Production of Mono-Hydroxylated Derivatives of Terpinen-4-ol by Bacterial CYP102A1 Enzymes

  • CYP102A1 from Bacillus megaterium is an important enzyme in biotechnology, because engineered CYP102A1 enzymes can react with diverse substrates and produce human cytochrome P450-like metabolites. Therefore, CYP102A1 can be applied to drug metabolite production. Terpinen-4-ol is a cyclic monoterpene and the primary component of essential tea tree oil. Terpinen-4-ol was known for therapeutic effects, including antibacterial, antifungal, antiviral, and anti-inflammatory. Because terpenes are natural compounds, examining novel terpenes and investigating the therapeutic effects of terpenes represent responses to social demands for eco-friendly compounds. In this study, we investigated the catalytic activity of engineered CYP102A1 on terpinen-4-ol. Among CYP102A1 mutants tested here, the R47L/F81I/F87V/E143G/L188Q/N213S/E267V mutant showed the highest activity to terpinen-4-ol. Two major metabolites of terpinen-4-ol were generated by engineered CYP102A1. Characterization of major metabolites was confirmed by liquid chromatography-mass spectrometry (LC-MS), gas chromatography-MS, and nuclear magnetic resonance spectroscopy (NMR). Based on the LC-MS results, the difference in mass-to-charge ratio of an ion (m/z) between terpinen-4-ol and its major metabolites was 16. One major metabolite was defined as 1,4-dihydroxyp-menth-2-ene by NMR. Given these results, we speculate that another major metabolite is also a mono-hydroxylated product. Taken together, we suggest that CYP102A1 can be applied to make novel terpene derivatives.

ANALYSIS OF THE LiF:Mg,Cu,Si TL AND THE LiF:Mg,Cu,P TL GLOW CURVES BY USING GENERAL APPROXIMATION PLUS MODEL

  • In this paper, we used computerized glow curve deconvolution (CGCD) software with several models for the simulation of a TL glow curve which was used for analysis. By using the general approximation plus model, parameters values of the glow curve were analyzed and compared with the other models parameters (general approximation, mixed order kinetics, general order kinetics). The LiF:Mg,Cu,Si and the LiF:Mg,Cu,P material were used for the glow curve analysis. And we based on figure of merits (FOM) which was the goodness of the fitting that was monitored through the value between analysis model and TLD materials. The ideal value of FOM is 0 which represents a perfect fit. The main glow peak makes the most effect of radiation dose assessment of TLD materials. The main peak of the LiF:Mg,Cu,Si materials has a intensity rate 80.76% of the whole TL glow intensity, and that of LiF:Mg,Cu,P materials has a intensity rate 68.07% of the whole TL glow intensity. The activation energy of LiF:Mg,Cu,Si was analyzed as 2.39 eV by result of the general approximation plus(GAP) model. In the case of mixed order kinetics (MOK), the activation energy was analyzed as 2.29 eV. The activation energy was analyzed as 2.38 eV by the general order kinetics (GOK) model. In the case of LiF:Mg,Cu,P TLD, the activation energy was analyzed as 2.39 eV by result of the GAP model. In the case of MOK, the activation energy was analyzed as 2.55 eV. The activation energy was analyzed as 2.51 eV by the GOK model. The R value means different ratio of retrapping-recombination. The R value of LiF:Mg,Cu,Si TLD main peak analyzed as $1.12\times10^{-6}$ and $\alpha$ value analyzed as $1.0\times10^{-3}$. The R of LiF:Mg,Cu,P TLD analyzed as $7.91\times10^{-4}$, the $\alpha$ value means different ratio of initial thermally trapped electron density-initial trapped electron density (include thermally disconnected trap electrons density). The $\alpha$ value was analyzed as $9.17\times10^{-1}$ which was the difference from LiF:Mg,Cu,Si TLD. The deep trap electron density of LiF:Mg,Cu,Si was higher than the deep trap electron density of LiF:Mg,Cu,P.

Empowerment and Ethical Sensitivity of Nurse in the Hospital (간호사의 임파워먼트와 윤리적 민감성)

  • Purpose: This study was to identify the degree of empowerment and ethical sensitivity of staff nurses and to examine the relationship between empowerment and ethical sensitivity. Method: Data were collected from 235 staff nurses in 4 hospitals in Gyeonggi Province using a questionnaire and collected data was analyzed by the SPSS PC program. Results: The degree of empowerment was 2.76(${\pm}$ .35) out of a possible score of 4 and that of ethical sensitivity was 0.71(${\pm}$ .11) out of a possible score of 1. The relationship of between empowerment and ethical sensitivity showed statistically significant positive correlation(r=.34, p=.00). For general characteristics, there was a significant difference in empowerment according to age(F=13.18, p=.00), educational background(t=-2.09, p=.04) and clinical practice career(F=15.15, p=.00) and in ethical sensitivity according to age(F=4.01, p=.02). In characteristics related to ethics, there was a significant difference in empowerment according to experience of ethics instruction in clinical practice(t=2.25, p=.03), attitude toward the nursing profession(F=7.96, p=.00) and ethical standards(F=9.39, p=.00) and in ethical sensitivity according to attitude toward the nursing profession(F=2.94, p=.03). Conclusion: Findings suggest that a systemic and effective training program reflecting the above general and related ethics characteristics be developed to enhance empowerment and ethical sensitivity.

Carbon Reduction Services of Evergreen Broadleaved Landscape Trees for Ilex rotunda and Machilus thunbergii in Southern Korea

  • This study quantified carbon reduction services through direct harvesting of Ilex rotunda and Machilus thunbergii, which are the typical urban landscape tree species in southern Korea. A total of 20 open-grown tree specimens (10 specimens for each species) were selected reflecting various sizes of stem diameter at breast height of 1.2 m (DBH) at a regular interval. The study measured biomass for each part of the tree specimens including roots to compute total carbon storage per tree. Annual carbon uptake per tree was also calculated analyzing the DBH growth rate of stem disk specimens. Quantitative models were developed using DBH as an independent variable to easily estimate storage and annual uptake of carbon by tree growth for each species. All the models had a high goodness-of-fit with R2=0.95-0.99. The difference in carbon reduction services between DBH sizes increased with increasing DBH. The storage and annual uptake of carbon from a tree with DBH of 10 cm were 13.5 kg and 2.4 kg/yr for I. rotunda, and 19.1 kg and 3.6 kg/yr for M. thunbergii, respectively. The tree of this size stored the amount of carbon equivalent to that emitted from a gasoline use of approximately 24 L for I. rotunda and 34 L for M. thunbergii, respectively. The study provides actual measurement data to quantify carbon reduction services of urban open-grown landscape trees for the warm-temperate species that have been little known until now.

Candidate Gene Analysis to Rice Bacterial Leaf Blight Resistance of Korean Races of Xoo (Xanthomonas oryzae) in Rice Genetic Resources by GWAS Analysis

  • Bacterial leaf blight (BLB), caused by X. oryzae pv. oryzae(Xoo), is one of the most destructive diseases of rice due to its high epidemic potential. Understanding BLB resistance at a genetic level is important to further improve the rice breeding that provides one of the best approaches to control BLB disease. In the present investigation, a total of 10,000 accessions of rice germplasm were tested to resistance degree of four Korean isolated races (K1, K2, K3 and K3a) of Xoo by bioassay and a diverse 268 accessions was selected to the genome-wide association study (GWAS) using high quality 34,724 SNPs to identify the associated with resistance loci. LOC_Os04g53160 of chromosome 4 was significantly associated with K1 race resistant. LOC_Os11g46230 and LOC_Os11g47150 of chromosome 11 were highly associated with K2 and K3 races as 23.7 and 27.4 of -log(P) value, but K3a resistant loci was weakly associated at LOC_Os03g55270 of chromosome 3. The results of the GWAS validate known gene of BLB resistant and identified novel loci of R genes that provide useful targets for further investigation to help the breeding system and identified gene and QTL provide valuable sources for further functional characterization.

A Study on the Ecological Indices for the Assessment of the Function and Maturity of Artificial Reefs (인공어초의 기능도와 성숙도 평가를 위한 생태학적 지수에 대한 연구)

  • We reviewed foreign evaluation systems based on the macrobenthic and macroalgal communities and developed a system, composed of a set of ecological indices able to evaluate the functionality (FI, Functional Index; estimation of stability and productivity) and maturity (MI, Maturity Index; comparisons with biological parameters of natural reefs) of artificial reefs by comparing the status in the adjacent natural reefs in Korean coastal waters. The evaluation system was applied to natural and artificial reefs/reef-planned areas (natural reefs), established in the 5 marine ranching areas (Bangnyeong-Daechung, Yeonpyung, Taean, Seocheon and Buan) in the west coast of Korea. The FI ranged between 31.6 (Bangnyeong-Daechung) and 72.5% (Buan) and MI did between 53.1 (Seocheon) and 76.9% (Taean) in average. The evaluation of artificial reefs by the two indices, showed the most appropriate status in Taean. The FI between the adjacent artificial and natural reefs were in significant linear relationship ($r^2=0.83$, p=0.01). This indicated the local status of biological community may be critical in determining the functionality of the artificial reefs. We have suggested an integrative but preliminary evaluation system of artificial reefs in this study. The output from the evaluation system may be utilized as a tool for environment/resource managers or policy makers, responsible for effective use of funds and decision making. Given the importance, we need to use the options to enhance and improve the accuracy as follows: (1) continuous validation of the evaluation system and rescaling the criteria of indicators, (2) vigorous utilization of observation and experience through the application and data accumulation and (3) development and testing of brand-new indicators.

Pharmacokinetic Analysis of Montelukast in Healthy Korean Volunteers by High Performance Liquid Chromatography-Tandem Mass Spectrometry

  • A rapid and specific high performance liquid chromatography-tandem mass (LC/MS/MS) method for the analysis of montelukast in human plasma has been developed and validated. After cold acetonitrile-induced precipitation of the plasma samples, montelukast and glipizide (internal standard, IS) were eluted on a reverse-phase $C_{18}$ column by isocratic mobile phase consisted of 10 mM ammonium formate buffer (adjusted to pH 3.5 with formic acid) and acetonitrile (3:97, v/v). Acquisition was performed with multiple reaction monitoring (MRM) mode by monitoring the transitions: m/z 587.2${\rightarrow}$ 423.2 for montelukast and m/z 446.0${\rightarrow}$321.2 for IS. Ranges of concentration for calibration curves (10-1000 ng/mL) showed correlation coefficients ($r^2$) were better than 0.9948. Precision of intra- and inter-day ranged from 3.70 to 11.68% and from 3.04 to 12.95%, accuracy of intra-day and inter-day ranged from 93.34 to 102.75% and from 100.79 to 107.63%, respectively. The described method provides a fast and sensitive analytical tool for determining montelukast levels in plasma, and was successfully applied to a pharmacokinetic study in 16 healthy human subjects after oral administration of 10mg tablet formulation of montelukast sodium under fasting conditions.

A Study on Self Care and the Problems in Patient with Ostomy (장루보유자의 자가 간호정도 및 문제점에 관한 연구)

  • This study was conducted to confirm the degree of self care and problems in patients with ostomy. The subjects were 80 patients with ostomy who were visiting to OPD in K University Medical Center and were members in the Ostomy Association of Korea. Data were collected from August, 2004 to September. The research tools were a self care measure for ostomy patient developed by Park (1996) and a problem measure for ostomy patient developed by Lee (1996). Data were analyzed by t-test, ANOVA, and Pearson's correlation. The results were as following: 1. Male (65.8%) and female (34.2%) had the ostomy in the most. Age was the most 61 years old and over. 68.5% of the subjects had managed the ostomy by themselves. 2. The mean score of self care in ostomy was 3.57 score. Hygiene of skin care around ostomy in the behavior of self care was the highest as mean 4.47 score. 3. The problems were as following: 1) expense 2) a decline on Activity of Daily Living 3) discomfort in travel 4) worry about ostomy 5) trouble of workplace 4. In the relationship between self care and religion there was significant difference in religion(t= 2.727, P=.008). 5. The relationship between self care and problem with ostomy was found to have statistically significant negative correlation(r= -.237, P=.041).

Meta-Analytic Approach to the Effects of Food Processing Treatment on Pesticide Residues in Agricultural Products (식품가공처리가 농산물 잔류농약에 미치는 영향에 대한 메타분석)

  • A trial of combining and quantifying the effects of food processing on various pesticides was carried out using a meta-analysis. In this study, weighted mean response ratios and confidence intervals about the reduction of pesticide residue levels in fruits and vegetables treated with various food processing techniques were calculated using a statistical tool of meta-analysis. The weighted mean response ratios for tap water washing, peeling, blanching (boiling) and oven drying were 0.52, 0.14, 0.34 and 0.46, respectively. Among the food processing methods, peeling showed the greatest effect on the reduction of pesticide residues. Pearsons's correlation coefficient (r=0.624) between weighted mean response ratios and octanolwater partition coefficients ($logP_{ow}$) for twelve pesticides processed with tap water washing was confirmed as having a positive correlation in the range of significance level of 0.05 (p=0.03). This means that a pesticide having the higher value of $logP_{ow}$ was observed as showing a higher weighted mean response ratio. These results could be used effectively as a reference data for processing factor in risk assessment and as an information for consumers on how to reduce pesticide residues in agricultural products.

A Relationship between Interdental Papilla Existence and the Distance from Contact Point to Interdental Alveolar Crest in the Maxillary Anterior Dentition of Korean adults (한국인 상악 전치부에서 치간유두 존재와 치아접촉점과 치간골 거리와의 관계)

  • As the public becomes concerned with looking younger and healthy, aesthetic considerations will become more relevant to dental treatment planning. The purpose of this study was to evaluate the relationship between interdental papilla existence and the distance from contact point to interdental alveolar crest in the maxillary anterior dentition of korean. Fifty-nine Korean adult consist of adults. 34 males and 25 females participated in the study. Papilla Index(PI) was recorded. The distance between contact point and interdental alveolar crest was measured by sounding with Williams probe. Measurement were carried out in 257 maxillary anterior interproximal area. The results showed that mean PI was 1.95 and mean distance between contact point and interdental alveolar crest was 5.07mm. The correlation between the papilla index and distance was negative and statistically significant(r=-0.819; p=0.000). A high negative correlation existed between PI and distance from contact point to alveolar crest. When the distance between contact point and alveolar crest was 4mm, the papilla got lost on a half of all cases. When the distance was 5mm, the papilla was present almost 11%. When the distance was 6mm, the papilla was present 4%. When the distance was 7mm or more, the papilla was lost in all cases.
