Chaenomeles sinensis (Thouin) Koehne fruit (Rosaceae) has been used as a traditional medicine in Korea, Japan and China to treat sore throat, diarrhea and inflammation. The ethanol extract of C. sinensis fruit was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. Among those fractions, the n-butanol fraction showed the most potent DPPH radical scavenging and superoxide quenching activities. To verify antioxidant activities, the n-butanol fraction was checked the activities of superoxide dismutase (SOD) and catalase activities, and intracellular ROS levels and oxidative stress tolerance in Caenorhabditis elegans. Furthermore, to see if increased stress tolerance of worms by treating of the n-butanol fraction was due to regulation of stress-response gene, we quantified SOD-3 expression using transgenic strain. Consequently, the n-butanol fraction elevated SOD and catalase activities of C. elegans, and reduced intracellular ROS accumulation in a dose-dependent manner. Moreover, the n-butanol fraction-treated CF1553 worms exhibited significantly higher SOD-3::GFP intensity.
The objective was to examine the antifungal activity of Pinus densiflora extract for the control of turfgrass fungal diseases. Antifungal activities of the various fractions of n-hexane, methylene chloride (Ch), ethyl acetate (EtOAc), and n-butanol from P. densiflora were evaluated against Rhizoctonia solani AG1-1B, R. solani AG2-2IV, Sclerotinia homoeocarpa, R. cerealis, Pythium spp., and Colletotrichum graminicola. The Ch and EtOAc fractions showed antifungal activity against Pythium sp. and C. graminicola in paper disc assay. The effective concentration to produce 50% mycelial inhibition ($EC_{50}$) using five discriminatory concentrations of pinosylvin (1) from the Ch fraction of P. densiflora was evaluated on R. solani AG1-1B, R. solani AG2-2IV, R. cerealis, and S. homoeocarpa. S. homoeocarpa showed the highest sensitivity with the lowest mean $EC_{50}$ value ($8.426{\mu}g/mL$) among the four pathogens. Among the three Rhizoctonia pathogens, R. cerealis had the highest mean $EC_{50}$ value ($99.832{\mu}g/mL$) and R. solani AG2-2IV, with the lowest sensitivity, had the lowest $EC_{50}$ value ($39.696{\mu}g/mL$). These results suggested that pinosylvin (1) from P. densiflora could be a valuable lead compound in the improvement of a novel antifungal agent.

Antioxidant effects and nuclear factor E2-related factor-2 (Nrf-2) signal pathway of methanol extract and 4 fractions [n-hexane, methylene chloride, ethyl acetate (EtOAc), and n-butanol fractions] from Petasites japonicus were investigated. The EtOAc fraction showed highest polyphenol and flavonoid contents among other fractions. In addition, EtOAc fraction showed stronger scavenging activity against superoxide anion radical than other fractions. Furthermore, we investigated antioxidants effects of the EtOAc fraction under cellular system using $LLC-PK_1$ cells. The EtOAc fraction dose-dependently increased the antioxidant protein expressions of heme oxygenase 1 (HO-1) and thioredoxin reductase 1 (TrxR1) known to be involved in oxidative stress, through activation of Nrf-2. The treatment of EtOAc fraction ($100{\mu}g/mL$) led to the elevation of the high expression of Nrf-2-dependent factor such as HO-1 and TrxR1. These results indicated that the EtOAc fraction of P. japonicus showed high antioxidant activity by regulation of Nrf-2 signaling pathway.

Scutellariae Radix has been widely used as oriental herbal medicine for the treatment of bacterial infections in the respiratory or the gastrointestinal tract. In partition experiment for better understanding of herbal medicine with various solvents, baicalein or wogonin have more hydrophobic characteristics than baicalin or wogonoside. Unexpectedly, methylene chloride could extract more for baicalin or wogonoside over other active ingredients. New compound from baicalin is discovered casting frointier area on herbal medicine in the future. Application study with new molecule hydrolyzed from baicalin is on the way for better treatment of the patient against specific disease. The baicalin modified with reaction has been shown weak antibacterial activity against Streptococcus pyogenes 308A and Pseudomonas aeruginosa 1771. The minimum inhibitory concentration (MIC) of the baicalin modified compound against those strains were about $600\;{\mu}g/mL$, respectively. In vitro antitumor experiment, $EC_{50}$ of baicalin modified with reaction was more than $300\;{\mu}g/mL$ and $EC_{50}$ of baicalein was $100\;{\mu}g/mL$. Among these compounds, baicalin exhibited high level of antitumor activity. $EC_{50}$ of baicalin was less than $33.3\;{\mu}g/mL$.
Fallen needles (8.5kg) of Larix kaempferi were separately collected, extracted with 95% EtOH. EtOH extract was evaporated under reduced pressure, concentrated then successively fractionated with a series of hexane, methylene chloride, ethylacetate and water on a separatory funnel. Then, each fraction was freeze dried. A portion of ethylacetate and water soluble powder were packed on a column chromatography (Sephadex LH-20) eluting with aqueous MeOH and EtOH-hexane mixture. Spectrometric analyses such as NMR and FAB-MS including TLC were performed to characterize the structures of isolated compounds. 5 compounds were isolated from the fallen needles of Larix kaempferi. The antioxidative activities of each fraction and isolated compounds were done by DPPH radical scavenging test.

As an ongoing study about Allium hookeri (Liliaceae), this study was performed to evaluate the anti-oxidative effect of the leaves of this plant. Ethanol extract of A. hookeri leaves was successively partitioned as methylene chloride, ethyl acetate, n-butanol and $H_2O$ soluble fractions. The ethyl acetate soluble fraction showed the most potent DPPH radical scavenging and superoxide quenching activities among those fractions. To prove antioxidant activity of ethyl acetate fraction of A. hookeri leaves, we checked the activities of superoxide dismutase (SOD) and catalase, and intracellular ROS level and oxidative stress tolerance in Caenorhabditis elegans. In this study, the inhibitory effect of Atriplex gmelinii C. A. Mey. against the activity of MMP-2 and MMP-9 secreted from phorbol-12-myristate-13-acetate (PMA)-stimulated HT-1080 cells was evaluated by gelatin zymography and enzyme-linked immunosorbent assay (ELISA), reverse transcription polymerase-chain reaction (RT-PCR), and Western blot assay. Specimens of the halophyte A. gmelinii were extracted twice for 24 hr with methylene chloride ($CH_2Cl_2$), and then twice with methanol (MeOH), in turn. Each extract significantly inhibited the enzymatic activities in gelatin zymography and MMP ELISA kit, and expression of MMP-2 and 9 in mRNA and protein levels. Two crude extracts were combined and then the combined crude extracts were fractionated into n-hexane, 85% aqueous methanol (85% aq.MeOH), n-butanol (n-BuOH), and water ($H_2O$) fractions, according to solvent polarity. Among solvent-partitioned fractions, the 85% aq.MeOH fraction showed the strongest inhibitory effect against MMP-2 and -9 in gelatin zymography and MMP ELISA kit. In RT-PCR, all solvent-partitioned fractions significantly suppressed mRNA expression of MMP-2 and -9. On the other hand, in Western blot assay, all solvent-partitioned fractions except $H_2O$ significantly reduced expression levels of protein. HT 1080 cell migration was most significantly inhibited by the n-BuOH fraction followed by the 85% aq.MeOH and $H_2O$ fractions. These results suggest that A. gmelinii could be used as a potential source to inhibit tumor cell metastasis.
Robinia pseudoacacia L. (Leguminosae) is widely distributed in Asia, North America and Europe. The root bark has been traditionally used for hemostasis, arthritis and hypertension. Therefore, this study was conducted to identify the biological activity and the bioactive constituents of the root bark. We found that the methanol extract obtained from the root bark of R. pseudoacacia reduced the level of ROS and NO production in LPS-induced inflammation of RAW 264.7 cell line. Among the fractions, methylene chloride fraction showed the highest inhibitory activity against the inflammation. Seven constituents (1-7) were isolated from this fraction, and the chemical structures were determined to be medicarpin (1), (-)-vestitol (2), indole 3-carboxaldehyde (3), 3-acetylindole (4), liquiritigenin (5), 4(1H)-quinolone (6) and 8-methoxyononin (7). Among the isolates, medicarpin (1), (-)-vestitol (2), 3-acetylindole (4) and liquiritigenin (5) inhibited ROS and NO production in a dose-dependent manner. This is the first study to show the anti-inflammatory activity of the root bark of R. pseudoacacia, and it is suggested that the four constituents (1, 2, 4, and 5) could play a role in the biological activity.
We investigated the chemical components of red onion powder dried using the low temperature vacuum method and the inhibitory effects of solvent extracts of the dried red onion powder on the growth of HT-1080 human fibrosarcoma and HT-29 human colon cancer cells and $H_2O_2$-induced oxidative stress. The moisture content of the dried red onion powder was 17.95%, while the vitamin C content was 96 mg/100 g and the total phenols content was 39.1 mg/mL. The inhibitory effects of acetone with methylene chloride (A+M) and methanol (MeOH) extracts of the red onion powder on the growth of HT-1080 and HT-29 cancer cells increased in a dose dependent manner (p<0.05). The inhibitory effect was greater on the growth of HT-29 cells, while the A+M extracts had a higher inhibitory effect than the MeOH extracts. Treatment with the hexane, 85% aq. methanol, butanol and water fractions of the extract led to significant inhibition of the growth of both cancer cell lines (p<0.05). Among the fractions, the hexane and 85% aq. methanol fractions showed a greater inhibitory effect. To determine the protective effect on $H_2O_2$-induced oxidative stress, a DCFH-DA (dichlorodihydrofluorescin diacetate) assay was conducted. All fractions, including the crude extracts of dried red onion, appeared to lead to a significant reduction in the levels of intracellular reactive oxygen species (ROS), and these reductions occurred in a dose dependent fashion (p<0.05). Among the fractions, the 85% methanol fraction showed the greatest protective effect on the production of lipid peroxides.

The effect of dried mackerel extract on the production of nitric oxide (NO) and cytokines, including interleukin-6 (IL-6), tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), and interferon-${\gamma}$ (IFN-${\gamma}$), was investigated. All extracts and fractions from dried mackerel significantly reduced NO production induced by lipopolysaccharide (LPS). Among the extracts, acetone+methylene chloride (A+M), n-hexane, and 85% aqueous methanol (MeOH) showed the strongest inhibitory effects. The 85% aqueous MeOH fraction at a concentration of $10{\mu}g$ significantly decreased LPS-induced IL-6 and TNF-${\alpha}$ production after 6 hr of incubation. In the case of LPS-induced IFN-${\gamma}$ production, the 85% aqueous MeOH fraction decreased the production of IFN-${\gamma}$ afer 6, 24, and 72 hr of incubation in a dose-dependent manner. The results show that an 85% aqueous MeOH fraction inhibits the production of NO and proinflammatory cytokines (IL-6, TNF-${\alpha}$, IFN-${\gamma}$), suggesting that this fraction acts as a potent immunomodulator.
