  • An OTHBVS cell line from HepG2 was established. This cell line stably expresses the human hepatitis B virus (HBV) middle S protein that includes the preS2 region which is important for HBV particle entry into the hepatocyte. To establish this cell line, the middle S open reading frame (ORF), with a promoter located in the 5' region and enhancer located in the 3' region, was cloned downstream from the metallothionine (MT) promoter of the OT1529 vector. In this vector, expression of the middle S protein was constructed to be regulated by its own promoter and enhancer. Expression of the large S protein which contains the preS1 region in addition to the middle S protein was designed to be regulated by the MT promoter. When extracts of OTHBVS cells were examined with an S protein detection kit (RPHA, Korea Green Cross Co.), an S protein was detected. Total mRNA of OTHBVS cell examined by northern blot analysis with an S ORF probe revealed small/middle S transcripts (2.1 kb). When the MT promoter was induced by Zn, large S transcripts (2.4 kb) were detected. The GP36 and GP33 middle S proteins were presumably detected, but large S proteins were not detected by immunostain analysis using anti-preS2 antibody.

  • Controlling the production of diverse cell/tissue types is essential for the development of multicellular organisms such as animals and plants. The Arabidopsis thaliana root, which contains distinct cells/tissues along longitudinal and radial axes, has served as an elegant model to investigate how genetic programs and environmental signals interact to produce different cell/tissue types. In the root, a series of asymmetric cell divisions (ACDs) give rise to three ground tissue layers at maturity (endodermis, middle cortex, and cortex). Because the middle cortex is formed by a periclinal (parallel to the axis) ACD of the endodermis around 7 to 14 days post-germination, middle cortex formation is used as a parameter to assess maturation of the root ground tissue. Molecular, genetic, and physiological studies have revealed that the control of the timing and extent of middle cortex formation during root maturation relies on the interaction of plant hormones and transcription factors. In particular, abscisic acid and gibberellin act synergistically to regulate the timing and extent of middle cortex formation, unlike their typical antagonism. The SHORT-ROOT, SCARECROW, SCARECROW-LIKE 3, and DELLA transcription factors, all of which belong to the plant-specific GRAS family, play key roles in the regulation of middle cortex formation. Recently, two additional transcription factors, SEUSS and GA- AND ABA-RESPONSIVE ZINC FINGER, have also been characterized during ground tissue maturation. In this review, we provide a detailed account of the regulatory networks that control the timing and extent of middle cortex formation during post-embryonic root development.

  • The softening of the femented vegetables and the fruits was resulted from the degradation of pectin substances, cellulose, hemicellulose by polygalacturonase(PG), pectinesterase(PE), Cx-cellulase, $\beta$-galctosidase. The conversion of insoluble pectin to soluble pectin in cell wall-middle lamella was a major factor in the changes of firmness. Ca2+ was substantially increased firmness. However, Ca2+ could be removed from cell wall by chelating agents such as oxalic acid and citric acid. And Ca2+ was replaced with Na+ by ion exchange reaction. Ca2+ deficient tissue was vulnerable to attack by PG. Preheating treatment and Ca2+ addition is most effective in inhibiting the vegetable food softening and in increasing middle lamella-cell wall regidity, which PE activation by preheating treatment and Ca2+ addition could created more anionic carboxyl groups for cationic materials binding such as Ca2+ and chitosan and for polypectategel formation. Excessive demethylation by PE was associated with loosening of middle lamella cell wall components and softening.

  • The purpose of this study was to analyze differences in psychological characteristics according to cell phone dependency and use purpose in cell phone users of adolescences. Psychological characteristics of adolescents were consisted of attention aggression, somatic symptoms, social withdrawal, depression variables. The subjects were 2,158 cell phone owned second graders of middle school. A data of the Korean Child-Youth Panel Survey(KCYPS) was employed for the this study. Dependency and use purpose of cell phone were classified by cluster analysis into four groups with similar characteristics : 'high dependency-multipurpose', 'low dependency-social', 'low dependency-unsocial' 'middle dependency-peer interaction' groups. Major findings were that: the group differences in psychological characteristics were statistically significant. The 'low dependency-social' group showed highest on adjustments, and the other groups were maladaptive. Finally, implications for future research were discussed.

  • Urticating spicules and poison -secreting cells of the last instar larva in Euproctis flava BREMER was studied histologically. Three kinds of cells in the epidermis of tubercles on the lst to 8th abodominal segments are classified according to the arrangement of their nuclei : smallepidermal cells, large gland cells, and elongated trichogen cells. As a result of Mallory's triple straining , the epicuticle , the papila-like structure apart form the tubules inside which are gathered at the base and connected with a middle layer cell through a canal in the cuticle, and the peripheral of the urticating spcicule are yellow. However, the inside of the spicule , the tubules within the papilla-like structure, the canal in the cuticle , nuclei in the pidermal cells and the thin exocuticle are red although the thich endocuticle is blue. Particularly , the large nuclei in the middle layer cells are bright red, the cytoplasms of which are little and stained red, too, and the inside of the spicules apt to be stained red when they are broken. The contents therefore seem to be continuous between the spicules and the large cells. Presumably , the large cell at the middle layer is not te tormogen cell which Tsutsumi (1958) has described , but the gland cell which secretes the poison-substance into spicules as Pawlowsky and Stein 91927) and Tonkes (1933) pointed out. Whether the pisonous substance is secreted from the gland cell into the cytoplasmic processes of the trichogen cells which stick large middle layer cells during the formation of the new spicule as Tsutsumi (1958) has observed, or the gland cell makes a new connection with the spicule after the spicule is formed is not clear.
