Synthetic Lethal Mutations with spmex67 of Schizosaccharomyces pombe in the Mediation of mRNA Export

  • Mex67p/Tap are evolutionally conserved mRNA export factors. To identify mutations in genes that are functionally linked to mex67 with respect to mRNA export, we used a synthetic lethal genetic screen in Schizosaccharomyces pombe. Three synthetic lethal mutants were isolated and mutations in these mutants defined separate complementation groups. These mutants exhibited the accumulation of poly A$\^$+/ RNA in the nucleus, with a decrease in the cytoplasm under synthetically lethal conditions, suggesting that the mutations cause an mRNA nuclear export defect. In addition, the S. pombe genes that were found to be involved in mRNA export did not suppress the synthetic lethality of these mutants. These results indicate that the isolated mutants contain mutations in new genes, which are involved in mRNA export from the nucleus.

Comparative Analysis of Filtering Techniques for Vegetation Points Removal from Photogrammetric Point Clouds at the Stream Levee (하천 제방의 영상 점군에서 식생 점 제거 필터링 기법 비교 분석)

  • This study investigated the application of terrestrial light detection and ranging (LiDAR) to inspect the defects of the vegetated levee. The accuracy of vegetation filtering techniques was compared by applying filtering techniques on photogrammetric point clouds of a vegetated levee generated by terrestrial LiDAR. Representative 10 vegetation filters such as CIVE, ExG, ExGR, ExR, MExG, NGRDI, VEG, VVI, ATIN, and ISL were applied to point cloud data of the Imjin River levee. The accuracy order of the 10 techniques based on the results was ISL, ATIN, ExR, NGRDI, ExGR, ExG, MExG, VVI, VEG, and CIVE. Color filters show certain limitations in the classification of vegetation and ground and classify grass flower image as ground. Morphological filters show a high accuracy of the classification, but they classify rocks as vegetation. Overall, morphological filters are superior to color filters; however, they take 10 times more computation time. For the improvement of the vegetation removal, combined filters of color and morphology should be studied.

Schizosaccharomyces pombe nup97, which Genetically Interacts with mex67, is Essential for Growth and Involved in mRNA Export

  • We have isolated previously three synthetic lethal mutants in Schizosaccharomyces pombe, which genetically interact with mex67, in order to identify the genes involved in mRNA export. A novel nup97 gene was isolated by complementation of the growth defect in one of the synthetic lethal mutants, SLMex3. The nup97 gene contains one intron and encodes an 851 amino-acid protein that is similar to nucleoporins, Nppl06p in S. pombe and Nic96p in Saccharomyces cerevisiae. The nup97 gene is essential for vegetative growth, and nup97 null mutant harboring pREP41X-Nup97 showed $poly(A)^+$ RNA export defect when expression of nup97 is repressed in the presence of thiamine. These results suggest that nup97 is involved in mRNA export from the nucleus to cytoplasm.

  • Safe and cheap emulsion explosives have recently replaced the existing CD explosives in order for people to reduce the prime cost and to prevent the safety accidents from happening in construction and civil engineering sites. However, the emulsion explosives have been in reality fared with difficulties in terms of the blasting force when using them in the tunnel constructed in the rock mass composed of hard rock. In this regards, this study is to verify their blasting efficiency and possibility of construction by applying MegaMEX, one of the high performance Emulsion explosives, to the rock mass of hard rock. In terms of their blasting efficiency such as advance ratio and fragmentation, it has turned out that they have overcome the limit of the existing Emulsion explosives and they have had the equivalent level of MegaMITE, one of the GD(Gelatin dynamite) types of explosives while they have been also advantageous to the environmental aspects.

  • This paper represents a new powertrain simulation methodology using the object-oriented programming paradigm. The advantage of the object-oriented is the module interchangeability and simulation flexibility. Powertrain subsystems and controller modules are implemented using the MEX files in MATLAB Simulink in this paper, preserving module interchangeability. Currently, the required CPU time on a 75MHz Pentium PC is about three times the real time. It is anticipated that the "Automotive Powersystem Toolbox" being developed in this research would be of much utility in designing subsystem controllers as well as in designing subsystem mechanicals.chanicals.

  • The sp%pl null mutant was constructed to study the function of fission yeast Schizosaccharomyces pombe spThp1, which is homologous to budding yeast Saccharomyces cerevisiae THP1. Tetrad analysis showed that the spThp1 is not essential for vegetative growth. The spThp1 null mutant also showed no massive poly(A)+ RNA export defect. However, spThp1 null is genetically associated with spMex67 null. These results suggest that spThp1 is involved in mRNA export out of the nucleus.

  • The web ontology language(OWL) has been used by ontology designers to construct ontology. In order to derive hidden information(concept subsumption, concept satisfiability and realization) of OWL ontology, a number of OWL reasoners have been introduced. But most reasoners simply report these information without process for any arbitrary entailment and unsatisfiable concept derived from a OWL ontologies. In this paper, we propose Minimum Expression Axiom Set(MEXS) detection and storing for debugging unsatisfiable concepts in ontology. In order to detect MEXS, we need to find axiom to cause inconsistency in ontology. Therefore, our work focused on two key aspects: given a inconsistency ontology, identifying the roots of axioms to occur unsatisfiable and derived axioms from among them; and extracting MEXS. Our results can be applicable to all application, which is at the basis of the description logic.
