유부의 가공공정중 미생물 분석과 저장 수명 평가 (Microbial Analysis of Processing and Evaluation of Shelf life of Fried Bean Curd)

  • 유부의 가공공정에 대한 위해 분석을 통한 위생관리 방안을 모색하기 위하여 가공공정별 시설.설비와 작업장 환경 및 원재료와 가공공정별 시료에 대한 미생물 변화와 오염원인을 분석하였으며, 포장유부를 $10^{\circ}C$에서 저장하면서 저장수명을 예측하고 이의 적절한 평가를 위한 품질지표를 도출코자 하였다. 가공공정의 효율성을 도모하고 최종제품의 저장수명을 개선하기 위해서는 가공공정 특히 사용용수, 침지, 성형, 절단, 유탕 이후의 공정에서의 작업장 환경과 시설.설비에 대한 위생 관리 대책과 침지와 성형공정에서의 미생물 증식 억제방안의 마련이 필요하였다. 유부의 저장수명을 예측하기 위한 품질지표로는 조직감 같은 관능지표와 육안적 곰팡이 발생 및 일반세균수가 매우 유용하게 이용될 것으로 판단되었으며, 유부의 저장수명은 최대 6일로 예측되었다. 이러한 연구는 유부의 가공조건 설정과 저장수명 평가에 대해 적절한 기초자료로 활용될 수 있을 것으로 기대된다.

Challenging the Hypothesis of de novo Biosynthesis of Bile Acids by Marine Bacteria

  • Bile acids are essential molecules produced by vertebrates that are involved in several physiological roles, including the uptake of nutrients. Bacterial isolates capable of producing bile acids de novo have been identified and characterized. Such isolates may provide access to novel biochemical pathways suitable for the design of microbial cell factories. Here, we further characterized the ability of Maribacter dokdonensis, Dokdonia donghaensis, and Myroides pelagicus to produce bile acids. Contrary to previous reports, we did not observe de novo production of bile acids by these isolates. Instead, we found that these isolates deconjugated the amino acid moiety of bile acids present in the growth medium used in previous reports. Through genomic analysis, we identified putative bile salt hydrolases, which could be responsible for the different bile acid modifications observed. Our results challenge the hypothesis of de novo microbial bile acid production, while further demonstrating the diverse capacity of bacteria to modify bile acids.

  • Bacterial strains which degrade Arabian Light crude oil were isolated by enrichment culture from oil-spilled soil. The strain A54 was finally selected after testing emulsifying activity and oil conversion rate. Strain A54 was identified as a Acinetobacter sp. based on the morphological, biochemical and physiological characteristics. It appears to be highly specialized for growth on Arabian Light crude oil in minimal salts medium since it showed preference for oil or degradation products as substrates for growth. It was found that it could grow on at least fifteen different hydrocarbons. The optimum cultural and environmental conditions were as follows; 25$\circ$C for temperature, 7,5 for pH, 2.0% for NaCl concentration and 2.0% for crude oil concentration. Additionally, the optimal concentration of NH$_{4}$NO$_{3}$, and K$_{2}$HPO$_{4}$, were 12.5 mM and 0.057 mM, respectively. Cell growth and emulsifying activity as a function of time were also determined. Crude oil degradation and the reduction of product peaks were identified by the analysis of remnant oil by gas chromatography. Approximately 63% of crude oil were converted into a form no longer extractable by mixed organic solvents.

Effectiveness of Phytogenic Feed Additive as Alternative to Bacitracin Methylene Disalicylate on Hematological Parameters, Intestinal Histomorphology and Microbial Population and Production Performance of Japanese Quails

  • This study was conducted to evaluate the effects of phytogenic additive and antibiotic growth promoter in laying Japanese quails. One hundred and sixty five quails were divided into three groups of 5 replicates and 11 quails (8 females and 3 males) in each replicate. Treatment 1 was fed control diet, treatment 2 was fed control diet supplemented with 0.05% bacitracin methylene disalicylate as antibiotic growth promoter and treatment 3 was fed control diet supplemented with 0.1% phytogenic feed additive (PFA) for two periods of 3 weeks each from 37 to 42 weeks of age. Results showed that egg production, eggshell strength, eggshell weight, villus height and villus height to crypt depth ratio were significantly (p${\leq}$0.05) increased and feed consumption, feed conversion ratio, albumen, Haugh unit, cholesterol, low-density lipoprotein, alanine transaminase, gamma glutamyltransferase, alkaline phosphatase, high-density lipoprotein, triglyceride, number of goblet cell, crypt depth and intestinal bacterial population of Coliforms, Salmonella and E. coli were significantly (p${\leq}$0.05) decreased in PFA fed group. It is concluded that addition of PFA containing phytomolecules and organic acids as main ingredients could significantly improve the production parameters and the general health of laying quails as an alternative to antibiotic growth promoters.

  • 본 연구에서는 ethanol 과 일부 유기산(adipic acid, citric acid, acetic acid)을 첨가하였을 때 물김치 발효에 관여하는 미생물 생육의 변화를 측정하였으며 그 결과를 요약하면 다음과 같다. Ethanol 및 유기산의 첨가가 물김치 서식 미생물에 미치는 영향을 직접적으로 살펴보기 위하여 pH 4.3 의 buffer 용액에 ethanol 과 유기산을 농도별로 첨가하고 Lactobacillus plantarum, Leuconostoc mesenteroides, Kluyveromyces marxianus을 접종하여 균의 생육에 미치는 영향을 살펴 본 결과 L. plantarum 와 Leu. mesenteroides에서는 ethanol 농도가 높을수록 그리고 유기산 종류에서는 adipic acid를 첨가했을 때 성장 억제 효과가 가장 크게 나타났다. 비교 균주에서는 L. plantarum이 Leu. mesenteroides보다 유기산과 에탄올에 영향을 받아 생육이 더욱 억제되었다. Ethanol에 대해서는 L. plantarum이 Leu. mesenteroides보다 숙성 72 시간째에 40.0% 정도 생육이 억제되었으나 유기산의 경우에는 종류에 따라 생육억제 정도가 달랐다. Acetic acid의 경우 L. plantarumol 약 40.0% 정도, citric acid의 경우는 L. plantarum이 약 15.0% 정도 생육이 억제되었지만 adipic acid의 경우는 L. plantarum 와 Leu. mesenteroides 모두에게 억제 효과가 있어 무첨가군에 비해 50.0∼60.0% 정도 생육이 감소되었다. K. marxianus에서도 ethanol 농도가 높을수록 성장 억제에 효과적이나 유기산에 대해서는 차이를 보이지 않았다. Ethanol 및 유기산이 각 균에 어떠한 영향을 주었는가을 알기 위해 균체내 물질의 누출을 260nm에서의 흡광도를 측정하여 관찰한 결과 L. plantarum 에서는 ethanol 2.0% 첨가 에서와 adipic acid 0.1 를 첨가한 실험군에서 24 시간 후의 흡광도가 가장 많이 증가되었다. Leu. mesenteroides에서는 ethanol 첨가시 유의적인 차이가 보이지 않았고 유기산 중 adipic acid 첨가시 흡광도가 많이 증가되었다. K. marxianus 에서는 ethanol 농도가 높을수록 흡광도가 증가되었고 유기산 중 citric acid 에서 많이 증가되었다.

Permanent Mycoplasma Removal Removel from Tissue Culture Cells: A Genetic Approach

  • Mycopasma contamination of tissue culture cells easily evades detection and, thus, represents a continous therat to cell biologists. In case where infected cell can not simply be replaced, attempts have to be made to eradicate mycoplacma from the tissue culture cells. A variety of anti-microbial agents have been shown to be toxic to mycoplasma strains ; however, cell associated mycoplasma are often protected from antibiotics at concentrations shown to be effective in vitro. Antibiotic concentrations high enough to be lethal to cell as sociated mycoplasmas frequently are also detrimentrations to the host cells, while moderately increased antibiotic levels tolerated by the host cells often lead to only temporary growth suppression and/or to the emergence of mycoplasma strains resistanct even to high concentrations of the antibiotis applied. Hare, a genetic approach for the elimination of mycoplasma from tissue culture cells that overcomes thens limitations is described. By expression of a selection marker conferring resistance to an otherwise toxic agent, Acholeplasma laidlawii infected BHK-21 cells used as the model system were enabled to temporarily tolerate antibiotic concentrations high enough to be lethal to cell associated mycopalsma while leaving the host cells unharmed. Upon successful mycoplasma eradicated, cultvation of the cured host cells in the absence of the selective agent yielded revertant cell clones that had regained susceptibillity to the toxic agent. Cressation of the selection marker expression was shown to result from the loss of the selection marker DNA, which is a consequence of the fact that the stable and permanent integration of foreign DNA in eucaryotic cell chrosomes is highly inefficient. Thus, the cells were cured from mycoplasma yet remained biochemically unaltered.

연속류식 미생물연료전지의 유기물 제거 및 전기 발생 특성 (Characteristics of Organic Material Removal and Electricity Generation in Continuously Operated Microbial Fuel Cell)

  • 양성자 교환막 미생물연료전지(PEM-MFC)의 경우 양극의 표면적을 기준으로 유기물 제거능력을 산출하면 유기물 부하에 관계없이 $3.0gCOD/m^2$ 수준으로 나타났다. 또 안정적인 전압이 관찰된 시기의 쿨롱 효율은 22.4~23.4 %로 높지 않은 수준이었다. 양성자 교환막은 양성자뿐만 아니라 초산도 통과시키는 것으로 확인되었다. 양성자 교환막을 사용하지 않은 상향류식 미생물연료전지(ML-MFC)의 경우 다공성 RVC 전극을 사용한 관계로 전극의 외부면적당 유기물 제거능력은 $9.3{\sim}10.1gCOD/m^2{\cdot}d$로 나타났다. 이는 양성자 교환막을 사용한 경우에 비하여 3배 정도 높은 수준이다. 그러나 RVC 양극의 비표면적 차이에 따른 유기물 제거 능력 차이는 크지 않았다. ML-MFC의 경우 전기 발생이 안정적이지 못하였으며, 쿨롱 효율도 3.6~3.7 %로 매우 낮은 수준이었다. 전기 발생량이 안정적이지 못한 것은 음극에 성장한 미생물의 영향으로 판단된다. 이를 해결하기 위해 음극부의 공기주입량을 증가시키면 일시적으로 전기 발생이 증가하였으나 오래 지속되지 못하였다.

[ $G_1$ ] Phase Arrest of the Cell Cycle by a Ginseng Metabolite, Compound K, in U937 Human Monocytic Leukamia Cells

  • We recently reported that the ginseng saponin metabolite, compound K (20-O-$\beta$-D-glucopyra-nosyl-20(S)-protopanaxadiol, IH901), inhibits the growth of U937 cells through caspase-dependent apoptosis pathway. In this study, we further characterized the effects of compound K on U937 cells and found that, in addition to apoptosis, compound K induced the arrest of the G1 phase. The compound K treated U937 cells showed increased p21 expression; an inhibitory protein of cyclincdk complex. The up-regulation of p21 was followed by the inactivation of cyclin D and the cdk4 protein, which act at the early $G_1$ phase, and cyclin E, which acts at the late $G_1$ phase. Furthermore, compound K induced the activation of JNK and the transcription factor AP-1, which is a downstream target of JNK. These findings suggest that the up-regulation of p21 and activation of JNK in the compound K treated cells contribute to the arrest of the $G_1$ phase.

김치유산균(Weissella koreensis HO20, Weissella kimchii HO22)으로 발효한 쌀가루의 이화학적 특성 및 이를 이용한 절편의 제조 (Fermentation of rice flour with Weissella koreensis HO20 and Weissella kimchii HO22 isolated from kimchi and its use in the making of jeolpyeon)

  • Demand for a rice cake, a popular traditional food in Korea, is rising, but its industrial-scale production is extremely difficult due to its short shelf-life caused by starch retrogradation and microbial spoilage. By means of the sourdough fermentation technique, we attempt to develop rice cakes with a longer shelf-life. Heterofermentative lactic acid bacteria (Weissella koreensis HO20, Weissella kimchii HO22) isolated from kimchi were used to ferment wet-milled rice flour for their abilities to produce exopolysaccharides and to inhibit the microbial spoilage of rice cakes. After 24 hr of fermentation at $25^{\circ}C$, viable cell counts in rice dough increased from $10^6$ CFU/g to $10^8$ CFU/g and total titratable acidity increased from 0.05% to 0.20%, whereas pH decreased from 6.5 to 5.1. Fermented rice flour showed significantly lower peak, trough, and final viscosities as well as breakdown and setback viscosities measured by rapid viscoanalyzer. Both lactic acid bacteria showed in vitro antifungal activity against Penicillium crustosum isolated from rice cakes. The antifungal activity remained constant after the treatments with heat, proteinase K and trypsin, but fell significantly by increase of pH. Rice cakes made of fermented rice flour were found to retard mycelial growth of P. crustosum. The degree of retrogradation as measured by the hardness of the rice cake was significantly reduced by the use of fermented rice flour. The results suggest that use of fermented rice flour has a beneficial role in retarding starch retrogradation and in preventing fungal growth, hence extending the shelf-life of rice cakes.

Nutrient intake, digestibility and performance of Gaddi kids supplemented with tea seed or tea seed saponin extract

  • Objective: An experiment was conducted to determine the nutrient intake, digestibility, microbial protein synthesis, haemato-biochemical attributes, immune response and growth performance of Gaddi kids fed with oat fodder based basal diet supplemented with either tea seed or tea seed saponin (TSS) extract. Methods: Eighteen male kids, $7.03{\pm}0.16$ months of age and $19.72{\pm}0.64kg$ body weight, were distributed into three groups, $T_0$ (control), $T_1$, and $T_2$, consisting of 6 animals each in a completely randomized design. The kids were fed a basal diet consisting of concentrate mixture and oat fodder (50:50). Animals in group III ($T_2$) were supplemented with TSS at 0.4% of dry matter intake (DMI), and group II ($T_1$) were supplemented with tea seed at 2.6% of DMI to provide equivalent dose of TSS as in $T_2$. Two metabolism trials were conducted, 1st after 21 days and 2nd after 90 days of feeding to evaluate the short term and long term effects of supplementation. Results: The tea seed ($T_1$) or TSS ($T_2$) supplementation did not affect DMI as well as the digestibility of dry matter, organic matter, crude protein, neutral detergent fibre, and acid detergent fibre. Nutritive value of diet and plane of nutrition were also comparable for both the periods. However, the average daily gain and feed conversion ratio (FCR) were improved (p<0.05) for $T_1$ and $T_2$ as compared to $T_0$. The microbial protein supply was also higher (p<0.05) for $T_1$ and $T_2$ for both the periods. There was no effect of supplementation on most blood parameters. However, the triglyceride and low density lipoprotein cholesterol levels decreased (p<0.05) and high density lipoprotein-cholesterol level increased (p<0.05) in $T_2$ as compared with $T_0$ and $T_1$. Supplementation also did not affect the cell mediated and humoral immune response in goats. Conclusion: Tea seed at 2.6% of DMI and TSS at 0.4% DMI can be fed to Gaddi goats to improve growth rate, FCR and microbial protein synthesis.
