This study was designed to investigate the effect of plant as a botanical air purification on the indoor pollution by formaldehyde. Three indoor plants such as Dracaena marhginata, Spathiphyllum and Dracaena reflexa, were placed in the artificially contaminated reactor under laboratory condition. Both plant and soil effects on removal of formaldehyde from contaminated indoor air were observed. Reductions in the formaldehyde levels appeared to have been associated with soil medium factors as well as plant factors. The effect of soil on formaldehyde reduction was high in the early stage of the experiment and the results suggest that sorption could be more important factor than microbial degradation in the initial dissipation of contaminants in the soil. It was suggested that the effect of plant on formaldehyde reduction might be related to the plant species, total leaf surface area of plant, degree of contribution of soil medium, and exposed concentration level. The results of this study showed that air purification using plants is an effective means of reduction on indoor formaldehyde level, though, utilization of soil media with high sorption capacity and/or supplementary purifying aids were also suggested when the source is continuous or exposed concentration level is high.
The aim of this review was to assess current knowledge related to the occupational exposure limit (OEL) for fluid aerosols including either mineral or chemical oil that are generated in metalworking operations, and to discuss whether their OEL can be appropriately used to prevent several health risks that may vary among metalworking fluid (MWF) types. The OEL (time-weighted average; 5 mg/$m^3$, short-term exposure limit ; 15 mg/$m^3$) has been applied to MWF aerosols without consideration of different fluid aerosol-size fractions. The OEL, is also based on the assumption that there are no significant differences in risk among fluid types, which may be contentious. Particularly, the health risks from exposure to water-soluble fluids may not have been sufficiently considered. Although adoption of The National Institute for Occupational Safety and Health's recommended exposure limit for MWF aerosol (0.5 mg/$m^3$ ) would be an effective step towards minimizing and evaluating the upper respiratory irritation that may be caused by neat or diluted MWF, this would fail to address the hazards (e.g., asthma and hypersensitivity pneumonitis) caused by microbial contaminants generated only by the use of water-soluble fluids. The absence of an OEL for the water-soluble fluids used in approximately 80-90 % of all applicants may result in limitations of the protection from health risks caused by exposure to those fluids.
Mycoplasma is well recognized as one of the most prevalent and serious microbial contaminants of biologic manufacturing processes. Conventional methods for mycoplasma testing, direct culture method and indirect indicator cell culture method, are lengthy, costly and less sensitive to noncultivable species. In this report, we describe a new TaqMan probe-based real-time PCR method for rapid and quantitative detection of mycoplasma contamination during manufacture of biologics. Universal mycoplasma primers were used for mycoplasma PCR and mycoplasma DNA was quantified by use of a specific TaqMan probe. Specificity, sensitivity, and robustness of the real-time PCR method was validated according to the European Pharmacopoeia. The validation results met required criteria to justify its use as a replacement for the culture method. The established real-time PCR assay was successfully applied to the detection of mycoplasma from human keratinocyte and mesenchymal stem cell as well as Vero cell lines artificially infected with mycoplasma. The overall results indicated that this rapid, specific, sensitive, and robust assay can be reliably used for quantitative detection of mycoplasma contamination during manufacture of biologics.
Cometabolic air sparging (CAS) is a new and innovative technology that uses air sparging principles but attempts to optimize in situ contaminant degradation by adding a growth substrate to saturated zone. CAS relies on the degradation of the primary growth substrate and cometabolic substrate transformation in the saturated zone and in the vadose zone for volatilized contaminants. In this study, we have investigated to determine MTBE degradation pattern and microbial activity variation if using propane as a primary substrate at the condition of considering air injection rate and air injection pattern. Laboratory-scale two-dimentional aquifer physical model studies were used and the experimental results were represented that the optimal conditions were as air injection rate of 1,000 mL/min and pulsed air injection pattern (15 min on/off). Over 1,000 mL/min air injection rate and continuous air injection pattern was no affected to increase DO concentration. On the other hand, Injection of propane and propane-utilizing bacteria degraded MTBE partially. And also, injection of propane- and MTBE-utilizing bacteria effectively degraded MTBE and TBA production was observed.
To properly manage and remediate groundwater contaminated with chlorinated hydrocarbons such as trichloroethylene (TCE), it is necessary to assess natural attenuation processes of contaminants in the aquifer along with investigation of contamination history and aquifer characterization. This study evaluated natural attenuation processes of TCE at an industrial site in Korea by delineating hydrogeochemical characteristics along the flow path of contaminated groundwater, by calculating reaction rate constants for TCE and its degradation products, and by using geochemical and reactive transport modeling. The monitoring data showed that TCE tended to be transformed to cis-1,2-dichloroethene (cis-1,2-DCE) and further to vinyl chloride (VC) via microbial reductive dechlorination, although the degree was not too significant. According to our modeling results, the temporal and spatial distribution of the TCE plume suggested the dominant role of biodegradation in attenuation processes. This study can provide a useful method for assessing natural attenuation processes in the aquifer contaminated with chlorinated hydrocarbons and can be applied to other sites with similar hydrological, microbiological, and geochemical settings.
서울메트로 지하철 1~4호선 이용객은 하루 평균 450만명이 지하철을 이용(2010년 상반기 수송인원 현황)하고 있으며 이것은 서울시 인구 57%에(2009년도 서울시 인구 총1,046만명) 가까운 사람들이 지하철을 이용하고 있는 것으로 나타남에 따라 전동차에 쾌적한 실내공기질 유지 및 관리는 무엇보다 중요하다. 특히 전동차내 실내공기질에 영향을 미치는 요인으로는 승객의 재채기, 호흡과 같은 생체 활동 및 피부각질 등에 의한 부유 미생물 작용에 있다(권순박 등, 2011). 바이러스와 같은 미생물 오염 물질이 승객의 인체의 호흡기, 점막부위, 피부 등에 접촉 할 경우 과민성 질환, 아토피 피부염, 전염성 질환, 알레르기성 질환, 호흡기 질환을 유발할 수 있는(환경부-국립환경과학원, 생활속 세균 이렇게 관리하세요) 산업 보건상의 문제로 대두되고 있어 적절한 환기 시스템 도입과 제습을 위한 냉방 등이 필요한 실정이다. 시민들의 관심과 보조를 맞추어 지하철 2호선 출 퇴근 및 평상시 전동차 객실내 부유 미생물을 측정한 결과 부유세균과 부유진균은 2011년도 85~385$cfu/m^3$, 67~98$cfu/m^3$로 기준치(별도의 법적 기준은 없으나, 부유세균은 국내 실내기준 800$cfu/m^3$, 부유진균은 세계보건기구의 가이드라인 150$cfu/m^3$) 보다 낮은 수준으로 나타났으나 유동인구가 많은 출 퇴근 시간대에는 미생물 오염 물질이 승객에게 쉽게 전염 될 가능성이 있을 것으로 사료됨에 따라 부유 미생물 오염방지를 위한 공기조화설비 성능개선 등 실질적인 개선방안을 찾는 것이 필요하다. 이에 본 연구에서는 2004년 이후 부유 미생물 제어장치에 대한 연구 논문을 분석하여 부유미생물이 승객들에게 미치는 영향, 환기량 증대방안 등의 다양한 인자에 대한 분석을 통하여 전동차내 공기질을 실질적으로 개선할 수 있는 방법을 연구 하였다.
Although, microbial arsenic mobilization by dissimilatory arsenate-reducing bacteria (DARB) and the practical use to the removal technology of arsenic from contaminated soil are expected, most previous research mainly has been focused on the geochemical circulation of arsenic. Therefore, in this review we summarized the previously reported DARB to grasp the characteristic for bioremediation of arsenic. Evidence of microbial growth on arsenate is presented based on isolate analyses, after which a summary of the physiology of the following arsenate-respiring bacteria is provided: Chrysiogenes arsenatis strain BAL-$1^T$, Sulfurospirillum barnesii, Desulfotomaculum strain Ben-RB, Desulfotomaculum auripigmentum strains OREX-4, GFAJ-1, Bacillus sp., Desulfitobacterium hafniense DCB-$2^T$, strain SES-3, Citrobacter sp. (TSA-1 and NC-1), Sulfurospirillum arsenophilum sp. nov., Shewanella sp., Chrysiogenes arsenatis BAL-$1^T$, Deferribacter desulfuricans. Among the DARB, Citrobacter sp. NC-1 is superior to other dissimilatory arsenate-reducing bacteria with respect to arsenate reduction, particularly at high concentrations as high as 60 mM. A gram-negative anaerobic bacterium, Citrobacter sp. NC-1, which was isolated from arsenic contaminated soil, can grow on glucose as an electron donor and arsenate as an electron acceptor. Strain NC-1 rapidly reduced arsenate at 5 mM to arsenite with concomitant cell growth, indicating that arsenate can act as the terminal electron acceptor for anaerobic respiration (dissimilatory arsenate reduction). To characterize the reductase systems in strain NC-1, arsenate and nitrate reduction activities were investigated with washed-cell suspensions and crude cell extracts from cells grown on arsenate or nitrate. These reductase activities were induced individually by the two electron acceptors. Tungstate, which is a typical inhibitory antagonist of molybdenum containing dissimilatory reductases, strongly inhibited the reduction of arsenate and nitrate in anaerobic growth cultures. These results suggest that strain NC-1 catalyzes the reduction of arsenate and nitrate by distinct terminal reductases containing a molybdenum cofactor. This may be advantageous during bioremediation processes where both contaminants are present. Moreover, a brief explanation of arsenic extraction from a model soil artificially contaminated with As (V) using a novel DARB (Citrobacter sp. NC-1) is given in this article. We conclude with a discussion of the importance of microbial arsenate reduction in the environment. The successful application and use of DARB should facilitate the effective bioremediation of arsenic contaminated sites.
20세기에 들어 산업, 군사 및 다양한 목적으로 비인화성 용매인 PCE와 TCE의 사용량이 증대하였다. 주의를 필요로 하는 물질임에도 불구하고 부주의한 사용과 보관으로 인해 토양, 퇴적토, 지하수에 심각하게 오염되었다. High-chlorinated ethenes은 호기성 박테리아의 oxygenation에 의해 분해되지 않는다. PEC및 TCE의 완전한 탈염소화는 혐기성조건에서만 관찰되어지며, 지난 10연년간의 연구에 의해서 탈염소화 혐기성 미생물의 수의 보고는 증가되었다. 혐기성 조건에서 탈염소화 미생물에 의해 PCE와 TCE는 less-chlorinated ethenes 또는 무해한 ethene으로 전환이 가능하다. 본 연구는 lactate를 electron donor로 이용해 PCE에서 ethene까지 완전히 탈염소화하는 혐기성 배양을 수행했다. PCE로 오염된 퇴적토 시료로부터 혐기성 미생물 배양을 성공했다. PCE가 ethene까지 완전히 분해되는 것이 관찰되었다. 추가적으로 혐기성 미생물 배양액에서 1,2-cis-dichloroethene (cis-DCE)와 vinyl chloride (VC)의 축적이 일어남을 관찰하였다. 혐기성 미생물 배양액에서 Dehalococcoides 16S rRNA gene sequences에 특이적으로 반응하는 primer를 이용한 DGGE를 통해 미생물 군집을 분석하였다. 결론적으로, 우리의 연구에서 PCE를 감소시키는 배양액을 배양했으며, 이 배양엑에는 Dehalococcoides sp. 존재하는 것을 확인하였다.
철을 이용한 반응벽체 (permeable reactive barrier, PRBs) 기술은 유기 화합물로 오염된 지하수를 환원적 반응에 의해 정화시키는 공법이다. 벽체의 매질로 주로 사용되는 영가 철은 반응이 진행됨에 따라 점차 2가 및 3가 철로 산화되어 제거능이 점차 저감된다. 자연계에 존재하거나 동정된 철 환원 박테리아는 산화된 Fe(III)를 Fe(II)로 환원시키는 능력을 가지고 있으며 이와 같이 환원된 Fe(II)는 반응 표면적을 넓히고 다시 할로겐 유기 화합물을 환원적으로 제거할 수 있도록 한다. 본 연구는 철 환원 박테리아로 순수균인 Shewanella algae BrY에 의한 산화철의 환원 경향을 aqueous phase와 solid phase로 나누어 관찰하고 환원된 철이 TCE 제거에 미치는 영향을 iron(II,III) oxide와 iron(III) oxide를 대상으로 하여 파악하는 것을 목표로 하였다. 박테리아는 배지 내에 존재하는 Fe(III)를 우선적으로 사용하여 Fe(II)로 환원시켰으며 선택성은 떨어지지만 입자상의 산화철 표면에 존재하는 Fe(III)도 환원시켰다. 또한 동량의 산화철이 존재할 때 iron(II,III) oxide에 비해 박테리아가 전자수용체로 사용할 수 있는 Fe(III)가 풍부한 iron(III) oxide의 환원이 더 잘 일어남을 알 수 있었고, 환원된 Fe(II)는 박테리아 또는 다른 철 산화물과 침전을 형성하였으며 TCE와의 반응속도 및 제거 능력을 향상시키는 것으로 판단된다.
한강에서 채취한 원수에 염소소독제인 hypochlorite를 $10{\mu}g/m{\ell}$의 농도로 투여한 후 1시간~14일까지 TOC (total organic carbon), 잔류염소량 및 탁도 등을 측정하고 THMs (trihalomethanes), HANs (haloacetonitriles), HKs (haloketones), chloral hydrate 및 HAAs (haloacetic acids) 등의 염소소독 부산물의 생성율을 조사하였다. 그 결과 잔류염소량은 투여후 1시간 경과 시 $6{\mu}g/m{\ell}$ 이상에서 14일째에 $1.23{\mu}g/m{\ell}$으로 감소하였으며 TOC 및 탁도는 큰 차이가 없었다. 7일 후 발생한 총 소독부산 물의 농도는 $101.3ng/m{\ell}$ (789.6 nM)이며 이 중 THMs이 69%로 가장 큰 비중을 차지하였다. 그 외에 HAAs가 19%, chloral hydrate가 10% 정도 검출되었으며, HANs와 HKs 및 chloropicrin 등은 미량 검출되었다. THMs 중에서는 chloroform이 $61.5ng/m{\ell}$로 총 THMs 중 약 89% 정도를 차지하였으며 HANs 중에서는 DCAN이 95%인 $0.72ng/m{\ell}$, HAAs 중에서는 TCAA가 50% 등으로 가장 높은 비율로 검출되었다. 각 부산물의 발생량의 상관관계를 조사한 결과 THMs과 HANs의 경우 THMs의 농도가 $40ng/m{\ell}$인 지점을 경계로 HANs과 경쟁적 발생관계가 있음이 나타났다. HAAs의 경우는 특별한 경향성을 나타내지 않았으나 전체적으로 초기에 산화상태가 큰 화합물에서 산화상태가 작은 화합물로 점차 변화하는 것으로 관찰되었다.
