Effect of Plant-growth-promoting Bacteria Inoculation on the Growth and Yield of Cucumber(Cucumis sativa L.) (식물생육촉진 세균이 오이 생육 및 수량에 미치는 영향)

  • We studied the effect of inoculation of microorganisms known to produce plant growth promoting substances, on the growth and yield of cucumber(Cucumis sativa L.), through a field experiment. The microorganisms used were isolated from the forest soil and consisted of Micrococus sp., Baccilus sustilis, Enterobacter agglomerans, Baccilus megaterium, Pseudomonas putida, Cellulomonas sp. and Staphylococus xyposus. Fotr the multiplication, microorganisms were cultured in liquid media of Pseudomonas P and Sabouraud dextrose. Inoculation of microorganisms was done by spraying the culture media after the culture of them to soil and cucumber plants, three times during the growth of cucumber at the rate of 10l/ha. The inoculation of microorganisms tended to promote the growth of cucumber plant and increase the yield of it. No sign of significant improvement of soil chemical and physical properties were observed after the harvest of crop. The population of bacteria and actinomycetes tended to be higher in the inoculated plots than in not inoculated plots, while opposite was the case in the population of fungi.

Trends of Perception and Information Needs on Food Additives of Children and Parents by Analyzing the Safety Assessment Reports of Food Additives in 2008~2013 (어린이와 학부모의 식품첨가물에 대한 인식 및 정보요구도 추이 분석: 2008~2013 식품첨가물 섭취 안전성 평가 연구 결과를 중심으로)

  • This study was performed in order to grasp the trends of elementary school children and their parents on their purchasing behavior of processed foods, awareness of food additives and its education experience by analyzing the safety assessment reports of food additives in 2008~2013. The most important factor in purchasing processed foods was safety in both groups followed by nutrition in parents and taste in children, respectively. While purchasing foods, the first item that is checked has been shifted from food additives to the origin of the products. Parents still perceived food additives as the most hazardous factor for food safety; however, recently, children began to regard microbial contaminants as being most hazardous, which is regarded as a desirable educational effect. The most concerned food additives were preservatives, synthetic seasoning and colorants in both groups. However, the awareness level on food additives still remained low as 3.0~3.1/5.0 for parents and 2.4~2.9/5.0 for children. Educational experience on food additives increased in children from 12% in 2008 to 25% in 2013; however, it decreased in parents from 23% in 2008 to 15% in 2013. Information needs for food additives by education and promotion were very high both in parents (4.2~4.5) and children (3.8~4.1). Both groups had an interest in the safety, legal standards of food additives, and foods with food additives, in order. The most reliable resource institutions on food additives were university/research institute and hospital for parents, but, hospital and government for children. The preferred media on food additives were TV and the internet for parents, and school newsletter and TV for children. Overall, the above results demonstrated that the perceptions on food additives did not change much with parents during the last 6 years; however, children's perceptions began to show improvement with the increase of educational experience. Hence, the government needs to make efforts to increase the trust level of consumers by developing educational tools and providing educational experiences including mass media for the promotion of risk communication on food additives.

Low Ruminal pH Reduces Dietary Fiber Digestion via Reduced Microbial Attachment

  • In vitro rumen incubation studies were conducted to determine effects of initial pH on bacterial attachment and fiber digestion. Ruminal fluid pH was adjusted to 5.7, 6.2 and 6.7, and three major fibrolytic bacteria attached to rice straw in the mixed culture were quantified with real-time PCR. The numbers of attached and unattached Fibrobacter succinogenes, Ruminococcus flavefaciens and Ruminocococcus albus were lower (p<0.05) at initial pH of 5.7 without significant difference between those at higher initial pH. Lowering incubation media pH to 5.7 also increased bacterial numbers detached from substrate regardless of bacterial species. Dry matter digestibility, gas accumulation and total VFA production were pH-dependent. Unlike bacterial attachment, maintaining an initial pH of 6.7 increased digestion over initial pH of 6.2. After 48 h in vitro rumen fermentation, average increases in DM digestion, gas accumulation, and total VFA production at initial pH of 6.2 and 6.7 were 2.8 and 4.4, 2.0 and 3.0, and 1.2 and 1.6 times those at initial pH of 5.7, respectively. The lag time to reach above 2% DM digestibility at low initial pH was taken more times (8 h) than at high and middle initial pH (4 h). Current data clearly indicate that ruminal pH is one of the important determinants of fiber digestion, which is modulated via the effect on bacterial attachment to fiber substrates.

Isolation and characterization of an antifungal substance from Burkholderia cepacia, an endophytic bacteria obtained from roots of cucumber.

  • In order to develop a new microbial fungicide for the control of vegetable diseases using endophytic bacteria, a total of 260 bacterial strains were isolated from fresh tissues of 5 plant species. After they were cultured in broth media, their antifungal activities were screened by in vivo bioassays against Botrytis cinerea(tomato gray mold), Pythium ultimum(cucumber damping-off), Phytopkhora infestans(tomato late blight), Colletotrichum orbiculare(cucumber anthracnose), and Blumeria graminis f. sp. hordei(barley powdery mildew). As the results of screening, 38 bacterial strains showed potent antifungal activities against at least one of 5 plant pathogens. A bacterial strain EB072 displayed potent disease control activities against 3 plant diseases. Among the bacterial strains with a potent antifungal activity against cucunlber anthracnose, three bacterial strains, EB054, EB151 and EB215, also displayed a potent in vitro antifungal activity against C. acutatum, a fungal agent causing pepper anthracnose. A bacterial strain EB215 obtained from roots of cucumber was identified as Burkholderia cepacia based on its physiological and biochemical characteristics and 165 rRNA gene sequence. An antifungal substance was isolated from the liquid cultures of B. cepacia EB215 strain by ethyl acetate partitioning, repeated silica gel column chromatography, and invitro bioassay, Its structural determination is in progress by various instrumental analyses.

Protection of Tobacco Plants from Bacterial Wilt with an Avirulent Isolate of Pseudomonas solanacearum (비병원성 Pseudomonas solanacearum을 이용한 담배 세균성마름병의 방제)

  • Microbial antagonism between virulent and avirulent isolates of Pseudomonas solanacearum was studied in relation to the control of bacterial wilt of tobacco. In nutrient broth media or in soil, the avirulent isolate of P. solanacearum grew faster than did the virulent one. Inhibitory effect of avirulent isolate against growth of virulent one was negligible in mixed culture of the two isolates. The disease severity of bacterial wilt was significantly reduced when the roots of cultivar BY 4 susceptible to bacterial wilt was dipped in suspension of an avirulent isolate for 6 hours prior to transplanting to the soil infested with virulent bacteria. When the seedlings of tobacco were poured with the suspension of an avirulent isolate onto the soil in pre-planting pots 24 hours before ransplanting, there was a significant reduction in disease severity in the field. However, the reduction was noticed until early July, but after middle of July, no difference between the avirulent isolate-treated and non-treated plants was found in severity of the bacterial wilt.

  • A microbiological hydrogen production process was optimized. Anaerobic photosynthetic bacteria like Rhodospirillum rubrum which is known to produce hydrogen from carbon monoxide efficiently and remove sulfur was used. To evaluate the potenital of this microorganism the optimization of media fermentation condition light intensity and light requirement for CO conversionwas tried in batch cultures and the continuous fermenter was also applied for this process. The gas residence time on CO conversion was sought out to get high conversion of carbon monoxide to hydrogen. Through this study the possibility of microbial synthtics gas concersion process was proposed.

Application of a Modified Sublimation Method to Screen for PAH-Degrading Microorganisms (다환 방향족 탄화수소(PAH) 분해 미생물 탐색을 위한 승화법의 개발)

  • Recent studies have described various microorganisms that can degrade PAH, however, there are currently limited methods available to screen for PAH-degrading microorganisms. To screen for PAH-degrading microorganisms, a sublimation method (Alley, Jeremy F. and Lewis R. Brown. 2000. Appl. Environ. Microbiol. 66, 439-442) was modified to produce a simple screening system. In our results, there were several bacterial species capable of pyrene degradation including genera, Coryenbacterium, Gordonia, Rhodococcus, and Streptomyces, which have been screened from 350 bacterial isolates of commercial gasoline and oil-spilled sediment by the sublimation method. The main advantage of this method is that it (i) safely deposits an even, thin and visible layer of PAH onto the agar surface without the use of solvents and (ii) the quantity of PAH sublimed onto the agar can be easily controlled. Overall, this sublimation method may be an effective and simple technique to screen for PAH-degrading microorganisms.

  • Abstract: In order to improve the system for biological nitrogen oxidizing process in sewage and wastewater, a bacterium having high abilities to oxidize of nitrogen was isolated from wastewater and polluted soils. The strain was identified to Bacillus sp. CH-N, based on the physiological and biochemical properties. Characteristics and oxidizing ability of both ammonia and nitrite were examined for the strain, Bacillus sp. CH-N. The strain showed the oxidizing rate about 80% to 90% on the sewage and wastewater after 48 h culture. The nitrogen oxidizing rate was increased in proportion to the initial concentration of glucose. The microorganism, Bacillus sp. CH-N cell immobilized on ceramic carrier were evaluated for the oxidation of ammonia in culture media.

Bioconversion of Citron oil by Co-Culture of E. coli EC3, EC4, and EC6 (E. coli 형질전환주의 공동배양에 의한 유자정유의 생전환)

  • E. coli transformants EC3, EC4. and EC6. harboring citron oil degrading pathway genes, were co-cultured in M9 media with citron oil as a sole carbon source at 28$^{\circ}C$. Each co-culture(EC3+EC4, EC3+EC6, EC4+EC6 and EC3+EC4+EC6) showed three to four times higher cell growth than each transformant single culture. Microbial conversion products from the co-cultures were determined by GC-MS. Linalool. 4-terpineol and ${\alpha}$-terpineol were the major common products from co-cultures. Various minor products also were detected and important in flavor characteristics of cultures.

Study on the Microbial Degradation of Nicotine (1) -Isolation and Identification of Nicotinophiles (미생물에 의한 니코틴 분해 연구(1) -니코틴 분해세균의 분리 및 동정)

  • From 143 sources of collected samples, the distribution of nicotinophiles were investigated and the biological characteristics as well as the rate of nicotine degradation were determined for the selected 34 strains which could grow successfully in the nicotine media, and one of the most effective strains was chosen and identified at the species level. Nicotinophils were distributed abundantly in the soils rich with organic materials, tobacco seed and root. The selected 34 strains were classified into 7 genus and identified with 4 strains of Arthrobacter, 11 strains of non-pigmented Pseudomonas, 2 strains of pigmented Pseudomonas, 6 strains of Alkaligenes, 5 strains Chromobacter 2 strains of Listeria and 4 strains of Achromobacter. Pseeudomonas and Alkaligenes were better than other genus in the rate of nicotine degradation and tobacco seed and root were also good sources for the isolation of effective nicotinophiles. Amnog 34 strains, strain NCT 27 which exhibited 97.l% of nicotine degradation rate was the best one for nicotine degradation and was indentified with Pseudomonas putida.
