HPLC 분석을 통한 한국산 밀크씨슬 중 실리마린과 실리빈의 정량 (Determination of Silymarin and Silybin Diastereomers in Korean Milk Thistle using HPLC/UV Analysis)

  • Silymarin (SM) and silybin diastereomers (SD) in milk thistle (Silybum marianum) were determined using high-performance liquid chromatography and quantified using a reverse-phase column in a gradient elution system. UV detection was performed at 288 nm. The content of SM and SD in milk thistle was 3.236 and 0.553 mg/g DW, respectively. Determining the presence and quantifying the content of SM and SD in milk thistle are vital for the pharmaceutical industry to identify optimal sources for developing health supplements or therapeutics.

  • Silybum marianum is an annual or biennial plant from the Asteraceae family. It can grow in low-nutrient soil and drought conditions, making it easy to cultivate. From the seed, a specialized plant metabolite called silymarin (flavonolignan complex) is produced and is known to alleviate the liver from hepatitis and toxins damages. To infer the phylogenetic placement of a Korean milk thistle, we conducted a chloroplast assembly and annotation following by a comparison with existing Chinese reference genome (NC_028027). The chloroplast genome structure was highly similar with an assembly size of 152,642 bp, an 153,202 bp for Korean and Chinese milk thistle respectively. Moreover, there were similarities at the gene level, coding sequence (n = 82), transfer RNA (n = 31) and ribosomal RNA (n = 4). From all coding sequences gene set, the phylogenetic tree inference placed the Korean cultivar into the milk thistle clade; corroborating the expected tree. Moreover, an investigation the tree based only on the ycf1 gene confirmed the same tree; suggesting that ycf1 gene is a potential marker for DNA barcoding and population diversity study in milk thistle genus. Overall, the provided data represents a valuable resource for population genomics and species-centered determination since several species have been reported in the Silybum genus.

Protective Effects of Milk Thistle (Silybum marianum) against Aflatoxin B1 in Broiler Chicks

  • Aflatoxin-contaminated feed cause mortality, suppression of the immune system, reduced growth rates and losses in feed efficiency. This research study was planned to investigate the immunomodulatory and growth promoting effect of milk thistle as feed additive against aflatoxin $B_1$ in broiler chicks at NWFP Agricultural University Peshawar, Pakistan. Two hundred and forty (240) day old broilers chicks were randomly assigned into four major groups AfF, aflatoxin free feed; Aflatoxin $B_1$ was present in the feed at the levels of 80-520 ${\mu}g/kg$ of the feed in the remaining three groups. Aflatoxin contaminated feed was provided for 5 weeks. Group AfB was supplemented with toxin binder "Mycoad" at 3 g/kg of feed and group AfT was supplemented with milk thistle at10 g/kg of feed. Each group was further sub divided into two sub-groups, vaccinated against ND (Newcastle disease), IB (Infectious bronchitis) and IBD (Infectious bursal diseases) according to recommended schedule of vaccination or non vaccinated. Each sub group carried three replicates with 10 chicks per replicate. Chicks were reared in pens in an open sided house. Supplementary heat was provided to all the chicks during brooding period. Mean body weight gain and dressing percentage were significantly (p<0.05) higher in group AfF, followed by AfT, AfB and Af. Weight gain and dressing percentage was the same in group AfB and AfT, while it was significantly lower in group Af. Feed intake, breast, thigh and leg weight were found significantly (p<0.05) higher in group AfF, followed by AfB, AfT and Af. Significantly lower (better) FCR value was recorded in group AfT. Water intake was significantly (p<0.05) higher in group AfT and AfF as compared to other groups. Mortality was significantly (p<0.05) higher in group Af. Mean bursa and thymus weights were found significantly (p<0.05) higher in group AfF, AfB and AfT followed by Af, while higher spleen weight was recorded in group AfT. Mean antibody titer against ND, IB and IBD was significantly (p<0.05) higher in group AfT, as compared to other groups. It is concluded that milk thistle at 10 g/kg of feed could effectively be utilized as immunostimulant and growth promotant in the presence of immunosuppressant aflatoxin $B_1$ in the feed.

밀크씨슬 오일에 의한 화학적 손상모발의 개선 효과 (Effects of Milk Thistle Oil on Chemically Damaged Hair Improvement)

  • 본 연구는 밀크씨슬 오일을 첨가한 모질 개선 제형제를 제조하여 손상된 모발에 도포하여 모질 개선 효과를 알아보고자 하였다. 밀크씨슬 오일 함량을 0 g, 1 g 2 g, 3 g 으로 다르게 하여 제형제를 제조하였다. 제조한 제형제로 탈색한 시료 모발에 도포 후 열처리와 자연방치를 한 후에 시료 별로 도포 전과 후를 측정하여 비교 분석하였다. 모질 개선 효과를 알기 위해 인장강도, 메틸렌블루를 이용한 흡광도, 광택을 측정하였고 하였다. 모질 개선 효과를 알기위한 인장강도 측정결과는 7레벨과 9레벨 모든 시료의 인장강도가 증가함을 알 수 있었다. 메틸렌블루를 이용한 흡광도 분석결과 도포 전, 후의 변화는 도포 후의 값이 감소함을 알 수 있었다. 광택 측정결과는 모든 시료에서 큰 차이가 없음을 알 수 있었다. 밀크씨슬 오일이 손상된 모발에 모질 개선 효과가 있는 것을 알 수 있었다. 차후 다양한 오일 종류와 연구방법으로 모질개선 연구가 필요하다.

The Pharmacological Properties of Silymarin and Its Constituents

  • Silymarin is a standardized extract obtained from the seeds and fruits of Silybum marianum L., or commonly called milk thistle, a member of Carduus marianum family that contains mix of flavonolignans. Some epidemiological and preclinical studies revealed that S. marianum L. has been used for herbal remedies for centuries for its pharmacological activity. In this review, pharmacological studies in vitro and in vivo of silymarin are discussed thoroughly stressing on antioxidant, antimicrobial, antiviral, and anti-carcinogenic aspects of silymarin. In addition, the protective influences of silymarin on some organs such as heart, liver, bone, and neuron tissue are reviewed as well. This review would be useful for further study regarding the potential of natural plant, notably silymarin, and its therapeutic potential in the prevention and treatment of diseases.

  • Herbal plants or traditional Oriental medicine have been considered as a potential resource of new drug development worldwide. However, traditional Korean medicine has given little effort to the field of new drug development. This study reports on a plant-derived hepatotherapeutic drug, silymarin, which has been popularly used in many countries. It was discovered as an active compound from Silybum marianum (milk thistle) which has been known as a medicinal plant having hepatoprotective properties in both European and Asian countries. While it has been used as an herbal prescription in Asia, its active compounds or scientific mechanisms were intensively studied in Europe. Currently, silymarin is one of the most powerful herbal extracts in the world, and its usage is being expanded to many other medical purposes. This report would be helpful for providing an informative example of herbal-derived drug development to Oriental doctors or scientists in the Oriental medicinal field.

인간 전립선 암세포 PC-3 세포에서 Silibinin의 세포주기조절을 통한 세포사멸 유도 효과 (Silibinin Inhibits Cell Growth and Induces Apoptosis through Cell-cycle Arrest in PC-3 Prostate Cancer Cells)

  • Silibinin은 milk thistle에서 분리된 주된 생리활성 성분으로 강력한 항산화제, 항암활성에 대해서 보고되어 있다. 하지만 항암활성에 대한 정확한 기전에 대해서는 밝혀져 있지 않다. 본 연구에서는 인간 전립선 암세포주인 PC-3 세포를 이용하여 세포사멸의 기전을 조사하였다. MTT assay를 통해 세포독성을 확인하였고, PI 염색을 통해 세포주기를 확인하고, Annexin-V/PI 염색을 통한 세포사멸을 확인하였다. 뿐만 아니라 western blot을 이용하여 세포주기 및 세포사멸에 관련된 단백질 발현 정도를 확인하였다. 본 연구의 결과에서 silibinin은 인간 전립선 암세포주인 PC-3 세포에서 세포주기관련 단백질의 발현을 조절하여 세포주기 진행을 억제함으로써 세포사멸을 유도 함을 알 수 있었다.

Remedy effects of dandelion and milk thistle on fatty liver hemorrhagic syndrome in laying hens

  • Fatty liver hemorrhagic syndrome (FLHS) is a metabolic disorder found in caged layer hens and causes reduced egg production and sudden death. Dandelion (Taraxacum coreanum, TC) and milk thistle (Cirsium japonicum var. ussuriense, CJ) are well known wild herbs inhabiting Korean peninsula and presenting antioxidative effects. This study investigated alleviate effects of these herbal mixture (6:4, w/w) composed of dried powder of TC and CJ against fatty liver in laying hens. The herbs mixture 5.0, 10.0, 20.0 or 40.0 g/kg feed was provided via feed admixture for 3 weeks to laying hens having FLHS. FLHS was induced by intramuscular injection of β-estradiol (2 mg/kg bw) 2 times per week for 3 weeks and supply with high caloric feed. Egg production rate was reduced from 76.2% at pre-treatment to 49.4% at 1 week and further decreased according to β-estradiol treatment. Increment of serum alanine aminotransferase (ALT), aspartate aminotransferase (AST), total cholesterol (T-Chol) and total bilirubin (T-Bil) and decrement of serum superoxide dismutase (SOD) and glutathione peroxidase (GPX) with fatty liver were found by the treatment of β-estradiol. Supplementation of TC and CJ mixture via feed admixture recovered the reduction of egg production and attenuated serological changes and gross and pathological lesions of fatty liver with the best amelioration effects at 5 and 10 g TC and CJ mixture per kg feed. In conclusion, TC and CJ mixture attenuates FLHS by means of antioxidative effects. Further mechanistic study is required to explain TC and CJ's amelioration effects against FLHS in laying hens.

Luteolin 5-O-glucoside from Korean Milk Thistle, Cirsium maackii, Exhibits Anti-Inflammatory Activity via Activation of the Nrf2/HO-1 Pathway

  • Luteolin 5-O-glucoside is the major flavonoid from Korean thistle, Cirsium maackii. We previously reported the anti-inflammatory activities of luteolin 5-O-glucoside in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. In this study, we determined the anti-inflammatory mechanisms of luteolin 5-O-glucoside through the inhibition of nitric oxide (NO) production in vitro and in vivo. Results revealed that luteolin 5-O-glucoside dose-dependently inhibited NO production and expression of iNOS and COX-2 in LPS-induced RAW 264.7 cells. Luteolin 5-O-glucoside also significantly inhibited the translocation of $NF-{\kappa}B$, the activation of MAPKs, and ROS generation in LPS-induced RAW 264.7 cells. In addition, protein expressions of Nrf-2 and HO-1 were also upregulated by luteolin 5-O-glucoside treatment. Moreover, luteolin 5-O-glucoside inhibited ${\lambda}-carrageenan-induced$ mouse paw edema by 65.34% and 48.31% at doses of 50 and 100 mg/kg body weight, respectively. These findings indicate potential anti-inflammatory effect of luteolin 5-O-glucoside particularly by downregulating $NF-{\kappa}B$ and upregulating HO-1/Nrf-2 pathway.

Activating Transcription Factor 3 is a Molecular Target for Apoptotic Effect of Silymarin in Human Colorectal Cancer Cells

  • Apoptosis has been regarded as a therapeutic target because apoptosis is typically disturbed in human cancer. Silymarin found in the seeds of the milk thistle (Silybum marianum) has been reported to exert anti-cancer properties through apoptosis. This study was performed to investigate the molecular target for silymarin-mediated apoptosis in human colorectal cancer cells. Silymarin reduced the cell viability and induced an apoptosis in human colorectal cancer cells. ATF3 overexpression increased PARP cleavage by silymarin. Increased ATF3 expression in both protein and mRNA was observed in silymarin-treated cells. In addition, silymarin increased the luciferase activity of ATF3 promoter. Inhibition of JNK and IκK-α blocked silymarin-mediated ATF3 expression. The results suggest that silymarin induces apoptosis through JNK and IκKα-dependent ATF3 expression in human colorectal cancer cells.
