Effect of Variable Feed Allowance with Constant Protein Input on Water Quality in Channel Catfish Production Ponds

  • This study was carried out to evaluate the effect of feeding higher protein feeds with lesser amount, but feeding the constant total protein input for all treatments, on water quality and nitrite toxicity in channel catfish ponds. There was no significant difference in survival rate among treatments $(P>0.05)$. Specific growth rate (SGR) for Treatment 1$(28\%\;protein\;and\;100\%\;of\;satiation)$ was significantly higher $(P>0.05)$ than for Treatment 3$(36\%\;protein\;and\;87.5\%\;of\;satiation)$, but not significantly higher than for Treatment 2 $(32\%\;protein\;and\;77.8\%\;of\;satiation)$ at constant digestible energy (DE), 3.08kcal/g (treatments 1, 2 and 3). At constant DE/P (treatments 4, 2 and 5), no significant difference in SGR was observed among treatments. Feed conversion ratio (FCR) slightly improved or improved as dietary protein level increased from $28\%$ to $32\%$ and feed allowance decreased by $12.5\%$, but did not improve as dietary protein level increased from $32\%$ to $36\%$ and feed allowance decreased by $22.2\%$, at constant DE and constant DE/P. There was no significant difference in water quality variables, such as total ammonia nitrogen (TAN), nitrite, chlorophyll a, soluble phosphorous concentrations among treatments, but significant difference in water quality variables over time as amount of feed fed increased $(P<0.0001)$. There was a trend toward increase in TAN and nitrite over time. A strong linear regression was observed between mean total ammonia nitrogen and nitrite for all treatments Y (Nitrite) =$0.04\times (TAN)+0.01$, $R_2=0.89$. Methemoglobin percent in the blood of catifish was not significantly different among treatments. And its mean value was $7.5\%$, which was relatively low, so that it was not serious problem in catfish production pond under these experiment conditions. There was the stronger linear regression between the percentage of Methemoglobin and the molar ratio of nitrite to chloride rather than nitrite alone: $Y\;(Methemoglobin\;\%)\;=\;58.45\;\times\;(NO^{2-}/Cl^-)\;+\;0.41,\;R^2=0.60$. These results indicate that deterioration of water quality has no strong impact on poor weight gain for $36\%$ dietary protein in this study.

Changes in Hematological Responses and Antioxidative Enzyme Activities of Japanese Eel Anguilla japonica Exposed to Elevated Ambient Nitrite (아질산에 노출된 뱀장어(Anguilla japonica)의 혈액학적 반응과 항산화효소의 활성 변화)

  • The study was performed to investigate changes in hematological responses and antioxidative enzyme activities (superoxide dismutase, SOD; catalase, CAT) of Japanese eel Anguilla japonica following exposure to 0 (control), 2.33, 4.60, 6,64 and 8.78 mM nitrite-N in fresh water for 48 h. Hematological parameters such as plasma nitrite, electrolytes, cortisol, glucose, glutamate oxaloacetate transaminase (GOT), glutamate pyruvate transaminase (GPT), hemoglobin (Hb), methemoglobin (metHb) and NADH-methemoglobin reductase (NMR) were measured. Plasma nitrite, cortisol, metHb and NMR increased directly with increasing ambient nitrite concentration, while Hb content showed a progressive decline. Levels of plasma potassium, GOT and GPT of the eel exposed to 6.64 mM ambient nitrite were significantly higher than the control fish. The activity of SOD and CAT in plasma, gill and liver of the eel following exposure to nitrite were augmented by increasing ambient nitrite. Levels of plasma nitrite, metHb, NMR, cortisol, glucose and antioxidative enzyme activities of the eel exposed to 2.33 mM ambient nitrite were significantly higher than the control fish. This study suggested that the eel acutely exposed to elevated ambient nitrite causes nitrite-induced stress responses, changes in antioxidative enzyme activities and hematological parameters.

Regulation of circulating Mg2+ concentration in rats by ATP depletion (흰쥐에서 ATP 결핍에 의한 혈중 Mg2+ 농도조절)

  • Since intracellular free $Mg^{2+}$ ($[Mg^{2+}]_i$) appears to be tightly regulated following cellular energy depletion, we hypothesized that the increase in $[Mg^{2+}]_i$ would result in $Mg^{2+}$ extrusion into circulation. Extracellualr $Mg^{2+}$ contents ($[Mg^{2+}]_o$) were measured in rat erythrocytes, the perfused heart and liver, and plasma in the anesthetized rat. Animals were injected intraperitoneally with sodium nitrite ($NaNO_2$) and plasma $Mg^{2+}$ was measured after the injection and then 10 and 20 minutes later. An increase in circulating (plasma) $Mg^{2+}$ ($[Mg^{2+}]_c$) and methemoglobin was observed in animals injected with $NaNO_2$ (30 mg/Kg). The time course of the effects demonstrated that $[Mg^{2+}]_c$ and methemoglobin continued to increase 10 minutes after the $NaNO_2$ injection. Under these conditions, there was a sustained increase in $[Mg^{2+}]_c$, but not in methemoglobin, which was inhibited by pretreatment with potassium cyanide (KCN, 4 mg/Kg), indicating that an increase in $[Mg^{2+}]_c$ was accompanied by ATP depletion. Injection of rotenone (0.9 mg/Kg) or 2,4-dinitrophenol (15 mg/Kg) also induced an increase in $[Mg^{2+}]_c$. Reduced respiration rate from 100/min to 10/min during 30 minutes also caused a time-dependent rise in $[Mg^{2+}]_c$. These increase in $[Mg^{2+}]_c$ were inhibited by pretreatment with KCN. In addition, ATP depletion by $NaNO_2$ or KCN sustainedly increased the $[Mg^{2+}]_o$ in rat erythrocytes. $Mg^{2+}$ efflux was stimulated by KCN in the perfused heart and liver, but not by $NaNO_2$. These results suggest that the activation of $Mg^{2+}$ effluxes into the circulation is directly dependent on the ATP depletion-induced increase in $[Mg^{2+}]_i$ and heart, liver and erythrocytes have a major pool of $Mg^{2+}$ that can be mobilized upon cellular energy state.

Pharmacokinetics of a New PEG-hemoglobin SBI for Single Intravenous Administration to Rats (흰쥐에 있어서 PEG-헤모글로빈 SBI의 단회정맥투여 약물동태)

  • An intravenous pharmacokinetics for a new red cell substitute, PEG-hemoglobin SB1, was studied in SD rats. Total-hemoglobin and its metabolite methemoglobin in the plasma were determined using a spectrophotometer. The limit of quantitation was 0.01g/dL and the C.V for interday assay reproducibility was less than 6%. Upon intravenous administration of anticipated clinical dose, 10 ml(0.7 gHb)/kg, plasma concentration curve of total-hemoglobin was well described by one-compartment model. The $t_{\frac{1}{2}},{\;}CL_{t}$, Vd and $AUC^{0-48hr}$ were $8.23{\pm}0.96{\;}hr,{\;}0.06{\;}{\pm}{\;}0.01 {\;}dL/hr/kg,{\;}0.66{\pm}0.05{\;}dL/lg{\;}and{\;}13.6{\;}{\pm}{\;}1.01g{\cdot}hr/dL$, respectively, in male rats(n=5, $mean{\;}{\pm}{\;}SD$). Those parameters in female rats were $9.21{\;}\pm{\;}2.31{\;}hr,{\;}0.06{\;}{\pm}{\;}0.01{\;}dL/hr/kg,{\;}0.79{\pm}{\;}0.08{\;}dL/kg{\;}and{\;}13.0{\;}{\pm}{\;}2.36{\;}g{\cdot}hr/dL$, respectively. Similar kinetic profiles between males and females were also obtained from other parameters. Small amount of methemoglobin, an oxidative metabolite of SB1, was detected in the plasma of both sexes, where the $AUC^{0-48{\;}hr,m}$ and $t_{{\frac{1}{2}},m}$ were approximately $1.5{\;}g{\cdot}hr/dL$ and 20 hr, respectively. The present work provides a critical kinetic data for the effective clinical applications of PEG-hemoglobin SB1.

The Quantitative Determination of Reversible and Irreversible Oxidative Damages Induced by Smoking Cessation and Supplementation of Antioxidative Vitamins in Korean Male Smokers (한국 남자 흡연자의 금연과 항산화제 보충에 따른 체내 가역적.비가역적인 산화 손상도 변화의 정량적 측정 연구)

  • The effect of oral vitamin e (800IU/day) and C (500mg/day) supplementation for 10 days and/or smoking cessation for 5 days on oxidative damage to the red blood cells (RBC) of male smokers (22.2$\pm$0.2 years old) was studied. RBC were tested for their ability to protect against smoking-induced oxidative damage by measuring heme proteins (carboxyhemoglobin, hemoglobin, methemoglobin, oxyhemoglobin), hemolysis and thiobarbiturinc acid reactive substances (TBARS). Plasma levels of vitamin c, A, E, $\beta$-catotene, total cholesterol, glutamic pyruvic transaminase(GPT) and glutamic oxaloacetic transaminase(GOT) were also analyzed. In experiment one, a comparison was made of heme proteins and lipid damage to RBC, plasma antioxidant status (indexed by plasma levels of vitamin C, E, A and $\beta$-carotene) between smokers(n=56) and non-smokers (n=16). No differences were found in plasma antioxidant status, heme protein damage and TBARS concentration of RBC. In experiment two, 46 fasting male smokers from experiment one were divided into 4 groups. The groups were smoking with placebo group(SP, n=14), smoking cessation with vitamins supplementatin group (SV, n=13), smoking cessation with placebo group (NSP, n=9) and smoking cessation with vitamins supplementation group (NSV, n=10). After supplementing antioxidant vitamins, significant increases were seen in plasma vitamins supplementation group (NSV, n=10). After supplementing antioxidant vitamins, significant increases were seen plasma vitamin C (p<0.05) and vitamin E levels (p<0.05). The plasma vitamin E level was highest in the NSV group. Vitmain E and C supplementation provided some protection against heme proteins and lipid damage by lowering methemoglobin, hemolysis and TBARS concentration of RBC. Smoking cessation significantly decreased TBARS of RBC and plasma total cholesterol concentration. Supplementing vitamin E and C with smoking cessation considerably lowered plasma total cholesterol. These results point to a special association among smoking, oxidative damage and plasma antioxidant vitamin status. They indicate that increases in plasma antioxidant status can be detected after the supplementation of vitamin C and E and that smoking cessation had an additional effect on plasma vitamin E level. The present data suggest that improved antioxidant status induced by antioxidant supplementation or smoking cessation may help prevent oxidative damage in smokers.

Toxicological Studies on the Humic acid (Humic acid의 독성학적 연구)

  • The author obtained three kinds of humic acid from the soil of suburbs in the Wonju city, sediments of Wonju stream and Maeji reservoir in 29 April 1986. The yield of each humic acid was 7.23 g, 3.35 g and 4.61 g per sample 200 g. In the physicochemical characters, the COD varied from 65.9 ppm to 54.9 ppm and BOD showed 6.7~15.9 ppm, but the standard humic acid was COD 58.9 ppm and BOD 6.5 ppm in 0.01 % solution. Doses of the humic acids for 20% methemoglobin formation are as follows; 0.303 mg/m$\ell$ in St. A humic acid, 0.602 mg/m$\ell$ in St. B humic acid, 0.84 mg/m$\ell$ in St. C humic acid and 0.105 mg/m$\ell$ in standard humic acid.

A Spetrophotometric Cyanmethemoglobin Method for the Estimation of Serum Hemoglobin Levels (혈청(血情)해모구로빈 측정(測定)에 관(關)한 연구(硏究))

  • Hemoglobinemia is found in hemolytic anemia, paroxysmal nocturnal hemolobinuria and paroxysmal hemoglobinuria. Recently increasing use of extracorporeal circulation in cardiac surgery has stimulated the development of more precise methods for rapidly evaluating the formation and clearance of extracoporeal hemoglobin. A spectrophotometric method of analysis seemed to offer the best solution to the requirements. The method presented is on the basis of hemoglobin derivatives converted to cyanmethemoglobin, and the absorbance is measured in a spectrophotometer at 540 and 680 nm. The blank value ismeasured at 680 nm while the absorbance at 540 nm measures the hemoglobin derivatives. This method estimates all the hemoglobin derivatives such as oxyhemoglobin, carboxyhemoglobin, methemoglobin, sulfhemoglobin and hemialbumin. The method was tested in recovery experiments, which is given table 1. A good degree of correlation was obtained in a comparision with the method described by Crosby and Furth for non-hemolyzed serum. The spectrophotometric technique described offers many advantages in speed and simplicity over the chemical procedure.

  • Physical compositions of solid waste in Wonju, shopping area were investigated with combustibles 78.13% (papers 35.89%, foods 14.41% etc.) incombustibles 21.87% (glass and ceramics 11.02%, metals 6.0% etc.) in 1991, solid waste in apartment area were investigated with combustibles 84.27% (foods 34.29%, papers 22.58% etc.), incombustibles 15.73% (glass and ceramics 8.77%, metals 4.85% etc.) and residence area were characterised with combustibles 70.37% (foods 33.55% , papers 10.53% etc.) and incombustibles 29.63% (ash of briquet 17.29%, glass and ceramics 7.49% etc.). Water qualities of a leachate from municipal landfill of Wonju city were analysed pH 8.0~8.4, total suspended solid 102~140 mg/1, CN ̄ 0.003~ 0.008 mg/1, NO$_2$-N 0.108~0.294 mg/1 and phenols 0.46~1.12 mg/1. Volume of the leachate for 20% methemoglobin formation were 0.2~0.4 mg/ml in Octever, 0.3~0.4 ml/ml in December sampling, 0.2~0.3 ml/ml in St.1 and 0.4 ml/ml in St.3.

  • Nitrate accumulated in vegetables can, via $NO_2$ during storage and after their ingestion, cause the formation of methemoglobin and many of which are carcinogenic in human body. Especially such vegetables with the highest consumption in Korea as chinese cabbage and raddish frequently showed that high contents of nitrate and nitrite could accumulate. Therefore, pH and the content of nitrate and nitrite of Korean pickles made of upper vegetables, viz, Baiechu Kimchi, Yulmookimchi and Kakdooki were estimated during storage at the conditions of $5^{\circ}C$ and $25^{\circ}C Storage at $25^{\circ}C$ generally led to the more rapidly than that at $5^{\circ}C$. But changes of nitrite level showed very low variation in all tested Kimchi.

  • In order to evaluate the anticancer activity of 1,4-naphthoquinone derivatives, several 1,4-naphthoquinone derivatives were newly synthesized and subjected to mouse leukemia p-388 cell line by MTT cytotoxicity assay. Among the several 1,4-naphtho-quinone derivatives, YC-001 has showed the most potent anticancer activity. To determine the safety of YC-001, hematotoxicity was tested. YC-001 induced hemolysis increased with both concentration and time dependent manner. The mechanism of hemolysis considered to be the generation of oxygen free radicals and lipid peroxydation of erythrocyte membrane which composed of phospholipids. Also methemoglobin, oxidized form of hemoglobin, was formed by YC-001.
