Industrial development has increase consumption of crude oil and environmental pollution. A large number of microbial lipolytic enzymes have been identified and characterized to date. To development for a new lipase with catalytic activity in degradation of crude oil as a microbial enzyme, Acinetobactor sp. B2 was isolated from soil samples that were contaminated with oil in Daejon area. Acinetobactor sp. B2 showed high resistance up to 10 mg/mL unit to heavy metals such as Ba, Li, Al, Cr, Pb and Mn. Optimal growth condition of Acinetobactor sp. B2 was confirmed $30^{\circ}C$. Lipase was purified from the supernatant by Acinetobactor sp. B2. Its molecular mass was determined to the 60 kDa and the optimal activity was shown at $40^{\circ}C$ and pH 10. The activation energies for the hydrolysis of p-nitrophenyl palmitate were determined to be 2.7 kcal/mol in the temperature range 4 to $37^{\circ}C$. The enzyme was unstable at temperatures higher than $60^{\circ}C$. The Michaelis constant $(K_{m})\;and\;V_{max}$ for p-nitrophenyl palmitate were $21.8{\mu}M\;and\;270.3{\mu}M\;min^{-1}mg\;of\;protein^{-1}$, respectively. The enzyme was strongly inhibited by $Cd{2+},\;Co^{2+},\;Fe^{2+},\;Hg^{2+},\;EDTA$, 2-Mercaptoethalol. From these results, we suggested that lipase purified from Acinetobactor sp. B2 should be able to be used as a new enzyme for degradation of crude oil, one of the environmental contaminants.

In this study, the dynamics of living cells (LC) and dead cells (DC) in a laboratory-scale biofilm membrane bioreactor for waste gas treatment was examined. Toluene was used as a model pollutant. The bacterial cells were enumerated as fluoromicroscopic counts during a 140 operating day period using BacLight nucleic acid staining in combination with epifluorescence and confocal laser scanning microscopy (CSLM). Overall, five different phases could be distinguished during the biofilm development: (A) cell attachment, (B) pollutant limitation, (C) biofilm establishment and colonization, (D) colonized biofilm, and (E) biofilm erosion. The bioreactor was operated under different conditions by applying different pollutant concentrations. An optimum toluene removal of 89% was observed at a loading rate of 14.4 kg $m^{-3}d^{-1}$. A direct correlation between the biodegradation rate of the reactor and the dynamics of biofilm development could be demonstrated. This study shows the first description of biofilm development during gaseous toluene degradation in MBR.
The biodegradable bacteria for fats and oils were isolatcd from soil and wastewater. The isolated strain was designated as LW-27 which had high COD removal rate and biodegr2idability on fats and oils, and was identified as pseudomonas chlrorapihis. The cell viability of LW-27 which produced by vacuum drying at $45^{\circ}C$ for 24 hours was 82%. When the wastewater was mixed with LW-27 agent (0.1g/ day) on the activated sludge unit, the removal rates of COD, BOD and n-hexanc extract of the effluents were about 92.9%, 94.8% and 98.0%, respectively.
Haloaliphatic hydrocarbons have been widely used as solvents and ingredients of pesticides and herbicides. However, when these compounds contaminate the environment, they can be very hazardous to animals and humans because of their potential toxicity and carcinogenicity. Therefore, lots of studies have been made for microbial degradation of those pollutant chemicals. In this study, 11 bacterial strains capable of degrading 1,2-dichloroethane (1,2-DCA), 2-chloropropionic acid (2-CPA), 2,3-dichloropropionic acid (2,3-DCPA), and 2-monochloroacetic acid (2-MCA) by hydrolytic dechlorination under aerobic conditions were isolated from wastewaters and rice paddy soil samples. Their morphological and biochemical characteristics and their degradation capabilities of haloaliphatic hydrocarbons were examined. On the basis of the 16S rDNA sequences, 8 different kinds of microbial species, including Pseudomonas plecoglossicida, Xanthobacter flavus, Ralstonia eutropha, were identified. All of the isolated strains can degrade MCA. In particular, strains UE-2 and UE-15 degraded 1,2-DCA, and strain CA-11 degraded 2,3-DCPA, which are hardly degraded by other strains.
This study evaluated the fermentation characteristics and protein degradation dynamics of wet green tea grounds (WGTG) silage. The WGTG was ensiled with distilled water (control), or lactic acid bacteria (LAB), enzyme (E), formic acid (FA) and formaldehyde (FD) prior to ensiling. Three bag silos for each treatment were randomly opened at 0, 3, 7, 14, 28 and 60 days after anaerobic storage. For all the treatments, except for FA, there was a rapid decline in pH during the first 7 days of ensiling. LAB treatment had higher lactic acid content, lower ammonia-N ($NH_3$-N) and free-amino nitrogen (FAA-N) contents than other treatments (p<0.05). E treatment had higher lactic acid, water-soluble carbohydrates (WSC) and non-protein nitrogen (NPN) content than the control (p<0.05). FA treatment had higher $NH_3$-N and FAA-N content than the control (p<0.05). FD treatment had lower NPN and FAA-N content than the control, but it did not significantly inhibit the protein degradation when compared to LAB treatment (p>0.05). Results indicate that LAB treatment had the best effect on the fermentation characteristics and protein degradation of WGTG silage.
Degradation of polychlorinated biphenyls(PCBs) and organochlorine pesticides by Alcaligenes aquamarinus has been studied and also degradation product of PCB-42 was investigated by TLC and GC. The less chlorinated members of PCBs such as Aroclor 1016 was degraded readily by the strain and rates of the microbial degradation of several organochlorine pesticides were found to decrease in the order of p,p'-DDT, r-BHC and Thiolix. Approximately 40 percent of PCB-42 was degraded when incubated with non-autoclaved soil for 25 days at $25^{\circ}C$. The yellow compound from PCB-42 was tentatively identified as p-chlorobenzoic acid.

The overall indicator of microbial activity in the fermentation-extinction reaction of food waste by using bio wood-chips were investigated by considering adenosine tri-phosphate(ATP). Degradation rate of organic compounds, which was represented by chemical oxygen demand(COD) and total nitrogen(TN), was increased with the concentration of adenosine tri-phosphate during fermentation-extinction reaction of food waste by using bio-wood chips. With this view, the ATP would be one of the overall evaluation indicator of organic degradation in the species of bio-wood chip for the fermentation-extinction of food waste.

This study was performed to evaluate efficiency of soil microbial inoculator for active composting of food waste. In addition the number of microorganisms in roil microbial inoculator and the effect of seeding in the process of composting were investigated. food waste samples collected from a refectory were analyzed for physical-chemical properties. The samples were adjusted to moisture content of 65% by saw dust and seeded with soil microbial inoculator of 10% by the weight in case of reactor B. The number of microorganisms, aerobic bacteria, actinomyces, yeast, and fungi in soil microbial inoculator were over $2.98{\times}10^9/g$, $3.93{\times}10^7/g$, $1.21{\times} 10^5/g$, and $5.79{\times}10^7/g$, respectively. During the process of composting, the highest temperatures were $63.4^{\circ}C$ at reactor A(unseeded control)after 10 days and $66.8^{\circ}C$ at reactor B(seeded compost) after 4 days. The pH values of reactor A and B rapidly increased after 3 days and after first few days during composting period, respectively. The highest $CO_2$ concentrations were 6.1%(after 10 days) and 10.8%(afer 4 days) in reactor A and B, respectively. The degradation rates of organic matter(rd) between reactor A and B increased by 17.1% and 64.5%, respectively Consequently, the effects of Inoculation on comporting parameter such as temperature increasing, pH change, chemical properties, and the degradation rates of organic matter(rd) were higher in seeded compost than in unseeded control.

The respiratory quinone profile was used as a tool for the study on microbial community structure in the mine tailings of abandoned mines over the depth. For the study, the area of Jingok mine located in Bongwha, Korea has been selected. The distributions of Cd, Cu, Pb, Al, Fe and Mn showed the following common patterns; the highest values in the upper part of mine failings (0-20 cm), rapid decrease with increasing depth. The dominant quinone species of the mine tailings were UQ-9 followed by UQ-10, suggesting that microbes had contributed to heavy metal degradation. The quinone contents in mine tailings ranged from 5.0 to 24.9 nmol/kg. The microbial diversity in the upper part of mine tailings (0-40 cm) was higher than that of lower part of mine tailings (100-120 cm).

Electrokinetic bioremediation was conducted on phenanthrene-contaminated soil to study the effects of soil temperature and pH on microbial population and removal efficiency at different current densities from 0.63 to 3.13 mA cm$^{-2}$ . Microorganism used in the biodegradation of phenanthrene was Sphingomonas sp. 3Y, which was isolated from a diesel-contaminated site. The microorganism was successfully penetrated into the contaminated soil by electrokinetic phenomena and the highest microbial population was observed in the middle region of soil specimen where soil pH was near neutral. Therefore, phenanthrene removal occurred mainly at anode and middle parts of soil specimen due to a relatively high microbial population. Also, the highest removal efficiency of 68.8% was obtained at 1.88 mA cm$^{-2}$ while low degradation was detected at 3.13 mA cm$^{-2}$ . It was presumably because the soil temperature at 1.88 mAcm$^{-2}$ was close to the appropriate temperature of about 30'c while the temperature increase to above 45$^{\circ}C$ at 3.13 mA cm$^{-2}$ inhibited the microbial activity severely.
