In vitro Antiviral Activities of Korean Marine Algae Extracts against Fish Pathogenic Infectious Hematopoietic Necrosis Virus and Infectious Pancreatic Necrosis Virus

  • To investigate the antiviral activity of marine algae against fish pathogenic viruses, which are often the causes of viral disease in aquaculture, the 80% methanolic extracts of 21 species collected from the coast of Korea were screened for their in vitro antiviral activities on infectious hematopoietic necrosis virus (IHNV) and infectious pancreatic necrosis virus (IPNV), using a flounder spleen (FSP) cell-line. Among them, Monostroma nitidum (10 ${\mu}g/mL$) exhibited the strongest inactivation on IHNV, showing a 2 log reduced virus titre as compared to the control in the determination of direct virucidal activity. In addition, Polysiphonia morrowii (100 ${\mu}g/mL$) remarkably reduced the virus titres of treated cells by 2-2.5 log, for both IHNV and IPNV, in the determination of cellular protective activity, implying the existence of substances that may modulate innate host defense mechanisms against viral infections. These results reveal that some marine algae could be promising candidates as sources of antiviral agents or as health-promoting feeds for aquaculture.

  • Houttuynia cordata root on non-lipid oxidative damage. The antioxidative efects of methanolic (MeOH) extract of Houttuynia cordata rooton non-lipid, including liposome oxidation, oxidation of deoxyribose, protein oxidation, chelating, scavenging,and 2'-deoxyguanosine (2'dG) oxidation were investigated. Houttuynia cordata root exhibited highantioxidative effect in a liposome model system. The inhibitory effect of MeOH extract on deoxyribosedamage exhibited antioxidative effect and it afforded considerable protection against damage to deoxyribose.In addition, MeOH extract at over 300extracts exhibited metal binding ability for hydrogen peroxide. Furthermore, the oxidation of 2'dG to 8-hydroxy-2-deoxyguanosine was inhibited by MeOH extracts, and scavenging activity for hydroxyl radicalexhibited a remarkable effect. The present results on biological model systems showed that MeOH extractswas effective in the protection of non-lipids against various oxidative model systems.

  • 본 연구에서는 주요 어류 병원성 그람음성세균인 E. tarda 및 V. anguillarum에 대하여 14 종의 생약 추출물의 항균활성을 검색하였다. 그 결과, 감초, 지유 및 칠피 추출물이 항균활성을 나타냄을 밝혔다. 그 중, 가장 우수한 항균활성을 나타낸 칠피 추출물에 대하여 활성지향적인 분리기법을 적용하여 활성분획물인 EtOAc분획물로부터 항균활성물질 1종을 분리하고 페놀성 화합물인 methyl gallate로 동정하였다. methyl gallate는 실험에 사용된 E. tarda 및 V. anguillarum의 균주들의 증식을 효과적으로 저해하였으며 이에 대한 MIC는 모두 1 mg/ml로 측정되었다.

Cytotoxic and antioxidant properties of four plants belonging to the genus Solanum

  • The aim of the study was to evaluate in vitro antioxidant and cytotoxic activities of the methanolic extracts of leaves of Solanum sisymbrifolium, Solanum anguivi multiflora, Solanum barbisetum and Solanum jasminoides. In the in vitro antioxidant screening using ABTS [2,2'-azino-bis (3-ethylbenzo-thiazoline-6-sulphonic acid) diammonium salt] method, the methanol extracts of Solanum jasminoides, Solanum barbisetum and Solanum anguivi multiflora exhibited potent antioxidant activity with $IC_{50}$ values 31.25 $\pm$ 0.35, 40.33 $\pm$ 0.57 and 54.33 $\pm$ 0.57 ${\mu}g$/ml, respectively. Solanum barbisetum also showed potent activity in DPPH [1,1-diphenyl-2-picryl hydrazyl] method with an $IC_{50}$ value of 55.33 $\pm$ 1.66 ${\mu}g/ml. In the cytotoxicity studies, the methanol extract of Solanum barbisetum exhibited moderate activity against Vero, HEp-2, HeLa and A-549 cell lines with $IC_{50}$ values in the range of 83.30 - 127.30 ${\mu}g$/ml. Solanum anguivi multiflora extract also showed moderate activity against Hep-2 cell line with $IC_{50}$ value of 80.13 ${\mu}g$/ml. Solanum barbisetum possessing both the activities requires further investigation.

  • A total of five compounds namely, 2-methylanthracene-9, 10-dione (1), 1-hydroxy-6-methylanthracene-9,10-dione (2), $\beta$-sitosterol (3), stigmasterol (4) and sitosterone (5) were isolated from the stem extracts of Polygonum lanatum for the first time. The structures of the isolated compounds (1-5) were established by extensive spectroscopic studies, including 2D NMR such as $^{1}H-^{1}H$ COSY, HSQC and HMBC studies. The crude extracts and purified compound (1) were screened for antimicrobial activity against a wide range of Gram-positive and Gram-negative bacteria and fungi by the disc diffusion method. The n-hexane and dichloromethane soluble partitionates of the methanolic extract revealed mild to moderate inhibition of growth of the test organisms. The cytotoxic potential of the extractives and compound 1 was also determined by using brine shrimp lethality bioassay, where the extractives demonstrated significant cytotoxic activities.

해조류 추출물의 In Vitro 항치매 활성 (In Vitro Screening for Anti-Dementia Activities of Seaweed Extracts)

  • 본 연구에서는 해조류 추출물 20종의 치매 예방 및 개선 소재로서의 가치를 검토하기 위해 총폴리페놀 함량, ABTS radical 소거능, acetylcholinesterase(AChE) 저해 활성, ${\beta}$-secretase 효소 억제 활성 및 신경세포 보호 효과를 비교하였다. 해조류 추출물의 총폴리페놀 함량과 ABTS radical 소거능을 측정한 결과 감태 추출물의 총폴리페놀 함량이 가장 높았으며, 검둥감태와 감태 추출물이 높은 ABTS radical 소거능을 나타내었다. AChE 저해 활성을 검토한 결과 감태, 검둥감태, 바위수염이 30% 이상의 높은 저해 활성을 보였고 그중 감태가 가장 높은 억제율을 나타내었다. 또한, ${\beta}$-secretase 효소 억제 활성은 검둥감태, 감태, 왜모자반 추출물에서 관찰되었다. $H_2O_2$에 의해 유도된 신경세포 독성에 대한 보호 효과는 감태, 검둥감태, 왜모자반 추출물 $100{\mu}g/mL$ 농도에서 관찰되었다. 이상의 결과로 미루어 보았을 때 감태 추출물이 치매 예방 및 개선제로서의 활용 가능성이 가장 뛰어난 것으로 생각되며, 추가로 생리활성 물질 구명 및 작용기전 입증을 위한 후속 연구가 진행되어야 할 것으로 생각한다.

원적외선 조사와 열처리가 탈지대두박 추출물의 항산화능에 미치는 영향 (Effect of Far-Infrared Irradiation and Heat Treatment on the Antioxidant Activity of Extracts from Defatted Soybean Meal)

  • 원적외선 조사 및 일반 열처리가 탈지대두박의 항산화능에 미치는 영향을 조사하기 위하여 $150^{\circ}C$에서 각각 5, 10, 15, 20, 40 그리고 60분간 처리한 후, 메탄올 추출물을 제조하여 총 페놀함량, DPPH 라디칼 소거능 그리고 환원력을 측정하였다. 이들 측정 결과, 무처리구 31.62 mg/ml, 11.6%, 0.068에 비해 원적외선을 $150^{\circ}C$에서 15분간 조사한 처리구는 각각 57.51 mg/ml, 53.1%, 0.147로 증가하였다. 한편, $150^{\circ}C$에서 15분간 일반 열처리한 시료의 경우는 각각 58.04 mg/ml, 65.2%, 0.160로 증가하였다. 그러나 원적외선 조사 및 일반 열처리를 20분간 이상하면 점차 감소하는 경향을 보였다. 이상의 결과는 적당한 조건에서의 원적외선과 열처리 조사가 탈지대두박에 존재하는 페놀 화합물의 추출을 촉진하며 항산화능을 향상시킴을 의미한다.

땅콩껍질 추출물의 항산화능에 대한 원적외선과 열처리 효과 (Effect of Far-Infrared Irradiation and Heat Treatment on the Antioxidant Activity of Extracts from Peanut (Arachis hypogaea) Shell)

  • 원적외선 조사 및 일반 열처리가 땅콩껍질의 항산화능에 미치는 영향을 조사하기 위하여 $150^{\circ}C$에서 각각 5, 10, 15, 20, 40 그리고 60분간 처리한 후, 메탄을 추출물을 제조하여 총 페놀함량, DPPH 라디칼 소거능 그리고 환원력을 측정하였다. 이 들 측정 결과, 무처리구 40.17mg/mL, $67.7\%$, 0.569에 비해 원적외선을 $150^{\circ}C$에서 5분간 조사한 처리구는 각각 42.30mg/mL, $76.3\%$, 0.639로 증가하였다. 한편, $150^{\circ}C$에서 5분간 일반 열처리 한 시료의 경우는 각각 43.52mg/mL, $79.3\%$, 0.623로 증가하였다. 그러나, 원적외선 조사 및 일반 열처리를 60분간 행하였을 경우에는 모두 대조구에 비해 감소하였다. 이상의 결과로 땅콩 껍질에 대한 적절한 원적외선 조사 및 열처리는 메탄올 추출물의 항산화 활성을 증가시킴을 확인할 수 있었다.

Screening and isolation of Antibiotic Resistance Inhibitors from Herb Materials IV- Resistance Inhibitors from Anetheum graveolens and Acorns gramineus

  • The hexane fractions from methanolic extracts of Anetheum graveolens L. (Umbelliferae) and Acorus gramineus Soland. (Araceae) revealed potent inhibitory activities against the resistance of multi-drug resistant Staphylococcus aureus SA2 when combined with ampicillin (Am) or chloramphenicol (Cm). As active principles, carvone and the liquid mixture containing carvone from Anetheum graveolens L. and a liquid mixture mainly consisting of benzoic acid phenyl-methyl ester (benzyl benzoate) from Acorus gramineus Soland. were identified. They showed resistance inhibition at the level of 20-50${\mu}g$/ml when combined with 100 or ${\mu}g$/ml of Am or Cm, respectively.

Chemical Constituents of the Fruiting Bodies of Clitocybe nebularis and Their Antifungal Activity

  • During a continuing search for antimicrobial substances from Korean native wild mushroom extracts, we found that the methanolic extract of the fruiting body of Clitocybe nebularis exhibited mild antifungal activity against pathogenic fungi. Therefore we evaluated the antifungal substances and other chemical components of the fruiting body of Clitocybe nebularis, which led to the isolation of nebularine, phenylacetic acid, purine, uridine, adenine, uracil, benzoic acid, and mannitol. Nebularine showed mild antifungal activity against Magnaphorthe grisea and Trichophyton mentagrophytes, and phenylacetic acid potently inhibited the growth of Pythium ultium and displayed moderate antifungal activity against Magnaphorthe grisea, Botrytis cinerea, and Trichophyton mentagrophytes. The other isolated compounds showed no antimicrobial activity.
