DETERMINATION OF THE APPARENT ILEAL DIGESTIBILITY OF PROTEIN AND AMINO ACIDS IN FEEDSTUFFS AND MIXED DIETS FOR GROWIG-FINSHING PIGS WITH THE MOBILE NYLON BAG TECHNIQUE

  • A series of experiments was conducted to determine the influence of various pepsin-HCL pretreatment factor, hereby the factors of duration of washing for the retrieved bags, inherent to the mobile nylon bag technique (MNBT), on apparent ileal digestibility of crude protein (AIDCP) and apparent ileal digestibility of dry matter (AIDDM). At last, the AIDCP and apparent ileal digestibility of amino acids (AIDAA) in maize, barley, wheat, rapeseed meal, cottonseed meal and three mixed diets were determined with the MNBT and ileo-rectal anastomis pigs (IRAT). For the MNBT techniques, bag measuring $25{\times}40$ MM and containing 0.75 g feedstuff samples, after pre-digestion in vitro, were introduced into the ileo-rectal anastomis pigs (IRAT) gastrointestinal tract through a duodenal cannula and recovered in the ileal digesta between 6 and 12 h. later. 1. The apparent ileal digestibility of dry matter (AIDDM) and crude protein (AIDCP) of the tested samples, with the exception of fish meal, determined by MNBT were not affected by the different pepsin-HCL pretreatment times in vitro between 2.5 h. and 4 h. 2. There was no significant (p > 0.05) difference of the AIDCP and AIDDM of maize determined by the MNBT among different pepsin concentration (0.03%, 0.07% and 0.1 %) treatment in vitro. 3. The AIDCP determined with the MNBT was affected by the washed and unwashed recovered bags from the ileal digesta. 4. The AIDCP and AID amino acids (AIDAA) of maize, barley, wheat, rapeseed meal, soya-bean meal, cottonseed meal and three mixed diets from the MNBT, with a solution of 0.01N HCL (PH 2) and 0.1% of pepsin concentration, a pepsin-HCL pretreatment time in vitro or 4h. and a washing time of the recovered bag from the ileal digesta compared well with those from the IRAT. The linear regression analysis showed a significant correlation (p < 0.01) of AIDCP and AIDDA between the IRAT and MNBT.

혼합 생균제 급여가 한국 토종닭의 생산성과 가슴살의 지방산 조성에 미치는 영향 (Growth Performance and Fatty Acid Profiles of Korean Native Chicken Fed Diets with Mixed Probiotics)

  • This study aimed to investigate the effects of dietary supplementation of mixed probiotics on growth performance and fatty acid profiles of Korean native chicken and to provide information regarding producers. Ninety six Korean native chicken (48 males and 48 females, Hanhyup-3-ho) were allocated to 16 wire cages with 4 treatments, 4 replicates and 6 chicks (3 males and 3 females) per wire cage and fed one of four diets containing 0, 0.5, 1 and 1.5% mixed probiotics for 6 weeks. There were no differences among treatments in growth performance of Korean native chicken. For fatty acid profiles, no statistically differences in each fatty acid, total saturated fatty acid and unsaturated fatty acids were observed in all treatments. In the present study, irrespective of statistically differences, inclusion of mixed probiotics tended to improve growth performance, reduce saturated fatty acid and increase unsaturated fatty acids in breast muscles from Korean native chicken compared to controls.

곰팡이 균종을 달리하여 제조한 혼합 곡류 누룩의 품질특성 (Assessment of the Quality Characteristics of Mixed-grain Nuruk Made with Different Fungal Strains)

  • 밀, 쌀 및 녹두를 분쇄 후 밀기울을 첨가하여 일정 비율(15:1:1:3)로 혼합한 곡물에 양조용 곰팡이의 균종을 달리하여 제조한 누룩의 품질 특성을 알아보고자 발효 시기별 일반성분 및 효소활성과 유기산 분석을 하였다. 누룩의 pH는 발효 시기별 큰 차이는 보이지 않았으며, 산도와 아미노산도는 두균을 혼합하여 제조한 누룩(AO-AK)이 가장 높게 나타났다. 전반적으로 두 균을 혼합한 누룩이 단일 균을 접종한 누룩보다 ${\alpha}$-amylase 및 acidic protease의 효소활성이 높게 나타났다. 누룩의 유기산은 acetic, citric, formic, fumaric, lactic, malic 및 oxalic acid 등이 확인되었다. 전체 유기산 총량은 혼용누룩(2,116.3 mg%), A. kawachii SC60 단용누룩(1,608.5 mg%), A. oryzae RIB1353 단용누룩(1,146.7 mg%) 순으로 혼용누룩의 유기산 생성량이 가장 높게 나타났다. 따라서 두 균주를 혼용하여 제조한 누룩이 단일균주를 사용한 누룩보다 유기산 함량, 효소활성 등 품질 특성이 우수한 것으로 여겨진다.

  • The application of Consomm prepared with different ingredients of ground beef and ground chicken ingredients to improve the taste preferences and reduce saving the food cost of Consomm was attempted by reviewing fundamental data and its examining the effect on the physicochemical characteristics of treatment. These treatments were prepared with the following different ratios (9:1, 7:3, 5:5, 3:7, 1:9 ; w/w) of ground beef to and ground chicken: 9:1, 7:3, 5:5, 3:7, and 1:9, w/w, respectively. Gelatin contents were increased significantly with by increasing the mixed ratio of ground chicken. The free sugars identified from Consomm were, glucose, fructose, and sucrose. Free sugar contents, increased significantly by with increasing the mixed ratio of ground beef. (Ed- what are the highlighted treatments? As many as 20 different kinds of free amino acids were detected infrom the Consomm and the amount of total amino acids waswere increased with by increasing the mixed ratio of ground chicken. The contents of arginine, glutamic acid and alanine contents were high in the free amino acids of Cconsomm prepared with different ingredients. The In the changes of mineral contents in the Consomm prepared with different ingredients, the mineral showed high contents increased according to in the following order of: K, Na, P, Mg, Ca, Fe, and Zn. From the The results of analysis results, in the Relatin contents, free sugars, free amino acids and mineral contents were all increased with by increasing the mixed ratio of ground chicken. The 7:3 treatment showed From above results, it was showed that B treatment prepared with a ratio of 1 ground beef to 3 ground chicken was the bestmost preferred in physicochemical qualities.

Effects of Halogenated Compounds, Organic Acids and Unsaturated Fatty Acids on In vitro Methane Production and Fermentation Characteristics

  • The objective of this study was to evaluate the effects of halogenated compounds, organic acids, unsaturated fatty acids and their mixtures on in vitro methane production and fermentative characteristics of mixed rumen microorganisms. Agents used in two in vitro experiments were bromoethanesulfonic acid (BES) and pyromellitic diimide (PMDI) as halogenated compound, fumarate and malate as organic acid, and linoleic acid and linolenic acid as unsaturated fatty acid sources. Ruminal fluid collected from a Holstein steer fed tall fescue and concentrate mixtures was incubated at $39^{\circ}C$ for 48 h with addition of those materials. Single supplementation of halogenated compounds, organic acids or unsaturated fatty acids decreased in vitro methane production (p<0.05). The second experiment was designed to investigate effects of combination of one of halogenated compounds and either organic acids or fatty acids on methane production. Lower concentration of methane and lower A:P ratio were observed with PMDI compared with BES (p<0.01). In general medium pH, VFA, total gas and hydrogen production, and dry matter degradability were affected by addition of the same compounds. In addition, PMDI+malate treatment resulted in the highest molar proportion of propionate, and lowest A:P ratio and methane production (p<0.01). Hydrogen production was highest in PMDI+linolenic acid and lowest in BES+malate treatment (p<0.01). PMDI+malate combination was the most recommendable in reducing methane production without too much influence on digestibility under conditions of present studies.

Effects of Mixed Bone and Brisket Meat on Physico-Chemical Characteristics of Shank Bone and Rib Extracts from Hanwoo

  • This study was conducted to investigate the effects of mixed bone and brisket meat on the quality characteristics and nutritional components of shank bone extract and rib extract from Hanwoo. The pH values were influenced by the raw bones, mixed bone, brisket meat and their interactions (p<0.05). The salinity, sugar content, turbidity, and essential amino acid values increased significantly with addition of mixed bone and brisket meat. All attributes of sensory evaluation score were the highest in T6 (Rib 500 g + Mixed bone 500 g + Brisket meat 400 g) (p<0.05). The mixed bone significantly increased the saturated fatty acids of shank bone extract (p<0.001). Thus, the addition of mixed bone and brisket meat had a positive effect on the quality and nutritional components in shank and rib extracts of Hanwoo cattle.

동해의 용존유기물 형광특성 및 아미노산 조성에 관한 연구 (Characteristics of Fluorescent Organic Matter and Amino Acids Composition in the East Sea)

  • 동해의 8개 정점에서 해수 및 추출된 용존 유기물의 형광특성과 아미노산 조성이 연구되었다. C-18 Sep-Pak cartridge에 의해 추출된 시료는 3차원 형광특성 분석에 따 라 생거대물질과 지구거대물질로 구분되었다. 전 조사 정점을 통하여 생거대물질(ex : 280 nm/em : 330 nm)은 표층이 높고 수온약층 아래에서 점차 감소하는 것으로 나타났 으며 이는 표층혼합층의 생물 활동에서 기인된 분해가능한 생거대물질이 수온약층 부 근 및 저층에서 활발한 미생물 분해과정에 의해 감소하는 것으로 사료된다. 한편 이와 는 역상관계를 보이는 지구거대물질 (ex : 330 nm/em : 430 nm)은 표층은 낮고 수온약 층 아래에서 증가하였는데 이는 표층에서 생성된 생거대물질 및 입자유기체가 생물 분 해 후 재축합 과정을 거쳐 난분해성의 지구거대물질로 전환된 것으로 사료된다. HPLC 를 이용하여 해수와 추출된 유존유기물의 아미노산 조성을 분석하였다. 분석결과 Glycine, serine 그리고 alanine등이 우점하였으며, 전체 농도의 50% 이상을 차지하는 것으로 조사되었다. 해수중의 용존 자유아미노산 농도는 표층이 0.7∼1.8um 범위로 저 층 0.2∼0.4um보다 높게 측정되었다. 추출된 유기물중 alanine의 D/L racemice ratio 측정결과 저층보다 표층이 상대적으로 낮은 값을 보였으며 이는 표층의 생거대물질이 연령이 젊고 재순환이 빠르며 생물 분해가능성이 큰 물질임을 시사하고 있다.

  • Paper Chromatography 법(法)으로 아미노산류(酸類)와 이노신산(酸) 및 수은중독(水銀中毒) 치료제(治療劑)인 BAL의 수은(水銀)에 대(對)한 친화력(親和力)의 크기를 비교(比較)하였다. 그 결과(結果) Cysteine Cystine 및 Methionine의 3종(種)의 합류황(合硫黃) 아미노산(酸)은 수은(水銀)과 쉽게 결합(結合)하였으나 다른 아미노산(酸)들과의 결합(結合)은 확인(確認)하지 못하였다. 수은(水銀)은 SH기(基)를 가지는 Sodium thiosulfate 및 BAL과는 쉽게 결합(結合)하였으며 이노신산(酸)과도 쉽게 결합(結合)하였다. 이들 함류황(含硫黃) 아미노산(酸) 류(類)보다 큰 것으로 판단(判斷)되었다.

Effects of Protease-resistant Antimicrobial Substances Produced by Lactic Acid Bacteria on Rumen Methanogenesis

  • Effects of protease-resistant antimicrobial substances (PRA) produced by Lactobacillus plantarum and Leuconostoc citreum on rumen methanogenesis were examined using the in vitro continuous methane quantification system. Four different strains of lactic acid bacteria, i) Lactococcus lactis ATCC19435 (Control, non-antibacterial substances), ii) Lactococcus lactis NCIMB702054 (Nisin-Z), iii) Lactobacillus plantarum TUA1490L (PRA-1), and iv) Leuconostoc citreum JCM9698 (PRA-2) were individually cultured in GYEKP medium. An 80 ml aliquot of each supernatant was inoculated into phosphate-buffered rumen fluid. PRA-1 remarkably decreased cumulative methane production, though propionate, butyrate and ammonia N decreased. For PRA-2, there were no effects on $CH_4$ and $CO_2$ production and fermentation characteristics in mixed rumen cultures. The results suggested that PRA-1 reduced the number of methanogens or inhibited utilization of hydrogen in rumen fermentation.

  • Peptides are formed in the rumen as the result of microbial proteinase activity. The predominant type of activity is cysteine ptoteinase, but others, such as serine proteinases, are also present. Many species of protozoa, bacteria and fungi are involved in ptoteolysis; large animal-to-animal variability is found when proteinase activities in different animals are compared. The peptides formed from proteolysis are broken down to amino acids by peptidases. Different peptides are broken down at different rates, depending on their chemical composition and particularly their N-terminal structure. Indeed, chemical addition to the N-terminus of small peptides, such as by acetylation, causes the peptides to become stable to breakdown by the rumen microbial population; the microorganisms do not appear to adapt to hydrolyse acetylated peptides even after several weeks exposure to dietary acetylated peptides, and the amino acids present in acetylated peptides are absorbed from the small intestine. The amino acids present in some acetylated peptides remain available in nutritional trials with rats, but the nutritive value of the whole amino acid mixture is decreased by acetylation. The genus Prevotella is responsible for most of the catabolic peptidase activity in the rumen, via its dipeptidyl peptidase activities, which release dipeptides rather than free amino acids from the N-terminus of oligopeptides. Studies with dipeptidyl peptidase mutants of Prevotella suggest that it may be possible to slow the rate of peptide hydrolysis by the mixed rumen microbial population by inhibiting dipeptidyl peptidase activity of Prevotella or the rate of peptide uptake by this genus. Peptides and amino acids also stimulate the growth of rumen microorganisms, and are necessary for optimal growth rates of many species growing on tapidly fermented substrates; in rich medium, most bacteria use pre-formed amino acids for more than 90% of their amino acid requirements. Cellulolytic species are exceptional in this respect, but they still incorporate about half of their cell N from pre-formed amino acids in rich medium. However, the extent to which bacteria use ammonia vs. peptides and amino acids for protein synthesis also depends on the concentrations of each, such that preformed amino acids and peptides are probably used to a much lesser extent in vivo than many in vitro experiments might suggest.
