Cultivation of Oyster Mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at Medium with Ca(OH)2 Treatment (수산화칼슘[Ca(OH)2] 처리 배지를 이용한 느타리버섯의 재배)

  • This research is aimed to investigate the cultivation method of oyster mushroom[Pleurotus ostreatus (Jacq. ex Fr.) Kummer] at the non-sterilized medium with $Ca(OH)_2$ treatment. Therefore, experiments were carried out to develop non-sterilization method of medium by addition of $Ca(OH)_2$ for omission of heat sterilization progress of medium. General components, minerals and amino acid in Jiri wild type No. 1 (Pleurotus ostreatus) and production cost were analyzed. For the purpose of omission of heat sterilization progress, treatment ratio of $Ca(OH)_2$ (purity 95%) was 5%(w/w) of dry medium. Initial pH of this medium was 11 and then the pH was changed by 9 after the uniform mixing of the medium for half an hour. The various germs occurred 50% and 100% at pH 8 and pH 7 of the non-sterilized medium, respectively. Production of oyster mushroom increased by $2,030\;ton\;ha^{-1}$ when the main raw material used corn pith instead of waste cotton. The time required of mycelium culture was 30 days when hypha was cultured at the non-sterilized medium, and pinhead occurred when 2 or 3 days was passed after the time required of mycelium culture. Occurrence of pinhead was most rapid at the condition of $22{\sim}26^{\circ}C$, 65% humidity and pH $6.5{\sim}7.0$ and required of $22{\sim}28$ days at $70{\sim}80\;mm$ thickness of non-sterilized medium. Ca content in 1st harvest oyster mushroom was higher than that in 2nd harvest one, and its difference was $30.3\;mg\;kg^{-1}$. Amino acid content by stipe thickness of oyster mushroom was ranged from 411.2 to $343.9\;mg\;kg^{-1}$ both in a pileus and a stipe of 1st harvest mushroom, and from 402.4 to $498.2\;mg\;kg^{-1}$ and from 442.6 to $470.4\;mg\;kg^{-1}$ in those of 2nd harvest one, respectively. The results of the present study suggest that the non-sterilization medium by addition of $Ca(OH)_2$ is usable with the cultivation of oyster mushroom.

Oxygen and Hydrogen Isotopic Compositions of Stream Waters in the Han River Basin (한강 수계 분지내 하천수의 산소, 수소 안정동위원소 조성)

  • Oxygen and hydrogen isotopic compositions of stream water in the Han river basin are expressed by the equation of $\delta$D=6.6$\delta$$^{18}$ O-7.4, which is not satisfy the meteoric water line ($\delta$D=8$\delta$$^{18}$ O+10). It might be depended on the local climatic condition and the evaporation effect in the Han river basin. The $\delta$$^{18}$ O and $\delta$D values of stream water in the Han river basin range from -8.2 to -10$\textperthousand$ (avg. -9.1$\textperthousand$) and -60 to -96$\textperthousand$ (avg. -69$\textperthousand$), respectively. The stream water from the South Han river (8$\delta$$^{18}$ O= -8.9~ -10$\textperthousand$, avg.-9.3$\textperthousand$ $\delta$D: -66~ -96$\textperthousand$, avg.-69$\textperthousand$) is slightly more depleted in $^{18}$ O and D than those of North Han river ($\textperthousand$$^{18}$ O= -8.4~ -9.7$\textperthousand$, avg. -9.2$\textperthousand$, $\delta$D= -64~ -95$\textperthousand$, avg. -69$\textperthousand$). It reflects more altitude effect than the effect of latitude and Inflow of the $^{18}$ O eniched S $O_4$$^{2-}$ and HC $O_3$- from the carbonate rock and sulfide minerals in the Taebagsan and Hwanggangri mineralized zone. The Main stream water of the Han river having $\delta$D: -60~ -76$\textperthousand$ (avg.-68$\textperthousand$) and $\textperthousand$$^{18}$ O= -8.2~-10$\textperthousand$ (avg.9.0$\textperthousand$) is enriched in $^{18}$ O compared to the South and North Han river waters, which is caused by the evaporation effect. Binary simple mixing ratio of the Main Han river water between South and North Han river waters was obtained to be 6 : 4 by the isotopic data, suggesting a strong influence of South Han river water to the Main Han river water.

Base-metal Mineralization in the Cretaceous Gyeongsang Basin and Its Genetic Implications, Korea: the Haman-Gunbug-Goseong(-Changwon) and the Euiseong Metallogenic Provinces (한국 경상분지 백악기 비철금속 광화작용과 그 성인적 의의: 함안-군북-고성(-창원) 및 의성 광상구를 중심으로)

  • The Cretaceous magmatism in the Gyeongsang Basin, Korea, led to the formation of two contrasting metallogenic provinces: the Haman-Gunbug-Goseong(-Changwon) (HGGC) and the Euiseong (EU). The mineralization in the HGGC metallogenic province represents copper, gold and iron of porphyry-related deposits that display close relationships in time and space with subvolcanic granitoids. Much of copper-gold-forming events in this province are consistently constrained to the period between ca. 89 and 81 Ma. The hydrothermal systems of copper-gold vein deposits in the HGGC province are associated with ore-forming fluids of high to intermediate temperature (300∼50$0^{\circ}C$) with high salinity (20∼55 equiv. wt. % NaCl). The ore-forming fluids become progressively more diluted by the incorporation of decreased quantities of magmatic water further from the nearby intrusion, suggesting significant input and fluid mixing of a meteoric water component to the magmatic fluids during the late stage of geothermal systems. In contrast, the EU metallogenic province is characterized by polymetallic vein deposits that are consistently constrained to a period of 78∼60 Ma. The geothermal systems of polymetallic vein deposits in the EU province are derived from a narrow range of intermediate temperature (200∼40$0^{\circ}C$) with relatively low salinity(1∼7 equiv. wt.% NaCl). It may represent a mixed fluid of magmatic and meteoric waters. The base-metal mineralization in the Gyeongsang Basin shows a close spatial and temporal distinction between the proximal environment derived from shallow-level granitoids in the southwestern HGGC province and the distal condition derived from volcanic environments in the northwestern EU province.

β-Glucan Content and Antioxidant Activity of Mixed Extract from Sarcodon aspratus and Rice Bran (능이버섯과 미강 혼합 추출물의 β-Glucan 함량 및 항산화 활성)

  • This study was to investigate the optimal condition of mixture ratio for development of functional food ingredient from Sarcodon aspratus and rice bran. First, $^{\circ}Brix$ was measured along with extraction time. Five kinds of mixtures of Sarcodon aspratus and rice bran (10:0, 7:3, 5:5, 3:7, 0:10) were extracted in $95^{\circ}C$ water over a one-hour period and the extraction yield was evaluated. We further evaluated ${\beta}-glucan$ content, DPPH radical scavenging activity, ferric ion reducing antioxidant power (FRAP), total phenolic content and total flavonoids content. As a result, both Sarcodon aspratus and rice bran showed a constant $^{\circ}Brix$ after 45 minutes of extraction time. The content of ${\beta}-glucan$ was highest in the Sarcodon aspratus and rice bran mixture with a ratio of 3:7. As the ratio of rice bran increased in all mixtures, the antioxidant capacity also increased. In conclusion, to create a functional food ingredient the optimal mixing ratio of Sarcodon aspratus to rice bran is 3:7.

The Effect of Seasoning on the Intestinal Absorption -Absorption by Passive Transport and the Effect of Red Pepper- (조미료가 창자 운동과 흡수기능에 미치는 영향 -소장의 피동적 흡수에 대한 고추의 영향-)

  • Numerous factors concern with the absorption of substances through the membrane of the gastrointestinal tract. To simplify the experimental condition, present work has been restricted to observe the disappearance rate of substance from the intestinal loop which was made in the jejunum, 70 cm apart from the pylorus of the adult rabbit. The purpose of the study is to clarify the absorption of urea through the jejunal wall is solely attributable to the concentration difference between the luminal fluid and plasma, and to observe the effect of adding red pepper upon the rate of absorption. The rabbits were anesthetized with nembutal, 35mg/kg I.V. Jejunal loop was made by ligating at 2 spots, 70 cm and 80cm apart from the pylorus. After rinsing with normal saline solution through the polyethylene tubing inserted from the end of the loop, 8 ml of test solution was placed through the same tubing. The test solution contained 200 mg% of urea and 150mg% of polyethylene glycol(M.W. 4,000) in normal saline solution. Right after placing the test solution the first specimen was taken through the tubing, and successive samplings were performed at 5, 10, 20, and 30 minutes. Logarithm of the difference of urea concentration between the luminal fluid and plasma was plotted against time elapsed after the onset of the experiment. If straight line is revealed, it would verify the nature of transport mechanism as diffusion, obeying the Fick's principle. The concentration of polyethylene glycol (PEG) was also measured in order to examine the change in the volume. PEG was used as the marker substance because it is not absorbable in the intestinal tract. Consequently the concentration of PEG relates inversely to the volume of the loop. Instantaneous concentration of urea in the loop times the volume will give the amount of urea remaining in the luminal fluid. The change in the amount of any substance is directly relate to the volume of the compartment and differs from the change in the concentration which is independent of the volume. After completion of the experiment without red pepper, it was added in the test solution and was centrifuged after thorough mixing. Supernatant of the mixture was placed in the loop and similar sampling were performed with the same time intervals that of previous run in order to observe the effects of the red pepper on the passive transport of the water soluble small substance, urea. The results obtained were as follows: 1. Logarithm of the concentration difference of urea between the luminal fluid and plasma was diminished exponentially as time elapsed. The decay constant in the experiment without red pepper was 0.0563/min. By adding red pepper in the test solution as much as the concentration rose to 4,000 mg% and 8,000 mg%, the decay constants were lowered to 0.0493/min and to 0.0506/min, respectively. The time interval by which the concentration difference dropped to one half of the initial value was prolonged. Without red pepper the half concentration time was 13.30 minutes, and by adding extract of red pepper, 15.31 minutes and 15.71 minutes were revealed. 2. The profile of the diminishing rate of tile amount of urea was quite different from that of the concentration because of the change in the volume of the loop during the observed period. 3. By adding the extract of red pepper, it slowed down the rate of absorption of urea in the intestinal loop, suggesting an increase in the diffusional barrier. 4. Larger dosage of red pepper brought an increase in the secretion of intestinal fluid with concomitant expansion of the luminal volume, and the retardation of the absorption of urea was noticed. This effect was largely dependent on the sensitivity of the individual animal to the red pepper, extract. The amount of urea remained after 10 minutes interval was 55.5% of the initial amount in the experiment without red pepper. On the other hand it was not consistent after administration of red pepper, showing 50.6% and 66.5% of the initial figures by adding 400 mg and 800 mg of red pepper in the test solution, respectively. It was postulated that symptom of diarrhea often encountered by taking a hot (red pepper) food might be attributable to the increase of secretion and the retardation of absorption in the intestinal tract.

Treatment of N, P of Auto-Thermal Thermophilic Aerobic Digestion Filtrate with Struvite Crystallization (Struvite 결정화 반응을 이용한 고온 소화 여과액의 N, P 처리 특성)

  • Recently, auto-thermal thermophilic aerobic digestion (ATAD) has a great attention for destruction of wasted sludge biomass in wastewater treatment plant. Reduction of sludge concentration has been successfully achieved with pilot scale ATAD and ceramic filtration process in field condition. However, high concentration of COD, total nitrogen (TN) and total phosphorus (TP) was observed in filtrate, which should be treated before recirculation of filtrate to biological wastewater treatment plant. This study was focused on removal of nitrogen and phosphorus contained in the filtrate of ATAD, using struvite crystallization method. The effect of operational and environmental parameters (such as, N, P and Mg ion concentration and molar ratio, pH, reaction time, agitation strength, seed dosage, and reaction temperature) on the treatment of TN and TP with struvite crystallization were evaluated. Magnesium (as $MgCl_26H_2O$) and phosphorus (as $K_2HPO_4$) ions were, if necessary, added to increase nitrogen removal efficiency by the crystal formation. Average concentration of $NH_4^+-N$ and $PO_4^{3-}-P$ of the filtrate were 1716.5 mg/L and 325.5 mg/L, respectively. Relationship between removal efficiencies of nitrogen and phosphorus and molar ratios of $Mg^{2+}$ and $PO_4^{3-}-P$ to $NH_4^+-N$ was examined. Crystal formation and nitrogen removal efficiencies were significantly increased as increasing molar ratios of magnesium and phosphorus to nitrogen. As molar ratio of $Mg^{2+}:PO_4^{3-}-P:NH_4^+-N$ were maintained to 2 : 1 : 1 and 2 : 2 : 1, removal efficiencies of nitrogen and phosphorus were 71.6% and 99.9%, and 93.8% and 98.6%, respectively. However, the effect of reaction time, mixing intensity, seed dose and temperature on the struvite crystallization reaction was not significant, comparing to those of molar ratios. Settled sludge volume after struvite crystallization was observed to be reduced with increase of seed dose and to be increased at high temperature.

Studies on the Various Utilization of Microbial Formulation for the Production of Vegetable Crops (원예작물(園藝作物) 생산성(生産性)에 미치는 미생물(微生物) 제제(製劑)의 복합적(複合的) 이용연구(利用硏究))

  • The carrier materials used for the development of bacterial inoculants to be effective in field were made with various carrier materials of two major forms, alginate bead and powder inoculants. Inoculants were prepared after mixing those carrier materials with Pseudomonas fluorescens SSL3 and Bacillus subtilis B5, and the treatment effects of each inoculants was investigated on cucumber, tomato, pepper and potato. Survival density of SSL3 and B5 in various carrier materials for duration of storage and the bead inoculants were better than the powder. In the powders, survival rate increased in carrier materials treated 5% skimilk. The growth condition of microorganisms in carrier materials is good at powder. When they were preserved in the long period, contamination is problem. Scanning(200 to 600nm) of the P. fluorescens SSL3 supernatant in centrifuged MKB broth incubated for 48h had two main peaks, pyochelin(300nm) and pyoverdin(400nm). The potato yield in field experiments of spring, treated with bead formulas showed increase of 22~29% in whole potato breeds as compared with control, because the bead formulas degraded, and released the antibiotic microorganisms in slow and constant rate. In the pot experiment, there were significant difference in soil, wheatbran, and bead formed wheatbran.

  • Storing the surplus energy from renewable energy resource is one of the challenges related to intermittent and fluctuating nature of renewable energy electricity production. $CO_2$ methanation is well known reaction that as a renewable energy storage system. $CO_2$ methanation requires a catalyst to be active at relatively low temperatures ($250-500^{\circ}C$) and selectivity towards methane. In this study, the catalytic performance test was conducted using a pressurized bubbling fluidized bed reactor (Diameter: 0.025 m and Height: 0.35 m) with $Ni/{\gamma}-Al_2O_3$ (Ni70%, and ${\gamma}-Al_2O_3$30%) catalyst. The range of the reaction conditions were $H_2/CO_2$ mole ratio range of 4.0-6.0, temperature of $300-420^{\circ}C$, pressure of 1-9 bar, and gas velocity ($U_0/U_{mf}$) of 1-5. As the $H_2/CO_2$ mole ratio, temperature and pressure increased, $CO_2$ conversion increases at the experimental temperature range. However, $CO_2$ conversion decreases with increasing gas velocity due to poor mixing characteristics in the fluidized bed. The maximum $CO_2$ conversion of 99.6% was obtained with the operating condition as follows; $H_2/CO_2$ ratio of 5, temperature of $400^{\circ}C$, pressure of 9 bar, and $U_0/U_{mf}$ of 1.4-3.

Seasonal Variation of Water Quality in a Shallow Eutrophic Reservoir (얕은 부영양 저수지의 육수학적 특성-계절에 따른 수질변화)

  • This study was carried out to assess the seasonal variation of water quality and the effect of pollutant loading from watershed in a shallow eutrophic reservoir (Shingu reservoir) from November 2002 to February 2004, Stable thermocline which was greater than $1^{\circ}C$ per meter of the water depth formed in May, and low DO concentration (< 2 mg $O_2\;L^{-1}$) was observed in the hypolimnion from May to September, 2003. The ratio of euphotic depth to mixing depth ($Z_{eu}/Z_{m}$) ranged 0.2 ${\sim}$ 1.1, and the depth of the mixed layer exceeded that of the photic layer during study period, except for May when $Z_{eu}$ and $Z_{m}$ were 4 and 4.3 m, respectively. Most of total nitrogen, ranged 1.1 ${\sim}$ 4.5 ${\mu}g\;N\;L^{-1}$, accounted for inorganic nitrogen (Avg, 58.7%), and sharp increase of $NH_3$-N Hand $NO_3$-N was evident during the spring season. TP concentration in the water column ranged 43.9 ${\sim}$ 126.5 ${\mu}g\;P\;L^{-1}$, and the most of TP in the water column accounted for POP (Avg. 80%). During the study period, DIP concentration in the water column was &;lt 10 ${\mu}g\;P\;L^{-1}$ except for July and August when DIP concentration in the hypolimnion was 22.3 and 56.7 ${\mu}g\;P\;L^{-1}$, respectively. Increase of Chl. a concentration observed in July (99 ${\mu}g\;L^{-1}$) and November 2003 (109 ${\mu}g\;L^{-1}$) when P loading through two inflows was high, and showed close relationship with TP concentration (r = 0.55, P< 0.008, n = 22). Mean Chl. a concentration ranged from 13.5 to 84.5 mg $L^{-1}$ in the water column, and the lowest and highest concentration was observed in February 2004 (13.5 ${\pm}$ 1.0 ${\mu}g\;L^{-1}$) and November 2003 (84.5 ${\pm}$29.0 ${\mu}g\;L^{-1}$), respectively. TP concentration in inflow water increased with discharge (r = 0.69, P< 0.001), 40.5% of annual total P loading introduced in 25 July when there was heavy rainfall. Annual total P loading from watershed was 159.0 kg P $yr^{-1}$, and that of DIP loading was 126.3 kg P $yr^{-1}$ (77.7% of TP loading. The loading of TN (5.0ton yr-1) was 30 times higher than that of TP loading (159.0 kg P yr-1), and the 78% of TN was in the form of non-organic nitrogen, 3.9 ton $yr^{-1}$ in mass. P loading in Shingu reservoir was 1.6 g ${\cdot}$ $m^{-2}$ ${\cdot}$ $yr^{-1}$, which passed the excessive critical loading of Vollenweider-OECD critical loading model. The results of this study indicated that P loading from watershed was the major factor to cause eutrophication and temporal variation of water quality in Shingu reservoir Decrease by 71% in TP loading (159 kg $yr^{-1}$) is necessary for the improvement of mesotrophic level. The management of sediment where tine anaerobic condition was evident in summer, thus, the possibility of P release that can be utilized by existing algae, may also be considered.

A Study on the Useful Trend of Plants Related to Landscape and How to Plant and Cultivate Through 'ImwonGyeongjaeji(林園經濟志)' ('임원경제지'를 통해 본 식물의 이용경향과 종예법(種藝法))

  • The result of a study on the useful trend of plants related to landscape and how to plant and cultivate through 'ImwonGyeongjaeji Manhakji'of Seoyugu is as follows: First, 'ImwonGyeongjaiji Manhakji', composed of total 5 volumes (General, Fruit trees, vegetables and creeper, plants, others) is a representative literature related to landscape which described the names of plants and varieties, soil condition, how to plant and cultivate, graft, how to prevent the insect attack etc systematically. Second, he recorded the tree planting as Jongjae(種栽) or Jaesik(栽植), and the period to plant the trees as Jaesusihoo(栽樹時候), transplanting as Yijae(移栽), making the fence as Jakwonri(作園籬), the names of varietieis as Myeongpoom(名品), the suitable soil as Toeui(土宜), planting and cultivation as Jongye(種藝), treatment as Euichi(醫治), protection and breeding as Hoyang(護養), garden as Jeongwon(庭園) or Wonpo(園圃), garden manager as Poja(圃者) or Wonjeong(園丁). Third, the appearance frequency of plants was analyzed in the order of flowers, fruits, trees, and creepers and it showed that the gravity of deciduous trees was 3.7 times higher than that of evergreen trees. The preference of flower and trees, fruit trees and deciduous trees and broad-leaved trees includes (1) application of the species of naturally growing trees which are harmonized with the natural environment (2) Aesthetic value which enables to enjoy the beauty of season, (3) the trend of public welfare to take the flowers and fruits, (4) the use of symbolic elements based on the value reference of Neo-Confucianism etc. Fourth, he suggested the optimal planting period as January(上時) and emphasized to transplant by adding lots of fertile soil and cover up the seeds with soil as high as they are buried in accordance with the growing direction and protect them with a support. That is, considering the fact that he described the optimal planting period as January by lunar calendar, this suggests the hints in judging the planting period today. For planting the seeds, he recommended the depth with 1 chi(寸 : approx. 3.3cm), and for planting a cutting, he recommended to plant the finger-thick branch with depth 5 chi(approx. 16.5cm) between January and February. In case of graft of fruit trees, he described that if used the branch stretched to the south, you would get a lot of fruit and if cut the branches in January, the fruits would be appetizing and bigger. Fifth, the hedge(fence tree) is made by seeding the Jujube tree(Zizyphus jujuba var. inermis) in autumn densely and transplanting the jujube tree with 1 ja(尺 : approx. 30cm) interval in a row in next autumn and then binding them with the height of 7 ja(approx. 210cm) in the spring of next year. If planted by mixing a Elm tree(Ulmus davidiana var. japonica) and a Willow(Salix koreensis), the hedge whose branch and leaves are unique and beautiful like a grating can be made.
