Presence of an Albumin-Like Protein on the Plasma Membrane of Adipocytes Isolated from Korean Cow

  • The main function of adipocytes in various species is to store nutrient energy in the form of triglycerides, and this function may he closely related with hormonal signaling through the plasma membrane of adipocytes. Using SDS-PAGE, two-dimensional gel electrophoresis, and a membrane biotinylation technique, we have identified a 55KDa protein (55K protein) from the plasma membrane fraction of adipocytes, with an isoelectric point (pI) of 8.1-8.3. However, this 55K protein was not observed with a two-dimensional gel electrophoresis carried out on plasma membrane fractions prepared from the liver, heart, and kidney tissues. An analysis of the 12 amino acids sequence at the N-terminal of the 55K protein showed that it has a similar sequence to that of bovine serum albumin.

Pontoon and Membrane Breakwater

  • A numerical study on the hydrodynamic properties of a floating flexible breakwater consisting of triple vertical porous membrane structures attached to a floating rigid pontoon restrained by moorings is carried out in the context of two-dimensional linear wave-flexible body interaction theory. The tensions in the triple membranes are achieved by hanging a clump weight from its lower ends. The clump weight is also restrained properly by moorings. The dynamic behavior of the breakwater was described through an appropriate Green function, and the fluid multi-domains are incorporated into the boundary integral equation. Numerical results are presented which illustrate the effects of the various wave and structural parameters on the efficiency of the breakwater as a barrier to wave action. It is found that the wave reflection and transmission properties of the structures depends strongly on the membrane length taking major fraction of water column, the magnitude of tensions on membrane achieving by the clump weight, proper mooring types and stiffness, the permeability on the membrane dissipating wave energy.

더덕으로부터 면역세포 활성 성분의 분리 (Isolation of Active Components on Immunocytes from Codonopsis Lanceolatae)

  • The purpose of this research was to isolate an active component on immunoytes from 70% MeOH extract of Codonopsis lanceolatae Radix(CLE). CLE was fractionated successively with hexane, methylene chloride, n-butyl alcohol and water, and then the water fraction was separated with molecularporous membrane tubing(m.w. 3,500). Each fraction(50mg/kg) was administered p.o. once a day for 7 days in BALB/c mice respectively. None of these fractions affected the apoptosis and mitochondrial transmembrane potential in thymocyte. Hexane and methylene chloride fractions decreased CD4$^{[-10]}$ CD8$^{+}$ single-positive cells, and the water fraction enhanced CD4$^{+}$ CD8$^{[-10]}$ single-positive cells in thymocyte. The proliferation of thymocytes was decreased by the fraction hexane, but was enhanced by the water fraction. Hexane, methylene chloride and butyl alcohol fractions suppressed the production of nitric oxide, which was not affected by the water fraction. Hexane and butyl alcohol fraction suppressed the phagocytic activity, but water fraction enhanced it. The components(m.w. 3,500 above) separated from the water fraction enhanced the proliferation of thymocyte, the population of CD4$^{+}$ CDB$^{[-10]}$ single-positive cells, and phagocytic activity in macrophage. These results suggest that the stimulative components of proliferation, TH population and phagorytc activity is in the water fraction, and the molecular weight is 3,500 above. 분류된 천연유기물질을 이용한 막 오염 특성 평가 (Effect of Fractionated Organic Matter on Membrane Fouling)

  • 낙동강 원수의 경우 소수성 물질이 43%, 친수성 물질이 39%, 반친수성 물질이 18%를 차지하고 있는 것으로 나타났으며, 소수성 물질을 세분화하여 조사한 결과 fulvic acid는 62%, humic acid는 38%로 나타났으며, 또한, 카르복실기는 64%, 페놀기는 36%를 차지하고 있는 것으로 나타났다. 친수성 물질을 세분화하여 조사한 결과 HPI-N이 44%, HPI-C가 56%를 차지하고 있었다. 친수성 유기물질 중 HPI-C에 비하여 HPI-N이 막 오염을 더욱 많이 유발하였으며, 소수성 유기물질에서는 HA가 FA에 비하여 막 오염을 더 많이 유발하였으며, 작용기에 따른 막 오염 현상을 살펴보면 카르복실기의 경우 페놀기에 비하여 투과 flux 감소가 크게 나타나고 있다. 공극의 크기가 커질수록 공극 속으로 전달되는 고분자 유기물이 많이 발생하며 이러한 고분자 유기물이 공극을 막아버리는 현상에 의하여 투과 flux 감소가 발생하는 것으로 나타났다. 또한 공극의 크기가 상대적으로 큰 막의 경우 투과되는 양이 공극이 작은 막 보다 상대적으로 많기 때문에 운전초기에 많은 양의 유기물이 막으로 전달되어 막의 표면과 공극에 흡착되어 막의 공극의 크기가 커짐에 따라 투과 flux 감소가 크게 나타났다.

  • Membranes obtained from the normal human RBC population were separated by continuous sucrose density gradient centrifugation and the density-fractionated membranes were then examined for changes in molecular markers. This study focuses on changes of (i) the membrane protein profile, (ii) differences in membrane-associsted enzyme activities, and (iii) the amount of autologous IgG bound. The following observations were made: (i) ratios for band 4. la over the sum of bands (4. la + 4.Ib) ranged from 0.58 to 0.79 for membranes of lowest density; (ii) significant changes in bound glyceraldehyde-3-phosphate dehydrogenase and acetvlcholinesterase activities were found; (iii) the amounts of autolosous IgG's attached to the red blood cells was highest in the membrane fraction of lowest density.

Effects of n-Alkanols on the Lateral Diffusion of Total Phospholipid Fraction Extracted from Brain Membranes

  • We investigated the effects of n-alkanols on the range and rate of the lateral diffusion of 1, 3-di(1-pyrenyl)propane in the model membranes of total phospholipid fraction extracted from synaptosomal plasma membrane vesicles. n-Akanols increased the range and rate of the lateral diffusion of 1, 3-di(1-pyrenyl)propane in the bulk model membrane structures (inner + outer monolayers) and the potencies of n-alknols up to 1-nonanol increased by 1 order of magnitude as the carbon chain length increases by two carbon atoms. The cut-off phenomenon was reached at 1-decanol, where further icnrease in hydrocarbon length resulted in a decrease in the lateral diffusion. However, significant changes in the 1'/1 value were not observed by methanol (from 100 to 2500 mM), ethanol (from 25 to 800 mM), and 1-propanol (from 10 to 250 mM) over entire concentration.

  • In order to confirm the biological function of ORF10 in cephabacin biosynthesis gene cluster of Lysobacter lactamgenus as an ATP-binding-cassette (ABC) transporter, the gene for ORF10 was amplified and subcloned into pET-28a(+) expression vector. After gene induction with 0.5 mM IPTG at 30~! and further cultivation at $30^~$ !. for 8 hr, a lot of the recombinant ORF10 protein was produced as soluble form in cytoplasmic fraction as well as a membrane protein in the membrane fraction as likely as other ABC transporters. Plant Inositol Signaling - Biochemical Study of Phospholipase C and D-myo-inositol -1,4,5-trisphosphate receptor

  • It is now generally accepted that a phosphoinositide cycle is involved in the transduction of a variety of signals in plant cells. In animal cells, the hydrolysis of phosphatidyl-4,5-bisphosphate catalysed by phosphatidylinositol - specific phospholipase C yields to D-myo-inositol - 1,4,5-trisphosphate and diacylglycerol, which are well known second messengers. The binding of InsP$_3$to a receptor located on the endoplasmic reticulum triggers a calcium release from the endoplasmic reticulum. We have detected and partially characterised key components of phosphoinositide signaling. First, tobacco microsomal fraction and plasma membrane PI-PLC. Consecutively, using a radioligand binding assay we have identified a $Ca^{2+}$ -dependent high affinity InsP$_3$binding site in microsomal membrane fraction vesicle preparation and then we have measured inositol-1,4,5-trisphosphate induced calcium release from tobacco microsomal fraction. These findings suggest that phosphoinositide signaling system is present and operates in the tobacco suspension culture.e.

인유 및 우유의 획분에 존재하는 표지효소들의 효소활성과 분포 (Enzymatic Activity and Distribution of Marker Enzymes between Human Milk and Bovine Milk with Their Separated Milk Fractions)

  • Human milk and bovine milk in normal stage were fractionated four parts : whey, skimmilk membrane, and casein pellet. The specific activity (nmole / mim / mg protein) and distribution ratio(%) of suborganella marker enzymes in each separated milk fraction were determined. Especially, neutral $Ca^{2+}$-ATPase, acid $Ca^{2+}$-ATPase, NADH-cytochrome C reductase, and acid phosphatase were higher in human milk. However, both $Ca^{2+}$-ATPases were not detected in all fractions of bovine milk. On the other hand, 5'-nucleotidase, phosphodiesterase I, alkaline phosphatase, and $\gamma$-glutamyl transpeptidase activities in bovine milk were higher than in human milk. Most of the marker enzymes were highly distributed in cream fraction of either human milk or bovine milk, and their specific activities were high to 24 fold from 3 fold when compared with that of whole milk. These results suggest that marker enzymes in mammary epitherial cell are transfered into cream fraction by the membrane rearrangement, and different biochemical reaction between human and bovine exists for milk secretion in mammary gland.

Effect of IGF-I Rich Fraction from Bovine Colostral Whey on Murine Immunity

  • Insulin-like growth factor-I (IGF-I) rich fraction, collected components between 1 kDa and 30 kDa, was fractionated from bovine colostral whey using an ultrafiltration membrane. IGF-I was confirmed in the collected IGF-I rich fraction by both SDS-PAGE and Western blotting. The concentration of IGF-I in the IGF-I rich fraction was 10 ng/mg protein. One hundred microliters of the reconstituted IGF-I rich fraction was intraperitoneally injected into ICR male mice for 2 weeks at 24 h intervals. The functions of peritoneal macrophages, including phagocytosis, interleukin (IL)-6 and tumor necrosis factor (TNF)-${\alpha}$ production, and nitric oxide and hydrogen peroxide production, were enhanced significantly by the administration of the IGF-I rich fraction in a dose-dependent manner (p<0.01). The proliferation of Concanavalin (Con) A-stimulated and Lipopolysaccharide (LPS)-stimulated splenocytes was also determined to have been enhanced significantly by the administration of the IGF-I rich fraction in a dose-dependent manner (p<0.01). Our results indicate that the administration of IGF-I rich fraction obtained from bovine colostral whey enhances both innate and acquired immunity for ICR male mice.
