제한절편 길이 다형성(RFLP) 분자마커를 이용한 납자루아과 담수어류 3종의 난과 치어 종 동정 기법 개발 (Development of a Species Identification Method for the Egg and Fry of the Three Korean Bitterling Fishes (Pisces: Acheilognathinae) using RFLP (Restriction Fragment Length Polymorphism) Markers)

  • 본 연구는 PCR 기반 RFLP (Restriction Fragment Length Polymorphism; 제한절편 길이 다형성) 분자기법을 활용하여 난 및 치어 대상 납자루아과 어류 3종의 동정을 좀 더 빠르고 정확하게 파악하고 납자루아과 어류의 종별 산란양상 및 번식생태 이해에 대한 기여가 목적이다. 본 연구를 위해 기존 선행된 문헌자료를 확인하고 납자루아과 어류가 2종 이상 동서하고 있는 지역을 확인하여 현지조사를 수행하였다. 현지조사 결과 확인된 납자루아과 어류는 묵납자루(Acheilognathus signifer), 줄납자루(A. yamatsutae) 및 각시붕어(Rhodeus uyekii)로 총 3종이 확인되었으며, 확인된 납자루아과 어류와 동서하고 있는 숙주조개(작은말조개; Unio douglasiae sinuolatus)를 채집하여 숙주조개 속 납자루아과 어류의 난 및 치어를 확보하였다. 현지조사 결과 확인된 납자루아과 어류 3종을 대상으로 미토콘드리아 DNA COI과 cyt b 유전자 염기서열을 비교하여 각각 종별로 특이성을 지닌 부위(단일염기변이; Single Nucleotide Variation: SNV)에 맞는 제한효소를 선정하였고, 숙주조개 속 난 및 치어를 대상으로 genomic DNA를 추출하여 PCR-RFLP 실험을 수행한 결과 현지조사 시 확인된 납자루아과 어류 3종의 독특한 제한절편 길이 양상을 전기영동을 통하여 확인하였다. 본 연구를 통해 묵납자루, 줄납자루 및 각시붕어의 종을 판별할 수 있는 RFLP 마커를 개발하였으며, 숙주조개 난 및 치어를 대상으로 정확한 종의 동정을 보다 빠르고 효과적으로 수행하여 각각 납자루아과 종별 산란양상을 보다 정확히 규명하고 향후 이들 자연개체군의 효과적인 유지, 관리 및 보전 방법 개발에 유용하게 활용될 수 있을 것으로 판단된다.

Identification and Characterization of Microbial Community in the Coelomic Fluid of Earthworm (Aporrectodea molleri)

  • Earthworms play an important role in soil fertilization, interacting continually with microorganisms. This study aims to demonstrate the existence of beneficial microorganisms living in the earthworm's immune system, the coelomic fluid. To achieve this goal, a molecular identification technique was performed, using cytochrome c oxidase I (COI) barcoding to identify abundant endogenic earthworms inhabiting the temperate zone of Rabat, Morocco. Then, 16S rDNA and ITS sequencing techniques were adopted for bacteria and fungi, respectively. Biochemical analysis, showed the ability of bacteria to produce characteristic enzymes and utilize substrates. Qualitative screening of plant growth-promoting traits, including nitrogen fixation, phosphate and potassium solubilization, and indole acetic acid (IAA) production, was also performed. The result of mitochondrial COI barcoding allowed the identification of the earthworm species Aporrectodea molleri. Phenotypic and genotypic studies of the sixteen isolated bacteria and the two isolated fungi showed that they belong to the Pseudomonas, Aeromonas, Bacillus, Buttiauxella, Enterobacter, Pantoea, and Raoultella, and the Penicillium genera, respectively. Most of the isolated bacteria in the coelomic fluid showed the ability to produce β-glucosidase, β-glucosaminidase, Glutamyl-β-naphthylamidase, and aminopeptidase enzymes, utilizing substrates like aliphatic thiol, sorbitol, and fatty acid ester. Furthermore, three bacteria were able to fix nitrogen, solubilize phosphate and potassium, and produce IAA. This initial study demonstrated that despite the immune property of earthworms' coelomic fluid, it harbors beneficial microorganisms. Thus, the presence of resistant microorganisms in the earthworm's immune system highlights a possible selection process at the coelomic fluid level.

한국 서해 남부해역에서 채집된 도화뱅어, Neosalanx anderssoni (뱅어과) 자치어의 분자 동정 및 첫 형태기재 (Molecular Identification and First Morphological Description of Larvae and Juveniles of Neosalanx anderssoni (Salangidae) Collected from the Southwestern Sea of Korea)

  • 본 연구는 2023년 4~5월 전라남도 영광군 칠산도 주변 해역에서 채집된 뱅어과(Salangidae) 자치어 총 6개체를 대상으로 분자 동정과 발달단계별 형태특징을 상세히 기재하였다. MtDNA COI 또는 16S 영역을 대상으로 분자 분석을 진행한 결과, 자치어 6개체는 도화뱅어(Neosalanx anderssoni) 성어와 유전거리 0~0.2%의 차이를 보여 도화뱅어로 동정되었다. 도화뱅어 자치어 6개체는 모두 측편형의 가늘고 긴 체형을 보였다. 전기자어~중기자어(10.24 mm NL, 13.37 mm SL) 시기에는 복강의 배쪽에 타원형의 흑색소포가, 복강 등쪽에는 옅은 점 모양의 흑색소포가 열을 이루었다. 반면 후기자어~치어 시기(22.12 mm SL, 28.43 mm SL)에는 복강 배쪽의 흑색소포가 사라지고 복강 등쪽의 흑색소포가 수적으로 증가하여 1열로 나타났다. 또한, 후기자어 시기부터는 꼬리지느러미에 크고 짙은 검은 반점 2개가 대칭적으로 나타났다. 본 연구 결과는 이전의 연구 결과(Kim and Park, 2002)와 달리 도화뱅어가 연안을 산란 또는 성육장으로 이용할 수 있음을 시사한다.

Sequence Divergence and Phylogenetic Investigation of the Nymphalidae (Lepidoptera: Papilionoidea) Occurring in South Korea

  • As a first step toward understanding the divergence and relationships of the Nymphalidae (Lepidoptera: Papilionoidea) occurring in South Korea, cytochrome oxidase subunit I (COI), 16S ribosomal RNA (16S rRNA), and elongation factor-$1{\alpha}$ (EF-$1{\alpha}$) that comprise 3,501-3,716 bp were either sequenced (55 species) or the sequences were obtained from GenBank (23 species). The concatenated sequence divergence of six nymphalid subfamilies ranked in the following order: Danainae (10.3%), Satyrinae (9.5%), Limenitidinae (8.0%), Apaturinae (7.0%), Nymphalinae (6.7%), and Heliconiinae (6.2%). As has been reported in previous large scale international studies, the subfamilial relationships of (((((Limenitidinae + Heliconiinae) + (Nymphalinae + Apaturinae)) + Satyrinae) + Libytheinae) + Danainae) were also confirmed, except for the switched positions between Danainae and Libytheinae, and supported all subfamilies and tribe monophylies. Unlikely consistent phylogenetic relationships among genera within the majority of tribes in Nymphalidae, a conflicting relationship within the subfamily Apaturinae was obvious, presenting Apatura as sister to either Mimathyma or (Mimathyma + (Sephisa + (Hestina + Sasakia))), and both of these relationships are unconventional. Within the subfamily Limenitidinae, the genus Neptis was consistently revealed as a paraphyletic with respect to the genus Aldania, requiring further taxonomic investigation of the genus. Although limited, current sequence information and phylogenetic relationships are expected to be helpful for further studies.

A New Species of Eudactylopus (Copepoda: Harpacticoida) from the South Coast of Korea Based on Morphological and Molecular Evidence

  • A new species of Eudactylopus Scott A., 1909 is described from the southern coast of Korea. The specimens were collected using a light trap set overnight at the entrance near a pier. Eudactylopus yokjidoensis n. sp. is similar to E. andrewi Sewell, 1940 and E. spectabilis (Brian, 1923) in two key respects: similar length of proximal and distal inner setae on female P2 enp-2, and modification of two subapical setae on male P2 endopod. However, E. yokjidoensis can be differentiated from the two species by following morphological characteristics: in females, the length ratio of cephalothorax/2nd-4th thoracic somites combined is smaller in E. yokjidoensis than other two species (1 : 0.8 vs. 1 : 1); antennule has nine segments (vs. 7-segmented in E. andrewi); P2 to P4 each bears a process in medial distal margin of basis, while it is just smooth in E. spectabilis; in males; the length ratio of cephalothorax to 2nd-4th thoracic somites combined is smaller in E. yokjidoensis than other two species (1 : 0.6 vs. 1 : 1 in E. andrewi and 1 : 0.8 in E. spectabilis); and P5 exopod has a comb-like innermost seta, while it is bipinnate seta in E. spectabilis. To prove the Korean species of Eudactylopus to be new, full descriptions of both sexes are given here, and the claim is supported by distinct genetic differences between E. yokjidoensis and E. spectabilis (22.3-22.7%) in the mitochondrial gene cytochrome oxidase subunit I(mtCOI) sequence.

가다랑어 (Katsuwonus pelamis) 자어의 국내 출현 (Occurrence of skipjack tuna (Katsuwonus pelamis) larvae in Korean waters)

  • Three specimens of larvae (5.2~7.8 mm in standard length (SL)), of the family Scombridae, were collected from the southeastern waters off Jeju Island in August, 2014. These specimens were identified to Katsuwonus pelamis have melanophores distributed on the 1st dorsal-fin spines and post ventral margin on caudal peduncle. An analysis of the cytochrome oxidase submit I (COI) sequences (652 base pairs) of mitochondrial DNA showed that our specimens must be K. pelamis, because their sequences were concordant with those of the adult K. pelamis (genetic distance= 0.000~0.002), followed by Auxis rochei (genetic distance= 0.061~0.063) and Euthynnus affinis (genetic distance= 0.077~0.079). During the larval stage, K. pelamis differed from Thunnus spp. species in melanophores distribution period of 1st dorsal-fin spines, lower jaws and ventral margin on caudal peduncle. In conclusion, occurrence of K. pelamis larvae means Korean waters are potential spawning ground of K. pelamis.

Biodiversity of Hawaiian Peyssonneliales (Rhodophyta): Sonderophycus copusii sp. nov., a new species from the Northwestern Hawaiian Islands

  • Specimens of red algae corresponding to the peyssonnelioid genus Sonderophycus were collected at Kure Atoll, Hawai'i, at a depth range of 88-94 m depth during mesophotic surveys of the Papahānaumokuākea Marine National Monument, Hawaiian Islands, and were analyzed using morphological and molecular approaches. Analyses of mitochondrial cytochrome oxidase subunit 1 and chloroplast rbcL DNA sequences demonstrated that the Hawaiian specimens were identical to one another yet distinct from the three other species currently recognized within the genus (S. capensis [Montagne] M. J. Wynne, S. coriaceus [Womersley & Sinkora] M. J. Wynne, and S. fervens Dixon), as well as the likely congener, Peyssonnelia caulifera Okamura, and are proposed here as a new species: Sonderophycus copusii A. R. Sherwood. Sonderophycus copusii is morphologically distinct from other members of the genus by the following combination of characters: the presence of occasional secondary perithallial growth, emergence of rhizoids from the hypobasal cuticle at a strongly acute angle, a lack of horizontally directed filaments in the lower perithallus, and the lack of a stipe. This is the first record of the genus Sonderophycus in the Hawaiian Islands. Sonderophycus copusii was documented as a dominant member of the algal community at Kure Atoll, and thus may play a significant ecological role in the deep-water benthic community of Kure Atoll, along the lines of reports of deep water peyssonnelioid beds in the Mediterranean, Red Sea, and Caribbean. This study further highlights the unexplored diversity of the Peyssonneliales in Hawai'i, and emphasizes more generally the degree of as yet undiscovered biodiversity of algae at mesophotic depths.

한국산 색가오리과(Dasyatidae) 어류 1미기록종, Pteroplatytrygon violacea (First Record of the Pelagic Stingray, Pteroplatytrygon violacea (Dasyatidae, Myliobatiformes) from Korea)

  • 매가오리목, 색가오리과에 속하는 Pteroplatytrygon violacea 1개체(전장 1,058 mm)가 2017년 7월 6일 제주도 북서쪽 연안에서 처음으로 유자망에 의해 채집되었다. 이 종은 넓은 주둥이를 갖는 점, 5쌍의 아가미구멍이 있는 점, 꼬리에 1개의 가시가 있는 점, 꼬리의 배쪽에 있는 피습이 꼬리끝까지 이르지 않는 점, 꼬리의 등쪽에는 피습이 없는 점, 그리고 배의 표면은 전체적으로 어두운 자주색을 띠는 것이 특징이다. 이러한 형태적 특징에 따라서 이 종을 P. violacea로 동정하였으며, 표본의 COI 유전자 염기서열을 이용하여 종동정을 확인하였다. 이 미기록종은 배 표면의 체색으로 인해 기존에 붙여진 이름에 따라 종명과 속명을 각각 "보라색가오리"와 "보라색가오리속"으로 제안한다.

우리나라 연안해역 난 및 자치어의 분포특성 (Distribution of Eggs and Larvae in Coastal Waters of Korea)

  • The distribution and abundance of fish eggs and larvae were investigated from February to December 2020 along the coastal waters of Korea. The eggs and larvae were identified using the mitochondrial DNA cytochrome c oxidase subunit I (mtDNA COI) and 16s rRNA gene. During the study period, eggs of overall 45 taxa belonging to 26 families were collected and larvae of overall 39 taxa belonging to 23 families were collected. In Yeongil Bay, eggs of Engraulis japonicus, which accounted for 83.9% of the total population, was the most dominant species, followed by Sardinops sagax (4.0%), Repomucenus valenciennei (3.8%) and E. japonicus larvae, which accounted for 34.9% of the total population. These were followed by Sebastiscus marmoratus (31.0%). In Gomso Bay, E. japonicus eggs accounted for 61.7% of the total population, followed by Sillago japonica (14.0%), Johnius grypotus (8.8%) and Pholis fangi larvae, which accounted for 53.5% of the total population, followed by Ammodytes personatus (34.1%). In Jinhae Bay, E. japonicus eggs accounted for 86.0% of the total population, followed by Leiognathus nuchalis (4.1%), Konosirus punctatus (3.7%) and E. japonicus larvae, which accounted for 48.7% of the total population, followed by Parablennius yatabei (21.6%).

한국산 1미기록종, Acanthaphritis unoorum (농어목, 꼬리점눈퉁이과)의 어란 및 자치어의 분자동정 및 형태기재 (Molecular Identification and Morphological Descriptions of the Eggs, Larvae and Juvenile of the Previously Unrecorded Species Acanthaphritis unoorum (Perciformes, Percophidae) in Korean Waters)

  • We presented detailed morphological descriptions of the eggs, larvae and juvenile of Acanthaphritis unoorum based on specimens collected with bongo nets from Korean waters during the period May 2017-July 2018. We collected 18 individuals including eggs (n= 4, 0.77-0.85 mm in egg diameter), preflexion larvae (n= 6, 4.11-6.31 mm in standard length, SL), flexion larvae (n= 4, 6.60-7.82 mm SL), postflexion larvae (n= 3, 8.94-13.46 mm SL), and one juvenile (n= 1, 14.67 mm SL). The mitochondrial (mt) DNA 16S rRNA sequences of the eggs, and the cytochrome c oxidase subunit I (COI) sequences of the larvae were identical to those of A. unoorum adults (genetic distances <0.01). The A. unoorum larvae and the juvenile that we collected were morphologically similar to those of Dactylopsaron dimorphicum, but the A. unoorum specimens were readily distinguishable by the presence of lateral melanophores. This is the first record of A. unoorum in Korean waters. We propose a new Korean name for A. unoorum: "O-ri-bu-ri-nuntung-i".
