  • In order to design industrial scale reactors and proceises for multi-phase biocatalytic reactions, it is essential to understand the mechanisms by which such systems operate. To il-lustrate how such mechanisms can be modeled, the hydrolysis of the primary ester groups of triglycerides to produce fatty acids and monoglycerides by lipased (glycerol-ester hydrolase) catalysis has been selected as an example of multiphase biocatalysis. Lipase is specific in its behavior such that it can act only on the hydrolyzed (or emulsified) part of the substrate. This follows because the active center of the enzyme is catalytically active only when the substrate contacts it in its hydrolyzed form. In other words, lipase acts only when it can shuttleback and forth between the emulsion phase and the water phase, presumably within an interphase or boundary layer between these two phases. In industrial applications lipase is employed as a fat splitting enzyme to remove fat stains from fabrics, in making cheese, to flavor milk products, and to degrade fats in waste products. Effective use of lipase in these processes requires a fundamental understanding of its kinetic behavior and interactions with substrates under various environmental conditions. Therefore, this study focuses on modeling and simulating the enzymatic activity of the lipase as a step towards the basic understanding of multi-phase biocatalysis processes.

Effects of Salt and Heparin on the Activation of Lipoprotein Lipase(LPL)

  • Lipoprotein lipase(LPL) is an acylglycerol hydrolase and is the extrahepatic enzyme responsible for the hydrolysis of triglyceride-rich plasma lipoproteins. LPL has been isolated from bovine milk by affinity chromatography on heparin-sepharose in 2M NaCl, 5mM barbital buffer, pH 7.4. Para-nitrophenyl butyrate(PNPB) was used as a substrate for the determination of LPL activity. Molecular weight of LPL was 55KD on 10% SDS-PAGE. When the effects of heparin on LPL activation were compared, LPL activity of heparin added group increased approximately 5 times higher than that of heparin non-added groups. These results indicated that heparin involved in the stabilization of LPL structure that led to increase enzyme activity. Furthermore, LPL activity increased about 4 times compared to the absence of heparin at various pH. LPL was stabilized when heparin was added either low or high salt concentrations. With the presence of heparin, NaCl concentration did not affect LPL activity at pH range 6∼9.

계절에 따라 여러 지역의 원유에서 분리된 내냉성 미생물의 효소 활성 (Enzyme Activity of Isolated Psychrotrophic Bacteria from Raw Milk of Different Regions on Season)

  • 지방분해효소 활성이 가장 높은 균은 가을철에 집유한 B 지역의 원유로부터 분리한 Acinetobacter genomospecies 10(match %: 99.90)이었으며, 단백질분해효소 활성이 가장 높은 균은 봄철에 B 지역의 원유로부터 분리한 Serratia liquefaciens(match %: 99.39)였다. Acinetobacter genomospecies 10(match %:99.90)는 전형적인 생장곡선의 형태인 시그모이드 곡선의 형태를 나타냈으며, 배양시간별 조효소 활성은 배양시간이 증가할수록 지방분해효소 활성이 증가하다가 정지기 후반부에서부터 사멸기에 도달하기까지는 배양시간이 길수록 활성이 감소하였다. pH에 따른 지방분해효소 활성은 pH 8.5에서 가장 높았고 온도는 $45^{\circ}C$에서 가장 높은 활성을 나타내었다. Serratia liquefaciens(match %: 99.39)의 생장곡선은 전형적인 시그모이드 형태를 나타내었고, 단백질분해효소 활성은 배양 12시간 후에 가장 높았고, 18시간 후에는 다시 감소하는 양상을 나타냈다. 단백질 분해효소의 pH 영향은 2.5에서 활성이 가장 낮았고 pH 8.5에서 가장 높은 활성을 나타내었다. 또한 반응온도에 의한 효소활성은 $35^{\circ}C$에서 가장 높았다.

Complete Genome Sequence of Chryseobacterium mulctrae KACC 21234T : A Potential Proteolytic and Lipolytic Bacteria Isolated from Bovine Raw Milk

  • Chryseobacterium mulctrae KACC 21234T is a novel species isolated from raw bovine milk. Psychrotrophic bacteria are considered contaminants and are hypothesized to originate from the environment. In this investigation, the C. mulctrae KACC 21234T genome was determined to be 4,868,651 bp long and assembled into four contigs with a G+C ratio of 33.8%. In silico genomic analyses revealed the presence of genes encoding proteases (endopeptidase Clp, oligopeptidase b, carboxypeptidase) and lipases (phospholipase A(2), phospholipase C, acylglycerol lipase) that can catalyze the degradation of the proteins and lipids in milk, causing its quality to deteriorate. Additionally, antimicrobial resistance and putative bacteriocin genes were detected, potentially intensifying the pathogenicity of the strain. The genomic evidence presented highlights the need for improved screening protocols to minimize the potential contamination of milk by proteolytic and lipolytic psychrotrophic bacteria.

Characterization of Lipid Binding Region of Lipoprotein Lipase

  • Lipoprotein lipase (LPL) I san enzyme that catalyzed the hydrolysis of triacylglycerols of chylomicrons and VLDL to produce 20acylglycerols and fatty acids. The enzyme, LPL, is localized on the surface of the capillary endothelium and is widely distributed in extrahepatic tissues including heart, skeletal muscle and adipose tissue. LPL has been isolated from boving milk by affinity chromatography on heparin-separose in 2 M NaCL, 5mM barbital buffer, pH 7.4. To elucidate the lipid-binding regin, LPL was digested with trypsin and then separated by gel filtration. Lipid binding region of LPL has been investigated by recombining LPL peptides with DMPC vesicles. Proteolytic LPL fragments with DMPC were reassembled and stabilized by cholate. Lipid-binding region of LPL was identified by a PTH-automated protein sequencer, as AQQHYPVSAGYTK. The analysis of the secondary structure of the lipid-binding peptides revealed a higher probability of $\alpha$-helix structure compared to the whole LPL protein. The prediction of hydrophobicity of lipid -binding region was highly hydrophobic (-1.1) compared to LPL polypetide(-0.4).

내냉성 미생물인 Acinetobacter genomospecies 10과 Serratia liquefaciens가 원유의 품질에 미치는 영향 (Effects of Psychrotrophic Bacteria Acinetobacter genomospecies 10 and Serratia liquefaciens on Raw Milk Quality)

  • 살균한 원유에 지방분해효소와 단백질 분해효소활성이 가장 높게 나타났던 균주를 접종하고 저장하면서 내냉성미생물이 생성하는 효소가 원유의 품질에 미치는 영향을 조사하였다. 지방분해효소활성이 높은 Acinetobacter genomospecies 10은 냉장저장기간 중 총 고형분과 유지방의 함량에 영향을 미치지 않았고, 지방의 분해는 냉장보관 14일째에 대조구와 비교하여 2.6배 이상의 유리지방산을 생성하였다. 생성된 지방산의 조성은 short chain free fatty acid(SCFFA), middle chain free fatty acid(MCFFA) 및 long chain free fatty acid(LCFFA)를 모두 생성하였으며, 특히 SCFFA와 MCFFA의 증가율이 높았다. 단백질 분해효소활성이 높은 Serratia liquefaciens은 원유의 총 고형분과 유단백질 함량에 영향을 미치지 않았고, 단백질 조성의 변화는 냉장저장기간 동안 케이신의 함량이 서서히 감소하다가 10일째부터 $\kappa$-케이신의 함량이 현저히 감소하였다. 유리아미노산은 대조구와 비교하여 냉장저장 14일째 2.8배 이상 생성되었으며 특히 소수성 아미노산으로 쓴맛을 내는 leucin, valine 등의 함량이 급격히 증가하였다.

Comparison of Physicochemical and Sensory Properties of Freeze-concentrated Milk with Evaporated Milk during Storage

  • This study was carried out to compare the changes of nutrients, sensory and chemical properties of freeze-concentrated and evaporated milks during storage. For pasteurization, the freeze-concentrated milk containing 27% of total solid was treated with 150 rpm ozone for 5 min, and 99% of microflora was eliminated. Also, the activities of protease and lipase decreased 93.31% and 96.15%, respectively, and phosphatase showed negative activity. Total bacteria count was maintained below$2.0{\times}10^4$CFU/ml. During storage, TBA absorbance was lower in freeze-concentrated milk than that in the evaporated milk. The production of short-chain free fatty acids and free amino acids increased proportionally to the storage period in both samples. While the short-chain free fatty acid production was lower in the freeze-concentrated milk compared with that in the evaporated milk, the production of individual free amino acid was similar in both samples. In sensory evaluation, cooked flavor and color were much lower in the freeze-concentrated milk than that in the evaporated milk. Overall acceptability score was higher in the freeze-concentrated than the evaporated milk. Based on above results, ozone treatment for the freeze-concentrated milk pasteurization was positive at the elimination of microflora and enzyme inactivation. During storage, the freeze-concentrated sample minimized the change of color and TBA absorbance, the production of short-chain free fatty acid and vitamins than the evaporated milk. Therefore, the freeze-concentrated milk process in the present study resulted in the positive effect in minimizing nutrient loss and keeping quality of milk during storage.

  • Most lipids in milk are dispetsed as the form of fat globules. Apical portion of plasma membrane is coated with fat globules, which are synthesized from mammary epithelial cells and then secreted into the lumen. The unique phenomenon in separation of the plasma membrane from the cell is observed only in mammary system. It has been suggested that milk fat globule membrane(MFGM) is formed from endoplasmic reticulum, Golgi apparatus, secretory granule to plasma membrane. For this reason MFGM is important for nuderstanding the structure and function of biological membrane. Because MFGM also plays an important role in inhibition of lipase action, stimulation of nutrient digestion and absorption, emulsion or function as natural liposome, study of the major components in MFGM will provide the opportunity for more broad industrial uses of MFGM in the future.

Lactobacillus plantarum KI134의 생리적 특성 및 이 균에 의한 우유 발효물의 항비만효과 (Physiological Characteristics and Anti-Obesity Effect of Milk Fermented by Lactobacillus plantarum KI134)

  • This study aimed to investigate the physiological characteristics and anti-obesity effects of milk fermented by L. plantarum KI134. The lipase, α-amylase, and α-glucosidase inhibitory activities of milk fermented by L. plantarum KI134 was 94.57±1.25%, 9.44±2.85%, and 2.74±1.24% (10 fold dilution), respectively. L. plantarum KI134 showed higher sensitivity to clindamycin and erythromycin in comparison to sixteen different antibiotics. It demonstrated the highest resistance toward ampicillin and vancomycin. The strain showed higher β-galactosidase, leucine arylamidase, valine arylamidase, acid phosphatase, β-glucosidase, and N-acetyl-β-glucosaminidase activities compared to other enzymes. It also did not produce carcinogenic enzymes, such as β-glucuronidase. The survival rate of L. plantarum KI134 in 0.3% bile was 96.90%. Moreover, the strain showed a 91.45% survival rate at a pH of 2.0. L. plantarum KI134 has resistance to Escherichia coli, Salmonella Typhimurium, Listeria monocytogenes, and Staphylococcus aureus at the rates of 70.00%, 68.18%, 59.05%, and 40.63%, respectively. L. plantarum KI134 (23.01%) showed higher adhesion ability than the positive control (16.32%) L. rhamnosus GG. These results demonstrated that milk fermented by L. plantarum KI134 demonstrated an anti-obesity effect under in vitro conditions, with confirmed potential as a probiotic.

원유에서 분리한 Enterococcus faecalis MD366의 생리적 특성 및 비만 억제 효과 (Physiological Characteristics and Anti-Obesity Effect of Enterococcus faecalis MD366 isolated from Raw Milk)

  • 본 연구는 원유로부터 비만 억제 능력이 있는 젖산균을 분리 및 동정하고, 이 균주의 생리적 특성을 규명하여 상업적으로의 이용가능성을 검토하고자 실시하였다. 이를 위해 Modified MRS 분별배지를 사용하여 노란색 집락을 형성하는 균주를 대상으로 각각 Anti-lipase activity와 Anti-adipogenic activity가 우수한 균주를 선발한 결과, MD366 균주가 최종 선발되었다. MD366 균주는 lipase 억제활성 $65.03{\pm}0.94%$, 지방분화억제활성 $27.4{\pm}1.4%$로나타났으며, 동정결과Enterococcus faecalis로 판명되었고, Enterococcus faecalis MD366으로 명명하였다. E. faecalis MD366의 최적 생장 온도는 $37^{\circ}C$ 이었으며, 담즙산과 산성의 pH에서 모두 우수한 생존력을 나타내었다. 효소활성은 전반적으로 낮았으나 acid phosphatase에 대해 비교적 높은 효소 활성을 나타내었다. 항생제 내성 실험 결과 neomycin, kanamycin, vancomycin에 내성이 있는 반면, novobiocin에 감수성을 나타냈으며, Escherichia coli, Salmonella Typhimurium과 Staphyloccous aureus에 대해 각각 80.4%, 60.2%와 65.4%의 억제 효과를 지니고 있는 것으로 나타났다.
