  • Acid dairy drinks(ADD) are a worldwide product existing in many variations: fruit milk drinks, yogurt drinks, soy milk, butter milk, whey drinks and kefir etc. These drinks are marketed with different shelf lives depending on processing: -Short shelf life(maximum 3 weeks, cold storage) - Long shelf life(2 to 9 months, pasteurized, sterilized or retorted) Acidic protein drinks tend to a separation or destabilization process in the absence of stabilizing system during the shelf life of the ADD. A phase separation results in sedimentation of large particles at the bottom of the package and/or the formation of a serum layer at the top(whey off). These beverages are usually composed of an acid dairy phase (fermented base)or a natural base(milk, soymilk etc.)with an acidic medium (fruit phase: pulp, fruit concentrate etc.) which can be flavored. Sugar and stabilizers are added. It has been proved since the late 1950's that adding high methoxy pectin (HM pectin)to acid milk drinks is the best way to prevent the formation of a sediment and/or the whey off. In this presentation, we explain about stabilization mechanism of ADD induced by pectin. Applications and market trend of ADD in Asia and Europe are explained.

Optimization of the Viability of Probiotics in a Fermented Milk Drink by the Response Surface Method

  • Growth promoters were added to skim milk to retain the viability of Lactobacillus acidophilus and Bifidobacterium longum to help the product meet the "therapeutic minimum" at the time of consumption. The experiments were divided into two parts. The first part of the study used chicory inulin, isomalto-oligosaccharides and sucrose to investigate the effects of sugars on the activity of L. acidophilus and B. longum. The results indicated that the addition of isomalto-oligosaccharides stimulated growth of L. acidophilus and B. longum, resulting in a higher level of the probiotics after one month storage and yielded better $\beta$-galactosidase activity during fermentation. The second part studied the effects of three growth promoters on the viability of the probiotic cultures and the response surface method was employed to find the optimal ratio for addition of the growth promoters. The optimal ratio for added calcium gluconate, sodium gluconate and N-acetylglucosamine in fermented milk drinks were established. The response surface method proved to be a very effective way of optimizing the activity of probiotic cultures when developing a new fermented milk drink.

  • Infanthood is a crucial period when mental and emotional developments take place along with physical growth. Adequate supply of nutrition is essential during this period and there is an urgent need for proper education and guidance on balanced nutrition. Therefore, this study was designed to assess to current status of lactation and to investigate mothers' perspectives on human milk and formular milk. As random samples for this study, a total of 352 mothers having infant under the age of 4 were surveyed. 27.3% of mother surveyed were breast-feeding, 38.0% were formula-feeding and 34.7% were mixed-feeding. According to the result of this study, the percentage of breast feeding was on the decrease, while the dependence on formular milk was on the rise. The main reason of formula-feeding was insufficient secretion of human milk on mothers part. The major factors that affect to select feeding method were employment status, degree of education, family income and age. Also, the interviewees desired the product fortified with special ingredients that are hot contained in formular milk currently available in the market. The most concerned problem of formula milk was indigestion and the interviewees wanted the formular milk to be improved digestibility.

Analysis of Physical Properties and Mechanical Energy Input of Cornmeal Extrudates Fortified with Dairy Products by Carbon Dioxide Injection (탄산가스 주입에 의한 유제품 강화 옥수수 압출성형물의 성질과 기계적 에너지 투입량의 분석)

  • Selected physical properties of cornmeal extrudates fortified with dairy products and extrusion process by $CO_2$ gas injection were analysed. Dairy products including whole milk powder, whey protein concentrate non-fat dry milk, and sodium caseinate were tested at the addition of 10% and 20%, based on cornmeal weight. $CO_2$ gas was injected to the barrel at the pressure of 0.7 and 1.4 MPa. Specific mechanical energy (SME) input was decreased by the addition of dairy products. Sodium caseinate had a little effect on decreasing the SME input, however whole milk powder tremendously reduced SME input when the concentration increased. An increase in milk product content resulted in increasing the piece density at the injection pressure of 0.7 MPa. At both 10% and 20% milk product content, the piece density was lowest at the injection pressure of 0.7 MPa. The sectional expansion index was highest at the injection pressure of 0.7 MPa. However, the specific length was constantly increased with the increase in $CO_2$ injection pressure. Water absorption index was decreased and water solubility index was increased by the addition of milk products. The injection pressure of $CO_2$ was optimum at 0.7 MPa. The addition of whole milk powder limited to puff the melt, but the other milk products tested resulted in puffing with $CO_2$ injection to 1.4 MPa.

Shelf Life of Freeze Dried Product of Lactic Acid Bacteria Fermented Food Prepared from Milk or Egg White Powder (우유 또는 난백분말로 만든 젖산균발효식품을 동결건조한 제품의 저장성)

  • Freeze dried products of lactic acid bacteria fermented food prepared from milk or egg white powder(EWP) were stored at $28^{\circ}C.\;5^{\circ}C$ and $-18^{\circ}C$ for 20 weeks. Properties of stored, freeze dried product and viable cell count. pH and organoleptic properties of stored, reconstituted product were investigated. (1) The viable cell count of reconstituted milk or EWP product stored at $5^{\circ}C\;or\;-18^{\circ}C$ was not changed markedly. However, the viable cell count of milk or EWP product stored at $28^{\circ}C$ was reduced during storage and it was changed substantially between 4 weeks and 5 weeks. However, pH of all samples stored at three different temperature was not changed. (2) Color of freeze dried product prepared from EWP became clearly brown at 16 weeks. (3) Appearance of reconstituted milk product stored at $5^{\circ}C\;or\;-18^{\circ}C$ for 20 weeks was not changed. However, homogeneity and solubility of reconstituted milk product stored at $28^{\circ}C$ for 20 weeks were reduced. Taste, odor and texture of reconstituted milk product stored at $28^{\circ}C$ for 20 weeks were markedly changed. (4) Viscosity of reconstituted EWP product stored for 20 weeks was slightly reduced. Solubility of reconstituted EWP product stored at $28^{\circ}C$ for 20 weeks was reduced and its taste and odor were markedly changed. Texture of reconstituted EWP product stored at $28^{\circ}C$ became rough.

Electrophoretical Properties of Transglutaminase Treated Milk Product Powders (Transglutaminase를 처리한 분말 유제품의 전기영동적 특성)

  • This study was performed to understand the behavior of protein mobility and intensity of enzymatic hydrolysis according to crosslinking of sodium caseinate, whey protein isolate, skim milk and whole milk powders with or without transglutaminase (TGase, w/w = 200 : 1) at $38^{\circ}C$. Whey protein was limited to crosslinking and skim milk was relatively more increased in high molecular polymer than whole milk. The degree of crosslinking decreased in the order of sodium caseinate>skim milk>whole milk>whey protein isolate. The SDS-PAGE results indicated that main bands of TGase treated samples had a high mobility and formed bands of molecular weights below 15 kDa by hydrolysis with pepsin after 10 min of reaction time. However, ${\beta}-lactoglobulin$ showed remarkable stability against pepsin hydrolysis treated with and without TGase. The high molecular polymers were easily hydrolyzed with digestive enzymes in vitro experiment. These results suggested that novel dairy products using TGase would have no special digestive problem in human body.

  • In Korea, there are a couple of risk assessment organizations: The Animal Plant and Fisheries Quarantine and Inspection Agency(QIA) and the Korea Food and Drug Administration (KFDA). The major food laws include the Food Sanitation Act (FSA), the Livestock Product Processing Act (LPPA), and the Agricultural Products Quality Control Act (APQCA). Milk and dairy products are mostly controlled by the Food Sanitation Act and the Livestock Product Processing Act. This study was carried out to estimate the current standard methods of foodborne pathogens for dairy products, comparing the Livestock Products Processing Act with the Food Sanitation Act. The standard methods of foodborne pathogens for dairy products are composed by growth culture, isolation culture, and identification, however, standard methods of QIA and KFDA are different at the using of medium and inspection stage. Therefore, consolidation of risk management and risk assessment methods are regarded important to provide safe dairy products to consumer.

Application of Response Surface Methodology (RSM) for Optimization of Anti-Obesity Effect in Fermented Milk by Lactobacillus plantarum Q180

  • Obesity, a condition in which an abnormally large amount of fat is stored in adipose tissue, causing an increase in body weight, has become a major public health concern worldwide. The purpose of this study was to optimize the process for fermented milk for the production of a functional product with an anti-obesity effect by using Lactobacillus plantarum Q180 isolated from human feces. We used a 3-factor, 3-level central composite design (CCD) combined with the response surface methodology (RSM). Concentration of skim milk powder (%, $X_1$), incubation temperature ($^{\circ}C$, $X_2$), and incubation time (h, $X_3$) were used as the independent factors, whereas pH (pH, $Y_1$), anti-lipase activity (%, $Y_2$) and anti-adipogenetic activity (%, $Y_3$) were used as the dependent factors. The optimal conditions of fermented milk for the highest anti-lipase and anti-adipogenetic activity with pH 4.4 were the 9.5% of skim milk powder, $37^{\circ}C$ of incubation temperature, 28 h of incubation time. In the fermentation condition, the predicted values of pH, anti-lipase activity and anti-adipogenetic activity were 4.47, 55.55, and 20.48%, respectively. However, the actual values of pH, anti-lipase activity and anti-adipogenetic activity were 4.50, 52.86, and 19.25%, respectively. These results demonstrate that 9.5% of skim milk powder and incubation at $37^{\circ}C$ for 28 h were the optimum conditions for producing functional fermented milk with an anti-obesity effect.

Production of a Mozzarella Cheese Analogue by Using Raw Milk and Soy Milk Blends (원유를 두유에 첨가한 혼합액으로 Mozzarella Cheese Analogue 제조에 관한 연구)

  • The study was conducted to evaluate the feasibility of Mozzarella cheese analogue generation by using a mixture of soy milk and raw milk and to compare the quality of the resultant cheese with that of Mozzarella cheese manufactured using the traditional method. The mixtures showed increase in protein and decrease in lactose and SNF in a dose-dependent manner with the addition of soy milk. The Mozzarella cheese analogue had lower total solids content than the control cheese product, while the fat content was similar between both. The analogue cheese had lower ash content than the traditionally prepared cheese; the content was proportional to the amount of soy milk in the mixture. Higher soy milk quantities within mixtures also resulted in proportionally higher levels of fat content within analogue cheese. Water-soluble nitrogen content was lower in the analogue cheese than in the control cheese. While the WSN level increased in the control cheese, it was almost constant in the analogue cheese. The control cheese had much higher actual and predicted yield than the analogue cheese, while the analogue cheese had a higher stability level. The control cheese had a higher transfer rate than the analogue cheese, with the exception of lactose. Electrophoresis analysis showed bands for Mozzarella cheese analogues that were present in addition to the normal ${\alpha}$-casein and ${\beta}$-casein bands. Physical characteristic analysis showed that hardness was affected by the addition of soy milk to cheese, while cohesiveness and brittleness were affected by the addition of raw milk, and elasticity was barely affected by milk composition. The meltability of the control cheese was higher than that of the analogue cheese and increased during 30 days of storage at $4^{\circ}C$. Browning, oiling-off, and stretching characteristics were almost identical between the 2 types of cheeses. Sensory evaluation findings showed that the control cheese had much better body texture, appearance, and flavor than the analogue cheese.

  • The optimum conditions for lactic acid fermentation by Lactobacillus acidophilus in the media containing malt syrup, skim milk or skim milk and malt syrup were studied to develope marketable fermented beverages from barley. The optimum sugar concentration of malt syrup was $10^{\circ}$ Bx. Addition of skim milk to malt syrup enhanced bacterial growth and acid formation. The best flavor of the product was obtained from the media containing 5% skim milk and sterilized at $90^{\circ}C$ for 30 minutes. Optimum temperature and pH for the formentation were $40^{\circ}C$ and 6.0, and bacterial cells grew logarithmically upto 16 hours.
