken stored at 5$^{\circ}C$ was not affected by gamma irradiation but slightly increased with the increase in storage period. All samples stored at -20$^{\circ}C$ were no different in heme pigment content between nonirradiated and irradiated samples and slightly decreased as the freezing storage period increased. The SDS electrophoresis patterns were not significantly different between nonirradiated and irradiated samples. All samples stored in at 5$^{\circ}C$ showed a prominent breakdown of molecular weights ranging from 97,000 to 116,000 Daltons after 8 weeks' storage. TBA values increased according to the increment of irradiation dose level and storage period at both temperatures, 5$^{\circ}C$and -20$^{\circ}C$. However, The acid value decreased with increasing irradiation dose level. In the VBN value, nonirradiated chickens were four times higher than that of 7 kGy-irradiated one.
