Predicting the moment capacity of RC slabs with insulation materials exposed to fire by ANN

  • Slabs prevent harmful effects of fire that may occur in any floor. However, it is necessary to protect the slabs from fire. Insulation materials may be appropriate to protect reinforced concrete (RC) slab from elevated temperature. In the present study, a model has been developed in artificial neural network (ANN) to predict the moment capacity ($M_r$) of RC slabs exposed to fire with insulation material. 672 data were obtained for ANN model through author's prepared program. Input layer in model consisted of seven input parameters; such as effective depth (d), ratio of d'/d, thermal conductivity coefficient ($k_{insulation}$), insulation materials thickness ($L_{insulation}$), reinforcement area ($A_{st}$), fire exposure time ($t_{\exp}$), and concrete compressive strength ($f_c$). The predicted $M_r$ by ANN was consistent with the obtained $M_r$ by author. It is proposed to ease computational complexity in determining $M_r$ using ANN. The effects of using insulation material on the moment capacity in RC slabs were also investigated. Insulating material with low thermal conductivity has been found to be more effective for durability to high temperature.

  • In today's fashion Street is a birthplace of new generation fashion and Street Fashion have frequently influenced on high fashion. From this view point this study is a meaningful approach to forecast future fashion trend by examining the street style of youth who are regarded as Street Fashion leaders. The purpose of this study is to examine the Youth Fashion style in early 1990s and to pro-vide a reference to fashion designer and mer-chandiser in trend forecasting and product plan-ning. This study is focused on 1990s Youth Fashion style through the historical Street Fashion : 1950s Teddy boys & Mods 1960s Hippies & Skinhead and 1970s Punk. There were mainly two popular Street Fashion phenomena in early 1990s. There were mainly two popular Street Fashion phenomena in early 1990s. One is grunge fashion created by new gener-ation in opposition to existing generation society and impacting on hish fashion leaders. The other is Remix style that is mixed : sub culture fashion originated by young group in 1950-1970s with 1990s sensitivity. Both fashion styles origionated and led by youth of street have influenced on famous high fashion designeres as well as the general public. Street Fashion will continue to play an import-ant role in future fashion and more study and interest on Street Fashion should be taken by fashion forecaster and related.

Universal Rice Primer (URP)-PCR에 의한 곰팡이 종의 유전적 다양성 검정 (Genetic Divesity Analysis of Fungal Species by Universal Rice Primer (URP)-PCR)

  • 20 mer의 URP primers 가 한국 재래적미의 반복배열 DNA 염기서열로부터 고안되어 다양한 곰팡이 종의 PCR 다형성검출에 적용 되어 왔다. URP-PCR 방법은 PCR반응중 $55^{\circ}C$이상의 고온에서 annealing반응을 함으로 하여 재생적인 PCR결과를 얻을 수 있으며 효모균류에서 담자균류의 고등 균류까지 33속, 142 종, 1,489 균주의 다양한 곰팡이 종의 유전체 DNA에 적용되어 그 유용성이 평가 되어 왔다. 본 논문은 URP-PCR의 특성과 지금까지 다양한 곰팡이 종 다양성 검정결과를 종합하여 보고 한다.

  • The determination of DNA hybridization can apply the molecular biology research, clinic diagnostics, bioengineering, environment monitoring, food science and other application area. So, The determination of hybridization is very important for the improvement of DNA detection system. In this study, we report the characterization of the DNA hybridization by the electricalchemical methods. A new electrochemical biosensor is described for voltammetric detection of gene sequence related to probe oligonucleotide of bacterium Escherichia coli O157:H7. The biosensor involves the immobilization of a 18-mer probe oligonucleotide, which is complemetary to a specific gene sequence related to Escherichia coli O157:H7 on a gold electrode through specific adsorption. The probe oligonucleotide was used to determine the amount of target oligonucleotide in solution using mitoxantrone(MTX) as the electrochemical indicators. The cathodic peak currents $(I_{peak})$ of MTX were linearly related to the concentration of the target oligonucleotide sequence in the range $1[{\mu}M]{\sim}0.1[nM]$. The detection limit of this approach was 0.01[nM]. In addition, these indicators were capable of selectivity discriminating against various mismatching condition.

  • Attachment and growth of mouse flbroblast cells on block copolypeptides were studied in the pres once or absence of serum proteins. Cells are rapidly attached to she polymer surface within 30 min regardless of substrate in the presence of serum. The number of flbroblast cells attached on the poly mer surface coated by collagen was larger than thats on the bare surface. Attachment of cells Is as a whole suppressed to a low level by the addltion of sodium azide in the absence of serum. Thls suE gests that the active attachment of cells requires the biological metabolism taking place on polymer substrates. In the presence of serum protein, flbroblast cells are more rapidly grown on the bolck co polymer consisting of poly(T-benzyl L-glutamate) (PBLG) and polyoxypropylene(POP) than on other block copolymers. These results were in agreement wish the data obatlned by an Inverted ml croscope.

  • 본 실험은 이병 딸기의 조직에서 분리 동정된 시들음병균(Fusarium oxysporum f. sp. fragariae) 균주들의 유?거 변이를 random amplified polymorphic DNA(RAPD) marker들을 이용하여 조사하였다. 총 24개의 딸기 시들음병 균주들의 DNA를 주형으로 하여 16개의 random 10-mer primer들을 사용하여 증폭시킨 결과 총 231개의 marker들을 이용하여 유전적 변이를 조사해 본 결과 크게 RAPD I과 RAPD II의 2개 그룹으로 나눌 수 있었다. RAPD I그룹에 속하는 균주는 VCG A에 속하는 Y1, K1, K2, K3, K4, N2, N3, N4-1, N6-1, N6-2, N8, N9, N10, M1-2-1 균주, VCG B에 속하는 M4-1 균주 그리고 VCG C에 속하는 N1, Y2 균주들이었고, RAPD II그룹에는 VCG B에 속하는 M1-1, M2-2-1, M2-4-2, M3-2, M3-3-2 균주와 VCG D에 속하는 N1 1 균주가 속하였다. 이들 2그룹 간에는 31%의 유사성이 있었다.

  • 초 고성능 바이오 서열 분석 장비 기술의 발달로 대량의 바이오 정보가 쏟아져 나오고 있으며, 바이오산업의 발달로 개인별 유전체 정보에 의한 맞춤의학의 시대가 도래되고 있다. 수많은 서열에 대한 분석에는 많은 저장장치 및 주기억장치가 필요하므로 슈퍼컴퓨터 급의 서버와 대량의 데이터를 빠르게 처리할 수 있는 프로그램이 필요하다. 이러한 분석에는 염기서열 일치 검색과 이를 기반으로 하는 Alignment와 Assembly 분석이 있으며, 이를 수행하는 기존의 알고리즘 및 대부분의 프로그램들은 염기서열을 문자열로 취급하고, 해쉬 인덱스 테이블, Brujin 그래프의 사용, 버러우즈 휠러 변환(BWT) 등의 기법을 활용하여 효율적인 분석을 도모하였다. 본 논문에서는 염기서열을 문자열이 아닌 k-mer 묶음의 정수형 하나로 변환하여 검색함으로써 저장 공간의 크기를 약 28% 이상으로 줄이고 형 변환 상태에서의 검색을 수행할 수 있는 알고리즘을 제안한다. Assembly 분석 프로그램인 CalcGen 프로그램을 개발하여 본 알고리즘의 효용성 및 효율성을 실험을 통해 검증하였다. 이 연구의 결과는 향후 대량의 유전체 염기서열의 효율적 분석과 저장 및 처리에 또 하나의 새로운 접근 방법을 제안하는데에 그 의미를 둘 수 있다.

Effect of Gender-Specific Adult Bovine Serum on Gene Expression During Myogenesis

  • Gender specificity in muscle growth and development is well known. Genesis of muscle is dependent on proliferation and differentiation potential of resident myogenic satellite cells (MSCs) present in muscle fibers. Multipotential capacity of forming myocyte, osteocyte, and adipocyte like cell makes MSCs a unique stem cell. To understand the molecular mechanism involved in determination of muscle quality due to difference in hormone concentration of different gender of animals, MSCs were isolated from bovine skeletal muscle and cultured in male, female, and castrated serum supplemented media. DNA microarray used consisted of 24,000 spots with 70 mer oligo in each spot. A total of 88 genes were up-regulated and 551 genes were down-regulated by more than two fold. Among up-regulated gene, 33, 34, and 21 genes were found up-regulated in cells grown in male, female, and castrated serum, respectively. Interestingly, male serum showed 4, female 11 and castrated male showed 4 genes expressed highly in each gender. Further study on the highly up-regulated gene may unfold the mystery of gender specificity found in muscle development. Also, the identification of differentially expressed genes in gender-specific serum will add information on infrastructure of bovine genome research.

Spectroscopic and Electrochemical Detection of Thrombin/5'-SH or 3'-SH Aptamer Immobilized on (porous) Gold Substrates

  • Thrombin is a serine protease that catalyzes the conversion of soluble fibrinogen to insoluble fibrin, and thus induces physiological and pathological blood coagulation. Therefore, it is important to detect thrombin in blood serum for purposes of diagnosis. To achieve this goal, it has been suggested that a 15-mer aptamer strongly binds with thrombin to form a G-quartet structure of the aptamer. Generally, 5'-end thiol-functionalized aptamer has been used as an anti-thrombin binder. Herein, we evaluate the possibility of utilizing a 3'-SH aptasensor for thrombin detection using SPR spectroscopy, and compare the enhancement of the electrochemical signal of the thrombin-aptamer bound on a porous gold substrate. Although the two aptamers have similar configurations, in SPR analysis, the 3'-SH aptamer was a effective aptasensor as well as 5'-SH aptamer. Results from electrochemical analysis showed that the porous gold substrate acted as a good substrate for an aptasensor and demonstrated 5-fold enhancement of current change, as compared to gold thin film.

A Rapid Method for the Measurement of the Absolute Activity of Carbon-14 in Pea Plant Tissue

  • 완두유묘의 자엽 및 뿌리조직중 탄소-14 방사능을 Liquid scintillation counter로 신속히 측정하는 새로운 방법에 관하여 실험하였다. 식물조직을 액체공기에 처리하여 분쇄한 다음 Gel phosphor에 헌탁시켜, Liquid scintillation counter로 측정하였다. 이시료에 다시 내부표준물질을 가한 다음 계측효율을 구하여 Apparent absolute activity를 얻었다. 이렇게 얻은 헌탁시료의 절대방사능은 자기 흡수현상 때문에, 따로 Flask combustion 방법으로 구한 절대방사능 보다는 낮았으나, 항상 비례하였다. 따라서 동일한 실험조건하에서 상술한 비율을 얻으면, 완두조직중 탄소-14의 점대방사능은, 비교적 간편한 헌탁측정(suspension counting) 만으로 가능하였다. 본방법은 완두 외에 다른 식물조직중 탄소-14 분석에도 쓰일 수 있다.
