Nuclear Transfer using Human CD59 and IL-18BP Double Transgenic Fetal Fibroblasts in Miniature Pigs

  • Xenotransplantation involves multiple steps of immune rejection. The present study was designed to produce nuclear transfer embryos, prior to the production of transgenic pigs, using fibroblasts carrying transgenes human complement regulatory protein hCD59 and interleukin-18 binding protein (hIL-18BP) to reduce hyperacute rejection (HAR) and cellular rejection in pig-to-human xenotransplantation. In addition to the hCD59-mediated reduction of HAR, hIL-18BP may prevent cellular rejection by inhibiting the activation of natural killer cells, activated T-cell proliferation, and induction of $IFN-{\gamma}$. Transgene construct including hCD59 and ILI-18BP was introduced into miniature pig fetal fibroblasts. After antibiotic selection of double transgenic fibroblasts, integration of the transgene was screened by PCR, and the transgene expression was confirmed by RT-PCR. Treatment of human serum did not affect the survival of double-transgenic fibroblasts, whereas the treatment significantly reduced the survival of non-transgenic fibroblasts (p<0.01), suggesting alleviation of HAR. Among 337 reconstituted oocytes produced by nuclear transfer using the double transgenic fibroblasts, 28 (15.3%) developed to the blastocyst stage. Analysis of individual embryos indicated that 53.6% (15/28) of embryos contained the transgene. The result of the present study demonstrates the resistance of hCD59 and IL-18BP double-transgenic fibroblasts against HAR, and the usefulness of the transgenic approach may be predicted by RT-PCR and cytolytic assessment prior to actual production of transgenic pigs. Further study on the transfer of these embryos to surrogates may produce transgenic clone miniature pigs expressing hCD59 and hIL-18BP for xenotransplantation.

Effect of Nicotinic Acid on Fresh Semen Characteristics in Miniature Pigs

  • Objective of this study was to investigate the effect of nicotinic acid (NA) on the characteristics in fresh semen of miniature pig. We evaluated viability, acrosome reaction and mitochondrial integrity of sperm on 0, 3, 7 and 10 days during storage period with nicotinic acid. As results, the survival rate of sperm in 15 mM NA (day 3, $87.8{\pm}1.2%$; day 5, $84.0{\pm}2.7%$; day 7, $82.2{\pm}0.9%$) and 30 mM NA (day 3, $87.7{\pm}0.3%$; day 5, $84.4{\pm}2.5%$; day 7, $82.3{\pm}0.7%$) groups were higher than control and 5 mM NA groups in 3, 7 and 10 days of semen storage. The NA-treated sperm on 10 day was used day for observing acrosome integrity. The survival sperm with acrosome reaction was higher in 30 mM NA group (day 3, $2.7{\pm}0.2%$; day 5, $3.3{\pm}0.6%$; day 7, $11.4{\pm}0.3%$) than in the control, significantly (P<0.05). Moreover, the live sperm with mitochondrial integrity was higher in whole treatment groups of NA than control group, significantly (P<0.05). Specially, most mitochondrial integrity on 10 day of semen storage was significantly higher in 30 mM NA group ($90.2{\pm}1.6%$) than other treatment groups (control, $81.8{\pm}3.1%$; 5 mM NA, $83.4{\pm}3.0%$; 15 mM NA, $89.1{\pm}0.7%$, P<0.05). In conclusion, supplement of NA in liquid semen of miniature pig can improve and maintain semen quality, such as viability, acrosome reaction, and mitochondria integrity.

  • This paper presents an autonomous target tracking approach and technique for transmitting ground control station image periodically for an unmanned aerial vehicle using onboard gimbaled(pan-tilt) camera system. The miniature rotary UAV which was used in this study has a small, high-performance camera, improved target acquisition technique, and autonomous target tracking algorithm. Also in order to stabilize real-time image sequences, image stabilization algorithm was adopted. Finally the target tracking performance was verified through a real flight test.

  • Development of an engine with good fuel economy is very important for successful implementation of long endurance miniature UAVs (unmanned aerial vehicles). In the study, a 4-stroke glow-plug engine was modified to a gasoline-fueled spark-ignition engine. Engine tests measuring performance and friction losses were conducted to tune a simulation program for performance prediction. It has been found that excessive friction losses are caused by insufficient lubrication at high speeds. The simulation program predicts that engine power and fuel economy get worse with high altitude due to increasing portion of friction losses. The simulation results suggest quantitative guidelines for further development of a practical engine.

  • 인정바닥구조는 신청에서부터 인정서 발급까지 기본적으로 2개월 이상이 소요되며, 신청 제품이 많을수록 그 기간은 길어지게 된다. 본 논문은 이와 같은 완충재의 성능을 확인하기 위해 소요되는 기간과 비용을 획기적으로 단축할 수 있는 간편한 방법을 제시하기 위한 것이다. 완충재 설치 이후 습식으로 시공되는 경량기포콘크리트와 모르터를 사전에 일정 크기로 제작(축소모형 시험판)한 후 골조가 완성된 현장에서 바닥충격음을 측정하는 방법이다. 완충재 2개 제품에 대하여 축소모형 시험판을 이용하여 측정한 결과와 전체 세대를 시공한 후 측정한 결과를 비교한 결과 경량충격음은 축소모형 시험판이 훨씬 낮은 결과를 나타냈지만 중량충격음은 비슷한 결과를 나타냈다. 이로써 축소모형 시험판을 이용하여 바닥충격음을 측정하는 것은 경량충격음을 저감하기 위한 재료 선정 및 제품 시스템 구성 등에 활용될 수 있을 것이며, 현장에서 중량충격음 차단성능을 신속하게 검증할 수 있는 방법으로 활용할 수 있을 것으로 기대된다.

NEW DEVELOPED PORTABLE NEAR INFRARED (NIR) SYSTEM USING MICROSPECTROMETER

  • In recent years, a miniature spectrometer has been extensively developed due to the marriage of fiber optics and semiconductor detector array. This type of miniature spectrometer has advantages of low price and robustness due to the capability of mass production and no moving parts are required such as lenses, mirrors and scanning monochromator. These systems are ideal for use in teaching labs, process monitoring and field analyses. A portable near infrared (NIR) system has been developed for qualitative and quantitative analysis. This system includes a tungsten halogen lamp for light source, a fiber optics connected a light source, and a sample module to the microspectrometer, The size of spectrometer can be as small as 2.5 cm x 1.5 cm x 0.1 cm. Wavelength ranges can be chosen as 360-800 nm, 800-1100 nm and 1100-1900 nm depending on the type of detector. The software consists of various tools for multivariate analysis and pattern recognition techniques. To evaluate the system, long and short-term stability, wavelength accuracy, and stray light have been investigated and compared with conventional scanning type NIR spectrometer. This developed system can be sufficiently used for quantitative and qualitative analysis for various samples such as agricultural product, herbal medicine, food, petroleum, and pharmaceuticals, etc.

  • Developing robot hands with multi-degree-of-freedom is one of the topics that researchers have recently begun to improve the limitation by adding flexibility and dexterity. In this study, an articulated servo-pneumatic robot hand system with direct-drive joints has been developed whose main feature is the minimization of the dimension. The servo-pneumatic system is advantageous to fabricate a dexterous robot hand system due to the high torque-to-weight and torque-to-volume ratio. This enables the design of a finger joint with an integrated rotary vane type actuator which produces high output torque without reduction gears, being very robust. In order to control the servo-pneumatic finger joints, a miniature proportional valve that can be attached to the robot hand is required. In this paper, a flapper nozzle type 4/3-way proportional directional valve has been designed and tested. The experimental results show that the developed valve can control a finger joint satisfactorily without much vibratory joint movements and acoustic noises.
