Thermal Process for Enhancing Mechanical Strength of PLGA Nanoparticle Layers on Coronary Stents

  • Poly (lactic-co-glycolic acid) (PLGA) nanoparticles loading paclitaxel have been deposited on coronary stents by self-assembling properties of colloidal particles. The layers of the nanoparticles were enhanced to a sufficient mechanical strength by a thermal process under the proper temperature and humidity conditions. In vitro release studies proved the controlled paclitaxel release of the nanoparticle layers. This technique gives rise to a new range of applications for nanoparticles and drug-eluting stents.

  • Three hairpin polyamides were designed and synthesized by a haloform reaction and DCC/HOBt coupling reaction without amino protection and deprotection. Their anticancer activity were investigated with three kinds of cell lines-hepatic carcinoma, lung carcinoma and gastric carcinoma, and the values of $IC_{50}$ were at range of $10^{-7}~10^{-8}M$.

  • Peptidoglycan is an extensively cross-linked polymer essential for the integrity of the bacterial cell wall. Many antibiotics act by disruption of its biosynthesis and assembly, several are targeted against the cytoplasmic enzymes that synthesize the key intermediate UDP-N-acetylmuramyl pentapeptide. One such drug is fosfomycin, which inactivates the first enzymes in this pathway, UDP-N-acetylglucosamine enopyruvyl transferase (murZ).(omitted)

  • Bare metal stents which were used to treat coronary artery disease have several biochemical problems. Polymerbased drug-eluting stents (DES) have opened up a new paradigm in the treatment of in-stent restenosis. Many studies and research programmes have proved that DES can prevent restenosis. In our study, paclitaxel-loaded poly (lactic-co-glycolic acid) (PLGA) nanoparticles have been deposited along the three dimensional scaffold of coronary stents by a method using self-assembling properties of colloidal particles. We found that the nanoparticles were deposited uniformly and closely packed. The amount of paclitaxel was easily controlled by the drug content of the nanoparticles and the deposition count.

  • Using molecular dynamics simulation method, we studied the carbon nanotube (CNT) non-covalently interacting with a polymer. As the polymer coiled around the CNT, the diameter of CNT deformed by more than 40% of its original value within 50 ps. By considering three different polymers, we conclude that the interaction between the CNT and polymer is governed by the number of repeating units in the polymer, not by the molecular weight of polymer.

  • The genome sequencing has revealed a large number of proteins of unknown or little characterized functions that have been well conserved during evolution. It remains a great challenge to decipher the molecular and physiological functions of these proteins. One example of the evolutionarily conserved protein family with little understood function is the surE family.(omitted)
