Protective Effects of Lacticaseibacillus rhamnosus IDCC3201 on Motor Functions and Anxiety Levels in a Chronic Stress Mouse Model

  • Growing evidence indicates a crucial role of the gut microbiota in physiological functions. Gut-brain axis imbalance has also been associated with neuropsychiatric and neurodegenerative disorders. Studies have suggested that probiotics regulate the stress response and alleviate mood-related symptoms. In this study, we investigated the effects of the probiotic Lacticaseibacillus rhamnosus IDCC3201 (L3201) on the behavioral response and fecal metabolite content in an unpredictable chronic mild stress (UCMS) mouse model. Our study shows that chronic stress in mice for three weeks resulted in significant changes in behavior, including lower locomotor activity, higher levels of anxiety, and depressive-like symptoms, compared to the control group. Metabolomic analysis demonstrated that disrupted fecal metabolites associated with aminoacyl-tRNA biosynthesis and valine, leucine, and isoleucine biosynthesis by UCMS were restored with the administration of L3201. Oral administration of the L3201 ameliorated the observed changes and improved the behavioral alterations along with fecal metabolites, suggesting that probiotics play a neuroprotective role.

Effects of LED Light Conditions on Growth and Analysis of Functional Components in Buckwheat Sprout (LED 광 조건에 따른 메밀 새싹의 생육 및 기능성 물질 분석)

  • Buckwheat sprouts are a vegetable; a functional food should provide health benefit and enhance performance as high nutritionally important substances. Buckwheat noodles are the major buckwheat food in Japan, Korea and China. In addition, Buckwheat as preventive medicine has undergone a great advancement in the last decade. Comparison of the functional properties distribution and utilization in tatary buckwheat is required of understanding the metabolites. The study was conducted to identify the sorts of phenolic compounds and metabolites in tatary buckwheat seedling at 4, 7, and 10 days seedling under the different combinations of light-emitting diode (LED) such as blue, red, mix (red, blue, and white), dark, and natural lights in stem and leaves. After breaking the dormancy, buckwheat seeds were grown in culture room under lights for 14 hrs and the dark condition for 10 hrs, at $25^{\circ}C$ for 10 days. Length of buckwheat was gradually increased under all of the conditions. Using HPLC, rutin was highest at 7 days under mix and natural light in stem and leaf, respectively. Quercetin was highest at 4 and 7 days under natural light in both. Chlorogenic acid was highest at 7 days under mix and natural in stem and leaf, respectively. Taken Together, this study indicates that phenolic compounds and metabolites present in those plants could be helpful for the human health and nutritional additive.

Association of PAH-DNA adducts and Urinary PAH metabolites influenced by polymorphisms of xenobiotic metabolism enzymes in industrial wase incinerating workers (산업폐기물 소각장 근로자에서 요중 PAHs 대사산물과 혈중 aromatic-DNA adducts)

  • This study evaluated the concentrations of urinary metabolites of polycyclic aromatic hydrocarbons (PAHs) in industrial waste incineration workers. The effect of genetic polymorphisms of xenobiotic metabolism enzymes on urinary concentration of PAH metabolites was assessed. And, aromatic DNA adduct levels were also determined in total white blood cells. Fifty employees were recruited from a company handling industrial wastes located in Ansan, Korea: non-exposed group (n=21), exposed group (n=29). Sixteen ambient PAHs were determined by GC/MSD (NIOSH method) from personal breathing zone samples of nine subjects near incinerators. Urinary 1-hydroxypyrene glucuronide (1-OHPG), a major pyrene metabolite, was assayed by synchronous fluorescence spectroscopy after immunoaffinity purification using monoclonal antibody 8E11 (SFS/IAC). Multiplex PCR was used for genotyping for GSTMI/TI and PCR-RFLP for genotyping of CYP1A1 (MspI and Ile/Val). PAH-DNA adducts in peripheral blood WBC were measured by the nuclease P1-enhanced postlabeling assay. Smoking habit, demographic and occupational information were collected by self-administered questionnaire. The range of total ambient PAH levels were 0.00-7.00 mg/㎥ (mean 3.31). Urinary 1-OHPG levels were significantly higher in workers handling industrial wastes than in those with presumed lower exposure to PAHs (p=0.006, by Kruskal-Wallis test). There was a statistically significant dose-response increase in 1-OHPG levels with the number of cigarettes consumed per day (Pearson correlation coefficient=0.686, p<0.001). Urinary 1-OHPG levels in occupationally exposed smoking workers were highest compared with non-occupationally exposed smokers (p=0.053, by Kruskal-Wallis test). Smoking and GSTMI genotype were significant predictors for log-transformed 1-OHPG by multiple regression analysis (overall model R²=0.565, p<0.001), whereas smoking was the only significant predictor for log-transformed aromatic DNA adducts (overall model R²=0.249, p=0.201). Aromatic DNA adducts was also a significantly correlation between log transferred urinary 1-OHPG levels (pearson's correlation coefficient=0.307, p=0.04). However, the partial correlation coefficient adjusting for Age, Sex, and cigarette consumption was not significant (r=0.154, p=0.169). The significant association exists only in individuals with the GSTMI null genotype (pearsons correlation coefficient=0.516, p=0.010; partial correlation coefficient adjusting for age, sex, and cigarette consumption, r=0.363, p=0.038). Our results suggest that the significant increase in urinary 1-OHPG in the exposed workers is due to higher prevalence of smokers among them, and that the association between urinary PAH metabolites and aromatic DNA adducts in workers of industrial waste handling may be modulated by GSTMI genotype. There results remain to be confirmed in future larger studies.

Statistical Optimization of Culture Conditions for the Production of Aphicidal Metabolites of Beauveria bassiana Bb08 (Beauveria bassiana Bb08의 살충성 물질 생산을 위한 배양조건의 통계적 최적화)

  • For the maximal production of aphicidal metabolites produced by the Beauveria bassiana Bb08, statistical methods such as the Box-Behnken experimental design and response surface methodology were used. The fungal culture filtrate was sprayed towards 3-star aphids and the mortality was examined. After the statistical analysis of the aphid mortality, the optimal culture conditions were found to be a culture temperature of $26.2^{\circ}C$, medium pH 5.9, flask shaking speed of 209.0 rpm, and culture time of 5.9 days. The expected mortality on days 4, 5, and 6 after spraying the filtrate on to the aphids were 76.8%, 84.9%, and 89.4%, respectively. All 4 factors of the culture conditions significantly affected the production of the aphicidal metabolites, and the order of significance was temperature, pH, culture time and shaking speed.

  • Streptomyces produces many kinds of secondary-metabolites including antibiotics. Screening of a new compound and elucidation of a biosynthetic pathway for the secondary metabolites are very important fields of biology, however, there is a main problem that most of the identified compounds are already researched compounds. To solve these problems, a microarray system that is based on the data related to the biosynthetic genes for secondary-metabolites was designed. For the main contents of DNA microarray, the important genes for the bio-synthesis of aminoglycosides, polyenes group, enediyne group, alpha-glucosidase inhibitors, glycopeptide group, and orthosomycin group were chosen. A DNA microarray with 69 genes that were involved in the bio-synthesis for the antibiotics mentioned above was prepared. The usability of the DNA microarray was confirmed with the chromosomal DNA and total RNA extracted from S. coelicolor whose genomic sequence had already been reported.

Inter-laboratory Comparison for Analyses of Heavy Metals and Organic Solvent Metabolites in Biological Samples (생체시료 중 중금속 및 유기용제 대사물 분석의 실험실간 정도관리 프로그램)

  • The result of five year's experience on Inter-laboratory Comparison for Analyses of Heavy Metals and Organic Solvent Metabolites in Biological Samples was described. Since 1995, around a hundred laboratories in the Occupational Health have participated this program twice per year by the Industrial Safety and Health Law. Four metals in blood and five organic solvent metabolites in urine were examined. Reference samples were made by spiking standard materials to human blood or urine pools treated previously to give homogeneity and stability for a specific time periods. Some reference samples for organic solvent metabolites were made from workers' urine who were exposed to the organic solvents. Some items such as Lead in blood and Hippuric acid in urine showed good accordance between participants while the other items such as Mercury and N-methylformamide in urine showed poor proficient rate. The results were published in the internet or newspaper to help the consumer of the laboratory's service to get the information on them and to make competition between them. The inter-laboratory's comparison program have done greate role to improve the ability of analysis and reliability of analytical data produced from each laboratory.

The study of analytical method for sulfonamide antibiotics and their metabolites in environmental samples (환경 시료 중 설폰아미드계 항생제 및 대사체 분석방법 연구)

  • The aim of this study was to develop an analytical method for sulfonamide antibiotics (sulfadiazine, sulfamerazine, sulfamethazine, sulfamethoxazole, sulfathiazole) and their metabolites ($N^4$-acetylsulfadiazine, $N^4$-acetylsulfamerazine, $N^4$-acetylsulfamethazine, $N^4$-acetylsulfamethoxazole, sulfamethoxazole-$N^1$-glucuronide) in environmental samples. The solid phase extraction (SPE) with LC/ESI-MS/MS have been used for the analysis of target compounds, and the recoveries of SPE clean-up were at the range of 12-94% for C18 cartridge, 60-95% for HLB cartridge, 25-123% for MCX cartridge, and 70-90% for tandem HLB/MCX. By established method, detection limit, recovery, and relative standard deviation were 0.001~0.187 ng/mL, 66~115%, and 5~17%, respectively. This method was effective and sensitive to use for the simultaneous determination of sulfonamide antibiotics and their metabolites in environmental samples. Four sulfonamide antibiotics were detected at the range of 0.008~2.153 ng/mL. For metabolites, only $N^4$-acetylsulfamethoxazole was detected, but the concentration was under the MDL level.

  • Gas chromatographic method for the analysis of phorate(0,0-diethyl S-ethyl-thiomethyl phosphorithioate) and its metabolites in soil and vegetables was studied by using a mixed phase column(10% DC-200+8% Reoplex-400+2% QF-1 on Gas Chrom Q, $1.8{\times}2mm$ i.d, borosilicate glass column). This column can separate completely phorate and its four metabolites except phoratoxon sulfoxide. Retention time of standard mixture ranged 1.8 to 16.1 minutes at column temperature programming from 130 to $200^{\circ}C$ at $5^{\circ}C/min$ and detector sensitivity was also very high(0.05 to 1.05ng). Recoveries from soil and vegetables untreated but fortified with phorate and its three major metabolites at 0.05 and 0.5ppm levels were above 90% for phorate, phorate sulfoxide and phorate sulfone while recovery of phoratoxon metabolite was about 84%.

Comparative Analysis of Heavy Metal Exposure Concentrations and Volatile Organic Compound Metabolites among Residents in the Affected Area According to Residential Distance from a Coal-fired Power Plant (화력발전소 영향권 주민 거주지의 이격 거리별 중금속 및 휘발성유기화합물 대사체 노출 농도 비교 분석)

  • Background: There are concerns about the health effects of various environmental pollution exposures among residents living near coal-fired power plants (CFPP). Objectives: This study attempted to compare the concentrations of heavy metals in blood and urine and those of urinary volatile organic compound (VOC) metabolites according to the residential separation distance. Methods: Participants in the study totaled 334 people who have lived for more than 10 years in areas within 10 km of a CFPP. The separation distance was analyzed in quartiles by dividing it into Q1 (88 people), Q2 (89 people), Q3 (89 people), and Q4 (68 people). We explained the purpose of this study to the participants and collected blood and urine after obtaining signatures on a participation agreement. Results: The study participants were 102 males (30.5%) and 232 females (69.5%), with an average age of 71. The average length of residence and distance were 43.8 years and 4,800 meters. The geometric mean concentrations of Pb, Cd, and Hg in blood and As and Cd in urine were respective 1.35 ㎍/dL, 1.43 ㎍/L, 3.16 ㎍/L. They were 167.88 ㎍/g for creatinine and 1.58 ㎍/g creatinine. The metabolite concentrations of VOCs were 50.67 ㎍/g creatinine in t, t-muconic acid (t, t-MA), 10.73 ㎍/g creatinine in benzyl mercapturic acid, 317.05 ㎍/g creatinine in phenylglyoxylic acid, 123.55 ㎍/g creatinine in methylhippuric acid, and 190.82 ㎍/g creatinine in mandelic acid. The concentration of Pb in the blood and Cd and t, t-MA in the urine of residents within affected area of the CFPP showed statistically significant differences among distance groups. Conclusions: The concentration of urinary VOCs metabolites, especially t, t-MA, differed according to the distance groups of residents within the affected area of CFPP (p<0.05).

Dual Component Analysis for In Vivo T2* Decay of Hyperpolarized 13C Metabolites

  • Purpose: To investigate the exchange and redistribution of hyperpolarized $^{13}C$ metabolites between different pools by temporally analyzing the relative fraction of dual $T_2{^*}$ components of hyperpolarized $^{13}C$ metabolites. Materials and Methods: A dual exponential decay analysis of $T_2{^*}$ is performed for [1-$^{13}C$] pyruvate and [1-$^{13}C$] lactate using nonspatially resolved dynamic $^{13}C$ MR spectroscopy from mice brains with tumors (n = 3) and without (n = 4) tumors. The values of shorter and longer $T_2{^*}$ components are explored when fitted from averaged spectrum and temporal variations of their fractions. Results: The $T_2{^*}$ values were not significantly different between the tumor and control groups, but the fraction of longer $T_2{^*}$ [1-$^{13}C$] lactate components was more than 10% in the tumor group over that of the controls (P < 0.1). The fraction of shorter $T_2{^*}$ components of [1-$^{13}C$] pyruvate showed an increasing tendency while that of the [1-$^{13}C$] lactate was decreasing over time. The slopes of the changing fraction were steeper for the tumor group than the controls, especially for lactate (P < 0.01). In both pyruvate and lactate, the fraction of the shorter $T_2{^*}$ component was always greater than the longer $T_2{^*}$ component over time. Conclusion: The exchange and redistribution of pyruvate and lactate between different pools was investigated by dual component analysis of the free induction decay signal from hyperpolarized $^{13}C$ experiments. Tumor and control groups showed differences in their fractions rather than the values of longer and shorter $T_2{^*}$ components. Fraction changing dynamics may provide an aspect for extravasation and membrane transport of pyruvate and lactate, and will be useful to determine the appropriate time window for acquisition of hyperpolarized $^{13}C$ images.
