Development and Characterization of Membrane for Local Delivery of Cephalexin

  • Laminated films composed of drug-containing reservoir layer and drug-free membrane were prepared. Zero-order drug release with lag time was achieved by laminating drug-free film onto the reservoir layer, while burst effect was observed on cast-on film. The rate controlling membrane was either attached to or cast directly into the reservoir. The release rate was independent on the reservoir composition but dependent on the composition of rate-controlling membrane. In growth inhibitory test of cephalexin from Eudragit RS film to Streptococcus Mutans, the disk even after release test for 72 hours showed more bacterial growth inhibition than that of control. Permeation of drug through rat skin was proportional to the HPC fraction in the film. We could control the release of cephalexin from the film by changing the fraction of Eudragit RS, HPC and DEP content. Consequently, Eudragit RS/HPC film was found to be very effective system for local delivery of drugs.

  • The activity of phospholipase A$_2$ (PLA$_2$) was identified and characterized from cytosolic and membrane fractions of oat cells, respectively. PLA$_2$ activity was determined fluorometrically in the presence of serum albumin using phospholipids labeled at sn-2-acyl position with 10-pyrenyldecanoic acid. When the cell-free extracts of oat tissues were fractionated by ultracentrifugation at 100,000 x g and the PLA$_2$ activity was assayed, we found that most of the PLA$_2$ activity was revealed from the cytoplasmic fraction rather than from the membrane fraction. The activity of cytosolic PLA$_2$ was dependent on Ca$^{2+}$ concentration and the optimum concentration of Ca$^{2+}$ was found to be 100 $\mu$M. It was also found that PLA$_2$ could be translocated toward the membrane site from the cytosol upon increasing Ca$^{2+}$ concentration. These results might suggest that an increased [Ca$^{2+}$]$_i$ by phytochrome action could promote the translocation of the cytosolic PLA$_2$ toward the membrane site.

Effects of Egg Shell Membrane Hydrolysates on Anti-Inflammatory, Anti-Wrinkle, Anti-Microbial Activity and Moisture-Protection

  • This study was conducted to examine the effects of eggshell membrane hydrolysates (ESMH) on the anti-inflammatory, anti-wrinkle, anti-microbial activity, and moisture-protection for cosmetic use. Whole ESMH (before fractionation), and fraction I (>10 kDa), fraction II (3-10 kDa), and fraction III (<3 kDa) of the hydrolysates were assessed in this experiment. As lipopolysaccharide (LPS) and IFN-${\gamma}$ caused the inflammation on Raw264.7 cell, whole ESMH and fraction I showed to be effective in inhibiting the induction of cell inflammation depending on the concentration, and also showed outstanding effect to suppress the skin inflammation. Fraction I inhibited collagenase and elastase activities to a greater extent than the other fractions, while all fractions had antibiotic effects at concentrations of 10 mg/disc and 20 mg/disc. In addition, it showed the moisture protection effects of skin on the holding amount and losing amount of moisture in upper-inner arm of the human body with a relatively low loss rate in skin, which confirmed that the hydrolyzed fractions of ESM helps to form the superior protective layer of moisture. It was concluded that ESMH fractions with different molecular weights, especially the 10 kDa fraction, have anti-lipopolysaccharide, anti-IFN-${\gamma}$-induced inflammation, anti-collagenase and elastase activities, and thus can be used as a cosmetic agent to protect skin.

The Neuroprotective Potential of Cyanidin-3-glucoside Fraction Extracted from Mulberry Following Oxygen-glucose Deprivation

  • In this study, cyanidin-3-glucoside (C3G) fraction extracted from the mulberry fruit (Morus alba L.) was investigated for its neuroprotective effects against oxygen-glucose deprivation (OGD) and glutamate-induced cell death in rat primary cortical neurons. Cell membrane damage and mitochondrial function were assessed by LDH release and MTT reduction assays, respectively. A time-course study of OGD-induced cell death of primary cortical neurons at 7 days in vitro (DIV) indicated that neuronal death was OGD duration-dependent. It was also demonstrated that OGD for 3.5 h resulted in approximately 50% cell death, as determined by the LDH release assay. Treatments with mulberry C3G fraction prevented membrane damage and preserved the mitochondrial function of the primary cortical neurons exposed to OGD for 3.5 h in a concentration-dependent manner. Glutamate-induced cell death was more pronounced in DIV-9 and DIV-11 cells than that in DIV-7 neurons, and an application of $50{\mu}M$ glutamate was shown to induce approximately 40% cell death in DIV-9 neurons. Interestingly, treatment with mulberry C3G fraction did not provide a protective effect against glutamate-induced cell death in primary cortical neurons. On the other hand, treatment with mulberry C3G fraction maintained the mitochondrial membrane potential (MMP) in primary cortical neurons exposed to OGD as assessed by the intensity of rhodamine-123 fluorescence. These results therefore suggest that the neuroprotective effects of mulberry C3G fraction are mediated by the maintenance of the MMP and mitochondrial function but not by attenuating glutamate-induced excitotoxicity in rat primary cortical neurons.

Effects of Egg Shell Membrane Hydrolysates on UVB-radiation-induced Wrinkle Formation in SKH-1 Hairless Mice

  • This study was conducted to examine the effect of egg shell membrane hydrolysates (ESMH) on wrinkle, UV, and moisture protection for cosmetic use. ESMH were fragmented as whole ESMH (before fractioning), Fraction I (> 10 kDa), Fraction II (3-10 kDa), and Fraction III (< 3 kDa). In order to test whether fractionated ESMH can be used for functional cosmetic materials, we examined not only the level of hyaluronic acid and collagen production, but also the MMP-1 activity using a HaCaT and CCD-986Sk cell line. Our study treated each sample of fractionated ESMH with different concentrations (0.01, 0.1, 1 mg/mL). In our in vivo research, we used hairless mice that had been exposed to UV-B to induce wrinkles for 7 wk, then applied Fraction I to the treatment group for 5 wk and then tested skin thickness, minimum erythema dose and moisture content. In addition, Fraction I was high in collagen and HA biosynthesis and it was better than TGF-${\beta}$ in improving of the skin. When TNF-${\alpha}$ caused MMP-1 activity in the CCD-986Sk cells, the whole ESMH and Fraction I proved to be effective in hindering the induction of collagenase depending on the concentration, and also showed outstanding effects in the suppression of skin aging. We found that the treatment group mice's UV-B radiation-induced skin damage was largely mitigated compared to that of the non-treatment group mice. Thus, we have concluded that EMSH helps to mitigate UV-B radiation-induced wrinkles, collagen, HA, MMP-1 activity and can be used for functional cosmetic materials.

Apoptosis-Inducing Activity of HPLC Fraction from Voacanga globosa (Blanco) Merr. on the Human Colon Carcinoma Cell Line, HCT116

  • Voacanga globosa (Blanco), a plant endemic to the Philippines, is traditionally used especially by indigenous people of Bataan in the treatment of ulcers, wounds and tumorous growths. This study aimed to provide scientific evidence to therapeutic properties by determining cytotoxic and pro-apoptotic activity of HPLC fractions from leaves on HCT116 human colon carcinoma and A549 human lung carcinoma cell lines. Ethanolic extraction was performed on V globosa leaves followed by hexane and ethyl acetate partitioning. Silica gel column chromatography and high performance liquid chromatography (HPLC) produced MP1, MP2 and MP3 fractions. Cytotoxic activity of the fractions was determined through MTT assay against the cancer cell lines HCT116 and A549 and the non-cancer AA8 Chinese hamster ovarian cell line. Pro-apoptotic activities of the most active fractions were further assessed through DAPI staining, TUNEL assay and JC-1 mitochondrial membrane potential assay with HCT116 cells. While the MPI fraction exerted no significant activity against all cell lines tested, MP2 and MP3 fractions demonstrated high toxicity against HCT116 and A549 cells. The MP3 fraction induced formation of apoptotic bodies, condensed DNA and other morphological changes consistent with apoptosis of HCT116 cells and TUNEL assay showed significant increase in DNA fragmentation over time. In these cells, the MP3 fraction also induced mitochondrial membrane destabilization, which is generally associated with the beginning of apoptosis. Phytochemical analysis demonstrated the presence only of saponins and terpenoids in the MP3 fraction. The results indicate that the MP3 fraction exerts cytotoxic activity on HCT116 cells via induction of apoptosis triggered by loss of mitochondrial membrane potential crucial for cell survival.

Biochemical Characterization of an ABC Transporter Gene Involved in Cephabacin Biosynthesis in Lysobacter lactamgenus

  • An ATP-binding-cassette (ABC) transporter gene in the cephabacin biosynthetic gene cluster of Lysobacter lactamgenus was characterized. The amplified orf10 (cpbJ) gene was subcloned into pET-28a(+) vector and expressed in E. coli BL21(DE3) strain by 0.5 mM IPTG at $30^{\circ}C$. The membrane fraction of recombinant E. coli cells was separated by ultracentrifugation, and solubilized using 2.5% octyl-$\beta$-D-glucoside. Using the solubilized membrane fraction, the artificial proteoliposomes were reconstituted and analyzed for the biological activity of CpbJ protein. Upon measuring ATPase activity, the proteoliposome made from recombinant E. coli membrane proteins showed slightly higher activity than that from host E. coli membrane proteins. In the measurement of membrane transport activity, the reconstituted proteoliposome of recombinant E. coli membrane proteins exhibited higher activity when both substrates of cephalosporin C and L-Ala-L-Ser were applied, compared to the case of cephalosporin C or L-Ala-L-Ser only. It implies that the CpbJ protein is an ABC transporter secreting cephabacin antibiotics synthesized in cytoplasm.

NaA 제올라이트 분리막의 물/에탄올 투과증발 특성 (Pervaporation Characteristics of NaA Zeolite Membrane for Water/Ethanol Mixture)

  • 분리막 투과증발은 증류에 비하여 낮은 에너지를 요구하기 때문에 물/유기물 혼합물의 분리에 이용될 수 있으며 공비 혼합물이나 이성질체의 분리에 선호되는 방법이다. 제올라이트 분리막은 고분자 분리막에 비하여 열적, 기계적, 화학적 안정성이 우수하며 NaA 제올라이트 분리막은 큰 친수성을 나타내므로 물/유기물 혼합물로부터 효율적인 물 분리에 적합하다. 본 연구에서는 NaA 제올라이트 분리막을 이용한 투과증발 막분리 공정을 이용하여 물/에탄올 혼합물에서 선택적으로 물을 분리하고자 하였다. 에탄올의 공급 농도가 증가함에 따라 총투과플럭스는 감소하였으며 선택도는 증가한 후 높은 에탄올 농도에서 감소하는 것을 관찰할 수 있었다. 실험 온도의 증가에 따라 총투과플럭스는 증가하였으며 선택도 또한 증가하였지만 높은 에탄올 농도에서는 감소하였다. 160시간 이상의 장기간 실험을 통하여 총투과플럭스와 선택도가 안정적으로 일정함을 확인할 수 있었다. 또한, 본 연구를 통하여 얻어진 NaA 제올라이트 분리막은 증류 및 PVA 계열 고분자 분리막보다 우수한 성능을 나타냄을 알 수 있었다.

쥐 소뇌에 있어서 raf protein kinases 의 면역세포 화학적 분포와 단백질 양상 (Immunocytochemical distribution of raf protein kinases and protein pattern in rat cerebellum)

  • 본 연구에서는 SDS/polyacrylamide 젤 전기영동에 의한 쥐의 성장과정에 따른 소뇌의 단백질양상의 변화양상과 immunocytochemistry를 이용하여 c-raf a-raf kinase의 정상 소뇌에서의 분포에 대해 관찰 하였으며 western blot을 이용하여 소뇌의 단백질들에서 c-raf의 존재에 대해 살펴보았다. 단백질 양상에서 쥐의 성장에 따라 crude에선,ㄴ 49,200 dalton과 169,000 dalton 사이의 bands가 양적 증가를 보였으며 cytosolic fraction 에서는 37,800 dalton의 band가 양적 증가를 보이는데 비해 membrane fraction 에서는 260,600 dalton의 band가 증가하였다. 이러한 결과로 성장 발달에 따라 고분자 량의 물질들이 이들 소뇌 부위에서 기여하였을 것으로 추정할 수 있었다. Immunocytochemistry에 의한 분석에서는 c-raf와 a-raf가 소뇌의 피질주위에서 조롱박 세포(Purkinje cell) 의 세포질 특히 핵 주변부위에서 강하게 검출되었으며 a-raf에 비해 c-raf가 더 강하게 나타났었다. 그리고 그 외에 Nucleus embolifornis의 큰 neuronal cell의 세포질 부위의 나타남을 볼 수 있었다. Immunoblot에 의한 분석에서는 crude와 cytosolic fraction에서 raf protein kinase의 존재를 확인할 수 있었으며, 이상의 결과들을 종합해 보았을 때 소뇌의 정상의 많은 신경세포(neuronal cell)에 raf protein kinase가 분포되어 있으며 이들이 정상의 cell에서 기능을 가질 것으로 추정된다.

Growth Factor를 처리한 피부상피세포로부터 Protein Kinase C Isoenzyme의 검출 (Detection of Protein Kinase C Isoenzymes in the Growth of Human Epidermal Keratinocytes by Growth Factors)

  • Protein kinase C는 세포의 신호전달계에 관여하는 중요한 조절효소로서 여러 가지 세포의 분화와 증식과도 밀접한 관련이 있다. 신생아의 포피 keratinocyte를 농도 200 ng/ml의 human recombinant epidermal growth factor (hrEGF)와 human recombinant insulin-like growth factor-1 (hrIGF-1) 그리고 hrEGF와 hrIGF-1의 혼합액을 각각 첨가하여 24시간 배양한후 세포질과 세포막의 PKC단백질을 추출하여 그 농도를 측정하고, Western blot analysis를 이 용하여 각 growth factor들의 PKC isoenzyme에 대한 영향을 분석하였다. 세포질의 총 PKC 단백질의 농도는 hrIGF-1을 처리한 keratinocyte에서 가장 높았으며, 세포막에서는 대조군의 단백질 농도가 가장 높게 나타났다. EGF를 처리한 keratinocyte의 세포질에서 는 PKC-$\beta$II, -$\delta$, -$\theta$가 막성분에서는 PKC-$\alpha$, -$\beta$I, -$\delta$, -$\Im$, -$\theta$가 증가하였다. IGF-1을 처리한 군의 세포질성분에는 PKC-$\beta$I, -$\Im$, -$\theta$, 막성분에서는 PKC-$\alpha$, -$\beta$I, -$\delta$, -$\Im$, -$\varepsilon$, -$\theta$가 증가하였다 EGF와 IGF-1의 혼합처리 군에서는, PKC-$\alpha$, -$\beta$I, -$\Im$, -$\theta$이 세포질에서, PKC-$\alpha$, -$\delta$, -$\Im$, -$\varepsilon$, -$\theta$은 세포막에서 증가하였다.
