고온성 종균제를 이용한 제지폐수 처리 (A Study on the Treatment of Paper Mill Wastewater with the Addition of High Thermal Microbial Inoculants)

  • The Wastewater of toilet paper mill recycling recovered milk carton was used as a raw material for this study. According to the actual mill conditions, hydraulic retention time was adjusted to 12 hours and F/M (Food/Micro-organism) ratio was adjusted to 0.23. Temperature of aeration basin was varied from 2$0^{\circ}C$ to 5$0^{\circ}C$. The change of Micro-organisms and removal efficiency of pollutant were investigated at the varied temperature of basin. Aeration basin using high thermal microbial inoculants showed more removal efficiency of SS, COD than aeration basin using conventional microbial inoculants at high temperature. Floc consolidation of aeration basin using high thermal microbial inoculants added sludge was better than that of sludge from aeration basin using conventional microbial inoculants.

  • Ozone has the advantages for the strong oxidant agents. Ozone is widely used as a disinfectant and deodorant in water treatment and biosafety cabinets. We attempted deodorization and disinfection in the piggery using ozone treatment. It was found that ozone affects deodorization of ammonia and decrease of microbial organism in the air. Concentration of ammonia decreased 50 percents and microbial organisms in the air were decreased below 10 percents. It will be useful for the application of the ozone to environmental treatment in the piggery.

  • To analyze the riverine microbial community structure, genetic fingerprints and ecological indexes such as species abundances, diversity, evenness, dominance of targeted rivers in Gyeonggi Province were acquired and evaluated using terminal restriction fragment length polymorphism (T-RFLP) technique. Genetic fingerprinting technique such as T-RFLP, which is able to show the microbial community clearly unlike traditional culture-dependent techniques, was thought to be useful to analyse the riverine microbial ecosystem under various factors. Riverine ecosystem evaluation using visible organisms would give biased results with time, targeted organism and researcher. But, T-RFLP, which can exclude the subjected biases such as culture condition and identification, would be an option to understand natural ecosystem by including the microorganisms that defy culture but perform important functions.

Detection of microbial organisms on Apis mellifera L. beehives in palm garden, Eastern Thailand

  • Background: Honey bees play a crucial role in pollination and ecological balance. Apis mellifera L. colonies, especially those located in specific geographic regions, such as the palm garden in Eastern Thailand, are susceptible to potential threats from microbial contaminants. Understanding and detecting microbial organisms in these beehives is essential for the preservation of bee health, honey production, and the broader ecosystem. However, the problem of microbial infection and antibiotic-resistant bacteria is more severe and continuously increasing, resulting in a health, economic, and social crisis. The purpose of this study is to determine the prevalence of microorganisms in A. mellifera beehives in palm gardens in Rayong province, Eastern Thailand. Results: Ten swabs in transport media were swabbed and obtained from different parts of each beehive (1 swab per beehive), for a total of 10 hives. Traditional microbial culture-based methods, biochemical tests, and antimicrobial susceptibility (disc-diffusion) tests were used to detect microbial organisms and antibiotic resistance in bacteria. The swab tests from nine beehives resulted in the detection of Gram-positive bacteria (63.64%), Gram-negative bacteria (27.27%), and fungi/yeast (9.09%). These microorganisms are classified as a group of coagulase-negative Staphylococcus spp. and made up 40.91% of the bacteria discovered. Other bacteria found were Coryneform bacteria (13.64%), Pantoea spp. (13.64%), Bacillus spp. (9.09%), yeast (9.09%), glucose non-fermentative Gram-negative bacilli (9.09%), and Pseudomonas spp. (4.55%). However, due to the traditional culture-based and 0biochemical tests usually used to identify the microbial organisms in clinical specimens and the limitation of identifying some environmental microbial species, the results of the antimicrobial susceptibility test cannot reveal if the organism is resistant or susceptible to the drug. Nevertheless, drug-sensitive inhibition zones were formed with each antibiotic agent. Conclusions: Overall, the study supports prevention, healthcare, and public health systems. The contamination of microorganisms in the beehives may affect the quality of honey and other bee products or even the health of the beekeeper. To avoid this kind of contamination, it is therefore necessary to wear personal protective equipment while harvesting honey and other bee products.

  • The genera Bifidobacterium is a member of the normal intestinal flora in humans, and important in food industry. In order to test the genetic identity of this bacterial genera, four primers originated from rice genome (SRILS Microbial $UniPrimers^{TM}$ kit) were used in molecular typing of 7 Bifidobacterial species and 20 isolates from various source. SRILS Microbial $UniPrimers^{TM}$ kit were effectively applied to genomic fingerprinting of various organism such as plant, animal and microorganism. (omitted)

  • 비교적 효율적으로 이용되고 있지 않는 섬유성물질의 이용성을 높이고 섬유소 자화세균에 의한 섬유소 단세포단백을 생산할 목적으로 전보에서 분리, 동정한 Collulomonas flavigena GFB 24-1의 몇 가지 균학적 성질을 통한 이용성을 검토한 결과를 요약하면 다음과 같다. 1. 섬유질 자화세균 생육배지에서 배양한 결과 pH 7.0에서 50시간 배양하였을 때 균체증식이 가장 좋았고, 배양시 유리되는 단백질은 배양중 시간이 경과됨에 따라 증가하였다. 2. Cellulolytic activity는 pH 5~7에서 40~50시간, 배양하였을 때 가장 좋았다. 4. 생육배지에서 배양시 기질의 농도의 증가에 따라 6%까지는 균체증식이 직선적으로 증가하였으며 8%까지는 완만하게 증가하다가 9~10%농도에서는 감소하였다. 5. NaOH 처리한 볏짚은 기질로 하여 진탕 flask에서 배양한 결과 48기산 배양하였을 때 균체증식이 10.6mg/ml로써 가장좋았다.

Cheonggukjang Fermented with Bacillus subtilis SCGB574 Ameliorates High Fat Diet-Deteriorated Large Intestinal Health in Rat Model

  • Cheonggukjang is a traditional fermented food in Korea, which is known to exert beneficial effects on health. In this study, we evaluated the effects of cheonggukjang fermented by Bacillus subtilis SCGB 574 (B574) on high fat diet (HFD)-deteriorated large intestinal health. Rats were fed with HFD or HFD supplemented with 10.1% cheonggukjang (B574). Fecal microbiota was analyzed based on 16S rRNA gene sequences, and the fecal and serum metabolome were measured using GC-MS. Our results showed that SCGB574 intake significantly reduced body weight, restored tight junction components, and ameliorated inflammatory cell infiltration. SCGB574 also shifted gut microbiota by increasing the abundance of short chain fatty acid producers such as Alistipes and Flintibacter, although it decreased the abundance of Lactobacillus. Serum and fecal metabolome analyses showed significantly different metabolic profiles between the groups. The top five metabolites increased by SCGB574 were i) arginine biosynthesis, ii) alanine, aspartate, and glutamate metabolism; iii) starch and sucrose metabolism; iv) neomycin, kanamycin, and gentamicin biosynthesis; and v) galactose metabolism. These results showed that cheonggukjang fermented by SCGB574 ameliorates adverse effects of HFD through improving intestinal health.

  • With the establishment of rapid sequence analysis of 16S rRNA and the recognition of its potential to determine the phylogenetic position of any prokaryotic organism, the role of 16S rRNA similarities in the present species definition in bacteriology need to be clarified. Comparative studies clearly reveal the limitations of the sequence analysis of this conserved gene and gene product in the determination of relationship at the pathogenic strain level for which DNA-DNA reassociation experiments still constitute the superior method. Since today the primary structure of 16S rRNA is easier to determine than hybridization between DNA strands, the strength of the sequence analysis is to recognize the level at which DNA pairing studies need to be performed, which certainly applies to similarities of 97% and higher.

Zoogloea ramigera에 의한 생물고분자 생산에 관한 연구 (The Production of Biopolymer by Zoogloea ramigera)

  • 생물응집체 개발을 목적으로 Zoogloea ramigera115를 이용하여 균주배양 및 생물고분다 생성실험을 시시하였다. 다당류 최고 생성률을 얻을 수 있는 최적기질로 탄소원으로는 glucose, 질소원으로는 $NaNO_3$ 유기영양원으로는 yeast extract가 선택되었다. 초기 pH가 6일 때 가장 높은 세포성장과 점성을 나타내었다. 생성돈 생물고분자는 ultrasonication과 고속 원심분리를 거친 후 propanol 첨가에 의해 효율적으로 추출되었다. 생물고분자는 jae test에 의한 응집실험에서 높은 침강성을 보여 응집제 사용가능성을 확인하였다.

식품중 미생물 위해성평가 방법론 연구 (Study on the Methodology of the Microbial Risk Assessment in Food)

  • 최근 국내에서는 Escherichia coli O157:H7, Listeria monocytogenes등의 미생물에 대한 건강위해성이 강조되면서 미생물 위해성평가에 대한 필요성이 제기되고 있고, U.S.FPA, U.S.DA, FAO/WHO를 비롯한 국제기구 및 외국 유관기관들에서도 미생물 위해성평가 방법을 식품관리에 활용하고자 방법론 연구에 주력하고 있다. 미생물 위험성은 화학물질과 달리 인체건강에 대한 영향이 즉각적이고, 심각하게 나타나 정량적인 위해성평가가 용이하지 않고 유해화학물질과는 다른 평가방법이 요구된다. 식품중 미생물의 위해성평가는 크게 4단계로 구분되는데, 미생물관련질환 추세파악 및 미생물 관련질병에 관한 역학조사 등을 활용하는 위험성확인 단계와 실제 식품원료, 식품가공, 수송, 포장단계 중 식품의 물리적, 화학적 조건에 따른 미생물 변화를 고려하여 식품중 미생물에 대한 노출을 정량화하는 노출평가 단계, 미생물의 용량에 따른 질병발생에 근거하여 용량-반응관계를 규명하는 용량-반응평가 단계, 규명된 모델을 활용하여 모든 평가결과를 통합함으로 위해 도치 예측과 불확실성 분석 등을 수행하는 위해도결정단계로 구성되어 있다. 미생물 용량-반응평가는 크게 비역치(Nonthreshold)와 역치(Threshold) 평가 방법론으로 구분되는데, 비역치 평가방법론은 단일 병원균이 감염을 일으킬 수 있다는 것과 감염을 일으킬 수 있는 확률이 독립적이라는 가정을 전제로 하고, 역치평가방법론은 미생물이 감염을 일으키기 위해서 각기 개별 역치가 존재하는데 어느 정도의 미생물수가 모여 서로 작용해야 독성유발물질을 만들어 낸다는 가정을 전제로 한다. 현재 받아들여지고 있는 비역치 모델로는 Exponential, Beta-poisson, Gompertz, Gamma-weibull 모델 등이 있으며, 역치모델로는 Log-normal, Log-logistic모델 등이 있다. 본 연구에서는 인체 volunteer자료를 활용하여 용량-반응자료를 입력하고 용량-반응자료를 토대로 적합한 수학적 모델을 찾아내어, 선별한 모델의 적합도 검정을 실시하는 방법론 연구를 실시하였으며, 노출평가 자료와 용량-반응평가 결과를 연계하여 위해도를 결정하는 과정에 대해 연구하였다 이 밖에도 모델(Food MicroModel)을 이용하여 식품의 염도, 수분활성도, 온도, pH등의 조건에 따른 미생물의 성장률, 사멸률 등 변화를 예측할 수 있는 방법론 연구를 통해 식품의 최적 보관 조건등을 찾아내는 방법을 습득하였다. 미생물 위해성평가는 외국에서도 아직 초기 연구단계에 있으며 현재로서 사후조사자료인 역학자료보다 건강한 성인남자를 대상으로 한 volunteer 자료를 우선적으로 활용하고 있으나 노약자나 민감그룹에 대한 실험은 현실적으로 불가능하여 동물실험을 이용한 평가방법을 연구중에 있다. 추후 연구방향으로는 국내 volunteer들을 대상으로 한 미생물별 용량-반응결과를 토대로 population sensitivity를 비교할 수 있는 기초자료를 확보함으로써 미생물에 대한 인구집단의 반응 민감성 차이를 비교하고 시료채취 후 즉각적인 실험실적 분석이 가능토록하여 정확한 인체노출평가를 수행함으로써 미생물 위해성평가방법론을 식품미생물관리에 적용하는 것이다.
