An artificial symbiosis was established between diazotropic Azomonas insignis and strawberry (Fragaria x ananassa). The partnership was created by in vitro techniques through callus induction and organogenesis. The basis of this partnerships is the bacterial dependence on the plants metabolic activity, using maltose in the medium as a carbon and energy source which can be utilized by the plant cells only. The presence of bacteria in the intercellular spaces of the callus tissues and regenerated plants was proven by microscopic techniques. Nitrogenase activity could also be detected in the plant tissues. For successful and high frequency introduction of bacteria to the plant tissues, biolistic gun method was used. On the basis of the DNA transfer method, Azotobacter vinelandii bacteria were delivered directly into strawberry tissues by the particle bombardment. This was the first use of living bacteria as microprojectils for bombardment of plant tissues. The treatment was successful, the presence of bacteria in the developing callus tissue and regenerated plants were detected by light and electron microscopy.
Brassica oleraceae var capitata is a member of the Brassicaceae family and is widely used as an horticultural crop. In the present study, transcriptome analysis of B. oleraceae L. var capitata was done for the first time using eight-week old seedlings treated with $50{\mu}m$ MeJA, versus mock-treated samples. The complete transcripts for both samples were obtained using the GS-FLX sequencer. Overall, we obtained 275,570 and 266,457 reads from seedlings treated with or without $50{\mu}m$ MeJA, respectively. All the obtained reads were annotated using biological databases and functionally classified using gene ontology (GO), the Kyoto Encyclopedia of Genes and Genomics (KEGG). By using GO analyses, putative transcripts were examined in terms of biotic and abiotic stresses, cellular component organization, biogenesis, and secondary metabolic processes. The KEGG pathways for most of the transcripts were involved in carbohydrate metabolism, energy metabolism, and secondary metabolite synthesis. In order to double the sequenced data, we randomly chose two putative genes involved in terpene biosynthetic pathways and studied their transcript patterns under MeJA treatment. This study will provide us a platform to further characterize the genes in B. oleracea var capitata.
Since 2005, food waste has been separately collected and recycled to animal feed or aerobic compost in South Korea. However, the conventional recycling methods discharge process wastewater, which contain pollutant equivalent to more than 50% of food waste. Therefore, anaerobic digestion is considered as an alternative recycling method of food waste to reduce pollutant and recover renewable energy. Recent studies showed that hydrogen can be produced at acidogenic stage in two-stage anaerobic digestion. In this study, the authors investigated the effects of pretreatment time and pH low set value on continuous mesophilic hydrogen fermentation of food waste. Food waste was successfully converted to $H_2$ when heat-treated at $70^{\circ}C$ for 60 min, which was milder than previous studies using pH 12 for 1 day or $90^{\circ}C$. Organic acid production dropped operational pH below 5.0 and caused a metabolic shift from $H_2/butyrate$ fermentation to lactate fermentation. Therefore, alkaline addition for operational pH at or over 5.0 was necessary. At pH 5.3, the result showed that the maximum hydrogen productivity and yield of 1.32 $m^3/m^3$.d and 0.71 mol/mol $carbohydrate_{added}$. Hydrogen production from food waste would be an effective technology for resource recovery as well as waste treatment.
Nowadays, the nutritional deficits are rarely seen in Korea. However, an increased availability of the highly palatable energy dense, nutrient-poor foods increases the risks of obesity and deficits of vitamins and minerals in the general population. Also, optimum intake of vitamins and minerals, which varies with age and genetic back ground, might not suffice the poor, young, obese, and elderly people. Young girls and individuals participating in weight reductions and aesthetic components are prone to micronutrient deficiencies because they restrict food intake and specific micronutrient rich foods. An inadequate intake of vitamins or minerals is associated with reduced physical performance and exercise capacity, increased obesity, decreased cognitive function, increased DNA damages such as single- and double-stranded breaks or oxidative DNA lesions, and accelerated aging process and increased neuronal damages with mitochondrial oxidative decay. Most of these deleterious effects of the deficit could be prevented by a one tablet of multivitamins with a good balanced diet. High dose B vitamins are frequently administered to overcome the metabolic inadequacy to the people with the less functional enzymes with increased Km values for their coenzymes due to the single gene mutation or due to the single nucleotide polymorphisms. And some certain antioxidant vitamins are also used in large quantities to overcome the oxidative stress and to repair the damages. In this review, new nutritional concepts of some vitamins and minerals, which are widely used and useful for the children, will be discussed.

Obesity is increasing worldwide and has become a major health burden in Western societies affecting every third American and every fifth European. Obesity makes a major contribution to morbidity and mortality, predisposing individuals to cardiovascular disease and diabetes. Many new substances are currently being investigated for their usefulness in the pharmacotherapy of obesity. Most anti-obesity drugs can be divided into four groups: those that reduce food intake; those that alter metabolism; those that increase thermogenesis; and those that regulate hormone involved in feeding behavior. In this article we review these and other agents available in various countries for the treatment of obesity. Perhaps more importantly, we have focussed on areas of potential productivity in the future. Over the last 5 or so years, this impetus in obesity research has provided us with exciting new drugs targets involved in the regulation of feeding behavior and cellular mechanism involved in energy expenditure. Recent development in the quest for control of human obesity include the discovery of hormones, neuropeptides, receptors and transcription factors involved in feeding behavior, metabolic rate and adipocyte development. For developing new, perhaps even more specific pharmacological agents, further research is needed to understand the individual different genetic and physiological basis of obesity. It remains the hope of research scientists that in the not too distant future we shall see a new class of anti-obesity drugs arising logically from the molecular biology revolutions.
Effects of the addition of \beta-glucanase to barley-based layer diets were examined by feeding 200 Leghorn layers with corn-based (Control) and \beta-glucanase supplemented diets (Barley+ Enzyme). The results obtained are sumrrarized as follows. 1. There were no siginificant (P>0.05) differences in hen-day egg production(%) and average egg weight between two treatments, indicating that the \beta-glucanase supplemented barley could successfully replace the commonly used corn in the layer diets. 2. Although there was no statistical difference (P>0.05) between two treatments, the daily feed consumption was numerically high in layers fed the barly diet compared to the corn-based diet. 3. Availabilities of crude fat and crude fiber of the barley diet were significantly poor (P<0.05) as compared to corn diet. 4. The \beta-glucarase supplementation depressed the viscosity of barley diets and excreta from therm. 5. Both serum and egg yolk cholesterol were not significantly affected by the addition of \beta-glucarase in the barley based diet. Our data indicate that the barley grain supplemented with \beta-glucarase can be sucessfully used as an energy source of layer diet when there is a price advantage. Although some possibilities to produce low cholesterol egg were recognized in this study, further studies pertaining to long-term feeding experiment and elucidaton of the metabolic interrelationship between serum and yolk cholesterol, are required to confirm the result.
Undifferentiated suspension cells had the ability to transfer glucose and fructose actively, but the suspension culture cells were unable to transfer saccharide without previously splitting to monosccarides. The uptake of fructose showed the low- and high-affinity system compared to of glucose, which possessed only one saturable uptake system. In this paper, the low affinity system of the uptake of fructose has been studied intensively. Glucose did not inhibit the low affinity system of fructose competitively. The Km value was 47 mM for fructose, 7.4 mM for glucose and Vmax was 69 $\mu$mol/h.g fresh weight for fuctose, 9.8 $\mu$ mol/h.g fresh weight for glucose. Metabolizer inhibitors, both 50 $\mu$M of CCCP and DNP, inhibited 70% of the uptake of the low affinity system of fructose. The proton ions were accompanied by the uptake of fructose. The stoichiometry showed ratio of proton to fructose was 0.17. The mechanism ofthe uptake was fructose-proton-symport. The molecules of fructose accmululated inside 25 times more than outside. Therefore, the low affinity system of fructose was not mere diffusion, but depended on metabolic energy and thus transported actively. The importance of this system was discussed.
The objective of this study was to investigate the metabolic energy consumption rate of the psychophysical Maximum Acceptable Weight of Loads (MAWLs) for different manual materials handling tasks. Lifting activities with four different lifting frequencies (2, 5, 8, 11 lifts/min) for a lifting range (from floor to 76cm height) were studied. The oxygen consumption rate and heart rate were measured or recorded while subjects were lifting their MAWLs. It was found that the relationship between MAWL and frequency can be described best by the exponential function with the R-sq value 0.9865 for this study. Psychophysical MAWL decreased from 22.38 to 7.48 kg, while the oxygen consumption rate with the MAWL increased from 717.8 to $1114.7m{\ell}-O_2/min$ as the frequency increased from 2 to 11 lifts/min. Heart rate also increased from 104.5 to 120.7 bpm. The ratio of oxygen consumption for the MAWL to the Physical Work Capacity (PWC) ranged from 28 to 43%. The MAWLs were greater than or equal to the Maximum Permissible Limits (MPLs) when the frequencies were higher than 8 lifts/min. It seems that the MAWLs by psychophysical approach when the frequencies were higher than 8 lifts/min tend to be overestimated from the viewpoint of the physiological criterion of the oxygen consumption rates. From these findings it is suggested that the NIOSH Guideline should not be directly applied to Korean without reasonable modifications.
Obesity is caused by unbalance of energy intake and expenditure, which results in extra accumulation of adipose tissue. Obesity is directly related to metabolic diseases such as diabetes, hyperlipidemia, fatty liver and so on. To investigate the anti-obesity effects of Scutellariae Radix, 70% EtOH extract and water extract of it were tested by in vitro and in vivo studies of fat accumulation. 3T3-L1 preadipocyte cell line was used in a in vitro study of fat accumulation. After 3T3-L1 cells were induced to differentiate into adipocytes, S. radix extract were added and fat accumulation was measured by oil red O staining. In vivo study showed that weight and epididymal/ retro-peritoneal adipose tissues were significantly reduced in mice fed Scutellariae Radix extract compared with control group. Especially, mice fed Scutellariae Radix extract showed reduced serum triglyceride and glucose levels. When adipose tissues were analyzed by microscope, mean adipocyte size was significantly reduced in Scutellariae Radix extract-fed mice. Therefore, this study showed inhibitory effects of Scutellariae Radix on in vitro and in vivo fat accumulation.
The purpose of this study was to develop a computerized software program to help for reproductive management in dairy cows. The developed software program in the study is compatible with a window 95/98 or XP system. Data in the system were recorded, stored, and processed from two sources: 1) Data downloaded monthly from the database of the Korean Dairy Herd Improvement Association (milk yields, milk somatic cell counts, milk fat, protein, lactose and urea nitrogen content). 2) Data recorded by the farmer or veterinarians by the time (body condition score, heats, inseminations, veterinary diagnosis and treatments). These data indices after processing by computerized dairy management system were presented by numerical or graphical display. The presented data were obtained from three dairy farms with more than 50 milking cows. The presented reports of this program using milk fat, protein, urea nitrogen, and somatic cell counts enabled the dairy producer and veterinarians to monitor the protein-energy balance and feeding management practice, and for distribution of diseases (mastitis, metabolic and reproductive disorder) in individual cows. The presented analytical reports of this program also included herd average of reproductive indices such as day to first insemination, days open, and inseminations per conception. This software program will assist in analysis, interpretation and demonstrate the results of reproductive trials conducted in dairy herds.
