The Effect of PAA on the Characterization of PVA/SSA ion Exchange Membranes (Poly(vinyl alcohol)/sulfo-succinic acid 이온교환막에 poly(acrylic acid)첨가에 따른 특성 연구)

  • The ion exchange membranes prepared from the reaction between poly(vinyl alcohol) (PVA) which is known as the good methanol barrier in pervaporation membrane processes and sulfo-succinic acid (SSA) was used as the basic membranes. In order to improve the ion exchange capacity, poly(acrylic acid) (PAA) was added to this ion exchange membranes. The methanol permeabilities, ion conductivities, water contents and ion exchange capacity were measured for the resulting membranes with varying PAA contents. In general, methanol permeability and ion conductivity of PVA/SSA/PAA membranes were less than those of PVA/SSA membranes due to the reduction of free volumes resulted from crosslinking. The vehicle mechanism could be more dominant than jump mechanism for membranes in question.

  • High-pressure membrane system like nanofiltration(NF) and reverse osmosis(RO) was investigated as a part of water treatment processes to produce high quality potable water with low organic matter concentration through membrane module tests and design simulation. River water and sand filtration permeate in Busan D water treatment plant were selected as feed water, and NE4040-90 and RE4040-Fen(Toray Chemical Korea) were used as NF and RO membranes, respectively. Total organic carbon(TOC) concentrations of NF and RO permeates were mostly below 0.5 mg/l and the average TOC removal rates of NF and RO membranes were 93.99% and 94.28%, respectively, which means NF used in this study is competitive with RO in terms of organic matter removal ability. Different from ions rejection tendency, the TOC removal rate increases at higher recovery rates, which is because the portion of higher molecular weight materials in the concentrated raw water with increasing recovery rate increases. Discharge of NF/RO concentrates to rivers may not be acceptable because the increased TDS concentration of the concentrates can harm the river eco-system. Thus, the idea of using NF/RO concentrate as the raw water for industrial water production was introduced. The design simulation results with feed water and membranes used in this work reveal that the raw water guideline can be satisfied if the recovery rate of NF/RO system is designed below 80%.

The effects of Korean Red Ginseng-derived components on oligodendrocyte lineage cells: Distinct facilitatory roles of the non-saponin and saponin fractions, and Rb1, in proliferation, differentiation and myelination

  • Background: Abnormalities of myelin, which increases the efficiency of action potential conduction, are found in neurological disorders. Korean Red Ginseng (KRG) demonstrates therapeutic efficacy against some of these conditions, however effects on oligodendrocyte (OL)s are not well known. Here, we examined the effects of KRG-derived components on development and protection of OL-lineage cells. Methods: Primary OL precursor cell (OPC) cultures were prepared from neonatal mouse cortex. The protective efficacies of the KRG components were examined against inhibitors of mitochondrial respiratory chain activity. For in vivo function of Rb1 on myelination, after 10 days of oral gavage into adult male mice, forebrains were collected. OPC proliferation were assessed by BrdU incorporation, and differentiation and myelination were examined by qPCR, western blot and immunocytochemistry. Results: The non-saponin promoted OPC proliferation, while the saponin promoted differentiation. Both processes were mediated by AKT and extracellular regulated kinase (ERK) signaling. KRG extract, the saponin and non-saponin protected OPCs against oxidative stress, and both KRG extract and the saponin significantly increased the expression of the antioxidant enzyme. Among 11 major ginsenosides tested, Rb1 significantly increased OL membrane size in vitro. Moreover, Rb1 significantly increased myelin formation in adult mouse brain. Conclusion: All KRG components prevented OPC deaths under oxidative stress. While non-saponin promoted proliferation, saponin fraction increased differentiation and OL membrane size. Furthermore, among all the tested ginsenosides, Rb1 showed the biggest increase in the membrane size and significantly enhanced myelination in vivo. These results imply therapeutic potentials of KRG and Rb1 for myelin-related disorders.

Tissue integration patterns of non-crosslinked and crosslinked collagen membranes: an experimental in vivo study

  • Purpose: Non-crosslinked and crosslinked collagen membranes are known to exhibit distinct degradation characteristics, resulting in contrasting orientations of the adjacent tissues and different biological processes. The aim of this study was to conduct a histomorphometric assessment of non-crosslinked and crosslinked collagen membranes regarding neovascularization, tissue integration, tissue encapsulation, and biodegradation. Methods: Guided bone regeneration was performed using either a non-crosslinked (BG) or a crosslinked collagen membrane (CM) in 15 beagle dogs, which were euthanized at 4, 8, and 16 weeks (n=5 each) for histomorphometric analysis. The samples were assessed regarding neovascularization, tissue integration, encapsulation, the remaining membrane area, and pseudoperiosteum formation. The BG and CM groups were compared at different time periods using nonparametric statistical methods. Results: The remaining membrane area of CM was significantly greater than that of BG at 16 weeks; however, there were no significant differences at 4 and 8 weeks. Conversely, the neovascularization score for CM was significantly less than that for BG at 16 weeks. BG exhibited significantly greater tissue integration and encapsulation scores than CM at all time periods, apart from encapsulation at 16 weeks. Pseudoperiosteum formation was observed in the BG group at 16 weeks. Conclusions: Although BG membranes were more rapidly biodegraded than CM membranes, they were gradually replaced by connective tissue with complete integration and maturation of the surrounding tissues to form dense periosteum-like connective tissue. Further studies need to be performed to validate the barrier effect of the pseudoperiosteum.

  • 이온교환막을 이용한 전기적 탈염기술은 막모듈 내에 양이온교환막과 음이온교환막을 교대로 장착시키고 모듈의 양단 전극에 전압을 적용함으로써 물속에 용존되어 있는 양이온과 음이온들을 전기의 힘을 이용하여 선택적으로 투과시키는 원리를 기반으로 하는 청정공정 기술이다. 이온교환막 공정은 전통적으로 산/알칼리의 생산, 산업폐수의 중금속의 제거, 해수의 담수화, 반도체 산업의 초순수의 제조, 해수에서 식염의 제조, 발효산업의 유기산 및 아미노산의 회수 등 다양한 산업분야에서 응용되어 왔다. 최근에는 이러한 기존의 응용분야에서 벗어나 새롭게 응용분야가 넓어지고 있다. 이온교환막과 다공성 탄소전극을 결합한 막축전식 해수담수화기술, 해수와 담수의 염도차를 이용한 역전기투석식 해수발전 등의 새로운 선택분리기능 및 응용분야를 가진 이온교환막의 개발 및 공정에 관한 연구가 활발히 이루어지고 있다. 그러나 국내에서는 이온교환막이 아직 상용화되지 않고 있어 이온교환막을 이용한 응용연구가 활발하게 진행되지 못하고 있어 그 개발이 시급하다. 본 논문에서는 먼저 이온교환막을 이용한 전기투석식 탈염기술, 물분해 전기투석, 전기탈이온 공정에 관한 동향을 조사하였다. 아울러 미래의 이온교환막의 응용기술인 해수담수화기술로서 역삼투법과 경쟁하여 에너지를 낮게 소모할 것으로 예상되는 분리막을 이용한 막축전식 탈염기술과 무한한 신재생에너지원인 해수와 담수를 이용한 역전기투석 해수발전기술에 대해 기술의 원리들과 최근의 연구동향 등을 정리하였다.

The Immunosuppressive Potential of Cholesterol Sulfate Through T Cell Microvilli Disruption

  • Cholesterol (CL) is required for various biomolecular production processes, including those of cell membrane components. Therefore, to meet these needs, CL is converted into various derivatives. Among these derivatives is cholesterol sulfate (CS), a naturally produced CL derivative by the sulfotransferase family 2B1 (SULT2B1), which is widely present in human plasma. CS is involved in cell membrane stabilization, blood clotting, keratinocyte differentiation, and TCR nanocluster deformation. This study shows that treatment of T cells with CS resulted in the decreased surface expression of some surface T-cell proteins and reduced IL-2 release. Furthermore, T cells treated with CS significantly reduced lipid raft contents and membrane CLs. Surprisingly, using the electron microscope, we also observed that CS led to the disruption of T-cell microvilli, releasing small microvilli particles containing TCRs and other microvillar proteins. However, in vivo, T cells with CS showed aberrant migration to high endothelial venules and limited infiltrating splenic T-cell zones compared with the untreated T cells. Additionally, we observed significant alleviation of atopic dermatitis in mice injected with CS in the animal model. Based on these results, we conclude that CS is an immunosuppressive natural lipid that impairs TCR signaling by disrupting microvillar function in T cells, suggesting its usefulness as a therapeutic agent for alleviating T-cell-mediated hypersensitivity and a potential target for treating autoimmune diseases.

Analysis of Bacterial Community Composition in Wastewater Treatment Bioreactors Using 16S rRNA Gene-Based Pyrosequencing (16S rRNA 유전자 기반의 Pyrosequencing을 이용한 하수처리시설 생물반응기의 세균군집구조 분석)

  • Bacterial community composition in activated sludge wastewater treatment bioreactors were analyzed using 16S rRNA gene-based pyrosequencing for the four different wastewater treatment processes. Sequences within the orders Rhodocyclales, Burkholderiales, Sphingobacteriales, Myxococcales, Xanthomonadales, Acidobacteria group 4, Anaerolineales, Methylococcales, Nitrospirales, and Planctomycetales constituted 54-68% of total sequences retrieved in the activated sludge samples, which demonstrated that a few taxa constituted majority of the activated sludge bacterial community. The relative ratio of the order members was different for each treatment process, which was assumed to be affected by different operational and environmental conditions of each treatment process. In addition, activated sludge had very diverse bacterial species (Chao1 richness estimate: 1,374-2,902 operational taxonomic units), and the diversity was mainly originated from rare species. Particularly, the bacterial diversity was higher in membrane bioreactor than conventional treatment processes, and the long solids retention time of the operational strategy of the membrane bioreactor appeared to be appropriate for sustaining diverse slow growing bacteria. This study investigating bacterial communities in different activated sludge processes using a high-throughput pyrosequencing technology would be helpful for understanding microbial ecology in activated sludge and for improving wastewater treatment in the future.

  • For commercialization of polymer electrolytemembrane fuel cell (PEMFC), durability of membrane electrode assemblies (MEAs) has to be improved. Especially, long-term stability of MEA is one of the most important issues for frequent shut-down and start-up processes of PEMFC. The degradation of MEA could be attributed to chemical attack of hydrogen peroxide radicals that are formed at high cell voltages without any special treatment to remove residual hydrogen from anode gas channel after shut-down of the fuel cell. In this study, we investigated the long-term stability of MEA under different on/off operation conditions. Residential hydrogen gas was removed from the anode flow channel by purging air or nitrogen. Also, a dummy resistance was applied to the fuel cell to exhaust residential hydrogen at the anode. In these cases, MEA showed much more stable durability. Electrochemical characteristics of the fuel cell were measured byrepeating the on/off cycles with the hydrogen removal processes. Also, degradation of MEA components was examined by SEM, TEM and XRD analyses.

A Conclusive Review on Amyloid Beta Peptide Induced Cerebrovascular Degeneration and the Mechanism in Mitochondria

  • Promising evidence suggests that amyloid beta peptide ($A{\beta}$), a key mediator in age-dependent neuronal and cerebrovascular degeneration, activates death signalling processes leading to neuronal as well as non-neuronal cell death in the central nervous system. A major cellular event in $A{\beta}$-induced apoptosis of non-neuronal cells, including cerebral endothelial cells, astrocytes and oligodendrocytes, is mitochondrial dysfunction. The apoptosis signalling cascade upstream of mitochondria entails $A{\beta}$ activation of neutral sphingomyelinase, resulting in the release of ceramide from membrane sphingomyelin. Ceramide then activates protein phosphatase 2A (PP2A), a member in the ceramide-activated protein phosphatase (CAPP) family. PP2A dephosphorylation of Akt and FKHRL1 plays a pivotal role in $A{\beta}$-induced Bad translocation to mitochondria and transactivation of Bim. Bad and Bim are pro-apoptotic proteins that cause mitochondrial dysfunction characterized by excessive ROS formation, mitochondrial DNA (mtDNA) damage, and release of mitochondrial apoptotic proteins including cytochrome c, apoptosis inducing factor (AIF), endonuclease G and Smac. The cellular events activated by $A{\beta}$ to induce death of non-neuronal cells are complex. Understanding these apoptosis signalling processes will aid in the development of more effective strategies to slow down age-dependent cerebrovascular degeneration caused by progressive cerebrovascular $A{\beta}$ deposition.

Numerical Study on a Poly-Generation Based on Gasification for Retrofit of a Natural Gas Combined Cycle (복합계통 개조를 위한 가스화 폴리제너레이션 시뮬레이션 연구)

  • In this work, a simulation study on net 500 MW class of Poly-Generation was conducted for the retrofit of an aged natural gas combined cycle. An entrained gasifier which has a capacity of maximum $260,000Nm^3/h$, 50 MW class of a Polymer Electrolyte Membrane Fuel Cell, and H-class Gas Turbine were selected as key processes. After unit design for those employed processes was set up and combined, the simulation was carried out with Gate-Cycle software (Ver. 6.0) for two cases. The selected cases are a retrofit type (Poly-Gen 1) and a new type (Poly-Gen 2). It was found that the efficiency of the retrofit case is 2.7% lower than that of the new case.
