Functional Properties of the Microcrystalline Collagen Manufactured from Raw Pigskins (생돈피에서 제조된 미세결정성 콜라겐의 기능적 성질)

  • Microcrystalline collagen was manufactured from raw pigskins and its functional properties were measured. It showed a thixotropic behavior. The maximum viscosity was obtained at pH 3.5 and the viscosity increased with the increase of concentration in a nonlinear manner. The increase in temperature decreadsed the viscosity while the effect of temperature was greater at pH below 4.5. Foaming capacity was similar to that of gelatin but superior to that of egg white at 1%. The foam stability was inferior to that of egg white. Emulsifying capacity was lower than that of pork. Values from physical and chemical analysis were somewhat different from those of medical-grade microcrystalline collagen made from bovine cerium collagen.

The Brown-Rot Basidiomycete Fomitopsis palustris Has the Endo-Glucanases Capable of Degrading Microcrystalline Cellulose

  • Two endoglucanases with processive cellulase activities, produced from Fomitopsis palustris grown on 2% microcrystalline cellulose(Avicel), were purified to homogeneity by anion-exchange and gel filtration column chromatography systems. SDS-PAGE analysis indicated that the molecular masses of the purified enzymes were 47 kDa and 35 kDa, respectively. The amino acid sequence analysis of the 47-kDa protein(EG47) showed a sequence similarity with fungal glycoside hydrolase family 5 endoglucanase from the white-rot fungus Phanerochaete chrysosporium. N-terminal and internal amino acid sequences of the 35-kDa protein(EG35), however, had no homology with any other glycosylhydrolases, although the enzyme had high specific activity against carboxymethyl cellulose, which is a typical substrate for endoglucanases. The initial rate of Avicel hydrolysis by EG35 was relatively fast for 48 h, and the amount of soluble reducing sugar released after 96 h was $100{\mu}g/ml$. Although EG47 also hydrolyzed Avicel, the hydrolysis rate was lower than that of EG35. Thin layer chromatography analysis of the hydrolysis products released from Avicel indicated that the main product was cellobiose, suggesting that the brown-rot fungus possesses processive EGs capable of degrading crystalline cellulose.

Effect of a Nonionic Surfactant on the Adsorption and Kinetic Mechanism for the Hydrolysis of Microcrystalline Cellulose by Endoglucanase Ⅰ and Exoglucanase II

  • Effect of a nonionic surfactant, Tween 20 on the adsorption and kinetic mechanism for the hydrolysis of a microcrystalline cellulose, Avicel PH 101, by endoglucanase Ⅰ (Endo Ⅰ) and exoglucanase Ⅱ (Exo Ⅱ) isolated from Trichoderma viride were studied. The Langmuir isotherm parameters, amount of maximum adsorption (Amax) and adsorption equilibrium constant (Kad) for the adsorption, were obtained in the presence and the absence of nonionic surfactant. On the addition of Tween 20, the Kad and Amax values of Exo Ⅱ were decreased, while those of Endo Ⅰ were not affected. These indicate that the adsorption affinity of Exo Ⅱ on the cellulose is weakened by nonionic surfactant, and the surfactant enhanced desorption of Exo Ⅱ from insoluble substrate. The enzymatic hydrolysis of the cellulose can be described by two parallel pseudo-first order reactions using the percentages of easily (Ca) and hardly (Cb) hydrolyzable cellulose in Avicel PH 101 and associated rate constants (ka and kb). The Ca value was increased by adding Tween 20 for all enzyme samples (Exo Ⅱ, Endo Ⅰ and their 1:1 mixture) implying that the low-ordered crystalline fraction in the cellulose may be partly dispersed by surfactant. The ka value was not affect by adding Tween 20 for all enzyme samples (Exo Ⅱ, Endo Ⅰ and their 1:1 mixture). The kb value of Exo Ⅱ was increased by adding Tween 20, while that of Endo Ⅰ was not affected. This suggests that the surfactant helps the Exo Ⅱ desorb from microcrystalline cellulose, and increase the hydrolysis rate. These results were show that the increase of hydrolysis of cellulose by the nonionic surfactant is due to both the activation of Exo Ⅱ and partial defibrillation of the cellulose.

Surface Modification of Microcrystalline Cellulose (MCC) Filler for CO2 Capture (CO2 흡착 충전제 제조를 위한 microcrystalline cellulose (MCC) 입자 표면개질연구)

  • In this study, we performed surface modification of biodegradable microcrystalline cellulose (MCC) to use as a filler in polyethylene (PE) composite in food packaging application. We modified MCC surface with (3-trimethoxysilylpropyl)diethylenetriamine (TPDT) silane coupling agent, which has one primary amino group and two secondary amino groups per molecule, to introduce amino groups with a carbon dioxide adsorption capability in MCC. Effects of each of the reaction conditions such as amount of TPDT introduced, swelling time, reaction temperature, and reaction time on surface modification degree of MCC were investigated by changing a variety of above reaction conditions. The amount of TPDT grafted on MCC surface and formation of chemical bonds were confirmed by Fourier transform infrared spectroscopy (FT-IR), elemental analysis (EA), X-ray photoelectron spectroscopy (XPS), thermogravimetric analysis (TGA) and solid state $^{29}Si$ nuclear magnetic resonance (NMR) spectroscopy. We confirmed increase of grafted amount of TPDT on MCC with increasing reaction time, reaction temperature, and amount of introduced TPDT.

  • Chitin is known as biodegradable natural polymer. In spite of various application of chitin derivatives from waste marine sources, commercial use of chitin has been limited due to high resistance to chemicals and the absense of proper solvents. We chitin prepared through the decalcification, bleaching and deproteination from Protunus trituberculatus shells by change of Hackman's method. Also, Microcrystalline chitin made by hydrolysis that was reduce made of resistance solvents used by dilute hydrochloric acid, ultrasonic and hydrogen peroxide. Crosslinked chitin derivatives were preparaed from chitin with crosslink agents(epichlorohydrin, 1,3-dichloropropanol) follwed by crosslinkage at 6C position. The effects of these parameters on chitin dervatives were invastigated by IR, DSC, XRD, BET, PSA and SEM. SEM analysis showed that both chitin and crosslinked chitin had a particle shaped morphology.

  • We review our studies on the growth of microcrystalline silicon films by the standard PECVD technique. In situ spectroscopic ellipsometry studies allow the optimization of the complex film structure with respect to competing aspects of the growth process. Fine tuning the hydrogen flux, the ion energy, and the nature of the species contributing to deposition produces unique films with a fully crystallized interface with silicon nitride. These materials have been successfully incorporated in bottom gate TFTs which present mobility values in the range of 1 to 3 $cm^2/V.s$, and stable characteristics when submitted to a bias stress. The stability of these TFTs makes them suitable for driver applications in AMLCDs as well as pixel elements in OLED displays.

  • Hot filament CVD 방법에서 가스압을 증가시키는 방법을 사용하여 nanocrystalline 다이아몬드 막을 합성하였다. 메탄-수소 혼합가스를 사용하고 메탄함량, 유량, 기판온도합성시간은 각각 4%, 100sccm, 110$0^{\circ}C$, 10시간으로 일정하게 유 였다. 합성 변수로서 가스압을 40 Torr에서 300 Torr 구간에서 변화시켰다. High-resolution SEM으로 막 표면의 형상을 관찰하고, TEM, XRD, micro-Raman spectroscopy를 사용하여 합성된 막의 구조 및 특성을 분석하였다. 합성된 다이아몬드 막은 압력이 높아짐에 따라 mocrocrystalline 다이아몬드 막에서 점진적으로 nanocrystalline 다이아몬드 막으로 변화해갔으며, 가스압에 다라 비다이아몬드 상의 량이 증가하였다. 증착속도는 microcrystalline 다이아몬드 막이 형성되는 구간에서는 압력에 따라 1.1~1.3 $\mu\textrm{m}$/h까지 증가하다가 nanocrystalline 다이아모느 막이 형성되는 구간에서는 압력에 따라 감소하였다. 감소하였다.
