Anti-Rheumatoid Arthritis Effect of the Kochia scoparia Fruits and Activity Comparison of Momordin Ic, its Prosapogenin and Sapogenin

  • MeOH extract of Kochia scoparia was fractionated into $CHCl_3-$, EtOAc- and BuOH extracts and the last fraction were hydrolyzed by 3%-NaOH ($MeOH-H_2O$) to compare the bioactivities on antinociceptive and anti-inflammatory effects. Silica gel column chromatography of BuOH fraction afforded a large amount of $3-Ο-{\beta}-D-xylopyranosyl {\;}(1{\rightarrow}3)-{\beta}-D-glucuronopyranosyl$ oleanolic acid (momordin Ic, 4) and that of acid hydrolysate of BuOH fraction gave $3-Ο-{\beta}-D-glucuronopyranosyl oleanolic$ acid (momordin Ib, 3), its 6'-Ο-methyl ester (2) and oleanolic acid (1). Silica gel column chromatography of alkaline hydrolysate afforded a large amount of 4. MeOH extract and both EtOAc- and BuOH fractions were active in the rheumatoidal rat induced Freund's complete adjuvant reagent (FCA) whereas $CHCl_3$ fraction was inactive. Compound 1 and 4 showed significant activities in the same assay but oleanolic acid 3-Ο-glucuronopyranoside (3) showed no activity. These fashions were also observed in carrageenan-induced edema of the rat and in the antinociceptive activity tests undertaken in hot plate- and writhing methods. These results suggest that momordin Ic and its aglycone, oleanolic acid, could be active principles for rheumatoid arthritis.

Pathogenic bacteria causing rot in commercial soybean sprout cultivation

  • Soybean sprout pathogenic bacteria were isolated from the large, deep containers of a commercial factory. Over a period of one year, 40 pathogenic-like bacteria were isolated among a total of 732 isolates. In addition to bacteria previously reported to be associated with rotting, such as Pseudomonas putida and Erwinia carotovora, several other genera were also identified: Acinetobacter spp., Chryseobacterium spp., Klebsiella sp., Pantoea agglomerans, Bacillus sp. Fatty acid methyl ester (FAME) analysis using the Microbial ID (MIDI) system, and 16s rRNA sequence analysis, yielded identical results, confirming the identities of these microorganisms. Several types of selective media were not good for identification and determination of population structure in commercial environments, as colony type was not specific to the genus. There was no dominant bacterium, and we were not able to find the main bacterium responsible for soybean spout rot. Even though we did not identify a major target for controlling rot or screening for resistant cultivars, the results of this study indicated that bacterial rot of soybean sprout is endemic. In addition, it emerged that factory epidemics in summer are not caused by the bacteria isolated in this study.

Pseudomonas putida Strain 17 Isolated from Replant Soil Promotes Tomato Growth and Inhibits Conidial Germination of Soilborne Plant Pathogens

  • The induction of growth promotion on numerous crops by rhizobacteria is a well documented phenomenon. In case of tomato (Lycopersicon esculentum), fruit yield is higher in replant soil than that in fresh soil. To investigate what kind of rhizobacterium is involved, microbial community in rhizosphere and on rhizoplane of tomato plants from each soil was analyzed by dilution plating on selective media. Many Gram-negative bacteria and actinomycetes were isolated from tomato in replant soil. One Gram-negative rhizobacterium isolated was identified as Pseudomonas putida based on its biochemical characteristics, fatty acid methyl ester analysis and 16S rDNA sequence. This bacterium designated strain 17 inhibited the growth of Pseudomonas corrugata, and increased growth of tomato seedlings. In addition, its culture filtrate inhibited conidial germination of plant-pathogenic fungi such as Fusarium oxysporum f. sp. radicis-lycopersici, F. oxysporum f. sp. cucumerinum, and Nectria radicicola. Scanning electron microscopy revealed strain 17 colonized and persisted on the epidermal surfaces of tomato radicles and roots. These results suggest that P. putida strain 17 may serve as a biological control agent to suppress multiple soil-borne diseases for tomato plants. Increased microbial populations that suppress deleterious microorganisms including pathogens could be one of the major factors in increased tomato yield in replant soil.

Development of a Screening System for Plant Defense-Inducing Agent using Transgenic Tobacco Plant with PR-1a Promoter and GUS Gene

  • Pathogenesis-related protein-1a (PR-1a) is strongly induced in tobacco plants by pathogen attack, exogenous salicylic acid (SA) application and by other developmental processes. In order to develop a rapid screening system for the selection of plant defense-inducing compounds originated from various sources, we have transformed tobacco Samsun NN plants with a chimeric construct consisting of GUS $(\beta-glucuronidase)$. In the $T_1$ generation, three transgenic lines having stable GUS expression were selected for further promoter analysis. Using GUS histochemical assay, we observed strong GUS induction driven by PR-1a promoter in PR1a-GUS transgenic tobacco leaves in response to the exogenous application of SA or benzol (1,2,3) thiadiazole-7-carbothioic acid S-methyl ester (BTH), a SA­derivative compound. In addition, GUS expression was maintained locally or systemically in PR1a-GUS transgenic line $\#5\;T_2$ generation) until after 3 days when they were treated with same chemicals. Our results suggested that the PR1a-GUS reporter gene system in tobacco plants may be applicable for the large-scale screening of defense-inducing substances.

  • Phytochemical investigations of the EtOAc-soluble fraction of the whole plants of Salvia plebeia using repeated column chromatography with preparative HPLC led to the isolation of a new phenylbutanone glucoside, 4-{4-0-[6-(4-hydroxybenzoyl)-0-${\beta}$-D-glucopyranosyl]-3-hydroxyphenyl}-butan-2-one (salviaplebeiaside, 1) along with two known phenolic compounds, rosmarinic acid methyl ester (2) and luteolin-7-0-${\beta}$-D-glucoside (3). The structures of these compounds were determined on the basis of spectroscopic methods including 1D-, 2D-NMR and MS spectrometry and comparison of spectroscopic data with those of values reported in the literatures.

  • To characterize non-adrenergic non-cholinergic(NANC) nerve mediated contractile responses in circular smooth muscle of bovine reticular groove, we investigated NANC relaxation and contraction induced by electric field stimulation to enteric nerves. In the presence of atropine($1{\mu}M$) and guanethidine($50{\mu}M$), electric field stimulation at frequency of 1 to 16Hz(square pulses, 0.5ms duration, 70V) evoked clear-cut relaxations through stimulations. Transient 'rebound contraction' was occured when the stimulus was switched off. All of the responses (relaxation and rebound contraction) were dose-dependently blocked by Nw-nitro-$_{\small{L}}$-arginine methyl ester(L-NAME), an inhibitor of nitric oxide synthesis, and methylene blue, and inhibitor of soluble guanylate cyclase. Tetraethyl ammonium(TEA), a potassium channel blocker, did not block the NANC relaxations.

  • 본 연구에서는 전자 주개 물질(electron donor)인 regioregular poly(3-hexylthiophene)(P3HT)와 전자 받개 물질(electron acceptor)인 phenyl-$C_{61}$-butyric acid methyl ester (PCBM)을 혼합한 복합 박막 구조(Bulk Heterojunction)를 이용하여 태양전지를 제작하고 광활성층(Active layer)의 두께를 변화시키면서 광학적 특성 및 전기적 특성에 대해 분석하였다. 광활성층의 두께가 두꺼워 질수록 광흡수율이 높기 때문에 태양전지의 효율이 증가하여 200nm정도의 두께에서 가장 좋은 특성을 보였으며, 그 이상의 두께에서는 광흡수율이 높더라도 직렬저항(Series resistance)의 증가로 개방 회로 전압이 감소하는 것을 볼 수 있었으며, 최적화된 광활성층의 두께(190nm)에서 개방 회로 전압($V_{oc}$)은 0.6V, 단락 회로 전류($J_{sc}$)는 8.29mA, Fill factor(FF)는 0.59, 전력변환효율($\eta$)은 2.94%였다.

  • Fleshing scrap is a kind of wastes produced during leather making process and used in the test of manufacturing biodiesel. The early step of manufacturing biodiesel is fat recovery from fleshing scrap. Hence, we investigated the influence of the way of fat recovery on the fatty acid composition. We used three different recovery ways, that is chemical method by protein decomposition with acid/fat recovering, physical method by protein denaturalization with heat and vacuum/fat pressing, and biodiesel method by protein decomposition/fat recovering. The biological method yielded the best results in terms of appearance transparency. It was most effective to lower acid value. Also the recovered fat by biological method would be favorable methyl-ester reaction raw material for biodiesel because it contains more than 5% of oleic acid among unsaturated fatty acid.

Inhibition of Proinflammatory Cytokine Generation in Lung Inflammation by the Leaves of Perilla frutescens and Its Constituents

  • This study was designed to find some potential natural products and/or constituents inhibiting proinflammatory cytokine generation in lung inflammation, since cytokines such as tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interleukin-6 (IL-6) are pivotal for provoking airway inflammation. In our preliminary screening procedure, the 70% ethanol extract of the leaves of Perilla frutescens (PFE) was found to clearly inhibit TNF-${\alpha}$ production in the lung at 100 mg/kg, after intranasal lipopolysaccharide treatment of mice. Based on this result, ten constituents including phenylpropanoids (allyltetramethoxybenzene, caffeic acid, dillapiole, elemicin, myristicin, nothoapiole, rosmarinic acid methyl ester, rosmarinic acid) and monoterpenes (perilla aldehyde and perilla ketone) were successfully isolated from the extract. Among them, elemicin and myristicin were found for the first time to concentration-dependently inhibit IL-$1{\beta}$-treated IL-6 production from lung alveolar epithelial cells (A549) at concentrations of $10-100{\mu}M$. These findings suggest that the phenylpropanoids including elemicin and myristicin have the potential to be new inhibitory agents against lung inflammation and they may contribute, at least in part, to the inhibitory activity of PFE on the lung inflammatory response.

  • To develop an whitening cosmetics, we isolated the melanogenesis inhibitors from the unripe fruits of Poncirus trifoliata (Rutaceae). Isolated compounds were identified as poncirin(l), naringin(2), bis(2-methylheptyl)phthalate(3), avenalumic acid methyl ester(4) by comparison of physical and spectral data with those reported in the literature. Among the isolated compounds, bis(2-methylheptyl)phthalate showed most potent inhibitory effect on the melanogenesis in cultured B-16 mouse melanoma cell lines$(IC_{50},\;36.8\;{\mu}M)$ compared with kojic acid$(IC_{50},\;150\;{\mu}M)$.
