Chemical Investigations and Anti-inflammatory Activity of Fixed Oil of Butea monosperma Seeds

  • The fruit and seeds of Butea monosperma (Lam) Kuntze (Fabaceae) are useful in piles, anthelmintic, eye diseases, and inflammation in the Indian system of medicine. Hence, we have evaluated the anti-inflammatory activity of the fixed oil, mixed fatty acids, and unsaponifiable matter of B. monosperma against carrageenan-induced paw oedema and cotton pellet-induced granuloma in rats. The fixed oil, mixed fatty acids, and unsaponifiable matter of the oil exhibited significant anti-inflammatory activity on the tested experimental animal models. The unsaponifiable matter of the oil produced higher protection compared to fixed oil and mixed fatty acids. Phytochemical analysis of the fixed oil revealed the presence of steroids and terpenoids while unsaponifiable matter of the oil showed the presence of ${\beta}-sitosterol$. Also, four fatty acids were identified in the fixed oil by gas liquid chromatography. The anti-inflammatory activity of the fixed oil may be due to unsaponifiable matter or combination of unsaponifiable matter and mixed fatty acids.

  • Studies were carried out to investigate physicochemical properties of taro during the pretreatment process of making Toranbyung. Seventeen kinds of amino acids were found in taro. Aspartic acid and glutamic acid were most plentiful among those amino acids. Essential amino acids were about 42.7% of the total amino acids. Sulfur containing amino acids, mehtionine and cystine were about 2.7% The amylograph indicated that the initial temperature triggering gelatinization of both rice flour mixed with taro and glutinous rice flour mixed with taro increased as the amount of taro increased. But the maximum viscosity of rice flour mixed with taro did not increase as the amount of taroincreased while the maximum viscosity of glutinous rice flour mixed with taro increased as the amount of taro increased. And the peak temperature of both rice flour mixed with taro and glutinous rice flour miced with taro increased as the amount of taro increased. The alkali degree of insoluble ash was higher than that of soluble ash. Glutinous rice dough had more total sugar than rice dough. Yellowness of rice dough was higher than that of glutinous rice dough.

  • The male rats after weaning were fed with the mixed diets of rice and some cereals for three weeks in a ad-libitum method. The growthgain of rats were determined by feeding those diets and the contents of free basic amino acids level in plasma were analyzed by amino acid analyzer. The results were as follows; 1. Rice diets group was highest in growthgain and weightgain, the second was the mixed of 80% rice-20% barley, and the last was the 80% rice-20% wheat group. 2. It was similar in the contents of plasma free basic amino acids of every diet group. The contents of Lys was highest and Arg, His were low in order. The mixed diet of 80% rice-20% barley group was higher than the rice only diet group in the contents of Lys and His. but rice only group was highest in Arg. The mixed of 80% rice-20% wheat diet group was lowest in the contents of Lys, His, Arg. Therefore feeding mixed diets of rice and cereals. the contents of Lys was highest, the second was Arg and the last was His in the plasma free basic amino acids level.

  • Influences of plant phenolic acids and their possible metabolites(non-phenolic aromatic acids involved) in the rumen on the cellulolytic activity of mixed rumen populations were examined by a simple in vitro culture technique. Initial concentrations of aromatic acids were 1, 5, 10 and 20 mM/l. All the tested aromatic acids reduced microbial cellulose digestion especially at the higher initial concentration. P-Coumaric acid, ferulic acid and cinnamic acid, those having unhydrogenated propenoic side chain were more inhibitory than were 3-phenylpropinic acid and phloretic acid, those having hydrogenated propanoic side chain. Lag-time for cellulose digestion was prolonged by former three acids by 16 h. Apparent reduction in p-coumaric acid concentration was observed at 24 h when cellulose digestion began. Volatile fatty acid productions from cellulose fermentation were shifted by former three aromatic acids to produce more acetate and less propionate. This suggests that the selection of celluloytic organisms was induced by these aromatic acids.

  • This experiment was conducted to evaluate the effects of supplemental unsaturated fatty acids (UFA) on fermentation characteristics, especially on gas production, cellulose degradation and volatile fatty acid (VFA) concentration by mixed ruminal microorganisms. In order to attain this objective, unsaturated fatty acids including oleic acid (C 18:1), linoleic acid (C18:2) and arachidonic acid (C22:4) were added at varying level. Mixed ruminal microbes used in this experiment were obtained from the rumen of a cannulated Holstein cow. Medium pH values after 7 d incubation were significantly affected by type and level of unsaturated fatty acids (p<0.01). All of UFA inhibited total gas production, and especially treatment of arachidonic acid at the levels of 0.01% gave the lowest gas. production after 7 d incubation (p<0.01). Comparison of the population of protozoa revealed that UFA did not have any significant effect on the total protozoa number. The addition of UFA did not effect dry matter degradation. Volatile fatty acid (VFA) composition of the culture was influenced little by UFA, although the considerable amount of iso-type VFA were detected in UFA supplemented incubations. The ratio of acetic acids to propionic acids, however, was lower than control in all the treatments after 7 d incubation (p<0.01).

  • When L, bulgurzcus KFCC 35462 and S. uvarum KFCC 32021 were mixed cultured in soymilk, gorwth of L. bulgaricus was stimulated with growth of S. uvarunz little affected. Cultured in soymilk singly, L. bulgaricus produced only 0.07%) of acids. S. uvarum, cultured singly, produced 0.40% of ethanol and 0.18% of acids. Mixed culture of both produced 0.27% of acids and 0.42% of ethanol. Thus. S. uvarurrz was not much affected in growth and ethanol fermentation by L. bulgariccus, while L, bulgarzcus was stimulated by Sacch. uvarunz in growth but not in acid fermentation.

  • The hydroxide precipitation method is appropriate to distinguish free metal ions with complexed metal ions with amino acids. Optimum pH conditions of hydroxide precipitation were investigated using mixed amino acids which have similar composition ratio with hydrolyzed amino acids. When applied to soil samples immobilities of Hg, Cr, and Cu ion with mixed and hydrolyzed amino acids were reasonable. But those of Cd and Zn were not sufficient.

Stability Test and Quantitative and Qualitative Analyses of the Amino Acids in Pharmacopuncture Extracted from Scolopendra subspinipes mutilans

  • Objectives: Scolopendra subspinipes mutilans (S. subspinipes mutilans) is known as a traditional medicine and includes various amino acids, peptides and proteins. The amino acids in the pharmacopuncture extracted from S. subspinipes mutilans by using derivatization methods were analyzed quantitatively and qualitatively by using high performance liquid chromatography (HPLC) over a 12 month period to confirm its stability. Methods: Amino acids of pharmacopuncture extracted from S. subspinipes mutilans were derived by using O-phthaldialdehyde (OPA) & 9-fluorenyl methoxy carbonyl chloride (FMOC) reagent and were analyzed using HPLC. The amino acids were detected by using a diode array detector (DAD) and a fluorescence detector (FLD) to compare a mixed amino acid standard (STD) to the pharmacopuncture from centipedes. The stability tests on the pharmacopuncture from centipedes were done using HPLC for three conditions: a room temperature test chamber, an acceleration test chamber, and a cold test chamber. Results: The pharmacopuncture from centipedes was prepared by using the method of the Korean Pharmacopuncture Institute (KPI) and through quantitative analyses was shown to contain 9 amino acids of the 16 amino acids in the mixed amino acid STD. The amounts of the amino acids in the pharmacopuncture from centipedes were 34.37 ppm of aspartate, 123.72 ppm of arginine, 170.63 ppm of alanine, 59.55 ppm of leucine and 57 ppm of lysine. The relative standard deviation (RSD %) results for the pharmacopuncture from centipedes had a maximum value of 14.95% and minimum value of 1.795% on the room temperature test chamber, the acceleration test chamber and the cold test chamber stability tests. Conclusion: Stability tests on and quantitative and qualitative analyses of the amino acids in the pharmacopuncture extracted from centipedes by using derivatization methods were performed by using HPLC. Through research, we hope to determine the relationship between time and the concentrations of the amino acids in the pharmacopuncture extracted from centipedes.

Determination of Saturated and Unsaturated Fatty Acids Amount in Leukocyte Membranes from Subjects Fed with Solid and Fluid Oils

  • Modifications in dietary fatty acid intake might lead to a modification in membrane phospholipid fatty acid composition. The purpose of this study was to investigate relationship between different type of oil consumption and leukocyte membrane phospholipid composition. This study was carried out in subjects utilizing butter (n = 15), margarine (n = 15), fluid oil (n = 15) and mixed types of oils (n = 15) in total 60 subjects. Leukocytes were separated from total blood by dextran sedimentation method. Membrane lipids and proteins were isolated following the cell disruption. Fatty acids of membrane phospholipids were isolated by hydrolysation with phospholipase B under ultrasonic dismembranator. Free fatty acids were identified with gas chromatography at chloroform phase. The results obtained were compared with data obtained by chromatograms of the standards. Results more prominent values of arachidic, dihomo-$\gamma$-linolenic and palmitoleic acids were found in butter-or mixed oil-user groups; eicosadienoic, eicosamonoenoic, dihomo-$\gamma$-linolenic and behenic acids in fluid oil; heptanoic, valeric, eicosadienoic and linolenic acids in margarine groups. The fatty acid composition of mixed oil was similar to butter, while other two oils were so different. From this study, it was concluded that the type of oil consumption might have an influence on phospholipid components of plasma membranes.

Organic Acids and Fatty Acids of Honey Harvested in Kangwon Area (강원도산 잡화벌꿀의 유기산 및 지방산 특성에 관한 연구)

  • Physico-chemical properties including the composition of organic acids and fatty acids in native bee-honey and foreign bee-honey harvested in Kangwon area were studied with the emphasis on the honey which was collected form wild flowers nd mixed flowers sources for honey nectars. the major organic acids were considered as acetic acid formic acid and valeric acid in volatile acids and gluconic acid maleic acid malic acid quinic acid and citric acid in non-volatile fraction in both of native bee-honey and foreigr bee-honey. Some naturally occuring fatty acids({{{{ { C}_14{ } }}}}-{{{{ {C }_{20 } }}}}) were observed with the principal fatty acids of myristic aicd linolenic acid and palmitic acid and palmitic acid in the various honey, The characteristics of moisture content pH viscosity free acidity lactone and total acidity were also analyzed and discussed.
