수소 생성능이 있는 Clostridium butyricum과 이 균주의 대사산물을 이용하여 수소를 생성할 수 있는 광합성 세균의 혼합 배양에 의하여 수소 생성을 할 수 있는 양 균주의 혼합 배양 동력학적 연구를 수행하였다. 혐기성 Clostridia의 발효에서 생성되는 아세트산염과 부티르산염은 Rhodopseudomonas sphaeroides에 의해 이용되어 수소를 생성하게 되었고, 이들 양 균주를 혼합 배양한 결과 수소 생성을 저해하는 아세트산염과 부티르산염의 배지내 농도는 점차 감소하였다. 이들 두 균주의 혼합 배양에서의 균체의 혼합 비율, 혼합시기, 접종량을 조사함으로써 단독 배양시와 비교하면서 최적 조건을 조사하였고, 혼합시 균체량의 비율 Clostridium 속 세균1부에 대하여 Rhodopseudomonas 속 세균 6부의 비율로 혼합할 수 있음을 확인하였다.
본 연구에서는 bacterial cellulose를 생산하는 감귤에서 분리된 Gluconacetobacter sp. gel_SEA623-2를 이용하여 초기 초산농도 및 Lactobacillus plantarum KCCM 80077 유산균을 접종한 mixed culture가 bacterial cellulose 생산성에 미치는 영향을 알아보았다. 0.5%의 초산을 첨가한 mixed culture의 경우 초기 생균수가 $2.4{\times}10^6CFU/ml$에서 14일 동안 배양한 이후에 $1.1{\times}10^7CFU/ml$까지 증가하였고 total acidity가 0.5%의 초산을 첨가한 single culture와 비교하여 0.3% 가량 높게 측정되어 유산균에 의해 추가적으로 젖산이 생성된 것으로 판단된다. Single culture에서는 초기 초산농도가 0.0 및 0.5%일 때 1.0% 농도와 비교하여 상대적으로 높은 bacterial cellulose 생산성을 나타내기는 하였으나 유의적인 차이는 보이지 못하였다. 하지만 mixed culture에서는 0.5%의 초산을 첨가하였을 때 $37.83{\pm}6.81g/L$의 dry weight과 $10.33{\pm}0.58mm$의 thickness를 나타냄으로써 1%의 초산을 첨가한 single culture의 $28.40{\pm}1.23g/L$와 $7.50{\pm}0.50mm$의 생산량과 비교하여 유의적인 차이를 나타내었다(P<0.05).
The objective of this work was to study the effect of mixed culture solid substrate fermentation of C. tropicalis ZD-3 with A. niger ZD-8 on detoxification of cottonseed meal (CSM), and to investigate the effect of fermentation period, proportion of CSM in substrate, sodium carbonate, minerals and heat treatment on the reduction of free gossypol levels during mixed culture solid substrate fermentation of CSM. Experiment 1: Three groups of disinfected CSM substrate were incubated for 48 h after inoculation with either of the fungi C. tropicalis ZD-3, A. niger ZD-8 or mixed culture (C. tropicalis ZD-3 with A. niger ZD-8). One non-inoculated group was used as the control. Levels of initial and final free gossypol (FG), CP and in vitro CP digestibility were assayed. The results indicated that mixed culture fermentation was far more effective than single strain fermentation, which not only had higher detoxification rate, but also had higher CP content and in vitro digestibility. Experiment 2: CSM substrates were treated according to experimental variables including fermentation period, proportion of CSM in substrate, sodium carbonate, minerals and heat treatment, Then, the treated CSM substrates were inoculated with mixed culture (C. tropicalis ZD-3 with A. niger ZD-8) and incubated at $30^{\circ}C$ for 36 h in a 95% relative humidity chamber. After fermentation ended, FG and CP content of fermented CSM substrate was assayed. The results showed that the appropriate fermentation period was 36 h, and the optimal proportion of CSM in substrate was 70%. Addition of sodium carbonate to CSM substrate was beneficial for fermentative detoxification. Heat treatment could facilitate fermentative detoxification, and supplementation with minerals was instrumental in reducing gossypol levels during mixed culture solid substrate fermentation of CSM.
Objectives: Recently lactic acid formulation was known as the adjuvant therapy on atopic dermatitis(AD) symptoms because of little side effects. In this study, it was studied that Lactobacillus mixed culture appliment was effective on not on AD symptoms. Methods: The case-photos 30-40 times of AD symptoms from case were observed with literatures. The case-photos were acquired with environmental AD dermatitis experience program which is conducted in SUNCHANG RESERCH INSTITUTE OF HEALTH AND LONGEVITY from 2014.01 to 2014.08. Experience applicants were limited the oral administration and chemical external administration. Results: Lactobacillus mixed culture appliment was effective because of the mitigation or disappearance of AD primary symptoms like pruritus, erythema, edema, effusion, dry skin, scaly, scab etc. AD symptom degree was dependent on lactobacillus mixed culture appliment times, and classified as Reaction Period (RAP), Reduction Period (RDP), Efection Period (EP), Reproduction Period (RPP) on a single mixed culture appliment time. And AD symptoms which were appeared in RPP were classified as Rebound Period (RBP), Effection Period (EP), Subclinical Period (SCP). Conclusions: Lactobacillus mixed culture appliment is considered to be useful for AD symptoms and is estimated to be effective by different mechanism with oral administration or steroid ointment application. Lactobacillus mixed culture appliment is expected to induce a synergistic effects on AD symptom reliefs with the other oral administration.
Protein productivity by the cellulolytic fungi, Trichoderma viride(MTCC 800), Chaetomium globosum and Aspergillus terreus was compared in co-culture and mixed culture fermentations of cashewnut bran. Co-cultures were more effective in substrate saccharification, which ranged between $85{\sim}88%$ compared to the $62{\sim}67%$ saccharification shown by the monocultures. Maximum saccharification was induced by T. viride and C. globosum co-culture resulting in the highest 34% release of reducing sugars. The maximum 16.4% biomass protein and the highest protein productivity(0.58%) were shown by T. viride and A. terreus co-culture. A. terreus performed better in co-culture in the presence of T. viride rather than with C. globosum. Among the cellulolytic enzymes, FPase(Filter Paper Cellulase) activity was significantly higher in all the co-cultures and in the mixed culture than in their respective monocultures. Mixed culture fermentation involving all the three fungi was not effective in increasing the per cent saccharification or the biomass protein content over the co-cultures.
When cultivated aerobically, Aspergillus niger hyphae produced extracellular glucoamylase, which catalyzes the saccharification of unliquified potato starch into glucose, but not when grown under anaerobic conditions. The $K_m\;and\;V_{max}$ of the extracellular glucoamylase were 652.3 mg/l of starch and 253.3 mg/l/min of glucose, respectively. In mixed culture of A. niger and Saccharomyces cerevisiae, oxygen had a negative influence on the alcohol fermentation of yeast, but activated fungal growth. Therefore, oxygen is a critical factor for ethanol production in the mixed culture, and its generation through electrolysis of water in an electrochemical bioreactor needs to be optimized for ethanol production from starch by coculture of fungal hyphae and yeast cells. By applying pulsed electric fields (PEF) into the electrochemical bioreactor, ethanol production from starch improved significantly: Ethanol produced from 50 g/l potato starch by a mixed culture of A. niger and S. cerevisiae was about 5 g/l in a conventional bioreactor, but was 9 g/l in 5 volts of PEF and about 19 g/l in 4 volts of PEF for 5 days.
The differential enhanced degradation of cis- and trans-1,3-D was observed in the previous two studies performed by Ou et al. (1995) and especially Chung et al. (1999). This study was initiated to investigate the involvement of microorganisms in the differential enhanced degradation of the chemicals. As expected, microorganisms were responsible for the enhanced degradation of the chemicals. A mixed bacterial culture capable of degrading 1,3-D was isolated from an enhanced soil sample collected from a site treated with 1,3-D. Similar to the enhanced soil, the mixed culture degraded trans-1,3-D faster than cis-1,3-D. This mixed culture could not utilize cis- and trans-1,3-D as a sole source of carbon for growth. Rather, a variety of second substrates were evaluated to stimulate the differential enhanced degradation of the two isomers. As a result, the mixed culture degraded cis- and trans-1,3-D only in the presence of a suitable second substrate. Second substrates that had the capacity to stimulate the degradation included soil leachate, tryptone, tryptophan, and alanine. Other substrates tested, including soil extract, glucose, yeast extract, and indole (ailed to stimulate the degradation of the two isomers. Therefore, it appeared that the degradation of cis- and trans-1,3-D was a cometabolic process. The mixed culture was composed of four morphologically distinctive bacterial colonies.
복합미생물제재를 사용하여 인공폐수에서 염소화 페놀화합물의 생물학적 분해를 조사하기 위하여 다양한 염소화 페놀화합물로 오염된 토양에서 8종의 미생물을 분리하였다. 복합미생물제재의 주된 미생물인 Pseudomonas sp. BM은 Gram-negative, catalase- 그리고 oxidase-positive, rod 형이며 $41^{\circ}C$ 이상에서는 성장하지 않았다. Pseudomonas sp. BM은 단일 탄소원으로서의 PCP(pentachlorophenol) 500mg/l의 농도로 첨가된 최소배지에서 배양 3일 후에 99%의 분해효율을 보였다. 복합미생물제재에 여러 가지 종류의 유기탄소 및 질소원을 첨가한 후 PCP의 분해효율을 관찰하였을 때 큰 차이를 나타내지 못하였다. Pseudomonas sp. BM은 pH 5에서 8까지 넓은 적용범위를 가지고 있으나 pH 7에서 가장 좋은 활성을 나타내었다. 종류가 다른 염소화 페놀을 함유한 인공폐수에서 염소화 페놀화합물의 분해시험은 7일간 배양후 최저 분해효율 73%(2,4-DCP)에서 최대 96%(2-CP)까지 비교적 높은 활성을 보였다. 연속배양 실험에서, 활성 슬러지와 복합미생물제재를 첨가한 것의 PCP 분해효율을 비교해 보았을 때 활성 슬러지만을 첨가한 대조구에 비해 복합미생물제재를 첨가하였을 때는 약 2배의 효과를 나타내었다. 이와 같은 결과는 다양한 염소화 페놀이 함유된 폐수에 복합미생물제재의 적용이 가능함을 나타낸다.
This study was conducted to evaluate the influence of Lactobacillus bulgaricus and/or Sterptococcus thermophilus on the fermentation of yoghurt containing 5% (w/v) buckwheat sprouts. The results revealed that after 12 hours of fermentation the appropriate pH, titratable acidity and number of viable cells were attained. At that time, the rutin content of the buckwheat sprout-added yoghurt prepared by the mixed culture method was not changed, but the quercetin content increased greatly. Conversely, the rutin content of yoghurt that only contained Streptococcus thermophilus was decreased while the quercetin content was increased. The total phenol contents as well as the DPPH radical scavenging activities of both the mixed culture and Streptococcus thermophilus yoghurt did not differ significantly. Taken together, the results revealed that the use of a mixed culture of Lactobacillus bulgaricus and Streptococcus thermophilus during the preparation of buchwheat sprout-added yoghurt was desirable due to the decrease in pH and increase in titratable acidity and viable cells that occurred after 12 hr of fermentation. Moreover, phytochemicals in buckwheat sprouts such as rutin, quercetin and phenol compounds were comparatively increased during fermentation and influenced the antioxidant activity in buckwheat sprout-added yoghurt.
분리대두단백을 단백분해효소로 처리하여 가수분해시킨 후 B. bifidum으로 단독배양하거나 B. bifidum과 L. acidophilus 또는 B. bifidum과 L. bulgaricus로 혼합배양하여 frozen soy yogurt를 제조한 다음 그 품질특성 및 관능적 특성을 측정하였다. 혼합배양시에 단독배양보다 생균수는 더 많았고 담즙산 내성은 더 우수하였으나 내산성은 큰 차이가 없었다. 그러나 ${\alpha}-chymotrypsin$으로 처리하고 B. bifidum과 L. bulgaricus 혼합배양한 시료가 비교적 내산성 및 담즙산 내성이 우수하였다. 시료의 overrun은 B. bifidum과 L. bulgaricus 혼합배양시에 ${\alpha}-chymotrypsin$ 처리군에서 가장 높았으며 아이스크림 제조기에서 30분 조작으로 73.45%를 형성하였다. ${\alpha}-chymotrypsin$ 처리군을 제외한 모든 시료에 있어서 단독배양시에 혼합배양보다 녹아내리는 정도가 더 컸다. 시료의 관능평가 결과 bromelain 처리군과 ${\alpha}-chymotrypsin$ 처리군의 경우 B. bifidum과 L. bulgaricus 혼합배양시에 단독배양보다 콩 비린내와 쓴맛은 적었고 신맛은 더 좋았으며 전반적으로 바람직하다고 평가되었다. 특히 ${\alpha}-chymotrypsin$ 처리군의 B. bifidum과 L. bulgaricus 혼합배양시에 신맛이 좋았고 쓰지 않았으며 떫지 않았다고 평가되어 가장 바람직하다고 평가되었다.
