Genotypic Variation in Leaf Water Status of Soybean

  • Plant water status during growth is directly and indirectly associated with seed yield. The objective of the present study was to determine the genotypic differences in leaf water characteristics at an early growth stage of soybean [Glycine max (L.) Merrill] plants through the pressure-bomb technique. Measurements of water potential as well as relative water content (RWC) were made at the third leaf from the fully-expanded top leaf of eight different soybean genotypes grown for 31 to 35 days after field emergence. On the basis of the modified exponential model, pressure-volume (PV) curves were fitted well ($R^2$=0.92** to 0.99** for the curvi-linear region and R=0.67** to 0.96** for the linear region), indicating that a segmented model using PROC NLIN of SAS could be used effectively to estimate the leaf water characteristics. The regression analysis for the pressure-volume (PV) curve revealed genotypic variation in the solute potential at saturation (Ψ$_{s,sat}$ :-10.7 to -14.8 bar), solute potential at incipient plasmolysis (Ψ$_{s,ip}$ : -14.3 to -18.3 bar), RWC at incipient plasmolysis (RW $C_{ip}$ : 83.3 to 91.7%), high integrated turgor pressure from saturation to plasmolysis ( $_1$$^{b}$ : 0.39 to 0.81), and maximum volumetric modulus of elasticity ($\varepsilon$$_{max}$ : 150 to 445 bar).).

  • 기후변화에 따른 환경 스트레스 대응 기술과 영농의사결정 플랫폼 개발을 위해서는 환경 조건에 따른 작물의 반응을 이해하기 위한 시스템 개발이 매우 중요하다. 본 연구는 한국형 SPAR 시스템이 다양한 환경 조건에서 작물 생육 반응을 어떻게 정량화하고, 향후 작물 생육 모형 개발에 어떻게 연계될 수 있는지에 대해 방향을 제시하고자 수행되었다. 한국형 SPAR 시스템은 온도, $CO_2$ 농도 등의 기상요소와 양 수분 관리 등 재배요소를 동시에 정밀 조절할 수 있을 뿐 만 아니라 군락수준에서 광합성 및 호흡 등 작물의 생육 반응을 실시간으로 정량화하기에 최적화되어 있다. 본 시스템을 통해 수집된 군락 광합성 정보는 실제 작물의 환경조건에 따른 생육량 변동을 매우 유의하게 반영하여 향후 작물 생육 모형에 실질적으로 적용 가능한 환경-유전 요인간 특이적 반응 함수 개발에 크게 활용될 것으로 기대된다.

Functional characterization of gibberellin signaling-related genes in Panax ginseng

  • Gibberellins (GAs) are essential phytohormones for plant growth that influence developmental processes and crop yields. Recent functional genomic analyses of model plants have yielded good characterizations of the canonical GA signaling pathways and related genes. Although Panax ginseng has long been considered to have economic and medicinal importance, functional genomic studies of the GA signaling pathways in this crucial perennial herb plant have been rarely conducted. Here, we identified and performed functional analysis of the GA signaling-related genes, including PgGID1s, PgSLY1s, and PgRGAs. We confirmed that the physiological role of GA signaling components in P. ginseng was evolutionarily conserved. In addition, the important functional domains and amino acid residues for protein interactions among active GA, GID1, SCFSLY1, and RGA were also functionally conserved. Prediction and comparison of crystallographic structural similarities between PgGID1s and AtGID1a supported their function as GA receptors. Moreover, the subcellular localization and GA-dependent promotion of DELLA degradation in P. ginseng was similar to the canonical GA signaling pathways in other plants. Finally, we found that overexpression of PgRGA2 and PgSLY1-1 was sufficient to complement the GA-related phenotypes of atgid1a/c double- and rga quintuple-mutants, respectively. This critical information for these GA signaling genes has the potential to facilitate future genetic engineering and breeding of P. ginseng for increased crop yield and production of useful substances.

Investigating the Metabolism of Clubroot-Infected Plants by Integrating Metabolomic and Transcriptomic Approaches

  • Clubroot (Plasmodiophora brassicae) is a serious agricultural problem affecting Brassica crop production worldwide. It also infects the model plant Arabidopsis thaliana. During infection, this biotrophic pathogen manipulates the development and metabolism of its host leading to the development of galls in the root and hypocotyl. In turn, its own development is strongly influenced by the host. The aim of this study is to investigate the metabolism of clubroot-infected plants using a combination of transcriptomic and metabolomic approaches. We have used direct injection mass spectrometry to obtain a metabolic fingerprint of when changes in the metabolome occur and linked this with changes in host and pathogen gene expression. We have identified alterations in carbohydrate metabolism that occur during P. brassicae infection of A. thaliana plants. Transcriptomic analysis showed that host genes associated with sugar transport and metabolism were induced during gall formation and that the pathogen also expresses genes associated with these processes. We have examined the impact of inactivating host sucrose synthase, cytosolic invertase and sugar permeases on gall formation, identifying host genes that are required for gall formation. We have also explored how sugar status is changed in root tissue, developing and mature leaf during infection of wild type and mutant plants.

제2형 당뇨 동물모델에서 일반메밀과 쓴메밀의 항당뇨 효과 비교 (Anti-diabetic effects of common buckwheat and tartary buckwheat in type II diabetes animal model)

  • 메밀은 전 세계적으로 곡식, 싹, 차 등 다양한 형태로 이용되고 있으며, 주요 재배종으로는 일반메밀과 쓴메밀이 있다. 본 연구는 일반메밀과 쓴메밀 종실과 전초를 대상으로 항산화 및 항당뇨 활성 효과를 평가하였다. 루틴 함량은 쓴메밀 추출물이 일반메밀 추출물보다 44-48배 이상 높았으며, 퀘세틴은 전초에서만 검출되었다. 플라보노이드 및 폴리페놀 함량도 쓴메밀 추출물이 일반메밀 추출물보다 2.5-4.8배 높았다. STZ 투여로 제2형 당뇨가 유발된 동물모델에 일반메밀과 쓴메밀의 종실과 전초를 급여하여 공복혈당, 경구 내당능, 인슐린 내성 변화, 혈중 포도당 및 인슐린 농도 등을 통해 항당뇨 효과를 확인하였다. 종실의 경우는 일반메밀에 비해 쓴메밀 종실군에서 경구 내당능 효과가 뛰어났으며, 혈청 포도당 농도도 유의적으로 감소하는 것을 확인하였다. 전초의 경우, 일반메밀이나 쓴메밀 급여 실험군에서 모두 당뇨대조군에 비하여 유의적인 항당뇨 효과를 보였으며, 인슐린 저항성 지표인 HOMA-IR과 인슐린 민감성 지표인 QUICKI를 비교해본 결과에서도 종실과 전초 섭취군 모두 인슐린 저항성을 감소시키고, 인슐린 민감성을 증가시키는 것을 확인하였다. 이러한 결과들을 고려하면 루틴 함량 및 항산화활성이 쓴메밀 전초의 경우 항당뇨 효과와 밀접한 관계가 있으며, 루틴 함량과 항산화 활성이 높은 쓴메밀 종실 및 일반메밀과 쓴메밀 전초는 항당뇨 식의약 소재로 활용 가능성이 높은 것으로 판단된다.

Telomere association of Oryza sativa telomere repeat-binding factor like 1 and its roles in telomere maintenance and development in rice, Oryza sativa L.

  • Telomeres are specialized nucleoprotein complexes that function to protect eukaryotic chromosomes from recombination and erosion. Several telomere binding proteins (TBPs) have been characterized in higher plants, but their detailed in vivo functions at the plant level are largely unknown. In this study, we identified and characterized OsTRFL1 (Oryza sativa Telomere Repeat-binding Factor Like 1) in rice, a monocot model crop. Although OsTRFL1 did not directly bind to telomere repeats $(TTTAGGG){_4}$ in vitro, it was associated with telomeric sequences in planta. OsTRFL1 interacted with rice TBPs, such as OsTRBF1 and RTBP1, in yeast and plant cells as well as in vitro. Thus, it seems likely that the association of OsTRFL1 with other TBPs enables OsTRFL1 to bind to telomeres indirectly. T-DNA inserted OsTRFL1 knock-out mutant rice plants displayed significantly longer telomeres (6-25 kb) than those (5-12 kb) in wild-type plants, indicating that OsTRFL1 is a negative factor for telomere lengthening. The reduced levels of OsTRFL1 caused serious developmental defects in both vegetative and reproductive organs of rice plants. These results suggest that OsTRFL1 is an essential factor for the proper maintenance of telomeres and normal development of rice.

Yield and Nutritional Quality of Several Non-heading Chinese Cabbage (Brassica rapa var. chinensis) Cultivars with Different Growing Period and Its Modelling

  • The aims of the experiment, conducted over three years in the Central Europe field conditions, were (1) to investigate the effect of growing period (plantings in the middle and at the end of August: $1^{st}$ and $2^{nd}$ term, respectively) on yield and chemical composition of the non-heading Chinese cabbage (Brassica rapa var. chinensis) cultivars 'Taisai', 'Pak Choy White', and 'Green Fortune', and (2) to develop regression models to evaluate the changes in crop yields as a function of weather conditions. A highest marketable yield was obtained from 'Taisai' (65.71 and 77.20 $t{\cdot}ha^{-1}$), especially in the $2^{nd}$ term of production. Low yield, observed for 'Pak Choy White' was due to its premature bolting. Almost 39% ($1^{st}$ term) and 70% ($2^{nd}$ term) of plants of this cultivar formed inflorescence shoots before harvest. The highest dry matter level was observed in the leaf petioles of 'Taisai', while 'Green Fortune' was the most abundant of carotenoids and L-ascorbic acid. The content of soluble sugars was the lowest for 'Pak Choy White'. In a phase of harvest maturity, more of the analyzed constituents were gathered by plants from earlier plantings, and differences were as follows: 4.7% (dry matter), 26.3% (carotenoids) and 22.1% (L-ascorbic acid), in comparison to $2^{nd}$ term of production. Significant increase of soluble sugars level was observed for plants from later harvest. The regression model for marketable yield of Chinese cabbage cultivar 'Taisai' as a function of maximum air temperature can predict the yield with accuracy 68%. The models for yield or bolting of 'Pak Choy White', based on extreme air temperatures and sunshine duration, were more precise (98%). It should be pointed out that Taisai could be recommended for later growing period in Central Europe conditions with regard to maximum yield potential. 'Green Fortune' was notable for its uniform yielding. To obtained plants of higher nutritional value, earlier time of cultivation should be suggested. Described models can be successfully applied for an approximate simulation of Chinese cabbage yielding.

Epigenetic regulation of fungal development and pathogenesis in the rice blast fungus

  • Fungal pathogens have huge impact on health and economic wellbeing of human by causing life-threatening mycoses in immune-compromised patients or by destroying crop plants. A key determinant of fungal pathogenesis is their ability to undergo developmental change in response to host or environmental factors. Genetic pathways that regulate such morphological transitions and adaptation are therefore extensively studied during the last few decades. Given that epigenetic as well as genetic components play pivotal roles in development of plants and mammals, contribution of microbial epigenetic counterparts to this morphogenetic process is intriguing yet nearly unappreciated question to date. To bridge this gap in our knowledge, we set out to investigate histone modifications among epigenetic mechanisms that possibly regulate fungal adaptation and processes involved in pathogenesis of a model plant pathogenic fungus, Magnaporthe oryzae. For functional and comparative analysis of histone modifications, a web-based database (dbHiMo) was constructed first to archive and analyze histone modifying enzymes from eukaryotic species whose genome sequences are available. Based on the database entries, we carried out functional analysis of genes encoding histone modifying enzymes. Here I provide examples of such analyses that show how histone acetylation and methylation is implicated in regulating important aspects of fungal pathogenesis. Current analysis of histone modifying enzymes is followed by ChIP-seq and RNA-seq experiments to pinpoint the genes that are controlled by particular histone modifications. We anticipate that our work will provide not only the significant advances in our understanding of epigenetic mechanisms operating in microbial eukaryotes but also basis to expand our perspective on regulation of development in fungal pathogens.

Monitoring Onion Growth using UAV NDVI and Meteorological Factors

  • Unmanned aerial vehicles (UAVs) became popular platforms for the collection of remotely sensed data in the last years. This study deals with the monitoring of multi-temporal onion growth with very high resolution by means of low-cost equipment. The concept of the monitoring was estimation of multi-temporal onion growth using normalized difference vegetation index (NDVI) and meteorological factors. For this study, UAV imagery was taken on the Changnyeong, Hapcheon and Muan regions eight times from early February to late June during the onion growing season. In precision agriculture frequent remote sensing on such scales during the vegetation period provided important spatial information on the crop status. Meanwhile, four plant growth parameters, plant height (P.H.), leaf number (L.N.), plant diameter (P.D.) and fresh weight (F.W.) were measured for about three hundred plants (twenty plants per plot) for each field campaign. Three meteorological factors included average temperature, rainfall and irradiation over an entire onion growth period. The multiple linear regression models were suggested by using stepwise regression in the extraction of independent variables. As a result, $NDVI_{UAV}$ and rainfall in the model explain 88% and 68% of the P.H. and F.W. with a root mean square error (RMSE) of 7.29 cm and 59.47 g, respectively. And $NDVI_{UAV}$ in the model explain 43% of the L.N. with a RMSE of 0.96. These lead to the result that the characteristics of variations in onion growth according to $NDVI_{UAV}$ and other meteorological factors were well reflected in the model.

Variability in the Viral Protein Linked to the Genome of Turnip Mosaic Virus Influences Interactions with eIF(iso)4Es in Brassica rapa

  • Plants protect against viruses through passive and active resistance mechanisms, and in most cases characterized thus far, natural recessive resistance to potyviruses has been mapped to mutations in the eukaryotic initiation factor eIF4E or eIF(iso)4E genes. Five eIF4E copies and three eIF(iso)4E copies were detected in Brassica rapa. The eIF4E and eIF(iso)4E genes could interact with turnip mosaic virus (TuMV) viral protein linked to the genome (VPg) to initiate virus translation. From the yeast two-hybrid system (Y2H) and bimolecular fluorescence complementation (BiFC) assays, the TuMV-CHN2/CHN3 VPgs could not interact with BraA.eIF4E.a/c or BraA.eIF(iso)4E.c, but they could interact with BraA.eIF(iso)4E.a in B. rapa. Further analysis indicated that the amino acid substitution L186F (nt T556C) in TuMV-UK1 VPg was important for the interaction networks between the TuMV VPg and eIF(iso)4E proteins. An interaction model of the BraA. eIF(iso)4E protein with TuMV VPg was constructed to infer the effect of the significant amino acids on the interaction of TuMV VPgs-eIF(iso)4Es, particularly whether the L186F in TuMV-UK1 VPg could change the structure of the TuMV-UK1 VPg protein, which may terminate the interaction of the BraA.eIF(iso)4E and TuMV VPg protein. This study provides new insights into the interactions between plant viruses and translation initiation factors to reveal the working of key amino acids.
