BACKGROUND: Animal manures are one of the biggest sources of greenhouse gases and improper manage-ment of animal wastes contributes to the increasing greenhouse gases in the atmosphere. Con-verting greenhouse gases generated from animal manures to energy is one way of contributing to the net-zero carbon emissions. METHODS AND RESULTS: The potential for methane production from cow manure (CM) was studied by measuring the methane yield using the biochemical methane potential (BMP) test. In particular, the effect of co-digestion using rice straw (RM) on the methane production was studied. The methane yields from the co-digestion of CM and RS were statistically similar to that from the mono-digestion of CM or RS. But there was a synergy effect at the CM:RS ratio of 1:2 and 1:1. This can be attributed to the increased C/N ratio. The changed microbial community structure with the addition of substrates (CM, RS) probably led to the increase in the methane produc-tion. CONCLUSION(S): The methane production potential of the particular CM used in this study was not improved by the addition of RS as a co-substrate. The addition of substrates to the anaerobic sludge promoted the increase in the microbial species having synergetic relationship with methano-gens, and this can partially explain the increase in the methane production with the addition of substrates. Overall, there are needs for further studies to improve the methane yield from CM.
Stored animal manure is considered as a significant agricultural source of methane $(CH_4)$ and nitrous oxide $(N_2O)$ which have 23 and 297 times higher global warming effect when compared to carbon dioxide $(CO_2)$. Uncertainties caused by lack of understanding physical and biochemical environment in stored animal manure and by errors of emission measurement methods, even though many researches measuring $CH_4\;and\;N_2O$ emissions from stored manure have been conducted for a few decades. In this paper, general information of $CH_4\;and\;N_2O$ generation and emissions from stored animal manure and the measurement methods for quantifying $CH_4\;and\;N_2O$ emissions are discussed.

Methane (CH4) and nitrous oxide (N2O) are significant contributors to greenhouse gas (GHG) emissions from rice fields. Mid-summer drainage is a commonly practiced water management technique that reduces CH4 emissions from rice fields. Slow-release fertilizers gradually release nutrients over an extended period and have been shown to reduce N2O emissions. However, the combined effect of slow-release fertilizer and water management on GHG emissions remains unclear. This study compared GHG emissions from a rice paddy subjected to mid-summer drainage for 10 days (control) with that of a rice paddy subjected to prolonged mid-summer drainage for 20 days combined with slow-release fertilizer (W+S). Gas sampling was conducted weekly using a closed chamber method. During the rice cultivation period, cumulative CH4 and N2O emissions were reduced by 12.3% and 16.2%, respectively, in the W+S treatment compared to the control. Moreover, the W+S treatment exhibited a 1.9% increase in grain yield compared to the control. Under experimental conditions, slow-release fertilizers, in combination with prolonged mid-summer drainage, proved to be the optimal approach for achieving high crop yield while reducing GHG emissions. This represents an effective strategy to mitigate GHG emissions from rice paddy fields.
Methane is the second most emitted greenhouse gas after carbon dioxide. Despite lower emissions than those of carbon dioxide, methane receives significant attention owing to its more than 20-fold higher global warming potential. Consequently, the importance of research on methanotrophic bacteria, microorganisms capable of converting methane gas into high-value materials, is increasingly emphasized. In the case of methanotrophic bacteria, knowledge on episomal plasmids that can be used for genetic engineering remains lacking, which poses significant challenges to the engineering process. The replication origin sequences of natural plasmids within methanotrophic bacteria have been predicted through in silico methods. The basic characteristics of the replication origin, such as a high A/T ratio, repetitive sequences, and proximity to proteins related to replication, have been used as criteria for identifying the replication origin. As a result, a region with a sequence of 18 base pairs repeated eight times could be identified. The putative replication origin sequence thus identified generally takes the form of iterons, but it also possesses unique features such as the length of the gap between iterons and the repetition of identical iteron sequences. This information can be valuable for future design of episomal plasmids applicable to methanotrophs.

Molecular hydrogen (H2) and formate (HCOO-) are metabolic end products of many primary fermenters in the rumen ecosystem. Both play a vital role in fermentation where they are electron sinks for individual microbes in an anaerobic environment that lacks external electron acceptors. If H2 and/or formate accumulate within the rumen, the ability of primary fermenters to regenerate electron carriers may be inhibited and microbial metabolism and growth disrupted. Consequently, H2- and/or formate-consuming microbes such as methanogens and possibly homoacetogens play a key role in maintaining the metabolic efficiency of primary fermenters. There is increasing interest in identifying approaches to manipulate the rumen ecosystem for the benefit of the host and the environment. As H2 and formate are important mediators of interspecies interactions, an understanding of their production and utilization could be a significant starting point for the development of successful interventions aimed at redirecting electron flow and reducing methane emissions. We conclude by discussing in brief ruminant methane mitigation approaches as a model to help understand the fate of H2 and formate in the rumen ecosystem.

Optimization of the dietary formulation is the most effective way to reduce methane. Nineteen feed ingredients (brans, vegetable proteins, and grains) were evaluated for their potential to generate methane and change methanogen diversity using an in vitro ruminal fermentation technique. Feed formulations categorized into high, medium and low production based on methane production of each ingredient were then subjected to in vitro fermentation to determine the real methane production and their effects on digestibility. Methanogen diversity among low, medium and high-methane producing groups was analyzed by PCR-DGGE. The highest methane production was observed in Korean wheat bran, soybean and perilla meals, and wheat and maize of brans, vegetable protein and cereal groups, respectively. On the other hand, corn bran, cotton seed meal and barley led to the lowest production in the same groups. Nine bacteria and 18 methanogen 16s rDNA PCR-DGGE dominant bands were identified with 83% to 99% and 92% to 100% similarity, respectively. Overall, the results of this study showed that methane emissions from ruminants can be mitigated through proper selection of feed ingredients to be used in the formulation of diets.

Hydrogen-blending effects in flame structure and NO emission behavior are numerically studied with detailed chemistry in methane-air counterflow diffusion flames. The composition of fuel is systematically changed from pure methane to the blending fuel of methane-hydrogen through $H_2$ molar addition up to 30%. Flame structure, which can be described representatively as a fuel consumption layer and a $H_2$-CO consumption layer, is shown to be changed considerably in hydrogen-blending methane flames, compared to pure methane flames. The differences are displayed through maximum flame temperature, the overlap of fuel and oxygen, and the behaviors of the production rates of major species. Hydrogen-blending into hydrocarbon fuel can be a promising technology to reduce both the CO and $CO_2$ emissions supposing that NOx emission should be reduced through some technologies in industrial burners. These drastic changes of flame structure affect NO emission behavior considerably. The changes of thermal NO and prompt NO are also provided according to hydrogen-blending. Importantly contributing reaction steps to prompt NO are addressed in pure methane and hydrogen-blending methane flames.

The chamber method is widely used for measuring methane emission from paddy rice fields. The closed static chamber has advantages of easy installation and removal in the field and low manufacturing cost. However, the manual chamber method requires a lot of labor and has a limited sampling time and frequency. To overcome the disadvantages of the manual chamber, the auto-chamber system is used for measuring methane emission. We compared the differences in methane flux between the auto-chamber and manual chamber. To investigate methane emissions by the two methods, a chamber was installed for each of the following treatments : control without rice straw (NA), spring plowing after autumn rice straw application (SPRA) and autumn plowing after autumn rice straw application (APRA). The total methane emission was lowest in the control and highest in APRA with both methods. There was no significant difference in total methane emission between the methods, but dynamic fluctuation in methane with temperature change was accurately measured in the auto-chamber. Measuring methane emission with an auto-chamber system is expected to reduce uncertainty and increase accuracy, accompanied by labor reduction.

To analyze the characteristics of ozone formation, measurements of the concentrations of individual exhaust hydrocarbon species have been made under various engine operating parameters in a 2-liter 4-cylinder engine for natural gas and LPG. Tests were performed at constant engine speed, 1800 rpm for two compression ratios of 8.6 and 10.6, with various operating parameters, such as excess air ratio of 1.0~1.6, bmep of 250~800 na and spark timing of BTDC 10~$55^{\circ}$. It was found that the natural gas gave the less ozone formation than LPG in various operating conditions. This was accomplished by reducing the emissions of propylene($C_3H_6$), which has relatively high maximum incremental reactivity factor, and propane($C_3H_8$) that originally has large portion of LPG. In addition, the natural gas show lower values in the specific reactivity and brake specific reactivity. Higher compression ratio of the test engine showed higher non methane HC emissions. However, specific reactivity value decreased since fuel species of HC emissions increase. brake specific reactivity showed almost same values under high bmep, over 500kPa for both fuels. This means that the increase of non methane HC emissions and the decrease of specific reactivity with higher bmep affect each other simultaneously. With advanced spark timing, brake specific reactivity values of LPG were increased while those of natural gas showed almost constant values.

Water management impacts both methane ($CH_4$) and nitrous oxide ($N_2O$) emissions from rice paddy fields. Although irrigation is one of the most important methods for reducing $CH_4$ emission in rice production systems it can also $N_2O$ emissions and reduce crop yields. A feasibility study on the system of rice intensification (SRI) methods with respect to irrigation requirements, greenhouse gas (GHG) emissions was conducted for either 2 or 3 years depending on the treatment in Korea. The SRI methods (i.e. SRI and midsummer drainage (MD) with conventional practice (CT)) reduced the irrigation requirement by 49.0 and 22.0 %, respectively. Global warming contribution of GHG to different depending on the type of GHG. Therefore, the emission of $CH_4$ and $N_2O$ shall be converted to Global Warming Potential (GWP). The GHG emission from the conventional practice with midsummer drainage (MD) and the SRI plots, in GWP were reduced by 49.1 and 77.1 %, respectively. Application of SRI water management method could help to improve Korea's water resources and could thus contribute to mitigation of the negative effects of global warming.
