효모 Saccharomyces diastaticus YIY 345에서의 Human Lactoferrin 유전자 발현 및 분비 (Expression of Human Lactoferrin Gene and Secretion in Saccharomyces diastaticus YIY345)

  • 본 연구에서는 인간에게 유용한 여러 생리적 기능을 갖는 Human lactoferrin (hLf)을 대량생산할 목적으로 동물 세포의 단백질을 안정하게 생산 및 분비할 수 있는 진핵생물인 효모 sacduraromyces diastaticus YIY345에 hLf 유전자를 도입하여 hLf의 발현 및 분비를 시도하였다. 1. hLf의 발현 및 분비를 위하여 STA1 (S. diasticus glucoamylase) 유전자의 promoter 및 분비 신호와 hLf 전사 종결을 위한 GAL7(galactose utilizing gene) transcriptional terminator하에 재조합 plasmid pYEGLf를 제작하였다. 2. 제작한 plasmid를 선별하고 증폭하기 위하여 대장균에 형질 전환하였고 대장균 형질전환체로부터 plasmid를 분리하여 적당한 제한 효소로 처리하고 크기를 확인하였다. 최종적으로 선별된 plasmid는 효모 S. diastaticus YIY345에 형질 전환시켜 효모 형질전환체를 얻었다. 3. 효모 형질전환체를 Western hybridization으로 단백질 수준에서 분석한 결과 pYEGLf가 도입된 형질전환체에서 hLf가 생산되어 세포외로 분비되었다. 4. hLf가 발현된 효모 형질전환체의 배양상등액을 paper disc법을 이용하여 E.coli에 대한 항균 효과를 측정하였으나 관찰할 수 없었다. E.coli에 대한 hLf의 MIC (minimal inhibitory concentration)는 약 $3000{\mu}g/m{\ell}$에 비해 효모에서 분비하는 hLf의 양은 상당히 적으므로 항균효과를 측정하기 불가능하다. 그러므로 정제 단계를 거쳐 더 높은 농도로 처리해야 할 것이다.

Methicillin 저항성 Staphylococcus aureus에 대한 선복화(Inula britannica flowers) 추출물의 항균효과 (Antimicrobial Effect of Inula britannica Flower Extract against Methicillin-resistant Staphylococcus aureus)

  • 선복화(Inula britannica flowers) 메탄올 추출물을 이용하여 methicillin에 저항성이 있는 Staphylococcus aureus (MRSA)에 대한 항균효과를 검증하였다. 선복화 메탄올 추출물은 다량의 quercetin을 함유한 것으로 분석되었다. 세 종류의 MRSA에 대한 항균효과는 disc diffusion method에 의해 확인되었고, 최소저해농도(MIC) 및 최소사멸농도(MBC)는 각각 0.625 mg/ml-1.25 mg/ml, 2.5 mg/ml로 확인되었다. 이들 MRSA 중 S. aureus ATCC 33591가 가장 민감한 작용을 하였으며, 이 균에 대해서 작용기작을 확인하였다. 시간에 따른 저해 정도와 형태관찰을 통해 선복화 메탄올 추출물이 항균활성을 갖는 것을 확인할 수 있었다. 메탄올 추출물의 resistant gene에 대한 영향은 mRNA 상에서 mecA와 mecRI gene의 발현감소와 mecI gene의 발현증가를 통해 메티실린 내성을 감소시키는 것을 알 수 있었다. 따라서 선복화 메탄올추출물은 MRSA에 대한 항균효과를 가지는 천연 항균성 소재로서의 가능성을 확인할 수 있었다.

  • 밤나무의 밤꽃과 밤잎의 항균활성 효과를 조사하기 위하여, 시료를 80% methanol로 추출하여 용매분획을 하였다. 용매분획별 수율은 밤꽃에서 ethylacetate 1.94%, methanol 4.75% 및 water 7.81%였고, 밤잎은 ethylacetate 2.24%, methanol 5.03% 및 water 8.47%였다. 밤꽃과 밤잎의 methanol 분획 추출물은 Gram양성 및 음성의 모든 균주에서 강한 항균활성을 보였으며, ethylacetate 및 water 분획 추출물은 약한 항균활성을 보였다. 최소저해농도는 $60\;{\mu}g/disc$ 이상에서 항균력이 인정되었다. Bacillus. subtilis에 대한 생육저해 효과는 control에 비하여 밤꽂과 밤잎 추출물은 모두 낮은 생육저해 효과를 보였으며, 또한 각 추출물은 모두 배양 15시간까지는 현저히 낮은 생육저해를 보이다가 그 후는 약간 높아졌다. 밤꽃 추출물에서는 methanol 추출물이 가장 높은 생육저해효과를 보인 반면에, water 추출물은 다른 추출물에 비하여 낮은 생육저해를 보였으며, 밤잎 추출물은 각 추출물에서 거의 비슷한 생육저해를 보였다. 밤꽃과 밤잎의 methanol 분획 추출물을 B. subtilis에 각각 500 ppm씩 처리한 후, 주사전자현미경(SEM)으로 관찰한 결과, 추출물을 처리하지 아니한 경우는 전형적인 Bacilli의 형태를 보였지만, methanol 분획추출물을 처리한 경우는 세포의 형태가 일그러지고 손상을 입은 형태를 보였다.

한국산 고감염 Agrobacterium spp의 분리 및 연초의 형질전환에 이용 (Isolation of Hypervirulent Agrobacterium spp from Korea and Application for Transformation of Tobacco)

  • 한국에서 자생한 자연산 tumor 조직과 근권토양으로부터 형질전환율이 높은 hypervirulent Agrobacterium spp를 분리하기 위해서 Salix, Diospyros, Populus 및 Malus에서 형성된 tumor 조직과 근권토양을 채취하괴 Schroth 선택배지와 New and Kerr 배지를 이용하여 78 균주의 colony를 특성에 따라 분리하였다. 이중에서 48 균주가 당근 disc에서 tumor를 형성하였으며 tumor를 형성한 균주중 형성시기가 빠르며 크기가 커 hypervirulent 균주로 생각되는 A. tumefaciens SP101을 biotype 1, 그리고 A. tumefaciens SM042를 biotype 2로 동정하였다. 토양중에서 선발한 A. tumefaciens SM042와 disarmed A. tumefaciens PC2760에 kanamycin 저항성 유전자를 함유하고 있는 binary vector pGA643을 도입하여 conjugant인 A. tumefaciens SM643과 A. tumefaciens PC643을 kanamycin과 tetracycline이 함유된 최소배지에서 획득하였다. 연초의 형질전환을 위해서 conjugant Agrobacterium과 연초조직을 동시배양 후 2,4-D와 kanamycin이 함유된 선발배지에 치상하여 형질전환을 유도한 결과 A. tumefaciens SM643이 A. tumefaciens PC643보다 더 많은 캘러스가 형성되었다. 그러나 A. tumefaciens PC643을 사용한 형질전환 캘러스는 대부분 friable한 캘러스로 유도되었으며 정상적으로 식물체로 생장하였으나 A. tumefaciens SM643을 사용한 캘러스는 매우 딱딱하며 둥그런 형태의 캘러스와 friable한 캘러스가 혼재한 상태로 생장하였으며, 이중에서 friable한 캘러스는 정상적인 shoot가 형성되었으나 딱딱한 캘러스로 유도된 형질전환체는 식물체로 형성되지 않고 반구형 미색의 전형적인 tumor 캘러스로 생장하였다. 한편 형질전환시 캘러스를 유도하지 않고 직접 shoot를 형성시킨 결과 disarmed Ti-plasmid를 사용하지 않고 wild-type Ti-plasmid를 사용해도 정상적인 형질전환체를 획득할 수 있었다.

Comparison of Chemical Compositions and Antimicrobial Activities of Essential Oils from Three Conifer Trees; Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa

  • The chemical compositions, and antibacterial and antifungal effects of essential oils extracted from three coniferous species, Pinus densiflora, Cryptomeria japonica, and Chamaecyparis obtusa, were investigated. Gas chromatography mass analysis of the essential oils revealed that the major components and the percentage of each essential oil were 16.66% $\beta$-phellandrene and 14.85% $\alpha$-pinene in P. densiflora; 31.45% kaur-16-ene and 11.06% sabinene in C. japonica; and 18.75% bicyclo [2,2,1] heptan-2-ol and 17.41% 2-carene in Ch. obtusa. The antimicrobial assay by agar disc diffusion method showed that $2.2{\mu}g$ of Ch. obtusa oil inhibited most effectively the growth of Escherichia coli ATCC 33312 and Klebsiella oxytoca ATCC 10031, whereas the C. japonica oil gave weak antimicrobial activity. The minimal inhibitory concentration(MIC) values for bacterial strains were in the range of 5.45-21.8 mg/ml depending on essential oils, but most Gram-negative bacteria were resistant even at 21.8 mg oil/ml. P. densiflora oil showed the most effective antifungal activity and the MIC values for Cryptococcus neoformans B42419 and Candida glabrata YFCC 062CCM 11658 were as low as 0.545 and 2.18 mg/ml, respectively. Cryp. neoformans B42419 was the most sensitive to all essential oils in the range of 0.545-2.18 mg/ml. Our data clearly showed that the essential oils from the three conifers had effective antimicrobial activity, especially against fungi.

  • Objective: Evaluations of the in-vitro anti-bacterial activities of aqueous extracts of Acacia catechu (L.F.)Willd, Castanea sativa, Ephedra sinica stapf and Shilajita mumiyo against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) and gram-negative bacteria (Escherichia coli, klebsiella pneumoniae, Proteus mirabilis, Pseudomonas aeruginosa) are reasonable since these ethnomedicinal plants have been used in Persian folk medicine for treating skin diseases, venereal diseases, respiratory problems and nervous disorders for ages. Methods: The well diffusion method (KB testing) with a concentration of $250{\mu}g/disc$ was used for evaluating the minimal inhibitory concentrations (MIC). Maximum synergistic effects of different combinations of components were also observed. Results: A particular combination of Acacia catechu (L.F.) Willd, Castanea sativa, Ephedra sinica stapf and shilajita mumiyo extracts possesses an outstanding anti-bacterial activity. It's inhibiting effect on microorganisms is significant when compared to the control group (P<0.05). Staphylococcus aureus was the most sensitive microorganism. The highest anti-bacterial activity against gram-positive bacteria (Staphylococcus aureus, Streptococcus pneumonia) or gram-negative bacteria (Escherichia coli, Klebsiella pneumonia, Proteus mirabilis, and Pseudomonas aeruginosa) was exerted by formula number 2 (table 1). Conclusion: The results reveal the presence of anti-bacterial activities of Acacia catechu, Castanea sativa husk, Ephedra sp. and Mumiyo against gram-positive and gram-negative bacteria. Synergistic effects in a combined formula, especially in formula number 2 (ASLAN$^{(R)}$) can lead to potential sources of new antiseptic agents for treatment of acute or chronic skin ulcers. These results considering the significant anti-bacterial effect of the present formulation, support ethnopharmacological uses against diarrheal and venereal diseases and demonstrate use of these plants to treat infectious diseases.

Direct Lateral Lumbar Interbody Fusion : Clinical and Radiological Outcomes

  • Objective : According to the recent development of minimally invasive spinal surgery, direct lumbar interbody fusion (DLIF) was introduced as an effective option to treat lumbar degenerative diseases. However, comprehensive results of DLIF have not been reported in Korea yet. The object of this study is to summarize radiological and clinical outcomes of our DLIF experience. Methods : We performed DLIF for 130 patients from May 2011 to June 2013. Among them, 90 patients, who could be followed up for more than 6 months, were analyzed retrospectively. Clinical outcomes were compared using visual analog scale (VAS) score and Oswestry Disability Index (ODI). Bilateral foramen areas, disc height, segmental coronal and sagittal angle, and regional sagittal angle were measured. Additionally, fusion rate was assessed. Results : A total of 90 patients, 116 levels, were underwent DLIF. The VAS and ODI improved statistically significant after surgery. All the approaches for DLIF were done on the left side. The left and right side foramen area changed from $99.5mm^2$ and $102.9mm^2$ to $159.2mm^2$ and $151.2mm^2$ postoperatively (p<0.001). Pre- and postoperative segmental coronal and sagittal angles changed statistically significant from $4.1^{\circ}$ and $9.9^{\circ}$ to $1.1^{\circ}$ and $11.1^{\circ}$. Fusion rates of 6 and 12 months were 60.9% and 87.8%. Complications occurred in 17 patients (18.9%). However, most of the complications were resolved within 2 months. Conclusion : DLIF is not only effective for indirect decompression and deformity correction but also shows satisfactory mechanical stability and fusion rate.

전방 경추 미세 추간공 확대술 : 경추증에 대한 최소침습적 전측방 접근법 (Anterior Cervical Microforaminotomy : A Minimally Invasive Anterolateral Approach for Spondylotic Lesions)

  • Objective : Various surgical approaches have been implemented to fulfill the ideal goals of treatment for cervical spondylotic lesions. Conventional approaches are represented by anterior approach with or without fusion and posterior approach. The authors has applied newly developed anterior cervical microforaminotomy for these lesions on minimally invasive basis. Materials and Method : Twenty-one patients, with cervical HIVD, or stenosis, or both, underwent anterior cervical microforaminotomy between March, 1998 and April, 1999. Fifteen patients underwent unilateral decompression, and 6 bilateral decompression via unilateral foraminotomy. Operation of one level was performed in 16 patients, 2 levels in 4 patients, and 3 in 1 patient. The foraminotomy was accomplished by resecting the uncovertebral joint. Through this hole, compressed nerve root was decompressed by removing the spondylotic spur or disc fragment, and diagonal removing of posterior osteophyte from foraminotomy site to begining of contralateral nerve root made spinal cord decompression. Results : The outcome was excellent in 17 patients(81%) and good in 4 patients(19%) based on Odom's criteria. No complication was encounterd, and average post-operation hospital stay was 3.7 days. Conclusions : These results indicate that anterior cervical microforaminotomy provide adequate neural decompression, minimum postoperative discomfort and fast recovery.

다분절 퇴행성 경추질환에서의 전방 금속판 사용 유무에 따른 환자의 예후분석 (Outcome Analysis of the Patients with and without Anterior Plating in Multi-Level Degenerative Cervical Diseases)

  • Objectives : The rates of pseudarthrosis for two- and three level fusion have been reported to be 17-63 and 50% without anterior cervical plating. The purpose of this study is to assess the effects of anterior cervical plating in the treatment of multilevel degenerative cervical disease such mostly the additional risks associated with hardware implants and its benefits, fusion rate and radiographic results, and clinical outcomes. Methods : Forty-seven patients who underwent operations between 1993 and 1997 were retrospectively reviewed. The technique for operation was same for both groups(Smith Robinson with autologous iliac bone graft). Group I consisted of 35 consecutive patients treated with anterior cervical decompression and fusion with anterior cervical plate fixation. Group II consisted of 12 consecutive patients treated without plate fixation. We compared clinical outcomes by Prolo score, radiographic results in the rate of fusion, cervical lordosis by Gore angle, disc height by Farfan method, and surgical complications between two groups. Results : The favorable clinical outcomes(excellent and good) by Prolo score were observed with the use of anterior cervical plate fixation(89% vs 75%). The successful fusion rate of multilevel cervical fusion was as seen with anterior cervical plate fixation(97% vs 75%). The overall graft complication rate in multilevel fusions was decreased, with anterior cervical plate fixation, and the hardware related complications were relatively minimal without serious consequences. Conclusion : Anterior cervical plate fixation in the treatment of multilevel cervical disorders is an effective stabilizing method which provides increased bony fusion rate, decreased graft complications, maintained cervical lordosis, early mobilization of the these patients without serious hardware related complications.

Antimicrobial efficacy of endophytic Penicillium purpurogenum ED76 against clinical pathogens and its possible mode of action

  • This study was aimed to evaluate the antimicrobial activity of Penicillium purpurogenum ED76 on several clinically important microorganisms. The endophytic fungus P. purpurogenum ED76 was previously isolated from Swietenia macrophylla leaf. The antimicrobial efficacy of P. purpurogenum ED76 dichloromethane extract was determined via disc diffusion and broth microdilution assay. A kill curve study was conducted and the morphology of extract treated bacterial cells were viewed under scanning electron microscope. The dichloromethane extract showed significant inhibitory activity on 4 test bacteria and 2 test yeasts. The minimal inhibitory concentration of the extract ranged from 125 to $1,000{\mu}g/ml$, which indicates the different susceptibility levels of the test microorganisms to the fungal extract. The kill curve study has revealed a concentration-dependent inhibition for all test microorganisms. With the increase of the extract concentration, the microbial growth was significantly reduced. The scanning electron micrograph of dichloromethane extract-treated Staphylococcus aureus cells showed the total damage of the cells. The cell wall invagination of the bacterial cells also indicates the loss of cellular materials and metabolic activity. The gas chromatography mass spectrometry analysis of the extract also showed that the major compound was stigmasterol, which constitutes 45.30% of the total area. The dichloromethane extract of P. purpurogenum ED76 exhibited significant inhibitory activity on several clinically important bacteria and yeasts. The study proposed a possible mode of action that the extract cause significant damage to the morphology of S. aureus cells.
