  • Injectable RGD-bioconjugated Mussel Adhesive Proteins (RGD-MAPs) composite hydroxypropyl methylcellulose (HPMC) hydrogels provide local periodontal tissue for bone filling in periodontal surgery. Previously we developed a novel type of injectable self-supported hydrogel (2 mg/ml of RGD-MAPs/HPMC) based porcine nano hydroxyapatite (MPH) for dental graft, which could good handling property, biodegradation or biocompatibility with the hydrogel disassembly and provided efficient cell adhesion activity and no inflammatory responses. Herein, the aim of this work was to evaluate bone formation following implantation of MPH and collagen membrane in rabbit calvarial defects. Eight male New Zealand rabbits were used and four circular calvarial defects were created on each animal. Defects were filled with different graft materials: 1) collagen membrane, 2) collagen membrane with MPH, 3) collagen membrane with bovine bone hydroxyapatite (BBH), and 4) control. The animals were sacrificed after 2 and 8 weeks of healing periods for histologic analysis. Both sites receiving MPH and BBH showed statistically increased augmented volume and new bone formation (p < 0.05). However, there was no statistical difference in new bone formation between the MPH, BBH and collagen membrane group at all healing periods. Within the limits of this study, collagen membrane with MPH was an effective material for bone formation and space maintaining in rabbit calvarial defects.

  • This study attempted to evaluate the process of self-forming dynamic membrane formation on mesh filter in membrane bioreactor with a two-stage method of batch (agitation) and continues (aeration) stage at different sludge concentrations. Four concentrations of activated sludge including $6{\pm}0.4$, $8{\pm}0.5$, $10{\pm}0.3$, $14{\pm}0.3g/L$ were used to demonstrate the optimal concentration of sludge for treating municipal wastewater and reducing fouling in dynamic membrane bioreactor. The formation time and effluent turbidity were decreased in the batch stage when increasing the activated sludge concentration. The minimum values of formation time and effluent turbidity were 14 min and 43 NTU for the optimum mixed liqueur suspended solids of $8{\pm}0.5g/L$, respectively. To improve operational condition and fouling reduction in the aeration stage, critical fluxes were measured for all concentrations by flux-step method. With increasing the sludge concentration, the relevant critical fluxes reduced. The optimum subcritical flux of $30L/m^2/h$ was applied as operating flux in the second stage. The maximum COD removal efficiency of 98% was achieved by the concentration of $8{\pm}0.5g/L$. Compressibility index of self-forming dynamic membrane and transmembrane pressure trend remained somewhat constant until the optimal concentration of $8{\pm}0.5g/L$ and thereafter they increased steeply.

  • The secondary effects of pencycuron on cell membrane of Rhizoctonia solani AG4 were investigated by the observation of giant protoplast formation and lipid peroxidation. Compared to protoplasts of R. solani R-C (sensitive strain) and Rh-131 (non-sensitive strain) increased in their size by 2.0-3.5 times 12 h after incubation in potato-dextrose broth containing novozyme (7 mg/$m\ell$) and $\beta$-glucuronidase ($60\mu\textrm{g}/$\textrm{ml}) with 0.6 M mannitol (pH 5.2). The increase of protoplast size in R-C was slightly inhibited from $13.8\textrm{mg}/\textrm{ml}$ without pencycuron to 10.3 ${\mu}{\textrm}{m}$ with 1.0$\mu\textrm{g}$/$m\ell$ of pencycuron. However, the size of giant protoplast of Rh-131 was not affected by the pencycuron treatment. Both strains R-C and Rh-131 did not exhibit the lipid peroxidation 12 h after the application of 1.0 $\mu\textrm{g}$/$m\ell$ pencycuron. The remarkable peroxidation of membrane lipid was observed only in R-C 24 h after pencycuron application, but not in Rh-131. Althought the inhibition of giant protoplast formation and the membrane lipid peroxidation were observed only in the sensitive strain R-C by pencycuron, it is difficult to conclude that these are the primary mechanism of pencycuron. The mild activity of pencycuron on the inhibition of giant protoplast formation and late membrane lipid peroxidation in the fungicide-sensitive strain did not noincid with the dramatic activity of pencycuron in R. solani. Therefore, our results suggest that inhibition of giant protoplast formation and membrane lipid peroxidation is the secondary effect of pencycuron.

  • The purpose of this study is to evaluate the effects of bioresorbable membranes in guided bone regeneration of streptozotocin induced diabetic rats. 50 Sprague-Dawley rats were randomly categorized into 4 groups: Group 1 & 2 had 10 normal rats each and group 3 & 4 included 15 streptozotocin induced diabetic rats each. Defect measuring 7mm in diameter was formed on every rat calvarium. No membrane was used in groups 1 & 3 and membranes were used in groups 2 & 4. The rates were sacrificed at 2 and 4 weeks after defect formation. Routine histological specimens were prepared. Masson-trichrome and HE stain were done before light microscopy. Guided regenerative potential was evaluated by measuring the amount of new bone formation in the calvarial defect by histomorphometry. Following results were obtained. 1. New bone formation in the diabetic groups was significantly less that than in the normal groups regardless of membrane use(p<0.05). 2. In the comparison of new bone formation in the normal groups, membrane group showed significantly more bone formation(p<0.1). 3. When the amount of new bone formation was compared in the diabetic groups, more bone was formed in the membrane groups but the difference was not statistically significant.4. In the normal groups the amount of new bone formation was significantly greater at 4 weeks compared to that at2 weeks(p<0.05) but amount of bone regeneration at 4 weeks was not significantly greater than that at 2 weeks in both diabetic groups.

  • Purpose: It has been shown that the inorganic polyphosphate is effective for the regeneration of bones through the preliminary animal test of rabbits. The most effective concentration of the polyphosphate, however, is not known yet. Moreover, the effectiveness of carriers inside human body is not confirmed.. Materials and Methods: In this study, we examined the effect of the concentration of the inorganic polyphosphate on the process of the bone regeneration using the 6 weeks old rabbits with the weight of 2.0 kg in average. We performed the experiment using TR-ePTFE membrane(membrane) filled with collagen immersed in 4%, 8% of inorganic polyphosphate, respectively, following removal of the proper sized cortical bones from the rabbit calvaria. The experimental results were compared with the one of the following four groups: The negative control group for membrane only, the positive control group for membrane filled with collagen, the first experimental group for membrane filled with collagen immersed in 4% of inorganic polyphosphate, and the second experimental group for membrane filled with collagen immerse in 8% of inorganic polyphosphate. The fragments of the tissue with membrane obtained from each group of the sacrificed rabbits for 8 or 16 weeks sustained after surgery were then prestained by the Hematoxylin-Eosin stain and coated by resin to form non-decalcified specimens for the histologic examination and analysis. New bone formation was assessed by histomorphometric and statistical analysis. Results: 1. All groups have shown better bone regeneration at 16weeks than 8weeks. 2. Negative control group has shown more bone regeneration relative to the other groups at 8 and 16 weeks. 3. All experimental groups have shown better bone regeneration relative to positive control group. 4. At 16 weeks, the first experimental group has shown more bone regeneration compared to the second experimental group. Exophytic bone formation is not good at the first and the second experimental groups compared with negative control group. But, the use of 4% inorganic polyphosphate was more effective to bone formation than the use of 8% inorganic polyphosphate. Conclusion: With above results, it is suggested the use of inorganic polyphosphate with vehicle under TR-ePTFE membrane.

  • PVDF membrane formation via TIPS was performed for PVDF/DBP and PVDF/DMP systems. PVDF/DBP system showed solid-liquid phase separation behavior, while PVDF/DMP system has liquid-liquid phase separation characteristic as well as solid-liquid phase separation characteristic. PVDF contents and cooling conditions had great influence on structure, and the effects of each parameter were examined. Spherulitic structure was obtained due to the dominant PVDF crystallization. Diluent rejected to the outside of spherulite occupied the surface of the PVDF spherulites to result in the microporous spherulite formation and micro-void between spherulites. PVDF/DMP system had competitive solid-liquid and liquid-liquid phase separation depending on the cooling path.

  • The effects of relaxation and backwashing on fouling in ultrafiltration were investigated using full-scale membrane bioreactors (MBRs) which operated at a constant flux of 30 LMH. This paper also estimated the feasibility of using intermittent aeration strategies for minimizing the hydraulic resistance to filtration in comparison with the continuous aeration for running MBRs. Multiple cycles of filtration (14.5 min each) and relaxation (0.5 min each) were repeated. Similarly, a backwash was conducted by replacing a relaxation after each filtration cycle for the comparative performance test. The attached cake thickness on the membrane rapidly increased, caused by subsequent no aeration leading to easier combining with gel layer and the formation of heterogeneous layer on the membrane surface. During periodic backwashing, it is expected that gel and thin cake layer might sufficiently be removed by heterogeneous layer. After periodic backwashing, subsequent cake layer formation during time of no aeration was rapid than frequent no aeration, acting as a prefilter and preventing further irreversible fouling. Based on the Pearson correlation analysis, overall period fouling (dTMP/min) and average of all cycles (dTMP/min) were strongly correlated with the on-off period of aeration for operating MBRs.
