In the grilled fish paste stored at $5^{\circ}C$, the total aerobic bacterial counts were effectively reduced by 2.5 kGy or more. In the samples stored at $30^{\circ}C$, the total aerobic bacterial counts of the samples irradiated at 7.5 kGy were below to the limit of detection (2 log CFU/g). The TBA values of the irradiated samples were considerably higher than those of the controls but not proportional to the irradiation dose. It is apparent that an irradiation treatment causes very little textural degradation and the sensorial quality of the sample was maintained by an irradiation at 7.5 kGy or more.
In order to investigate mechanisms related to the microbial degradation of 3,4-dichloroaniline, it was incubated with a soil fungus, Chaetomium globosum and the following results were obtained. (1) 3,4-Dichloroacetanilide turned out to be the major metabolite, indicating that acetylation is the major scheme. (2) The presence of trace amounts of 3,4-dichloronitrobenzene, 3,3', 4,4'-tetrachloroazo-benzene, 3,4-dichloroaniline is suggestive of the aromatic amine oxidation as the minor pathway. (3) Other metabolites with m/e 112, 114, and 279 were also isolated, but their identities are under investigation. (4) Dechlorination occurring during incubation indicates the possibility of forming hydroxylated and other metabolites.

This research investigates the importance of the microbial metabolic pathways such as denitrification, iron reduction, and methanogenesis, in the degradation of organic matters of the sediments. There are statistically significant differences( P < 0.05) in the rates of denitrification, iron reduction, and methanogenesis according to the location: Site A has no plant, Site B is dominated by Scirpus, and Site C is dominated by Phragmites. Among them, Site C showed different methanogenesis rate depending on the sediments depth. The organic matter content increased from Site A to Site C. Site A had the smallest organic matter content whereas it showed the largest denitrification rate and iron reduction rate. Site C had the largest methanogenesis rate. Denitrification is the dominant pathways based on the assumption that anaerobic degradation of organic matter is mainly carried out through denitrification, iron reduction, and methanogenesis.
Recently the bioremediation technology has been widely used to recover the oil contaminated environments The application of bioremediation agents to oil polluted environments became common and thus many kinds of commercial products were imported into domestic market. In Korea, howcver. the standardization of bioremediation products quality is not yet established and results of efficacy test .ire scarce. In this study five oil spill bioremediation commercial products including microbial inoculants and en'cyme agents are tested for the oil degradation rate. From the results most products shows the strong oil emulsifying phenomena due to the contained chemical oil dispersant and the low oil degradation rate. Product D inhibited the oil degradability of microorganisms even in the natural sea water. From these results it could be concluded that in the near future the laboratory protocol and standardization of products quality for bioremediarion agents should be prepared to activate the effective application of bioremediation technology in Korea.
Sediment bacterial communities are critical to the biogeochemical cycle in river ecosystems, but our understanding of the relationship between sediment bacterial communities and their specific input streams in rivers remains insufficient. In this study, we analyzed the sediment bacterial community structure in a local river receiving discharge of urban domestic sewage by applying Illumina MiSeq high-throughput sequencing. The results showed that the bacterial communities of sediments samples of different pollution types had similar dominant phyla, mainly Proteobacteria, Actinobacteria, Chloroflexi and Firmicutes, but their relative abundances were different. Moreover, there were great differences at the genus level. For example, the genus Bacillus showed statistically significant differences in the hotel site. The clustering of bacterial communities at various sites and the dominant families (i.e., Nocardioidaceae, and Sphingomonadaceae) observed in the residential quarter differed from other sites. This result suggested that environmentally induced species sorting greatly influenced the sediment bacterial community composition. The bacterial co-occurrence patterns showed that the river bacteria had a nonrandom modular structure. Microbial taxonomy from the same module had strong ecological links (such as the nitrogenium cycle and degradation of organic pollutants). Additionally, PICRUSt metabolic inference analysis showed the most important function of river bacterial communities under the influence of different types of domestic sewage was metabolism (e.g., genes related to xenobiotic degradation predominated in residential quarter samples). In general, our results emphasize that the adaptive changes and interactions in the bacterial community structure of river sediment represent responses to different exogenous pollution sources.
Cometabolism technology was employed to degrade lignin wastewater in Sequencing Batch Biofilm Reactor. Cometabolic system (with glucose and lignin in inflow) and the control group (only lignin in inflow) were established to do a comparative study. In contrast with the control group, the average removal rates of lignin increased by 14.7% and total oarganic carbon increased by 32% in the cometabolic system with glucose as growth substrate, under the condition of 5 mg/L DO, $0.2kgCOD/(m^3{\cdot}d)$ lignin and glucose $1.0kgCOD/(m^3{\cdot}d)$. Functional groups of lignin are degraded effectively in cometabolic system proved by fourier transform infrared spectroscopy and Gas Chromatography-Mass Spectrometer, and the degradation products were amides (mainly including acetamide, N-ethylacetamide and N, N-diethylacetamide), alcohols (mainly including glycerol and ethylene glycol) and acids. Meanwhile, results of Polymerase Chain Reaction-Denaturing Gradient Gel Electrophoresis showed great differences in microbial population richness between cometabolic system and the control group. The Margalef's richness index and Shannon-Wiener's diversity index of microorganism in cometabolic system were 3.075 and 2.61, respectively. The results showed that extra addition of glucose, with a concentration of 943 mg/L, was beneficial to lignin biodegradation in cometabolic system.
A sundry system is one of popular systems for composting livestock manure, of which main honest is to utilize unlimited, clean, and free solar radiation. A sundry system with a composter of two horizontal screw-type concrete ducts at different height, was constructed and operated for three days for each test in May, 1993, to evaluate its composting performance. Four treatments of the mixture ratio of swine manure and saw dust (manure : sawdust= 1 : 1.25, 1 : 1, 1 : 0.7, 1 : 0.5) were implemented to evaluate the effect of the mixture ratio on degradation of the composting materials of a sundry system with a screw-type composter. Maximum temperature of the composting materials was over 5$0^{\circ}C$ at D1 or D2 (one or two days after operation starts) for each test. Mean C/N ratio and water contents of the materials were reduced by more than 15 and 20%, respectively. Microbial density of each test showed a typical variation with the lapse of the composting time. Mesophilic microorganism seemed to play more important role on degradation of the materials than thermophilic. A sundry system with a screw-type composter can be considered as a feasible system on basis of maturity data. The conclusion was completely reverse from that of Choi et at., although both adopted a sundry system. A further study is recommended to pursue the cause of better performance of the screw-type composter, whether it was due to affirmative weather or more efficient composter.

In the United States (U.S.), the monitored natural attenuation (MNA) approach has been used as an alternative remedial option for organic and inorganic compounds retained in soil and dissolved in groundwater. The U.S. Environmental Protection Agency (EPA) defines the MNA as“in-situ naturally-occurring processes include biodegradation, diffusion, dilution, sorption, volatilization, and/or chemical and biochemical stabilization of contaminants and reduce contaminant toxicity, mobility or volume to the levels that are protective of human health and the environment”. The Department of Soil Environment. National Institute Environmental Research (NIER) is in the process for demonstrating the MNA approach as a potential remedial option for the BTEX contaminated site in Uiwang City. The project is charactering the research site in terms of the nature and extend of contamination, biological degradation rate, and geochemical and hydrological properties. The microbial-degradation rate and effectiveness of nutrient and redox supplements will be determined through laboratory batch and column tests. The geochemical process will be monitored for determining the concentration changes of chemical species involved in the electron transfer processes that include methanogenesis, sulfate and iron reduction, denitrification, and aerobic respiration. Through field works, critical soil and hydrogeologic parameters will be acquired to simulate the effects of dispersion, advection, sorption, and biodegradation on the fate and transport of the dissolved-phase BTEX plume using Bioplume III model. The objectives of this multi-years research project are (1) to evaluate the MNA approach using the BTEX contaminated site in Uiwang City, (2) to establish a standard protocol for future application of the approach, (3) to investigate applicability of the passive approach as a secondary treatment remedy after active treatments. In this presentation, the overall picture and philosophy behind the MNA approach will be reviewed. Detailed discussions of the site characterization/monitoring plans and risk-based decision-making processes for the demonstration site will be included.
Estuarine sediments are frequently polluted with hydrocarbons from fuel spills and industrial wastes. Polycyclic aromatic hydrocarbons (PAHs) are components of these contaminants that tend to accumulate in the sediment due to their low aqueous solubility, low volatility, and high affinity for particulate matter. The toxic, recalcitrant, mutagenic, and carcinogenic nature of these compounds may require aggressive treatment to remediate polluted sites effectively. In petroleum-contaminated sediments near a petrochemical industry in Gwangyang Bay, Korea, in situ PAH concentrations ranged from 10 to 2,900 ${\mu}g/kg$ dry sediment. To enhance the biodegradation rate of PAHs under anaerobic conditions, sediment samples were amended with biostimulating agents alone or in combination: nitrogen and phosphorus in the form of slow-release fertilizer (SRF), lactate, yeast extract (YE), and Tween 80. When added to the sediment individually, all tested agents enhanced the degradation of PAHs, including naphthalene, acenaphthene, anthracene, fluorene, phenanthrene, fluoranthene, pyrene, chrysene, and benzo [a] pyrene. Moreover, the combination of SRF, Tween 80, and lactate increased the PAH degradation rate 1.2-8.2 times above that of untreated sediment (0.01-10 ${\mu}g$ PAH/ kg dry sediment/day). Our results indicated that in situ contaminant PAHs in anoxic sediment, including high molecular weight PAHs, were degraded biologically and that the addition of stimulators increased the biodegradation potential of the intrinsic microbial populations. Our results will contribute to the development of new strategies for in situ treatment of PAH-contaminated anoxic sediments.
Dental pulp is a loose, mesenchymal tissue almost entirely enclosed in the dentin. It consists of cells, ground substance, and neural and vascular supplies. Damage to the dental pulp by mechanical, chemical, thermal, and microbial irritants can provoke various types of inflammatory response. Pulpal inflammation leads to the tissue degradation, which is mediated in part by Matrix metalloproteinase leads to accelerate extracellular matrix degradation with pathological pathway We have now investigated the induction of MMPs and inflammatory cytokines by Lipopolysaccharide (LPS) control of inflammatory mediators by peroxisome proliferator-activated receptors (PPARs). Human dental pulp cells exposed to various concentrations of LPS ($1-10{\mu}g/ml$) revealed elevated levels of MMP-2 and MMP-9 at 24 hrs of culture. LPS also stimulated the production of ICAM-1, VCAM-1, $IL-1{\beta},\;and\;TNF-{\alpha}$. Adenovirus $PPAR{\gamma}\;(Ad/PPAR{\gamma})\;and\;PPAR{\gamma}$ agonist rosiglitazone reduced the synthesis of MMPs, adhesion molecules and pro-inflammatory cytokines. The inhibitory effect of $Ad/PPAR{\gamma}$ was higher than that of $PPAR{\gamma}$ agonist. These result offer new insights in regard to the anti-inflammatory potential of $PPAR{\gamma}$ in human dental pulp cell.
