Plant Regeneration from Mesophyll Protoplasts Culture of Solanum sisymbriifolium

  • The optimal culture conditions were studied for plant regeneration from mesophyll protoplasts of Solanum sisymbriifolium. Axenic seedlings of S. sisymbriifolium were used as a explant for protoplast culture. Many viable protoplasts were isolated by incubating leaf slices in an enzyme solution containing 0.25% Meicerase and 0.05% Macerozyme for 16 hr at $25^{\circ}C$ without shaking. Protoplast density of $5.0{\times}10^4\;ml^{-1}$ in Kao medium containing 5.0 mg/L NAA, 1.0 mg/L 2,4-D and 1.0 mg/L BA was optimal for colony formation. Most colonies were formed when protoplasts were cultured at $25^{\circ}C$ after initial culture at $30^{\circ}C$ for one week. On the MS agar medium with 1.0 mg/L zeatin, 38.4% of protoplast-derived calli differentiated shoots. These shoots rooted on 1/2MS medium with 5.0 g/L sucrose and 2.5 g/L gellan gum, and developed into whole plants.

  • This study was carried out to elucidate the physiological and ecological mechamism of the branch pattern and electronmicroscopic structure of photosynthetic organs in Zoysia japonica Steud. The angles between the internodes of rhizome and the branches of Z.iaponica were changed from 0 to 52 degrees. The rhizomes tended to branch from the 3th node to the 10th node. When the terminal bud was damaged by various environmental factors, the initial branching formation was appeared immediately to the 4th node. The internode growth of the rhizome started from the beginning of April and continued to the end of Octobor. It became evident that there is a division labor between two different kinds of photosynthetic cells of mesophyll and bundle sheath tissues in Z.japonica The thick-walled bundle sheath cells surrounds the leaf vascular bundles as the Kranz type and separates them from the mesophyll cells.

Selection of Glyphosate-Resistant Clones form MNNG-treated Mesophyll Protoplasts of Haploid Tobacco Plants (반수체 담배의 엽육 원형질체로부터 MNNG 처리에 의한 Glyphosate 저항성 클론의 선별)

  • Selection of glyphosate-resistant clones from MNNG-treated mesophyll protoplasts of haploid tobacco and their differentiation were studied. The protoplasts were treated with 0.1 to 100 $\mu\textrm{g}$/mL N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) for 30 min when they expanded to oval shapes. After the treatment, the protoplasts in 4-16 cell stages were transferred to the selective medium containing 1 mM glyphosate for the selection of the glyphosate-resistant colonies. The efficiency of the cell division of the protoplasts in the selective medium decreased as the MNNG concentrations in creased. Optimal MNNG concentration for induction of the glyphosate-resistant clones was 10$\mu\textrm{g}$/mL and mutation frequency was 2.66$\times$10-6. The stability of the glypohsate-resistance of the clones was examined by prolonged subculture in the medium with 1 mM glyphosate, and the resistant clones were survived more than 10 months. Among them one clone has been proliferating and greening and the others were proliferating without greening or greening with slower proliferating.

Physical Changes in Satsuma Mandarin Leaf after Infection of Elsinoë fawcettii Causing Citrus Scab Disease

  • Citrus scab disease is one of the destructive diseases that reduce the value of fruit for the fresh market. We analyzed the process of symptom development after infection with scab pathogen $Elsino{\ddot{e}}$ fawcettii in the susceptible satsuma mandarin leaves to observe the structural modification against pathogen. The cuticle and epidermal cells along with 3-5 layers of mesophyll tissue were degraded 1-2 days post inoculation. Surrounding peripheral cells of degraded tissues grew rapidly and then enveloped the necrotic area along with the growing conidia. Cross sections through the lesion revealed hyphal colonization in epidermis and mesophyll tissues. In response to the pathogen colonization, host cell walls were lignified, inner cells were rapidly compartmentalized and a semi-circular boundary was formed that separated the infected region from the non-infected region, and finally prevented the intercellular pathogen spread.

  • Leaf mesophyll protoplasts from five cultivars of tobacco (N. tabacum L.) were cultured. The protoplasts did not survive in culture medium containing Murashige and Skoog inorganic salts for over 6 days. NH4NO3 and FeSO4.Na2EDTA concentration of Murashige and Skoog medium were toxic in tobacco leaf mesophyll protoplast culture. Therefore we investigated optimum condition in Murashige and Skoog medium. High plating efficiency was obtained by reducing the concentrations of NH4NO3 and FeSO4.Na2EDTA to 1/3 and 1/10, respectively, on the supplemented with 5$\mu$M IAA, 0.5 $\mu$M 2.4-D 5 $\mu$M BAP. Plants were regenerated from protoplast-derived calluses.

Effect of Calcium Ion on Mesophyll Protoplast Culture of Arabidopsis thaliana (Arabidopsis thaliana의 엽육세포 원형질체배양에 미치는 칼슘이온의 영향)

  • The present study was performed to investigate the effect of calcium ion on the mesophyll protoplast culture of Arabidopsis thaliana. The mesophyll protoplase were isolated and cultured on an IMH medium supplemented with CaCl$_2$of various concentrations. When the protoplasts were cultured on the medium containing 0 to 12.5 mM CaCl$_2$, extreme vacuolization occurred without cell division. When the protoplasts were cultured with higher levels of CaCl$_2$ up to 50 mM, vacuolization decreased dose-dependent): and the number of plasma-rich cells increased. Cell division was induced when the protoplast were cultured on the medium with CaCl$_2$ higher than 25 mM. The highest plating efficiency (5-6%) was obtained with 50 mM CaCl$_2$. However the plating efficiency was markedly inhibited by 100mM CaCl$_2$ or above. These resole suggest that the relatively high concentration (50 mM) of calcium ion may be required for the culture of protoplasts.

  • A field experiment was conducted in order to investigate the seasonal changes of leaf grwoth and related characteristics in three cultivars of orchardgrass; Potomac, Kay and Sumas. The results were summarized as follows: 1. Leaf elongation was increased in a nearly linear phase during first and third cutting stages. It was increased slowly in early 10 days to 15 days after cutting and increased rapidly there-after during the rest cutting stages. In cultivars, Potomac was showed higher leaf elongation than other cultivars during all cutting stages. There was no difference of leaf width within cutting stages, but the leaf width of fall regrwoth was narrow. Sumas had relatively short and wide leaves. 2. Leaf dry weight and leaf area in first cutting stage were larger than others. Leaf area was increased rapidly form 15 days after cutting and leaf weight was increased rapidly from 20 days over all cutting stages. The increase in leaf area and dry weight were slow down after 30 days. 3. Number of epidermal cells was increased rapidly after cutting and the rate of increase was slow down after 30 days. In a cross section of leaf tissue, the part of mesophyll was occupied with about 60% of total area and larger area than other tissue, the part of mesophyll was occupied with about 60% of total area and larger area than other tissues. Leaf tissue had a large vacancy at early growth period after harvest and was filled gradually with mesophyll. This result was related to the increase of leaf dry matter.

Comparative Morphology of Plastids on Vegitative Tissue of Cannabis sativa L. (대마(Cannabis sativa L.) 영양조직의 색소체 비교)

  • Various plastids in leaf and bracteal tissues of Cannabis sativa L. were examined by electron microscopy. Young chloroplasts without starch grain in mesophyll cells were ellipsoidal, and osmiophilic globules within them were common in stroma. During the plastid differentiation, the mature chloroplasts in mesophyll were changed in shape depending on the numbers and sizes of starch grain in stroma. Electron-dense granular substances were occurred along the outer membrane of chloroplasts in mesophyll. Typical plastids with reticulate body were present in the glandular trichomes. Electron-grey material appeared along the surface of a plastid. A light area in reticulate body is considered to represent junction point of thylakoids.
