Physiological signals such as body temperature, heart rate, blood pressure and heart rate variability and biomechanical workload for stress analysis were investigated during the cherry tomato harvesting work in a greenhouse. The skin temperatures raised $0.05^{\circ}C$/min, $0.03^{\circ}C$/ min, and $0.08^{\circ}C$/min in standing, stooping and squatting postures, respectively. Breath rate significantly increased from 18 to 28 breaths/min during the cherry tomato harvesting work. As the heart rate during the work ranged from about 72 to 110 beats/min (bpm), the cherry tomato harvesting work appeared to be a light intensity task of less than 110 bpm. The worker's average energy consumption rate in three positions during 43 min working time was 65.74 kcal (91 kcal/h in 70 kg). This was a light intensity of work, compared to 75 kcal/h in 70 kg of basic metabolic energy consumption rate of a worker with 70 kg weight; The maximum shear force on the disk (L5/ S1) due to static workload in the cherry tomato harvesting work was 446 N in the stooping posture, 321 N in the squatting posture and 287 N in the standing posture. Acute stress index expressed with the heart rate variability, increased parasympathetic activation up to about 70 while workers were doing most agricultural work in this study. This study provided a system to measure quantitatively workers' physiological change, kinematics and kinetic factors without any restrictions of space in the greenhouse works.
Chemically induced hypoxia has been shown to induce a depletion of ATP. Since intracellular free $Mg^{2+}\;([Mg^{2+}]_i)$ appears to be tightly regulated following cellular energy depletion, we hypothesized that the increase in $[Mg^{2+}]_i$ would result in $Mg^{2+}$ extrusion following hormonal stimulation. To determine the relation between $Mg^{2+}$ efflux and cellular energy state in a hypoxic rat heart and isolated myocytes, $[Mg^{2+}]_i,$ ATP and $Mg^{2+}$ content were measured by using mag-fura-2, luciferin-luciferase and atomic absorbance spectrophotometry. $Mg^{2+}$ effluxes were stimulated by norepinephrine (NE) or cAMP analogues, respectively. $Mg^{2+}$ effluxes induced by NE or cAMP were more stimulated in the presence of metabolic inhibitors (MI). Chemical hypoxia with NaCN (2 mM) caused a rapid decrease of cellular ATP within 1 min. Measurement of $[Mg^{2+}]_i$ confirmed that ATP depletion was accompanied by an increase in $[Mg^{2+}]_i.$ No change in $Mg^{2+}$ efflux was observed when cells were incubated with MI. In the presence of MI, the cAMP-induced $Mg^{2+}$ effluxes were inhibited by quinidine, imipramine, and removal of extracellular $Na^+.$ In addition, after several min of perfusion with $Na^+-free$ buffer, a large increase in $Mg^{2+}$ efflux occurred when $Na^+-free$ buffer was switched to 120 mM $Na^+$ containing buffer. A similar $Mg^{2+}$ efflux was observed in myocytes. These effluxes were inhibited by quinidine and imipramine. These results indicate that the activation of $Mg^{2+}$ effluxes by hormonal stimulation is directly dependent on intracellular $Mg^{2+}$ contents and that these $Mg^{2+}$ effluxes appear to occur through the $Na^+-dependent\;Na^+/Mg^{2+}$ exchange system during chemical hypoxia.
The1hepatic cell death induced by acetaminophen (APAP) is closely related to cellular adenosine triphosphate (ATP) depletion, which is mainly caused by mitochondrial dysfunction. Adenosine monophosphate (AMP)-activated protein kinase (AMPK) is a key sensor of low energy status. AMPK regulates metabolic homeostasis by stimulating catabolic metabolism and suppressing anabolic pathways to increase cellular energy levels. We found that the decrease in active phosphorylation of AMPK in response to APAP correlates with decreased ATP levels, in vivo. Therefore, we hypothesized that the enhanced production of ATP via AMPK stimulation can lead to amelioration of APAP-induced liver failure. A769662, an allosteric activator of AMPK, produced a strong synergistic effect on AMPK Thr172 phosphorylation with APAP in primary hepatocytes and liver tissue. Interestingly, activation of AMPK by A769662 ameliorated the APAP-induced hepatotoxicity in C57BL/6N mice treated with APAP at a dose of 400 mg/kg intraperitoneally. However, mice treated with APAP alone developed massive centrilobular necrosis, and APAP increased their serum alanine aminotransferase and aspartate aminotransferase levels. Furthermore, A769662 administration prevented the loss of intracellular ATP without interfering with the APAP-mediated reduction of mitochondrial dysfunction. In contrast, inhibition of glycolysis by 2-deoxy-glucose eliminated the beneficial effects of A769662 on APAP-mediated liver injury. In conclusion, A769662 can effectively protect mice against APAP-induced liver injury through ATP synthesis by anaerobic glycolysis. Furthermore, stimulation of AMPK may have potential therapeutic application for APAP overdose.
Underreporting of dietary intake is common and might distort analysis and interpretation of dietary surveys. This study was designed to investigate the degree of underreporting and characterastics of under-reporting group in Korean college female students. Dietary survey of 1-day 24-hour recall method was conducted on 379 college students in Seoul and Chonan areas. Physical activity and life style were aquired from questionnnaires. Underreporting was defined as energy intake(EI) lower than 0.9BMR(based metabolic rate), since EI<0.98BMR is statistically judged as bias in 1-day 24 hour recall. BMR was calculated from Schofield's equation. Proportion of underreporting was 18.7% and it's not so different from one of American or European women. Intake of nutrients except vitamin A by underreporting group was lower than other groups(p<0.001). Proportions of subjects with nutrient intake level less than 75% of Korean RDA were more than 80% in protein, Ca, Fe, vitamin A, riboflavin, niacin, zinc as well as energy. Dietary quality of underreporting group was also worse than other groups. Proportion of subjects less than 3 food groups among 5 food group was higher in underreporting group. The number of foods eaten by underreporting group were also less than those of other groups. BMI and body weight were the largest in underreporting group(p<0.05) and the trial of weight reduction was shown higher trend(p<0.01). Different in PAC and other characteristics between underreporting group and other group were not significant. Not only dietary quantity but also dietary quality were worse in the underreporting group. Furthermore underreporting in college female students seemed to be affected by body weight and concern for weight reduction.
Dairy cow undergoes major adaptations in glucose and lipid metabolism to meet fetal and mammary nutrient requirements during the periparturient period. These adaptations are characterized by major changes in response to the homeostatic signal of insulin. In response to insulin, fatty acids are mobilized to meet energy demand. And cow develop insulin resistance during the late prepartum period to facilitate glucose sparing for the fetus and mammary gland. Insulin resistance is an important adaptation and may be partly responsible for the large increase in plasma nonesterified fatty acid (NEFA) that occurs prior to parturition and coincides with the characteristic decrease in dry matter intake (DMI). Modulation of insulin resistance by administration of thiazolidinedione (TZD) during the periparturient period, likely through the activation of $PPAR{\gamma}$, has substantial potential to minimize energy and immune-related metabolic disorders, and thus may result in increased productivity and improved health of dairy cows.

The current experiment was conducted to compare nutrient utilization in the aged laying hens with different eggshell strengths or different intensities of brown eggshell color. Two metabolic trials with 96-week-old Hy-Line Brown laying hens were conducted. Twelve hens with a strong eggshell strength (SES) and 12 hens with a weak eggshell strength (WES) were selected and used in the first experiment, whereas 12 hens with dark brown eggshells (DBE) and 12 hens with light brown eggshells (LBE) were selected and used in the second experiment. All hens were fed a common commercial layer diet. Diets included chromic oxide as an indigestible marker to determine the apparent total tract retention (ATTR) of the gross energy (GE), dry matter, acid-hydrolyzed ether extract, nitrogen, neutral detergent fiber, acid detergent fiber, ash, and minerals. The results indicate that the ATTR of Ca in the aged laying hens with SES (59.7%) was greater (p < 0.05) than in the aged laying hens with WES (48.6%). However, the ATTR of the GE and other nutrients was not different between the hens with SES or WES. There were no differences in the ATTR of GE and nutrients between the hens with DBE or LBE. In conclusion, aged laying hens with SES have a greater ability to utilize dietary Ca than those with WES, indicating that the extent of dietary Ca utilization determines the eggshell strength of aged laying hens. However, nutrient utilization in diets may have little impact on eggshell brown coloration in aged laying hens.
BACKGROUND/OBJECTIVES: Nobiletin (NOB), a citrus flavonoid, is reported to have beneficial effects on cardiovascular and metabolic health. However, there is limited research investigating the effect of long-term supplementation with low-dose NOB on high-cholesterol diet (HCD)-induced hypercholesterolemia and non-obese nonalcoholic fatty liver disease (NAFLD). Therefore, we investigated the influence of NOB on hypercholesterolemia and NAFLD in HCD-fed mice. SUBJECTS/METHODS: C57BL/6J mice were fed a normal diet (ND) or HCD (35 kcal% fat, 1.25% cholesterol, 0.5% cholic acid) with or without NOB (0.02%) for 20 weeks. RESULTS: HCD feeding markedly reduced the final body weight compared to ND feeding, with no apparent energy intake differences. NOB supplementation suppressed HCD-induced weight loss without altering energy intake. Moreover, NOB significantly decreased the total cholesterol (TC) levels and the low-density lipoprotein (LDL)/very-LDL-cholesterol to TC ratio, and increased the high-density lipoprotein-cholesterol/TC ratio in plasma, compared to those for HCD feeding alone. The plasma levels of inflammatory and atherosclerosis markers (C-reactive protein, oxidized LDL, interleukin [IL]-1β, IL-6, and plasminogen activator inhibitor-1) were significantly lower, whereas those of anti-atherogenic adiponectin and paraoxonase were higher in the NOB-supplemented group than in the HCD control group. Furthermore, NOB significantly decreased liver weight, hepatic cholesterol and triglyceride contents, and lipid droplet accumulation by inhibiting messenger RNA expression of hepatic genes and activity levels of cholesterol synthesis-, esterification-, and fatty acid synthesis-associated enzymes, concomitantly enhancing fatty acid oxidation-related gene expression and enzyme activities. Dietary NOB supplementation may protect against hypercholesterolemia and NAFLD via regulation of hepatic lipid metabolism in HCD-fed mice; these effects are associated with the amelioration of inflammation and reductions in the levels of atherosclerosis-associated cardiovascular markers. CONCLUSIONS: The present study suggests that NOB may serve as a potential therapeutic agent for the treatment of HCD-induced hypercholesterolemia and NAFLD.

The growth and health of the fetus and neonate are directly influenced by the nutritional and physiological status of sows. Sows are often under catabolic conditions due to restrict feeding program during pregnancy and low voluntary feed intake during lactation. The current restrict feeding program, which aims at controlling energy intake during gestation, results in an inadequate supply of dietary protein for fetal and mammary gland growth. Low voluntary feed intake during lactation also causes massive maternal tissue mobilization. Provision of amino acids and fatty acids with specific functions may enhance the performance of pregnant and lactating sows by modulating key metabolic pathways. These nutrients include arginine, branched-chain amino acids, glutamine, tryptophan, proline, conjugated linoleic acids, docosahexaenoic acid, and eicosapentaenoic acid, which can enhance conception rates, embryogenesis, blood flow, antioxidant activity, appetite, translation initiation for protein synthesis, immune cell proliferation, and intestinal development. The outcome is to improve sow reproductive performance as well as fetal and neonatal growth and health. Dietary supplementation with functional amino acids and fatty acids holds great promise in optimizing nutrition, health, and production performance of sows and piglets. The uptake of putrescine, spermidine and spermine by Fortner's hamster amelanocytic melanoma AMEL-3 cells was observed in this study to be time-dependent, temperature-sensitive, pH-dependent and saturable. Metabolic poisons nullified polyamine uptake, an indication that this is an energy-requiring mechanism. The presence of $Na^+$ ions was found to be requisite to full activity. Valinomycin, gramicidin, monensin and the calcium ionophore calcimycin were also observed to inhibit the process substantially. The transporter active site would seem to contain sulfhydryl groups. Other diamines and polyamine analogues, as well as cationic diamidines, suppressed putrescine uptake. The presence of the ornithine decarboxylase inhibitor DFMO in the culture medium induced putrescine inflows. Putrescine, in turn, induced the negative expression of the carrier, thus suggesting that this influx mechanism is governed by up/down regulation. The cationic diamidine CGP 40215A and its analogue CGP039937A competitively inhibited putrescine transport, with Ki values of 1.9 and $15{\mu}M$, respectively. The role of polyamine uptake in these cultures is discussed.
Sugarcane press residue (SPR), a byproduct from the sugar industry was evaluated for it's nutrient and energetic quality in broilers and layers. The composition of SPR included (% DM): CP-11.76 (methionine-2.21, cystine-1.05, lysine-4.85, threonine-5.48% of CP), EE-7.87 (palmitic acid-30.3, stearic acid-4.1, oleic aicd-17.2, linoleic acid-38.0, linolenic acid-5.4% of EE), CF-10.08, TA-21.08 (Ca-3.87, P-1.10, Mg-0.95%, Fe-3500, Mn-284, Zn-113, Cu-61.5, Co-5.0 ppm and AIA-4.93%) and NFE-48.35% indicating that SPR is a valuable source of both organic and inorganic nutrients for poultry. The metabolic trials revealed the average ME of SPR as 749, 842 and 1,270 kcal/kg, respectively in broilers and 844, 936 and 1,031 kcal/kg in layers, at 10, 20 and 30% inclusion levels, respectively. Further, the fortification of SPR incorporated diets with biotechnological products viz., lipid utilizing agents (lipase and lecithin) or NSP degrading enzymes and their combination did not improve the ME content of such diets.
