레이저 광 산란을 이용한 검출 시스템 및 레이저를 이용한 광학적 검출에 있어서 높은 발광 강도를 통해 궁극적으로 높은 효율의 광 산란 신호를 광검출기에서 얻기 위해서는 발광 레이저빔을 미세유체 칩의 채널 중앙에 집광하는 것이 매우 중요하다. 본 논문을 통해 레이저 광 산란을 이용한 세포 검출을 위해 PDMS 마이크로렌즈가 집적화된 PDMS 미세유체 칩을 소개하고자 한다. 기존에 제작된 PDMS 미세유체 칩 위에 간편히 정렬하여 올려놓아 사용함으로써 검출 효율을 증가시킬 수 있는 PDMS 마이크로렌즈를 제작하였다. PDMS 마이크로렌즈는 포토레지스트 리플로우와 PDMS 복제 몰딩에 의해 제작되었다. 이 제작 방법은 간단하며 높은 치수 정확성 및 좋은 마이크로렌즈의 성능을 제공한다. PDMS 미세유체 칩 위에 집적화된 PDMS 마이크로렌즈가 적혈구를 이용한 레이저 광 산란을 통한 세포 검출 실험에서 레이저 강도를 증가시켜 신호대잡음비 및 감도를 증가시킴을 검증하였다.
본 연구에서는 직경이 수백 nm로부터 수 ${\mu}m$에 이르는 균일한 크기의 액적을 생성하는 마이크로 플루이딕 플랫폼이 설계되었다. 미세한 액적을 생성하기 위하여 T-정션과 유동집속 장치가 마이크로�a푸이딕 채널로 통합되었다. 상대적으로 큰 수성 액적들이 상류의 T-정션에서 생성되어 유동집속 장치로 이송되는데, 여기에서 각각의 액적은 압력과 점성응력의 작용에 의하여 목표로 하는 크기로 잘게 쪼개진다. 이러한 구성은 내부 유체의 매우 느린 유량과 유동집속 영역에서 내부 및 외부 유체 사이의 높은 유량비를 가능하게 한다. 본 마이크로플루이딕 장치는 약 $1\;{\mu}m$ 크기의 직경을 가지는 액적들을 3%보다 작은 표준 편차로 생성할 수 있음이 제시되었다.
The real-time enrichment and detection of pathogens are serious issues and rapidly evolving field of research because of the ability of these pathogens to cause infectious diseases. In general, bacterial detection is accomplished by conventional colony counting or by polymerase chain reaction (PCR) after DNA extraction. As colony counting requires considerable time to cultivate, PCR is an attractive method for rapid detection. A small number of pathogens can cause diseases. Hence, a pretreatment process, such as enrichment is essential for detecting bacteria in an actual environment. Thus, in this study, we developed a microfluidic chip capable of performing rapid enrichment of bacteria and the extraction of their genes. A lectin, i.e., Concanavalin A (ConA), which shows binding affinity to the surface of most bacteria, was coated on the surface of magnetic particles to nonspecifically capture bacteria. It was subsequently concentrated through magnetic forces in a microfluidic channel. To lyse the captured bacteria, magnetic particles were irradiated by a wavelength of 532nm. The photo-thermal effect on the particles was sufficient for extracting DNA, which was consequently utilized for the identification of bacteria. Our device will help monitor the existence of bacteria in various environmental situations such as water, air, and soil.
본 논문은 미세유체채널에 채워진 유체를 이용하여 위상응답을 제어하는, 잉크젯 프린터로 인쇄된 미세유체채널 복합 좌 우향 전송선로(CRLH TL: Composite Right/Left Handed Transmission Line)를 제안한다. 제안된 CRLH TL은 종이 위에 은 나노입자 잉크를 이용하여 인쇄되었으며, Poly Methyl Methacrylate(PMMA)에 레이저 식각을 이용하여 제작된 미세유체채널은 잉크젯 프린터로 인쇄된 접착물질인 SU-8을 이용하여 CRLH TL 위에 부착되었다. 제안된 CRLH TL은 미세유체채널에 채워진 유체에 따라서 위상응답을 변화시킬 수 있으며, 미세유체채널에 각기 다른 유체가 흐를 때, 900 MHz에서 -10 dB 이하의 반사계수를 유지한 상태로 위상 지연, $0^{\circ}$ 위상, 위상 앞섬 특성을 모두 나타낼 수 있음을 확인하였다. 제안된 CRLH TL의 성능은 시뮬레이션 결과와 측정 결과를 통하여 성공적으로 증명되었다.
액적-기반 미세유체장치는 물질 합성 및 초고속 대용량 스크리닝 등 다양한 응용분야에서 변형 가능한 새로운 접근법을 이끌어 냈다. 그러나 단일의 액적생성기를 이용한 액적의 생성 속도가 매우 낮기 때문에 이를 상용화 하기 위해서는 생산속도를 높이기 위한 노력이 필요하다. 본 연구는 단일의 유동-집속 생성기를 병렬로 연결하여 단분산성 액적의 생성 속도를 높이는 방법에 관한 것이다. 이러한 액적생성기를 갖는 미세유체장치를 제작하기 위해 본 연구에서는 양면 임프린팅 방법을 이용하여 단층 엘라스토머 조각에3차원의 마이크로 채널을 갖는 3D 모놀리식 탄성중합체 장치(monolithic elastomer device, 3D MED)를 제작 할 수 있다. 이렇게 제작된 8개의 액적생성기가 연결된 3D MED를 이용하여 연속상과 분산상의 유체를 조절하여 단분산성 액적의 형성속도가 향상되었음을 증명하였다. 따라서 본 미세유체시스템을 사용하여 다양한 재료 또는 세포들을 함유하는 단분산성 액적을 형성하여 마이크로입자 제조 및 스크리닝 시스템과 같은 넓은 분야에 활용될 수 있을 것으로 기대된다.
There is a great demand to manipulate biological cell autonomously since biologist should spend much time to obtain skillful manipulation techniques. For this purpose, we propose a cell chip to control, carry, fix and locate the cell. In this paper, we focus on the cell rotator to rotate individual biological cell based on a micro fluidics technology. The cell rotator consists of injection hole and rotation well to rotate a biological cell properly. Under the variation of flow rate in injection hole, the angular velocity of a biological cell is evaluated to find the feasibility of the proposed rotation method. As a practical experiment, Zebrafish egg is employed. Based on this research, we find the possibility of non-contact rotation way that can highly reduce the damage of the biological cell during manipulation. To realize an autonomous biological cell manipulation, a cell chip with manipulation well and micro channel in this research will be utilized effectively in near future.
A method based on transient shear flow dynamics of red cell aggregates was developed to investigate reversible re-aggregation processes with decreasing shear flow. In the microchannel-flow aggregometry, the aggregated red blood cells that are subjected to continuously decreasing shear stress in microchannel flow were measured with the use of a laser-scattering technique. Both the laser-backscattered intensity and pressure were simultaneously measured with respect to time, resulting in shear stress ranging from $0{\sim}35\;Pa$ for a time period of less than 30 seconds. The time dependent recording of the backscattered light intensity (syllectogram) yielded an upward convex curve with a peak point, which reflected the transition threshold of aggregation in the RBC suspensions. Critical-time and critical-shear stress corresponding to the peak point were examined by varying the initial pressure-differential and the micro channel depth, and these results showed good potential for being used as new aggregation indices. In the present study, these newly proposed indices were also validated by differentiating the effect of fibrinogen on RBC aggregation and then these indices were compared to the conventional indices that were measured by a rotational aggregometer.
본 연구에서는 부피가 다른 두 유체의 효과적인 유동제어를 위하여 Stokes 유동 방정식을 기반으로 설계된 미세채널을 제안한다. 부피가 다른 두 유체가 압력 구동에 의하여 주입구를 통과하는 과정에서, 두 유동이 동시에 만나 주어진 부피비를 유지하며 흘러나간 후에 일시에 끝날 수 있도록 주입구의 폭과 길이를 조절하였다. 디자인된 미세채널은 비압축성, 뉴턴 유체의 과도상태 유동에 대한 전산유체역학 모사를 수행하였다. 또한, 소프트리소그래피를 통해 미세유체칩을 제작하고, 압력 구동에 의한 부피가 다른 두 유체의 유동 특성을 평가하였다.
Most microfluidic devices such as heat sinks for cooling micro-chips, DNA chip, Lab-On-Chip, and micro pumps etc. have microchannels of various size. Therefore, the design of practical microfluidics demands detail information on flow structure inside the microchannels. However, detail velocity field measurements are rare and difficult to carry out. In addition, as the microfluidics expands, accurate understanding of microscale transport phenomena becomes very important. In this research, micro-PIV system was employed to measure the velocity fields of flow inside a micro-channel. We carried out PIV measurements for several microchannels with varying channels width, inlet and outlet shape, filters, CCD camera and ICCD camera, etc. For effective composition of micro-PIV system, first of all, it is essential to understand optics related with micro-imaging of particles and the particle dynamics encountered in micro-scale channel flows. In addition, it is necessary to find the optimal condition for given experimental environment and? micro-scale flow to be investigated. The problems encountered in measuring velocity field of micro-channel flows are discussed in this paper.
파우더 블라스팅은 미세 유리가공법으로서 가공속도가 빠르고 저비용의 장점이 있지만 유리를 취성파괴 시키기 때문에 표면거칠기가 좋지않다. 블라스팅된 표면에 저압의 워터젯을 분사하여 표면에서의 연마 슬러리의 흐름을 통해 표면거칠기를 저감할 수 있다. 본 연구에서는 소다라임 유리에 블라스팅으로 마이크로 채널을 가공한 후 워터젯을 연속 적용하고, 마이크로 채널의 표면거칠기 및 단면 형상의 변화의 과정을 관찰하였다. 워터젯의 적용결과, 초기단계에서는 블라스팅에 의한 미세 요철이 제거되었고, 이후 표면하부의 크랙이 제거되어 평균 표면거칠기 50 nm 근방의 매끈한 표면을 얻을 수 있었다. 표면거칠기 저감에 동반하여 채널단면의 확장 과정도 함께 관측하였다. 마지막으로 제안한 방법에 의해 미세유체칩의 가공 결과를 제시하였다.
